1.Production and characterzation of mcAb to murine astrocyte and some biological properties of astrocyte product

Using hybridoma technique,17 hybridoma cell lines against astrocyte were ob-tained.The screening test of McAb showed:(1)Using immune enzymatic histochemistryfixed cell staining method all of 17 McAb reacted with astrocyte,(2) Using immuneenzymatic histochemistry viable cell staining method,eight of seventeen McAb react-ed with astrocyte.As to biological properties of McAb results were as follows:(1)Seven of eight McAb that showed positive reaction in viable staining assay have di-fferent degree of cytotoxicity to astrocyte,thymocyte and spleen cell in presence ofcomplement,one of eight has specific cytotoxicity only to astrocyte.(2) McAb(2C)possesses blocking effect on astrocyte,thymocyte and spleen cell proliferation respon-sive to various kind of mitogen.Furthmore,it has been found LPS may promote theproliferation of astrocytes and secreting interleukin 1(IL-1)-like substance This IL-1like substance seems to have a mutiple biological activities,i.e:(1)Thymocyte prolife-ration.(2) Fibroblast(L929 cell) proliferation and(3) Endogenous pyrogen activities.
2.Effects of Murine Thymic Epithelium Cultural Supernatant on Spontaneous and ConA-Stimulated Proliferation of Thymocytes

Cultivation of murine thymic epithelium has been established.Murine thymic epi-thelial cultural supernatant showed activity increasing both spontaneous ~3H-TdR inco-rporation and ConA-stimulated proliferation of thymocytes by the exponent of 1.5-2at dilution 1:8 to 1:32,the peak of the activity was present at 10-20 or 15 days ofcultivation.However,the thymic epithelial supernatant from cultures at 6-7 daysshowed activity suppressing both proliferations of thymocytes at dilution 1:8-1:16.Theresult showed thymic epithelial supernatant had no 1L-2 activity.
3.The establishment of ACTHR detecting method and assay of ACTHR on rat thymocytes

In this paper, it was reported that rat thymocytes harbor both high and low affinity ACTHR. The proper number of thymocytes used for deterting ACTHR varies from 1?10~6 /0.2 ml to 2 ?10~6 /ml, the maximum binding of ACTH to ACTHR occurs at 20 minutes. The IC_(50) for ACTH concentration is 800 nM or so. The saturation curves show that thymocyte ACTHRs are saturated at the concentration of 2.5 nM of ~(125)IACTH. There are high affinity ACTHR of 840 sites/cell on thymocyte (KD=0.15nM, Bmax = 2. 8 fmol/2 ? 10~6 cells); whereas low affinity ACTHRs are 12400 sites/cell (KD=4.2 nM, Bmax = 41.5 fmol/2 ? 10~6 cells).Moreover amputation stress can reduce thymocyte ACTHR remarkably.
4.The Induction of Suppressor Cells in Murine Spleensby Stress and Its Effect on the Responsive for the Mitogen

In this paper, it has been found that such stressors as operation or swimming in cold watermay suppress the mitogenic response of murine spleen cells to ConA or LPS, whichis closely related to the action of suppressor cells.Using the plastic-adherence method, the plastic-adherence cells in spleen(PAC)was removed or obtained. The difference of mitogenic responsivebefore and after the removal of PAC is very apparent in stressed mice, being much lower thannormal before the renioval of PAC and near to normal after removing PAC. It is interestingthat the lymphocyte transformation stimulated by LPS is suppressed predominantly after swim-ming stress instead of those by ConA in operation stress.It has also been observed that theindomethacin may block the induction of suppressor PAC by stress, which means that the pro-staglandins may play an important role in the induction and action of suppressor PAC.
5.The Predictive Factors of IFN Therapeutic Effectiveness in Hepatitis C

Objective: Our purpose was to study the predictive factors of interferon(IFN) therapeutic effectiveness. Methods:Genotypes of HCV, HCV RNA quantities, β2 microglobulin, 2′- 5′oligoadenylate synthetase, and peripheral blood lymohocyte subgroup were detected by using specific primer PCR assay, energy transference technique of signal primer, sheet chromatography, radioimmunoassay, and APAAP immunoenzyme bridge technique respectively in 20 blood samples of patients with chronic hepatitis C. Results:(1)The patients who had lower level of HCV RNA before treatment and had continuing decreased HCV RNA level after treatment had good response to IFN treatment in follow-up survey. (2)The patients with HCV-Ⅲ type had complete response much more than those with HCV-Ⅱ type (P<0.05).(3)The patients with lower level of 2′- 5′oligoadenylate synthetase before treatment might have good response to IFN treatment. (4) There was no significant change in β2 microglobulin level before and after treatment and in different response groups before treatment.(5) The CD+3, CD+4, and CD+4/CD+8 in peripheral blood increased after treatment. The patients in complete response group had higher level CD+3, CD+4, and CD+4/CD+8 than those in part response and non-response groups, but there was no significance in statisitcs. Conclusion:The HCV RNA quantities, HCV genotypes, 2′- 5′oligoadenylate synthetase level before treatment were predictive factors of IFN therapeutic effectiveness. The changes of HCV RNA level in treatment and follow-up survey had important significance in therapeutic evaluation, β2 microglobulin level, and lymphocyte subgroup before treatment were not used as predictive factors for IFN therapeutic effectiveness.
6.Changes of Th1/Th2 ratio in the PBMC and hepatocellular carcinoma tissues of patients with heptocellular carcinoma and its clinical significance

