1.Cross-sectional survey of adolescent sexual psychology and knowledge

BACKGROUND: The population of male and female adolescents in China accounts for 1/5. Adolescents have much psychological bewilderment,troublesome and even diseases on sexual issues.OBJECTIVE: To investigate the development status quo and the regulation of adolescents' sexual physiology, sexual psychology, sexual knowledge and sexual education.DESIGN: A cross-sectional survey by employing students of junior high school as subjects.SETTING: A clinic for adolescent of a tertiary hospital.PARTICIPANTS: Totally 960 students aged between 14 and 19 years old of three junior high schools in Dalian city were selected as subjects between November and December of 2001.METHODS: Self-complied questionnaire was used in the survey based on the sexual psychological education situation scale for junior and senior high school students. Classes were randomly selected for anonymous survey based on voluntary principle.Answers to sex-related questions and differences between male and female students.RESULTS: The average age of menarche in female students was 12.96years old and the average age of first emission in male students was 13.89years old, which were consistent with the results of similar national researches. The incidence and frequency of masturbation and sexual impulse was significantly higher in males than females. There were significant differences in sexual concept and the understanding of sexual knowledge between males and females. The desire to affiliate with the opposite sex was far more than the real occurrence.CONCLUSION: The understanding of sexual knowledge in adolescent is not satisfied. Students are also not satisfied with the sexual education provided by their schools. Hence, school sexual education for adolescents should be enhanced with the involvement of community.
2.Clinical analysis of deep fungal infections in ICU hospital

ObjectiveTo investigate the clincal status and countermeasures for intensive care unit (ICU) hospital deep fungal infections.Methods108 cases of ICU hospital deep fungal infections patients were divided into comprehensive treatment group and converntional treatment group,54 cases in each group.The morbidity and treatment effects were observed.ResultsThe cure rate of observation group was 96.3%,significantly higher than 79.6%in control group.The observation group complication rate was 9.2%,significantly lower than the control group 20.4%,the difference was significant (P < 0.05 ).ConclusionIn the ICU nosocomial fungal infection treatment,the scientific and effective means of comprehensive prevention and control best practices,could significantly improve patient outcomes,improve treatment,reduce hospital stay and reduce complications.
3.Effects of mycobacterium phlei on proliferation of dendritic cells derived from human umbilical cord blood in vitro

BACKGROUND: The central position of dendritic cells (DCs) has aroused increasing attention due to strong antigen presentation capability in anti-tumor. However, how to obtain enough functional DCs and reports regarding immunomodulator with low toxicity are few. OBJECTIVE: To investigate the effect of mycobacterium phlei F.U.36 (Utilins) on the proliferation and maturity of DCs derived from human umbilical cord blood in vitro.METHODS: The mononuclear cells were isolated from human umbilical cord blood using Ficoll-Hypaque method and cultured with Utilins, cytokine (human recombinant granulocyte/macrophage colonystimulating factor + recombinant human tumor necrosisfactor-α + recombinant human interleukin-4), or combination cytokine + Utilins, respectively. In addition, cells cultured with RPMI-1640 served as controls. Morphological features and growth of DCs were observed by an inverted microscope. Thephenotype changes of DCs, such CD1a and HLA-DR, were detected by flow cytometry at 9 days after culture. Moreover, DCs were stained by Wright-Giemsa and observed under oil lens. RESULTS AND CONCLUSION: Typical DCs with high expressions of CD1a and HLA-DR were obviously detected in all experimental groups except the control group. The positive rates of CD1a and HLA-DR in the Utilins group were higher than those in the control group, but lower than the cytokine group (P < 0.05). The HLA-DR positive rate of the combination group was higher than that of the cytokine group (P < 0.05). The results revealed that, Utilins can not only promote the proliferation of DCs derived from human umbilical cord blood in vitro but also cooperate with cytokines to induce the maturity of DCs.
4.EFFECTS OF THYROID HORMONE ON THE ULTRASTRUCTURE OF DISTAL NEPHRON IN RAT——A MORPHOMETRIC STUDY

An ultrastructural analysis of distal nephron has been investigated using morphometric method for rats with hyperthyroidism and hypothyroidism. The cells analysed were distal convoluted tubule cells (DCT cells), distal straight tubule cells (DST cells), intercalated cells (I cells) and principal cells (P cells) of collecting duct. The cells were sampled from superficial cortex. The results revealed that in DCT, DST and P cells the profile area of cells, the surface density of plasma membrane (Sv), the boundary length of cell surface [B(A)], the volume density (Vv) of mitochondria, and the length of microvilli were significantly decreased in the rat with hypothyroidism. However, in the rat with hyperthyroidism there were an obvious increase of profile area of cell and Vv of mitochondria in I cells. The results suggest that thyroid hormone is an important factor for maintaining the normal cellular structure in the distal nephron. The morphometry shows that the changes of DCT, DST and P cells are distinct in the rat with hypothyroidism, and the ultrastructural changes of I cell are obvious in the rat with hyperthyroidism.

