1.Chronic mountain sickness modulated cerebrovascular reactivity and biological effects on the related vascu-lar responses cytokines

Objective To investigate the influence of the chronic mountain sickness (chronic mountain sickness, CMS) on Cerebrovascular reactivity (CVR) and regulators of cerebrovascular responses. Methods Twenty-six CMS pa?tients and 23 healthy control group using transcranial Doppler ultrasound (Transcranial Doppler Ultrasoiund, TCD) as?sessment of CVR, enzyme-linked immunosorbent assay were applied to measure the serum levels of the endothelin (endo?thelin 1 , ET-1) and its receptor (endothelin receptor B, ETBR), endothelial nitric oxide synthase (endothelial nitric oxide synthase, eNOS) in CMS patients and healthy control. Results CVR (3.84 ± 3.01) was significantly lower in CMS patients than in the healthy control CVR (6.39 ± 6.87) (P <0.05); the serum concentration of ETBR in CMS patients was higher than in control [(386.07±281.57)μg/μL vs.(312.30±238.07)μg/μL] (P<0.05). Conclusions The cerebral circulation re?serve in CMS patients is significantly lower compared with healthy people. The regulation of vascular capacity by serum ET-1 and eNOS is similar between CMS patients and healthy control. The hypoxic vasodilatation in CMS patients is close?ly associated with cerebrovascular high expression of ETBR. This study may provide a scientific basis for the prevention and treatment of cerebral infarction in the patients with chronic mountain sicknes.

Objective To evaluate the effect of Gamma nail,PFN(proximal femoral nail)and recon-struction nail fixation in treatmen t of intertrochanteric fractures an d problems regarding the operative t echnique.Methods This study reviews the X-ray films an d complications of one hundred forty-five cases of in-tertrochanteric fractures treated with intramedullary nail at Beijing Jishuitan Hospital from March 1998t o October2001.Results All cases healed ,one case died eight months postoperatively.The distal fragment was broken intraoperatively in two cases.Ante roposterior and lateral and rotatio n displacement occured intraoperatively in fifteen cases.Limb discrepancy over one centimeter occurred in five cases.Conclusion Intramedullary nail is a good device to treat intertrochante ric fractures with vartous coxa varu s the common complication.Skillful in-tramedullary nail technique is the k ey to decrease the complications.In cidence of coxa varus in the stable ty peⅡis less than that in the unstable typeⅠafter the treatment.[
3.Chronic Toxic Effect of Herbicide Mixture on Mice Hepatic Function and Ultrastructure

Objective To observe the chronic hepatotoxicity of the commonly used herbicide mixture in the mice. Methods Low doses herbicide mixture diluted in mice drinking water. The parameters related to hepatic function and ultrastructural changes of the hepatocytes were examined after 18 weeks and 24 weeks of treatment. Results No changes was observed in every parameter after 18 weeks of treatment compared with the control group. After 24-week of treatment, the electron microscope revealed the ultrastructural changes in the hepatocytes, rough endoplasmic reticulum showed dilatation, mitochondria showed matrix muddy with distortion and disappearance of inner crista and the serum transaminase were higher than those in the control group(P

5.EFFECT OF LIPOPOLYSACCHARIDE ON EXPRESSION OF INTERLEUKIN-6 IN COCULTURED LUNG MICROVASCULAR ENDOTHELIAL CELLS

Rat lung microvascular endothelial cells (LMVEC) were cocultured with rat pulmonary arterial smooth muscle cells (PASMC). The cultures were divided into 5 groups: normal group (N), lipopolysaccharide (LPS) 2h (L 2), LPS 8h (L 8), LPS 16h (L 16 ), and LPS 24h (L 24 ). The activity of IL 6 in LMVEC and PASMC supernatants was determined by radio immunity method, and the expression level of IL 6 mRNA in LMVEC was detected with RT PCR. The results showed that the activity of IL 6 in LMVEC and PASMC was enhanced by LPS at L 2, reaching the higher level after 8 hour stimulation. Furthermore, the activity of IL 6 was stronger in homotypic groups than that of coculture groups. The expression of IL 6 mRNA in LMVEC was increased in L2 group, and highest in L6 group, but lowered in L12 group, though it was still stronger than that of N. It suggest that IL 6 mRNA and the activity of IL 6 in LMVEC and PASMC were enhanced in both homotypic and coculture groups by LPS. And the mRNA level and activity of IL 6 were higher in homotypic groups than that of coculture groups.
6.Construction of bait plasmid containing SIGIRR cytoplasmic tail and detection of self-activation in yeast two hybrid system

