1.Changes of L-selectin expression on peripheral blood polymorphonuclear leukocytes and their significance in rats with acute lung injury

AIM: To explore the changes of L-selectin expression on peripheral blood polymorphonuclear leukocytes (PMNs) and their significance in rats with acute lung injury (ALI). METHODS: ALI model in rat was established by intravonous injection of E. coli endotoxin (ET). The expression of L-selectin on peripheral blood PMNs was measured by immunofluorescence and flow cytometry.RESULTS: The contribution of L-selectin fluorescence signal was on the surface of PMNs membrane. The expression of L-selectin on poeripheral blood PMN was significantly lower at 5 min after injection of ET and the lowest during 15 min to 30 min, then gradually increased, but the expression of L-selectin on PMN was lower at 60 min after injection of ET than that of control animal.CONCLUSION: In physiological state, L-selectin were expressed on the surface of PMN membrane. The protein expression of L-selectin on PMNs reduced rapidly after injection of ET and the lowest at 15 min, then gradually increased. L-selectin may play a role in the development of ALI.
2.Correlation between flagellin and TNF-? in acute lung injuries induced by sepsis in rats

Objective The aim of present study was to observe the correlation between flagellin and tumor necrosis factor ?(TNF-?)in rats with sepsis-induced acute lung injury(ALI).Methods A total of 120 male healthy Wistar rats were randomly assigned into 2 groups:sepsis group which was reproduced by cecal ligation and puncture(CLP),and sham operated group which underwent laparotomy only.All the indexes were observed at the time points of 2h,4h,6h,12h,24h and 48h after operation.Changes in partial pressure of oxygen in artery(PaO2)were determined by blood gas analysis.Pathological changes in pulmonary-tissue were observed by light microscopy.The levels of flagellin in serum,bronchoalveolar lavage fluid and pneumono-homogenate,as well as TNF-? of serum,were determined by enzyme linked immunosorbent assay(ELISA).Results Rats with acute lung injury induced by sepsis was successfully reproduced.The PaO2 of sepsis group significantly decreased at 12h after injury and reached the lowest point at 48h after injury.PaO2 of sepsis group was remarkably lower than that of sham operated group at the time points of 12h,24h and 48h after injury(P

4.Inhibition of sialic acid related substances on Pseudomonas aeruginosa infection in rat trachea vulnerated by hydrochloric acid

Objective To investigate whether sialic acid disaccharides, specificity agglutinins and neuramidinases could inhibit Pseudomonas aeruginose infection in rat trachea vulnerated by hydrochloric acid. Methods One hundred and forty male Wistar rats were vulnerated by hydrochloric acid through intratracheal instillation and then inoculated by Pseudomonas aeruginose admixed Sial ?2,3-Gal, Sial ?2,6-GalNAc, Maackia amurensis agglutinin, Sambucus nigra agglutinin, specificity ?2,3-neuramidinase or non-specificity neuramidinase respectively. Thirty minutes and 48 h after inoculation, whole trachea of 8 rats in each group was weighed, homogenated, and quantitative bacterial culture was performed while whole trachea of another 2 was fixed in 10% methanol, embedded with paraffin and HE stained. Results Sial ?2,3-related disaccharide, agglutinin and neuramidinases could decrease colony count of trachea 30 min after inoculation remarkably. But only Sial ?2,3-Gal and neuramidinases could obviously decrease colony count 48 h later. Sial ?2,6-related substrances have not analogical effects. Conclusion Sial ?2,3-related substrances could interfere Pseudomonas aeruginose binding/multiplication in the trachea, especially Sial ?2,3-Gal and neuramidinase.
5.The expression of Toll-like receptor 5 in the lung tissues of rats with acute lung injury induced by flagellin

Objective To determine the expression of Toll-like receptor 5(TLR5) mRNA in the lung tissues of rats with acute lung injury induced by flagellin, and to investigate its potential role in the pathogenesis of the disease. Methods Flagellin was isolated and purified from Escherichia coli ATCC25922, subsequently identified by monoclonal antibody to flagellin. One hundred and eight SD rats were randomly divided into control group (n=36), flagellin challenged 1 group (n=36) and flagellin challenged 2 group (n=36). Rat model of ALI was reproduced by injecting flagellin. The expression of TLR5 mRNA in the lung tissues of rats was determined with in situ hybridization technique at six time points. Blood gas was monitored and pathological changes in the lung was observed at the same time. Result Flagellin was isolated and purified successfully and its molecular weight was approximately 65kD. Flagellin-induced acute lung injury model was successfully reproduced in rats. From 1h after flagellin injection, TLR5 mRNA expression was found to be increased in the lung tissues of rats with flagellin induced acute lung injury, and the expression was on the increase with the elapse of time and increase of the dose of flagellin. The PaO_2 levels in flagellin challenged 1 and 2 groups decreased. Interstitial edema, alveolar edema and inflammatory cells infiltration were also observed in flagellin challenged groups. Conclusion Flagellin can induce ALI in rats; the development of flagellin induced ALI in rats is related with TLR5 expression in the lung tissues of rats.
6.Relativity of beta adrenoceoptor changes between polymorphonuclear leukocytes and lung tissue in acute lung injury rats