Objective:To investigate the changes of Th1/Th2 ratio in the PBMC and hepatocellular carcinoma tissues of patients with heptocellular carcinoma and its clinical significance.Methods:Th1/Th2 subsets and cellular factor were detected by ELISPOT and double-sandwich ELISA.Results:Th1/Th2 ratio in the PBMC and hepatocellular carcinoma tissues of patients with heptocellular carcinoma,Th1 sebset expressing cellular factor IFN-?,IL-2,IL-12 were significantly lower,and Th2 sebset expressing cellular factor IL-4,IL-10 were significantly higher than those of the normal control group.PMBC in patients with heptocellular carcinoma producing Th1 sebset cytokins deduced by PHA and IL-12 were significantly higher and producing Th2 sebset cytokins were significantly lower than those in the three month after operation.Conclusion:Th1/Th2 ratio in the PBMC and hepatocellular carcinoma tissues were of imbalance and immune functional status of patients with hepatocellular carcinoma was inhibitory.Therefore,it was nessassory to enhance immune functional status of patients during treatment.
7.Effect of helicobacter pylori infection on hyperdynamic circulatory state and portal hypertensive gastrropathy in liver cirrhotic patients

Objective:To evaluate the effect of Helicobacter pylori infection on hyperdynamic circulatory state and portal hypertensive gastrropathy in liver cirrhotic patients.Methods:Helicobacter pylori infection,IL-1,IL-8,TNF-?,Endotoxin,NO,ET,anti-VacA and anti-CagA levels were determined in cirrhotic patients by using rapid urease test,()~(14)C-urea expiratory test,immunoimpression test,nitrite colorimetric test and ELISA.The parameters of portal venous system flow were determined by doppler ultrasonography.Results:HP prevalence in patients with liver cirrhosis was 77.1%,anti-VacA and antiCagA positive rate was 60.4% in common.IL-1,IL-8,TNF-?,Endotoxin,NO and ET levels in anti-VacA and anti-CagA positive group were significantly higher than in anti-VacA and anti-CagA negtive group.The amount of splenic venous flow(SVF) and superior mesenteric venous flow(SMVF)in anti-VacA and anti-CagA positive group was greater and the amount of portal venous flow(PVF) was smaller than those in negtive group.Conclusion:There was the effect of Helicobacter pylori infection on hyperdynamic circulatory state and portal hypertensive gastrropathy in liver cirrhotic patients.
8.Determination of Main Component and the Related Substances in Pirenoxinum Natricum Eye Drops by HPLC

OBJECTIVE:To establish a HPLC method for the determination of main component and the related substances in pirenoxinum natricum eye drops. METHODS: A Inertsil C8-3 column was used with pH2.2 perchloric acid-methanol (35∶65) as mobile phase at a flow of 1.0 mL?min-1. The detection wavelength was 230 nm and the sample size was 10 ?L. RESULTS: Pirenoxinum natricum and the related substances could be baseline separated. The calibration curve was linear in the range of 49.5~990 ng(r=0.999 9). The detection limits was 89.3 pg, and average recovery was 99.1%, RSD was 0.42%. CONCLUSIONS: This method is rapid, accurate sensitive and can be used for the quality control of pirenoxinum natricum eye drops.
9.Detection of food allergen-specific IgE and IgG in patients with chronic urticaria

Objective To evaluate the roles of food allergen detection in patients with chronic urticaria.Methods Food allergen-specific IgE and IgG were detected in 502 patients with chronic urticaria and 100 normal human controls.Double-blind,placebo-controlled food challenge (DBPCFC) test was performed in patients who were proved to be positive or negative for food-specific IgG or IgE.Results Food-specific IgE was detected in 37.45% (188/502) of the patients,with cashew,peanuts and soybeans as the most common allergens.The detection rate of food-specific IgG was 72.11% (361/502),with the major allergens being milk,egg,shrimp and crab.DBPCFC revealed false positivity and negativity in the detection of food-specific IgG and IgE.Conclusions Clinicians should pay attention to food allergen detection in the treatment of chronic urticaria,especially in the treatment of allergy mediated by IgG antibodies.
10.Expression and clinical significance of urotensin Ⅱ in lung cancer tissue

ObjectiveTo investigate the expression of urotensin Ⅱ(UⅡ) in the lung cancer tissue from surgical resection of lung cancer patients,and to detect the relationship between UⅡ expression and pathologie types and the clinical stages of lung cancer.MethodsThe expression rates of UⅡof 45 lung cancer tissues and 20 inflammatory pseudotumor were measured by immunohistochemical assay,and the relationship between UⅡ expression and the pathologic types and clinical stages of lung cancer was analyzed.ResultsUⅡwas mainly distributed in lung cancer cell cytoplasm,which was tan-yellow particles.The positive expression rate of UⅡin nonsmall cell lung cancer was 61.3% (19/31),which was higher than that in small cell lung cancer(7.1%,1/14)and pulmonary inflammatory pseudotumor( 15.0%,3/20) (P < 0.01 ).The positive expression rate of UⅡ was 100% in adenocareinoma.The positive expression rate of UⅡin staging Ⅲ non-small cell lung cancer( 85.7% )was higher than that of staging Ⅰ ( 16.7% ) ( P < 0.05).ConclusionUⅡ cxists in the cytoplasm of lung cancer cells,and the expression of UⅡis correlated with the pathological type and TNM staging of lung cancer.