Objective To construct the bait plasmid of pSos-single immunoglobin IL-1 receptor related protein (SIGIRR) in Cy-toTrap yeast two hybrid system ,and to test its self-activation .Methods The cDNA fragments of SIGIRR(480 -1 230 bp) were amplified from pReceiver-LV19-SIGIRR and ligated into the bait plasmid pSos to generate the plasmid pSos-SIGIRR .The pSos-SI-GIRR was identified by DNA sequencing and dual-site endonuclease digestion .Then the recombinant plasmid and control plasmid were introduced into the yeast cell cdc25H .The transformants were inoculated on plates of 25 ℃ /SD/Glucose(-UL) ,25 ℃/SD/Ga-lactose(-UL) ,37 ℃ /SD/Glucose(-UL) and 37 ℃ /SD/Galactose ,respectively and the proliferation ability of transformant was ob-served for 6 d .The Western blot was adopted to detect the expression of target protein .Results The pSos-SIGIRR vector was cor-rectly constructed and proved of no self-activation and toxic action .The Western blot showed that the target protein was expressed in a form of fusion protein of 170KD .Conclusion The bait plasmid containing SIGIRR cytoplasmic tail can be applied to the yeast two-hybrid system and lays the important foundation for seeking the interacting protein with SIGRR from the human lung cDNA li-brary in .
7.Cloning and identification of partial positive regulatory sequence of Na+/H+ exchanger-1 gene from human lung cancer cells

Objective To clone the partial positive regulatory fragment of Na+/H+ exchanger-1 (NHE-1) gene from human lung cancer cells. Methods After BamHⅠ and EcoRⅠ cut sites were added to the 5' ends of the upstream and downstream primers respectively, the partial positive regulatory sequence of NHE-1 gene was cloned with the length of 170 bp from genomic DNA of lung cancer cell line A549 cells with PCR method. The cloned fragment was ligated to plasmid pUC18. Finally, the constructed recombinant was identified with enzyme cut, PCR and DNA sequencing. Results The cloned fragment was about 170 bp in size and successfully ligated to pUC18 with identifiation of double enzyme cut and PCR. DNA sequencing approved that the fragment cloned was objective one with 168 bp in length. Compared with the reported sequence, two t were lost. Conclusion The positive regulatory fragment of NHE-1 gene from human lung cancer cells was successfully cloned.
8.Imaging Characteristics and Diagnostic Value of Primary Ureter Carcinoma

Objective To explore the imaging characteristics and the diagnostic value of primary ureter cancer.Methods 24 cases of primary ureter carcinoma underwent ultrasound(US), intravenous pyelography(IVP),retrograde pyelography and CT scan.Those imaging features were analyzed retrospectively.Results The postive rate of US,IVP,retrograde pyelography and CT was 45.8%(11/24),37.5%(9/24),88.8%(16/18)and 62.5%(10/16).Conclusion Ultrasound(US) is a more value examination for making the choice of cases; IVP is an essential method for understood function of the kidneys, showing carcinoma; retrograde pyelography is most value in the diagnosis of primary ureter carcinoma;CT scan is more important diagnostic values in area,properties and extent of tumor.
9.Effects of sodium nitroprusside (SNP) on TLR4 in U937 induced by LPS

Objective:To investigate the effects of nitric oxide donor,sodium nitroprusside(SNP)on the expression and production of TLR4 in U937 induced by LPS.Methods:U937 was induced to maturation by PMA and then stimulated by LPS.Then the cells were treated with SNP and divided into four groups:control group without stimulation of LPS,LPS group(10 ng/ml LPS+100 ng/ml rhLBP),low dose SNP group and high dose SNP group(50,500 ?mol/L SNP respectly).The mRNA and protein of TLR4 was determined by RT-PCR and Western blot.Results:The mRNA of TLR4 in SNP groups was lower than those in LPS group(0.308?0.050 and 0.138?0.0044 vs 0.342?0.098,P0.05).Conclusion:SNP might have a potential protective role in LPS induced inflammation such as sepsis and acute lung injury through inhibiting the mRNA and protein expression of TLR4.
10.Preparation of a new cationic liposome and determination of its gene transfection rate

Objective To preparation a new cationic liposome of which the positive component is a new cytofectin as a gene delivery vehicle for laboratory research of gene transfection and gene therapy in lung diseases. Methods The new cationic liposome was prepared by film. Electron microscopy and gel electrophoresis were employed to define the condition for transfection. The prepared cationic liposome was used in the transfection of a fluorescence plasmid to lung cancer cell line SPC and the rate of the transfection was evaluated. Results The prepared new cationic liposome was global microcapsule in size of 100 nm-1 ?m. The highest transfection rate of 60% was attained when the liposome was in a ratio of (6-8)∶1 with plasmid. Conclusion The new cationic liposome is prepared with cytofectin N 4-spermine cholesteryl carbamate as its positive component, and sound transfection activity is observed under a certain circumstance. It provides a basis for gene transfection and gene therapy.
