1.THE EXPRESSIONS OF VCAM-1 AND MHC CLASSⅡ MOLECULES IN THE EXPERIMENTAL AUTOIMMUNE ENCEPHALOMYELITIS RAT BRAIN VASCULAR ENDOTHELIAL CELLS

Objective To explore the surface molecule changes of the experimental autoimmune encephalomyelitis (EAE) rat brain vascular endothelial cells. Methods The expressions of vascular cell adhesion molecule\|1(VCAM\|1) and major histocompatibility complex (MHC) class Ⅱ molecules in the EAE rat brain vascular endothelial cells were tested by immunohistochemistry. The level of tumor necrosis factor (TNF) and interferon\|?(IFN\|?) in defferent clinic grade EAE were tested by MTT [3\|(4 5\|dimethythiazol\|zyl)\|2\^5\|diphenyl) tetrazolium bromide] method. Results There were inducible expressions of VCAM\|1 and MHC class Ⅱ molecules in the EAE rat brain vascular endothelial cells compared with control. The sera of EAE rats possessed the activities of TNF and IFN\|?.Conclusion\ In the CNS inflammation, the change surface molecules in the brain vascular endothelial cells may suggest that it plays important roles in EAE pathogenesis.\;[
2.A chemical method for preparing biotin-labeled nucleic acid probe

A chemical method to prepare biotin-labeled nucleic acid probe for nonisotopic hybridization is introduced. The method involves the bisulfite-catalyzed transamination reaction of cytosine residues in nucleic acids with ethylenediamine or hexanediamine. Primary amino groups on the cytosine derivatives are then reacted with biotiny-N-hydroxysuccinimide ester. Results of dot blot and Southern blot carried out with biotinlabeled ? DNA probe show that such nonisotopic probe method is sensitive, specific, repeatable and unexpensive.
3.The regulatory effect of Ginsenside on NK cell activity via pituitary-adrenal axis in operative stress

It was reported that immuno-regulatory mechanism of Ginsenoside on NK cell activity through pituitary-adrenal axis in operation stressed mice. Ginsenoside could modulate NK cell activity in mice against stress. On the other hand the level of plasma ACTH, corticosterone and the splenocyte ACTHR increased in stressed mice, as we know that ACTH, corticosterone could inhibite immunocyte function. However, Ginsenoside decreased the level of ACTH and corticosterone. This result suggests that the regulation effect of Ginsenoside on NK cell activity may be related to the action of pi tui taryadrenal axis under stress conditions.
4.Establishment of radioimmunoassay system for detecting corticotropinreleasing

The porcine CRF partly purified according to Vale's method used an immunogen to prepare the rabbit anti-CRF serum. The specific CRF radioimmunoassay was developed with the antiserum and ~(125)I-h-CRF. The antiserum revealed 20% tracer binding at 1: 1600 working dilution and was no cross-reactivity with relative peptides in RIA. The lower limit of detection of standard CRF was 25-50pg/ml in RIA. The concentration of CRF in plasma and brain tissue was measured by this method. The results are consistent with that of Sapolsky et al. Therefore the CRF RIA can be used for CRF detection in plasma and tissue extracts.
5.Studies on different conditions for denaturing DNA and RNA

The conditions for denaturation of DNA and RNA were invertigated. The results show that one minute to heat DNA at 100鈩?is sufficient for denaturation of DNA (several hundreds bp to 23kb). DNA degraded more obviously with longer neating time and the degradation decreased when DNA was dissdved in TE buffer. But no difference of degree of RNA degradation was found in there three conditions for RNA denaturation.
6.The safety of preoperative use of dexmedetomidine in preschool children

