OBJECTIVE:To study the protective effect of shuanghuanglian sterilized powder for injection on hepatic in?jury of model mouse induced by human cytomegalovirus(HCMV)infection.METHODS:The mice were randomized into nor?mal control group,infection model group,ganciclovir group and shuanghuanglian sterilized powder for injection group(high,middle and low dosage),all of which except the normal control were vaccinated with HCMV5d to establish the hepatic injury model,each group was administered with different drugs;10days later,the enzyme activity of ALT and AST in the eye sockets blood of each group was determined;The mice were put to death by cervical vertebra dislocation and the necrosis degree of livers from the dead mice was evaluated and the pathological observation was conducted.RESULTS:The activity of ALT and AST were normal in the normal control,and no significant changes were found in the livers of this group;Compared with the normal control,the plasma ALT and AST levels have been increased significantly and there was a significant pathological hepatic injury in the infection model group;Compared with the infection model group,significant decreases were found in the activity of ALT and AST in the high and middle dosage groups of shuanghuanglian sterilized powder for injection,and there were obvious alleviated pathologic hepatic injury while no significant differences were found in the plasma ALT and AST ac?tivity in the low dosage group;Compared with the infection model group,the ALT and AST activity of the ganciclovir group were lowed significantly;Compared with the ganciclovir group,no significant differences were found in the ALT and AST ac?tivity in the high and middle dosage groups of shuanghuanglian sterilized powder for injection.CONCLUSION:Shuanghuan?glian sterilized powder for injection does have protective effect on hepatic injury of model mouse induced by HCMV.

BACKGROUND:How to improve the stability of artificial cornea in the host and reduce the complications is the current key issues to be solved.Therefore,looking for an ideal biocompatible scaffold material is still the focus of the study.OBJECTIVE:To explore the biocompatibility of nano-hydroxyapatite (nHA)/polyvinyl alcohol (PVA) porous composite hydrogel and pure PVA hydrogel as artificial corneal materials.METHODS:Inverted microscope was used to observe cell growth of rabbit corneal stromal fibroblasts when cultured with nHA/PVA composite hydrogel extract or PVA extract for 48 hours.MTT method was used to detect the relative proliferation rate of rabbit corneal stromal fibroblasts cultured with nHA/PVA composite hydrogel extract or PVA extract.Systemic toxicity,allergies,pyrogen reaction and deaths were observed in New Zealand white rabbits at 48 hours after artificial corneal implantation.ELISA and hematoxylin-eosin staining were used to detect changes in serum inflammatory factors at 4 weeks after artificial corneal implantation.Slit-lamp examination was performed to observe corneal or conjunctival hyperemia/edema and corneal neovascularization at 1-4 weeks after corneal implantation.Corneal neovascularization time and neovascularization area were detected after the two materials were implanted.RESULTS AND CONCLUSION:The cells were sparse and grew slowly at 48 hours after culture in nHA/PVA composite hydrogel or PVA extract as compared with the blank control group.The cell growth in the nHA/PVA composite hydrogel group was better than that in the PVA group.The relative cell proliferation rate was significantly decreased at different time after culture in nHA/PVA composite hydrogel or PVA extract compared with the blank control group (P ＜ 0.05),and the relative cell proliferation rate in the nHA/PVA composite hydrogel group was significantly higher than that in the PVA group (P ＜ 0.05).There were two rabbits appearing to have allergic reaction,but no one presenting with pyrogen reaction and death in the nHA/PVA composite hydrogel group;and there were two rabbits appearing to have allergic reaction,and two appearing to have pyrogen reaction,but no death in the PVA group at 48 hours after implantation.The inflammatory factor levels were increased significantly in the nHA/PVA composite hydrogel group and PVA group compared with the control group (P ＜ 0.05),and inflammatory infiltration was milder in the nHA/PVA composite hydrogel group than the PVA group at 4 weeks after implantation (P ＜ 0.05).Corneal neovascularization appeared later in the nHA/PVA composite hydrogel group than the PVA group,and the neovascularization area was also smaller in the nHA/PVA composite hydrogel group than the PVA group (P ＜ 0.05).Taken together,the biocompatibility of nHA/PVA composite hydrogel is superior to that of pure PVA when they are used as artificial cornea materials,which provides a scientific basis for artificial cornea material innovation and extensive clinical application.

