Objective To investigate the role of the expression of p27 and cyclin E in the pathogenesis of Bowen's disease and squamous cell carcinoma (SCC). Methods The expression of p27 and cyclin E was assessed by immunohistochemical method in 16 patients with Bowen's disease, 53 patients with SCC and 25 normal controls. Results The expression of p27 was lower in Bowen's disease and SCC compared to that in the normal skin, and the expression level decreased with a decline in the differentiation of tumors, i.e. the lowest expression was observed in poorly differentiated SCC, followed by highly differentiated SCC and Bowen's disease. The expression of cyclin E was higher in Bowen's disease and SCC than that in the normal skin, and the expression level increased with a decline in the differentiation of tumors. The expression of p27 was inversely correlated with that of cyclin E among all the cases of Bowen's disease and SCC. Conclusion The abnormal expression of p27 and cyclin E may contribute to the pathogenesis of Bowen's disease and SCC.

Objective To observe the clinical effect of artificial ventilation combined continuous positive airway pressure(CPAP) with removal of tracheobronchial foreign bodies for children and to explore the possibility and security of the method. Methods 60 children with tracheobronchial foreign body, underwent total intravenous anesthesia ,were randomly divided into A group and B group. Each group had 30 cases. A group was given artificial ventilation with CPAP. The bronchofibroscope was connected to anesthesia machine with side hole after induction for 3 minutes, and high fresh gas flow(10 ~ 15L/min) was given to maintain continuous positive airway pressure. B group were given high frequency jet ventilation(HFJV) ,60 ~ 100 bpm. The mask ventilation was given in stand of bronchofibroscope when SpO2 < 90% and until SpO2 improved. MAP, HR, ECG, SpO2, PaO2, PaCO2 were monitored and recorded at time points: T0 (entered operation room), T1 (beginning of bronchofibroscopy), T2 (5 min after bronchofibroscopy), T3 (10 min after bronchofibroscopy), T4 (end of operation). The side effects, the rate of fail to bronchofibroscopy and the rate of intubations after operation in two groups were observed and recorded. Results The HR of post-anesthesia in two groups significantly decreased than those at T0 (P < 0.01), but no difference showed in HR between two groups(P > 0.05). SpO2 and PaO2 of post-anesthesia in two groups significantly increased than those at T0 (P <0. 01) ,PaO2 at T1 ,T2 ,T3 in A group were significantly higher than those in B group(P <0.05). PaCO2 gradually increased after bronchofibroscopy in two groups ,and the values in A group was significantly lower than in B group(P <0.05 or 0. 01). There were no significant differences in the rates of fail to bronchofibroscopy and of intubations after operation between two groups, but the total number of B group was higher. Conclusion Artificial ventilation with CPAP for children with removal of tracheobronchial foreign bodies was safe and practical, and has a better controllability, a minor effect to respiratory function, deserve popularizing.

Background and Purpose:Prostate cancer is one of the most common cancers and the second leading cause of cancer-related death in Europan and North American males. The incidence of prostate cancer has also been increasing during the past few decades in China. It is widely accepted that this heterogeneity, which results from the tumor progression driven largely by genomic instability(genetic and/or epigenetic alterations)of tumor cells in primary tumor, endows specific populations of tumor cells with the unique character needed for invasion, migration, and metastasis colony formation in other organs and only these subpopulations possessing thost character can survive the potentially destructive journey from the primary tumor to the sites of metastases. The purpose of the present study was to explore the genes associated with invasion and metastasis of human prostate cancer cell line PC-3M in nude mice. Methods:After PC-3M cells were inoculated into orthotopic site(prostate) in a male nude mouse for two months, tumor cells were isolated from the primary tumor and lymph node metastasis, separately.Cell invasion and adhesion ability in vitro were first compared between two cells. Then metastasis-related genes differentially expressed between them were analyzed by utilizing cDNA microarray technique. Results:The in vitro cell invasion and adhesion potential of tumor cells from lymph node metastasis was significantly higher than those from primary tumor by 2.5 fold and 1.5 fold,respectively.Metastasis-related genes differentially expressed between those two sublines were identified, all of them were up-regulated in the tumor cells from lymph node metastasis and could be categorilized: 1.genes encoding cellular matrix-degrading proteolytic enzyme including cathepsin and MMP.2.genes encoding transcription factors.3.genes related to heterotypic adhesion of tumor cells.4.genes encoding cell surface receptors. Conclusions:There are significant differences in invasion and adhesion potential between cells from primary tumor and those from lymph node metastasis. Some differentially expressed molecules might be playing pivotal roles in promoting tumor cells to migrate from primary tumors to distant metastases, which may be helpful to elucidate the possible mechanism of metastasis in prostate cancer.

The transposon vector containing enhanced green fluorescent protein (EGFP) was injected into early housefly (Musca domestica L.) eggs by microinjection method to realize stable gene expression in vivo for verification, and to study housefly gene function. A borosilicate glass micro injection needle suitable for microinjection of housefly eggs was made, the softening treatment conditions of housefly egg shells were explored, and a microinjection technology platform suitable for housefly was constructed with a high-precision microsyringe Nanoject Ⅲ as the main body. The recombinant plasmid PiggyBac-[3×P3]-EGFP containing the eye-specific 3×P3 promoter and EGFP and the stable genetic expression helper plasmid pHA3pig helper were microinjected into the treated housefly eggs. After emergence, the eye luminescence was observed, and the expression and transcription level of EGFP were detected. The results showed that the normal hatching rate of housefly eggs was 55% when rinsed in bleaching water for 35 s. The hardness of the egg shell treated for 35 s was suitable for injection and the injection needle was not easy to break. About 3% of the emerged housefly eyes had green fluorescence. Through further molecular detection, EGFP specific fragments with a size of 750 bp were amplified from DNA and RNA of housefly. Through the technical platform, the stable expression of reporter genes in housefly can be conveniently and effectively realized, and a bioreactor with housefly as the main body can be established, which provides certain reference value for subsequent research on housefly gene function.