Objective To explore the degree of cooperation and the effect of hemodynamics in preschool children who received preoperative use of dexmedetomidine.Methods One hundred preschool children who scheduled for lower abdominal operation were randomly divided into two groups,each 50patients.The treatment group was given bolus infusion dexmedetomidine 0.5 μ g/kg (20 ml),the control group was given bolus infusion 0.9% sodium chloride 20 ml.They were completed in 20 min.The cooperation was assessed by induction compliance checklist (ICC) score,and the level of mean arterial pressure (MAP),heart rate(HR),pulse oxygen saturation(SpO2),end-tidal pressure of carbon dioxide(PnCO2) at monitoring ward before bolus infusion (T1),end of bolus infusion (T2),before induction (T3),after induction (T4),incised skin (T5) and end of operation (T6) were compared.Results The scores ofICC in treatment group was significantly lower than that in control group [(2.73 ± 0.12) scores vs.(6.17 ± 0.91) scores] (P < 0.05).In treatment group,48 patients ICC score was 0,2 patients ICC score >5 scores,and in control group,there was 48,2 patients,there was significant difference(P < 0.05).The level of MAP,HR at T1,T2,T4between two groups had no significant difference (P > 0.05).The level of MAP,HR at T3,T5,T6 in treatment group were significantly lower than those in control group(P < 0.05).In control group,the level of MAP,HR at T3,T5,T6 were significantly higher than those at T1 (P < 0.05).In treatment group,the level of MAP,HR at T2-T6 had no significant difference compared with T1 (P > 0.05).The level of SpO2,PnCO2 between two groups and interclass had no significant difference (P >0.05).Conclusion Bolus infusion dexmedetomidine before operation can improve the induction of anesthesia and operation smoothly,and improve the patients early rehabilitation.
7.THE PRACTICE OF DIET PLANNING IN NURSERIES AND ITS EFFECT ON THE NUTRITIVE QUALITY OF THE DIET AND THE GROWTH LEVEL OF THE CHILDREN

In cooperation with a District Health Station of Beijing, 13 nurseries were selected for the study of the practice of diet planning. At the beginning, the health workers of the selected nurseries -were gathered together at the station to attend a short-term class. Lectures were given on the basic knowledge of child nutrition, the methods of dietary survey and diet planning, i.e. using the proper amount of the cheaper but more nutritive foods to make essentially balanced diets according to the RDA. The health workers of the nurseries meet and comunicate once a month at the health station to calculate the monthly food consumption and the average daily intake of the nutrients of the children in the past month and to make plan for the next month. The body weight of the children were measured every month and the height at three months intervals. The data collected in 12 months were summarized. The results indicated that the yearly average intake of most nutrients by the children increased and their collective growth level in weight and height increased correspondingly during the one year period of diet planning without any increase of the boarding expense of the children.
8.Changes of intestinal permeabiloty of postoperative fasting patients

Objective To measure intestinal permeability of postoperative fasting patients and evaluate their intestinal barrier function.Methods Twenty patients after moderate or minor operation ,10 male and 10 female,were included in the study .After 3 to 5 days of fasting,when their bowel functon returned to normal,10 ml solution of lactulose(2g) with mannitol (1g) were given orally,urine of 6 hours was collected.As a control group,20 healthy volunteers were also included.There were no statistical differences in sex and age between two groups.Results The L/M of the patients and healthy volunteers were 0.1989?0.1186 and 0.4398? 0.2165 respectively,the difference was significant.Conclusions The intestinal permenbility of postperative fasting patients was elevated when compared with that of volunteers.
9.Induction of a CTL response to hepatitis B viral nucleic acid vaccine in BALB/C mice

Objective:To discuss CTL response to the hepatitis B viral nucleic acid vaccine in mice.Methods:The recombinant plasmid containing HBV S gene was directly injected intramusculary into BALB/C mice.HBsAb in sera was checked with ELISA.The recombinant plasmid was transfected P815 cells by liposome and screened by G418.Splenocytes were isolated for investigating specific CTL response. Results:HBsAb in sera of mice was tested after pcDHBs injection.HBsAg was detected in culture medium of the transfected P815 cells. Compared mice injected with pcDNA3.1,the specific CTL cytotoxity activity of mice immunized with pcDHBs was significantly enhanced.Conclusion:HBV nucleic acid vaccine could stimulate humoral immunity and CTL response of the mice.
10.Clinical Therapeutic Observation of Pricking Cupping for Herpes Zoster at AcuteStage

ObjectiveTo observe the efficacy of pricking cupping in treating herpes zoster at acute stage.MethodSixty patients with herpes zoster at acute stage were randomized into a treatment group and a control group, 30 in each group. The treatment group was intervened by pricking cupping, while the control group was by oral administration of Western medicine. The total effective rate and the improvement of symptoms and pain at different stages were observed afterintervention.ResultThe total effective rate was 90.0% in the treatment group versus 86.7% in the control group, and the difference was statistically insignificant (P>0.05); after the first treatment course, the decreases of symptom score and pain indexin the treatment group were significantly superior to that in the control group (P<0.05); after the second treatment course, there were no significant differences in comparing the decreases of symptom score and pain index between the two groups (P>0.05).ConclusionPricking cupping is an effective approach in treating herpes zoster of the acute stage, as it can produce a comparatively higher total effective rate and also significantly improve the symptoms and pain.