OBJECTIVE:To prepare Frovatriptan succinate film-coated tablet,and investigate its in vitro dissolution behavior. METHODS:Using lactose monohydrate,microcrystalline cellulose,dioxide,silica,sodium carboxymethyl starch and magnesium stearate as accessories,Frovatriptan succinate tablet was prepared. Using opadry premix spray-coating liquid,Frovatriptan succinate film-coated tablet was prepared. Single factor test was used,using moisture,angle of repose,rigidity,friability,disintegration time and dissolution rate as indexes,to screen the formulation;using dissolution degree as index,coating material dosage was screened. The dissolution curves in vitro of self-made tablets and imported tablets in water,0.1 mol/L HCL,pH of 5.5,6.8 phos-phate buffer solutions were compared. RESULTS:The optimal formulation of Frovatriptan succinate uncoated tablet was as follow as frovatriptan succinate 3.91 mg,lactose monohydrate 99.18 mg,microcrystalline cellulose 33.06 mg,magnesium stearate 1.40 mg,sodium carboxymethyl starch 1.05 mg,silica 1.40 mg;optimal coating weighed quality was 2.0%-4.0%. In the 4 mediums, the dissolution behavior of self-made tablets and imported tablets were similar. CONCLUSIONS:Frovatriptan succinate film-coated tablet is prepared successfully,and its in vitro dissolution behavior is similar to the imported preparations.

ObjectiveThe purpose of this study is to describe the clinicopathologic characteristics and the survival of pulmonary carcinosarcoma.Methods Clinical data of 26 patients with pulmonary carcinosarcoma who underwent surgical treatment was analyzed retrospectively using Statistical Package for the Social Sciences ( Release 17.0,SPSS,Inc).Survival analysis was estimated using the Kaplan-Meier method and compared using the log-rank test.ResultsThe overall 5-year survival rate of the patients was 31.7％.The 5-year survival rate of patients with stage Ⅰ,stage Ⅱ and stage Ⅲ/Ⅳ disease was 69.7％,38.6％ and 9.8％ respectively ( P =0.03 ) Conclusionp-TNM was correlated with survival.Early diagnosis and radical operation is important to the survival of the patients with pulmonary carcinosarcoma.

As the improved understanding of the biological behavior of colorectal cancer and the development of diagnosis and surgical techniques,the prognosis of patients with locally advanced colorectal cancer has been improved significantly.Locally advanced colorectal cancers are best treated with multivisceral resections,but the procedure is high technique-demanding and the indications for the procedure should be strictly followed.In this article,the procedure of right colectomy combined with pancreatoduodenectomy for colon cancer was described in detail in order to share the experiences and skills with surgeons.

Objective To assess the feasibility and short-term effect of transanal everted extraction of laparoscopic rectal excision without abdominal incision.Methods Eleven patients were operated by transanal everted extraction of laparoscopic rectal excision without abdominal incision.Results All the operations were accomplished successfully,the average operative duration was (189.5±25.2) min,the intra-operative blood loss volume was (25.4±9.4) ml,the average time to flatus was (38.9±8.6) h,the average postoperative hospital stay was (12.7±2.5) d,and the number of lymph nodes harvested was 13.5±4.1.There were no post-operative complications.Conclusion The technique of transanal everted extraction of laparoscopic rectal excision without abdominal incision is safe and feasible,but the long-term effects need more observation.

Aim To investigate the effect of Shuanghuanglian on the the load of hepatic HCMV DNA in mice infected with HCMV.Method The model of mice hepatitis infected with HCMV was prepared. 10 days after treated with Shuanghuanlian,the hepatic HCMV DNA was detected qualitatively by polymerase chain reaction;the level of alanine aminotransferase (ALT) and aspartate aminotransferase(AST) in serum were assayed by spectrophotometer;and hepatic pathology was observed with HE staining.Results HCMV DNA load in infected model group was 3875.4760 copies/mg,while in high and middling dosage shuanghuanglian therapy group it was 8.3261 copies/mg、9.1476 copies/mg respectively;It decreased significantly compared with the former(P0.05);low dosage of shuanghuanglian(HCMV DNA load was 3812.8980 copies/mg) had no notable difference compared with infected model group;serum transaminase (ALT,AST) levels significantly elevated in infected model group; serum transaminase level decreased sharply and liver histological pathology was lighten after treated with high and middling dosage of Shuanghuanglian(P