This paper describes the analysis of insitu polymerization of monomeric reactants (PMR) polyimide resin solution by reversed phase ion pair suppression chromatography. the mobile phase consists of acetonitrile and water. The methyl sulphonic acid was used as an ion-pair reagent in the mobile phase. The addition of sodium perchlorate was required to increase the ionic strength of the mobile phase. The information was obtained on monomeric isomer distribution of PMR polyimide resin solution and purity of prepared materials.
6.Ankle proprioception after a stroke

Objective:To explore vibration, position and motion proprioception of the ankle joints after a stroke.Methods:Twenty-eight stroke survivors with impaired ankle proprioception were divided into a right-side stroke group ( n=18) and a left-side stroke group ( n=8). Twenty-two healthy volunteers constituted a control group. Vibration perception thresholds, passive and active joint angle resetting, and motion minimum thresholds were quantified among the stroke survivors on both the healthy and the affected side. With the controls the dominant and non-dominant sides were used. The differences in proprioception between the healthy volunteers and the stroke patients, between the affected side and the healthy side of the stroke patients, and between left- and right-side stroke patients were analyzed and compared. Results:Among the stroke survivors the vibration perception threshold on the affected side averaged (28.91±22.53)μm. The absolute difference in the perception of passive positioning was (5.49±5.39)° for 15° of plantar flexion and (4.48±3.89)° for 5° of dorsal extension. In active positioning plantar flexion was (5.23±4.34)° and for 30° of plantar flexion it was (3.26±1.73)°. The 5° dorsal extension error was (4.97±3.48)°. The motion perception thresholds between 20° of plantar flexion, 10° of plantar flexion and the neutral position were significantly higher, on average, than among the control group. The stroke group also had significantly higher motion perception thresholds than the control group.Conclusion:The vibration, position, and motion sense of the ankle joint on a stroke survivor′s affected side tend to be impaired, with the impairment of vibration and motion sensing tend to be more substantial. After stroke, there is also mild impairment of vibration, position and motion sensing in the healthy ankle joint. The impairment of proprioception caused by right cerebral hemisphere injury may be more serious than that caused by injury on the left.
7.Effect of genetic modified K562 cells on NK cell markers and function

Objective:To study the influence of different culture conditions on charcic and inhibition activity of nature killer cells ( NK) ,whether to join the modified K562 cells with IL-6 cytokine.Methods:According to the 5′end of the human IL-6 cDNA sequence,PCR primers designed to amplificate,express and transfect K562 cells cDNA library as a template for DNA.Genetic modified K562 cells as stimulating cells were prepared by expressing IL-6.To extract peripheral blood mononuclear cells( PBMC) from human peripheral blood.PBMC were explanted by genetic modified K562 stimulated.The expansion was initiated by CO-culture of PBMC and irradiate genetic modified K562 cell.The number of NK cell increased by directed induced generation of genetic modified K562 cell.Immunophenotypic analysis of NK cell surface markers was performed by flow cytometry (FCM).51Cr release assay was employed to measure the specific lysis skilling of NK cell target K562 cells.Results:We have constracted genetic modified K562 cells by genetic engineering.As stimulated cell added into the PBMC,an average of 760 ±18 fold expansion of CD56+CD16+CD3-cells was observed after 3 weeks of co-culture system.The NK cells population could proliferated more 91%±2% after expansion comparing with 6%± 0.4%in PBMC before expansion by FCM.The cytotoxical activity of NK cells which was induced by genetic modified K562 cell was the strongest than induced by IL-6 cytokine alone.The expanded NK cells lysed 92%±2% of K562 targets in a 5∶1 effector to target ratio.In this case,the NK cells induced by genetic modified K562 cells against tumor cells was more lethal.Conclusion:PBMC based in vitro expansion of natural killer cells was set up by genetic modified K562 cells.The cytotoxicity of NK cells was the strongest induced by genetic modified K562 cell treated.These results had important guiding significance for the the NK large number of amplification and used in clinical.
8.Effects of CICARE communication model combined with WeChat health education on self-management among patients with type 2 diabetes mellitus combined with pulmonary tuberculosis