AIM: To observe changes of beta - adrenoceptor (?-AR) and its relativity to polymorphonulear leukocytes (PMN) and acute lung injury (ALI) in rats. METHODS: ALI modle in rat was established by in- travonous injection of E. Coli endotoxin (ET). ? - AR was measured by radioligand binding assay with [3H]-dihy- droalprenlol. RESULTS: (1)Maxmal capacity of ? - AR (Bmax) in both lung tissue and PMN decreased significantly at the lst, 4th and 6th hours after injection of or. (2)No relativity was found between ?-AR Bmax changes of lung tissue and ?-AR Bmax changes of PMN. CONCLUSIONS: (1) Decrease of density of 3-AR in lung tissue and PMN may played a role in the development of ALI. (2) The ?-AN of PMN in circular bind can not be used as a relative index of ?-AR changes of lung tissue in rats with ALI.
7.Lipopolysaccharide-binding protein inhibitory peptide inhibits the binding of LPS to U937 cells

AIM:To investigate the inhibitory effect of P12,a kind of lipopolysaccharide(LPS)-binding protein(LBP) inhibitory peptide,on the binding of LPS to macrophage in vitro.METHODS:Human monocyte-like cell line(U937 cells) was grown in RPMI-1640 and stimulated with PMA in order to induce their differentiation to macrophage stage.The relative affinity of P12 to LPS was determined by enzyme-linked immunosorbent assay(ELISA).The effects of P12 on the binding of LPS to U937 cells were determined by flow cytometry analysis.The production of tumor necrosis factor-alpha(TNF-?) was measured by ELISA.RESULTS:The relative binding activity of P12 to LPS was higher than that of LBP in the same mass concentration.P12 inhibited the binding of FITC-conjugated LPS(FITC-LPS) to U937 cells.The productions of TNF-? was also significantly suppressed by P12.CONCLUSION:The results suggest that blockage of LBP at the inflammatory sites might attenuate LPS-induced circulatory shock.
8.Clinical analysis of pulmonary tuberculosis infection in renal transplantation recipients

Objective To investigate the diagnosis and treatment of pulmonary tuberculosis in renal transplantation recipients.Method The clinical data of 8 renal transplantation recipients suffering from pulmonary tuberculosis infection were retrospectively analyzed.Result Fever,cough and expectoration were the most common symptoms,however,lacking typicality.Images of chest Xray and CT scan were various and couldn't verify TB infection from pneumonia.Seven of 8 cases were diagnosed through invasive methods,either bronchofibroscope or fiberthoracoscopy.Immunosuppressants were decreased in all cases.Three-drug regimens,including isoniazide,rifampicin and ethambutol or pyrazinarnide,were administrated as anti-tuberculosis chemotherapy.All the cases were cured,without episodes like acute rejection and liver function impairment.Conclusion Routine examinations are not sufficient to diagnose pulmonary tuberculosis in kidney transplantation recipients.While,invasive methods like bronchofibroscope and fiberthoracoscope are helpful.When diagnosed,patients should receive normative anti-tuberculosis treatment and immunosuppressive agents adjustment,which can benefit the prognosis of pulmonary tuberculosis in renal transplantation recipients.

ObjectiveTo explore the clinical efficacy of minitype vascularized superficial peroneal artery's singleness perforator flap,and then to accurately repair skin and soft tissue defects of hands and foots. MethodsFrom November 2009 to January 2011,eight cases(one case of left foot,one case of right foot,three cases of left hand and 3 cases of right band,1.3 cm × 5.0 cm - 2.5 cm× 6.0 cm of skin and soft tissue defects)were treated by minitype vascularized superficial peroneal artery's singleness perforator flap.Based on preoperative applied anatomy papers, it was the superficial peroneal artery's perforator position in the middle of the lateral lower leg to the fibula head;We designed the flap based on the size and shape of skin defect,and then to analysize the flap design,lap cut,he vascular anastomosis of flap and recipient,effect and characteristics of survival. ResultsAll the flaps(1.5 cm × 5.2 cm-2.8 cm × 6.2 cm) survived and satified in shape and texture, they acquired good functional recovery postoperation;There was blister,dark purple but survival after the scab off on 1 patient;There was phalanx ostomyelitis and healed after treatment on 1 patient.ConclusionThe clinical effects were satisfactory for repairing small skin and soft tissue defect of hands and foots by minitype vascularized superficial artery's singleness perforator flap.