Objective To evaluate the significance of the expression of carcinoembryonic antigen (CEA) and carbohydrate antigen 19-9 (CA19-9) as prognostic indicators for patients with stage Ⅱ colorectal cancer. Methods A total of 285 patients with stage Ⅱcolorectal cancer who underwent potentially curative surgery were enrolled in the study. A high CEA level was defined as a level exceeding 5 ng/ml and a high CA19-9 level was defined as a level exceeding 37 U/ml. Patients were followed up every 3 months to review CEA and CA19-9 levels. Two years later, the chest and abdominal CT examination were performed every 3 months, and then every 6 months until 5 years. Results Out of these 285 patients, 99 (42.60%) patients had high CEA levels, 25 (8.77 %) patients had high CA19-9 levels, and 12 (4.21 %) patients had both high CEA and CA19-9 levels. The overall survival rates of patients with both high CEA and CA19-9 levels were significantly worse than those of others (P< 0.05). During the study, 51 recurrences were diagnosed. There were 22 hepatic recurrences, 10 pulmonary recurrences, 9 local recurrences, 8 lymph node recurrences, and 2 peritoneal recurrences. CT, CEA and CA19-9 were the first abnormal examinations in 30, 5 and 8 recurrent patients, respectively. In 27 % of recurrent patients, the recurrence was detected earlier by CEA and CA19-9 than that by CT. Conclusions The detection of preoperative CEA and CA19-9 levels is useful for predicting the prognosis after potentially curative surgery in patents with stage Ⅱ colorectal cancer. For early detection of occult recurrence of colorectal cancer, tumor markers are relevant.

Objective:To construct the "3+2" counterpart cut-through sectional training nursing major un-dergraduate curriculum system, which is oriented by vocational competence. Methods: The preliminary draft of"3+2" nursing undergraduate curriculum setting was established base on the literature review and expert group in-terview, and the 25 experts was conducted two rounds of expert questionnaire consultation using Delphi method. Results:Experts' opinions tended to be consistent after two rounds of consultation, the expert authority coefficient was 0 . 92 , the coordination coefficient of Kendall was 0 . 44 in the second round of expert consultation and finally established 5 curriculum groups, including total 28 courses of public elementary courses, professional basic cour-ses, professional core courses, professional oriented courses and centralized practice courses. Conclusion: It should construct the"3+2" counterpart cut-through nursing major undergraduate curriculum system, which is o-riented by vocational competence, and achieve effective connection between the knowledge structure and the quality of the nursing students, in order to provide the reference for perfecting the curriculum system of vocational educa-tion in our country.

Objective To investigate whether the gene polymorphisms of cytochrome P450(CYP) 2E1 are associated with the risk of anti-tuberculosis drug induced hepatotoxity (ADIH).Methods In this case control study, 339 patients who matched the diagnosis criteria of tuberculosis were included. The gcneral healthy status and liver biochemical parameters were checked in all these patients. Polymerase chain reaction-restriction fragment length polymorphism (PCR RFLP) technique was used to determine CYP 2Et polymorphisms. The statistic analysis were performed by using both univariate and multivariate Logistic regression analysis. Results The allele frequencies of CYP 2E1 7632T/A, 1019C/T and 1259G/C in 103 tuberculosis patients of ADIH group were 17.5%, 26.2%and 27.2 % respectively, while those in 236 tuberculosis patients of control group were 29.7 % ,39.4 %and 40.7%, respectively (x2 =5.539, P＜0.05; x2 =5.458, P＜0.05; x2 =5.628, P＜0.05). The results of univariate analysis demonstrated that the risk of concurrent ADIH was significantly higher in patients with wild genotypes of CYP 2E1-7632T/A, CYP 2E1-1259G/C, CYP 2E1-1019C/T than in patients with other genotypes. After adjusted for sex, occupation and alcohol consumption status, the results of multivariate Logistic regression analysis also showed that wild genotypes of CYP 2E1-7632T/A, CYP 2El-1259G/C, CYP 2E1-1019C/T were significantly associated with higher risk of ADIH. The results of interaction analysis indicated that the wild genotypes of CYP 2E1-7632T/A and CYP 2E1-1259G/C or CYP 2E1-1019C/T had synergetic effects on the development of ADIH.Conclusions The risk of concurrent ADIH is significantly higher in patients with wild genotypes of CYP 2E1-7632T/A, CYP 2E1-1259G/C, CYP 2E1-1019C/T compared to patients with othergenotypes. Wild genotypes of CYP 2E1-7632T/A and CYP 2E1-1259G/C or CYP 2El-1019C/T have synergetic effects on the development of ADIH.