Objective To explore the effects of CICARE communication model combined with WeChat health education on self-management capacity, focal absorption and indexes of glucose metabolism among patients with type 2 diabetes mellitus (T2DM) combined with pulmonary tuberculosis (PTB). Methods A total of 80 patients with T2DM combined with PTB at Chest Hospital of Xinjiang Uyghur Autonomous Region were selected by convenience sampling from 1st January 2016 to 31st December 2016. They were divided into control group (n=40) and observation group(n=40) with the method of the random number table. Patients in control group received conventional nursing and health education during hospitalization and at discharge. On the basis of control group, patients in observation group received nursing with the CICARE communication model along with health education by WeChat after discharge. The self-management capacity, focal absorption and effective rate, index of glucose metabolism of patients of both groups were compared six months after intervention.Results Before intervention, there were no statistically significant differences in self-management capacity and index of glucose metabolism (P> 0.05). Six months after intervention, the score of self-management capacity and effective rate of treatment in observation group was higher than those in control group. Besides, the fasting plasma glucose (FPG), 2-hour postprandial blood glucose (2hPG) and glycosylated hemoglobin in observation group were lower than those in control group with significant differences (P< 0.05). Conclusions The CICARE communication model combined with the communication and propagation function of WeChat can improve the self-management and clinical symptoms. Moreover, it has a synergistic action in improving treatment effects.
9.Contact tracing of a possible case of HIV sexual transmission by using Miseq platform.

OBJECTIVEAn approach for analysis of HIV quasispecies using Miseq high-throughput sequencing platform (hereinafter referred to as Miseq platform) was established and applied to contact tracing for a possible case of HIV sexual transmission.
METHODSFour plasma specimens were collected from 2 HIV infections (P1 and P2) suspected to be involved in the sexual transmission and 2 local HIV infections as controls (P3 and P4). The RNAs were extracted from the specimens and then reverse-transcribed into cDNA. After HIV subtyping, Miseq platform was performed to detect and sequence the HIV quasispecies (352 bp) in each specimen. The frequency of quasispecies was counted and ranked. Intrapersonal and interpersonal genetic distance and phylogenetic tree were calculated by using the top 5, 20, 100, 500, and all quasispecies, respectively.
RESULTSThe subtypes of HIV from all 4 specimens were CRF01_AE. 23 788 to 37 397 cleaned sequences representing 1 229 to 1 412 unique HIV quasispecies were obtained from these specimens by using Miseq platform. The average genetic distance (3.5%-4.5%) between quasispecies from specimens P2 and P1 was significantly lower than that (10.3%-19.6%) between quasispecies from P2 and the controls (P3 or P4). Phylogenetic tree analysis indicated that sequences from specimens P1 and P2 clustered together while sequences from P3 and P4 exhibited completely independent clusters. When the top 20 or more quasispecies from each specimen were analyzed, sequences from P1 showed a paraphyletic relationship with those from P2, which may indicated that the direction of HIV transmission was from P1 to P2.
CONCLUSIONWith the feature of convenient and economic operation, Miseq platform has high practical value in contact tracing of possible HIV transmission.

10.Effects of continuing care based on habit forming education on insulin treatment compliance in elderly patients with type 2 diabetes mellitus

Objective To explore the effects of continuous nursing based on habit forming education care on insulin treatment compliance in elderly patients with type 2 diabetes.Methods From May 2014 to April 2015 , 80 cases of elderly patients with type 2 diabetes treated with insulin were randomly divided into observation group and control group .The patients of control group were given routine health education and outpatient review.The patients of observation group were given health education and habit forming education before discharge.After discharge, the patients of observation group were given continuous follow-up, health education and network service .The treatment effects and insulin treatment compliance were compared between two groups 6 months after discharge .Results In the observation group , insulin injection compliance was significantly higher than that of the control group (P<0.05), but FPG, 2hPG and HbAlc were lower than those of the control group (P<0.05).There was no significant difference in incidence of hypoglycemia between two groups (P>0.05).Conclusions The continuous nursing based on habit forming education can improve the correct rate of insulin self-injection and treatment compliance , improve the clinical symptoms and reduce the incidence of adverse reactions in patients with self-injection of insulin .
