1.Assay of ginsenoside Rb_1,ginsenoside Rg_1 and notoginsenoside R_1 in Sanxue Mingmu Tablet

AIM: To develop a method for determining contents of ginsenoside Rb_1,ginsenoside Rg_1 and notoginsenoside R_1 in Sanxue Mingmu Tablet(Radix et Rhizoma Notoginseng,Pheretima,Herba Leonuri,Rhizoma Imperatae,etc.). METHODS: The contents of ginsenoside Rb_1,ginsenoside Rg_1 and notoginsenoside R_1 were separated by gradient elution on Hypersil ODS C_(18) column(5 ?m ? 4.6 mm?100 mm) as stationary phase;acetonitrile and water as gradient mixed mobile phase;detected at 203 nm wavelength;(25 ?C) of column temperature and 1.0 mL/min of mobile rate. RESULTS: Ginsenoside Rb_1,ginsenoside Rg_1 and notoginsenoside R_1 had good linearities(r=0.999 9;n=6) at the ranges of 1.480 0-14.800 ng;0.372 0-3.720 ng and 1.072 0-10.720 ng.The average recoveries were 98.44%(RSD=0.96%);99.02%(RSD=0.94%);97.95%(RSD=(1.02%)),respectively. CONCLUSION: This method is high in column efficiency and satisfactory for isolation and repeatability,the result is accurate.It can be adopted for quality control of Sanxue Mingmu Tablet.
2.Analysis of risk factors in different gender patients with atherosclerotic renal artery stenosis

Objective To analyze the relationship between traditional & emerging risk factors and the pathogenesis of atherosclerotic renal artery stenosis (ARAS) in different gender patients.Methods Selective renal artery angiography was performed immediately after routine coronary angiography in 2060 patients with suspected coronary artery disease.Traditional & emerging risk factors were recorded and compared in different gender patients with ARAS.Results Among the patients with ARAS, smoking (59.18% vs.12.50%, x2 =17.47,P<0.01) and diabetes(28.57% vs 46.87%, x2 =5.64, P<0.05) compared with both genders were significantly differences.There were significant differences in high density lipoprotein cholesterol, ApoA1, ApoB/ApoA 1, HbA1C and Hcy (t =2.62,2.07,-2.83,-2.37,3.74;P< 0.05) in the ARAS patients compared with both genders.Conclusion The main risk factors in different gender patients with ARAS were different.It is important to adopt appropriate control strategies.

Treatment by way of pasting TCM on acupoints has a long history and the clinical efficacy is determined. In this article, based on the concept of TCM wholism and meridian theory, the mechanism of acupoint pasting therapy was expounded from the aspects of medicine effects on local stimulation of the body, meridian conduction, medicine transdermal absorption. Combining with the research hot spots of modern medicine transdermal delivery system, metabolomics, and efficacy material, this article proposed a new study mode of systems biology of TCM.
4.Research Progress in Animal Models of IBS-D Disease Combined with Liver Stagnation and Spleen Deficiency

At present, TCM treatment of diarrhea-predominant irritable bowel syndrome (IBS-D) is based on the combination of disease differentiation and syndrome differentiation. Therefore, the establishment of IBS-D of liver stagnation and spleen deficiency syndrome combined with animal model as a combination of traditional Chinese and Western medicine innovation theory has become increasingly concerned about, and gradually become a new direction for the development of TCM experimental animal model. This article reviewed the research progress in IBS-D liver and spleen deficiency syndrome in recent years, discussed the establishment of IBS-D liver stagnation and spleen deficiency animal model and research ideas for the treatment of IBS-D, and provided references for mechanism research of TCM treatment for IBS-D and research and development of new medicine.
5.Changes in serum level of carboxy-terminal telopeptide of type I collagen in patients with coronary heart disease

Objective To investigate the serum level of carboxy-terminal telopeptide of type I collagen (ICTP) and explore its correlation with MMP-2 and MMP-9 in patients with coronary artery disease (CHD). Methods A total of 103 CHD patients treated in our hospital between October, 2013 and May, 2014 were enrolled, including 39 with stable angina pectoris (SAP), 39 with unstable angina (UA), and 25 with acute myocardial infarction (AMI), with 38 non-CHD volunteers as the control group. The serum levels of ICTP, MMP-2, and MMP-9 were detected in all the subjects using enzyme-linked immunosorbent assay (ELISA). Results No significant difference in serum levels of MMP-2, MMP-9, or ICTP was found between the control and SAP groups or between UA and AMI groups (P>0.05), but the latter two groups had significantly higher serum levels of MMP-2, MMP-9, and ICTP than the former two groups (P<0.05). Serum ICTP level was found to negatively correlated with the fibrotic area and positively with the lipid component in the plaques (P<0.05). Regression analysis revealed significant positive correlations of serum ICTP with MMP-2 and MMP-9 (P<0.05). Conclusions An elevated serum ICTP level is indicative of the presence of unstable plaques in CHD patients. Serum ICTP is more strongly correlated with MMP-2 than with MMP-9, and can be used as a non-invasive marker for assessing vulnerable plaques in patients with acute coronary syndrome.

7.Protective Effects and Mechanism of Modified Baishile Decoction on Hippocampal Neuronal Cell Injury Induced by Oxygen Glucose Deprivation Combined with Lipopolysaccharide

Objective To observe the protective effect of modified Baishile Decoction on hippocampal neuronal cells cultured in vitro;To explore its mechanism of treating post-stroke depression.Methods Hippocampal neuronal cells from mammary rats were isolated and cultured in vitro,cell injury was induced by oxygen glucose deprivation combined with lipopolysaccharide.The cells were divided into normal group,model group,blank serum group(10%)and modified Baishile Decoction containing serum group(10%).Invertedmicroscope was used to observe cell morphological changes,CCK-8 method was used to detect cell survival rate,Hoechst33342 staining was used to observe apoptosis,ELISA was used to detect Glu,5-HT,TNF-α,IL-1β,and IL-6 contents in cell supernatant,the expressions of purinergic P2X7 receptor(P2X7R)and NOD-like receptor protein 3(NLRP3)were detected by immunofluorescence staining.Results Compared with the normal group,the hippocampal neurons in the model group showed significant changes in cell morphology,the cell survival rate significantly decreased(P<0.01),the cell apoptosis increased(P<0.01);Glu,TNF-α,IL-1β,IL-6 contents in cell supernatant significantly increased(P<0.05,P<0.01),5-HT content significantly decreased(P<0.01),P2X7R and NLRP3 expressions in hippocampal neuronal cells significantly increased(P<0.01).Compared with the model group,the morphology of hippocampal neurons in modified Baishile Decoction containing serum group was significantly improved,the cell survival rate significantly increased(P<0.01),the cell apoptosis reduced(P<0.01);Glu,TNF-α and IL-1β content in cell supernatant significantly reduced(P<0.05,P<0.01),5-HT content significantly increased(P<0.01),and P2X7R and NLRP3 expressions in hippocampal neuronal cells significantly decreased(P<0.01).Conclusion Modified Baishile Decoction may exert a protective effect on oxidative glucose deprivation combined with lipopolysaccharide induced hippocampal neuronal inflammation damage by inhibiting the P2X7R/NLRP3 signaling pathway,regulating neurotransmitter secretion,and inhibiting inflammatory factor release,thus treating post-stroke depression.
8.Study on the Rationality Evaluation Indicator System of Antibiotics with Delphi Method

OBJECTIVE: To provide scientific evidence for establishing perfect rationality evaluation indicator system of antibiotics. METHODS: On the basis of literature research, Delphi method was used to conduct several rounds of consultations on 30 experts in related fields with E-mail and questionnaire field investigation. After several rounds of consultations, rationality evaluation index system for antibiotics was determined. The effective recovery rate of expert consultation questionnaire was used to express the positive coefficient of experts; the authoritative coefficient was used to express the authoritative degree of experts; the index importance assignment and the full score ratio were used to reflect the concentration degree of experts’ opinions; the coefficient of variation and the coefficient of coordination were used to express the coordination degree of experts’ opinions. RESULTS & CONCLUSIONS: After two rounds of consultations, response rates of the questionnaire in two rounds of surveys were 100% and 96.67%, indicating experts were highly motivated. The authoritative coefficients were 0.91 and 0.88, indicating experts had a high degree of authority. Finally, an evaluation index system for rationality of antibiotics was established, which included four first-level indicators (indications, drug selection, medication process, management indicators) and 35 second-level indicators. Among them, the mean value of importance scoring of first-level indicators ranged from 4.28 to 5.00, the full score ratio from 0.93 to 1.00, the coefficient of variation from 0.00 to 0.15 and the coefficient of coordination was 0.446 (P<0.001). While, the mean value of importance scoring of second-level indicators ranged from 3.83 to 4.79; the full score ratio from 0.67 to 1.00; variation coefficient from 0.10 to 0.26 and the coefficient of coordination was 0.115 (P<0.001), which indicating the system was reliable.
