Objective To evaluate the differences of serum RANTES(regulated on activation, normal T cell expressed and secreted), MCP-1 (monocyte chemoattractant protein), and SDF-1β (stromal cell-derived factor-1β) in patients with acquired immunodeficiency syndrome (AIDS) and healthy people. Methods 38 AIDS patients who were admitted to a hospital between January 2010 and January 2015 were as AIDS groups, 38 healthy persons were as a healthy group, serum levels of RANTES, MCP-1, and SDF-1β in two groups were detected, and the subgroup analysis was carried out according to the viral load. Results Serum levels of RANTES, MCP-1, and SDF-1β in AIDS group were (1 392.55±227.69)pg/mL,(450.91±103.04)pg/mL, and(104.82±22.52)pg/mL respectively, all were significantly higher than those in healthy group([120.58±55.87] pg/mL, [74.25±33.62] pg/mL, and [39.04±11.43]pg/mL respectively)(all P<0.05).Among AIDS patients with HIV viral load 4≤Log(VL)<5 and Log(VL)≥5, serum RANTES were (1 470.34±155.01)pg/mL and (1 408.29±181.54)pg/mL respectively, which were both significantly higher than patients with HIV viral load Log(VL)<4([1 183.12±174.54]pg/mL);serum MCP-1 and SDF-1β levels in AIDS patients with HIV viral load 4≤Log (VL)<5 were (537.93±89.32)and(149.31±18.05)pg/mL respectively, which were significantly higher than patients with HIV viral load Log(VL)≥5([410.26±80.57] pg/mL, [81.53±20.31]pg/mL respectively) and HIV viral load Log(VL)<4([381.71±77.26] pg/mL, [72.90±21.62]pg/mL respectively), differences were both statistically significant(both P<0.05).Conclusion Serum levels of RANTES, MCP-1, and SDF-1β are significantly increased in AIDS patients, which are related to the level of viral load.

Objective To detect the serum levels of C-reactive protein (CRP),procalcitonin (PCT),and interleukin-6(IL-6) and evaluate their prognostic values in liver cirrhosis with systemic inflammatory response syndrome(SIRS).Methods A total of 56 patients with liver cirrhosis and systemic inflammatory response syndrome were divided into two groups, improved group(n =39)and death group(n=1 7).Serum CRP、PCT、IL-6 levels were measured at the first day, third day and seventh day after SIRS. Results CRP,PCT and IL-6 levels of two groups were no statistically significant difference in the first day (P >0.05).PCT and IL-6 levels of two groups were no statistically significant difference in the third day (P >0.05).CRP levels in improved group were lower than those in death group in the third day (P <0.05).CRP,PCT and IL-6 levels in improved group were lower than those in death group in the seventh day (P <0.05).There was no statistically significant difference of CRP,PCT and IL-6 at the first day, third day and seventh day in death group (P >0.05).CRP levels at the third day and seventh day were lower than those at the first day in improved group (P <0.05).PCT and IL-6 levels at the seventh day were lower than those at the first day in improved group (P <0.05).Conclusion Poor prognosis was showed at sustained high levels of CRP,PCT and IL-6.Compared with PCT and IL-6, CRP might be more rapid in showing condition improved.

Objective To establish an efficient HPLC method for the determination of uric acid in plasma of hyperuricemia model mice, and the evaluation of uric acid lowering effect of Lesinurad. Methods The Laballiance Series Ⅲ HPLC system was adopted with Kromasil C18 column (100 mm × 4.6 mm,5 μm).The mobile phase consisted of methanol-0.5% acetic acid (10:90) for isocratic elution with a flow rate of 0.4 mL/min.The detection wavelength was set at 283 nm.The established HPLC method was used to detect the plasma uric acid level of mice at 0.5,1.0,and 2.0 h time points after which being ip injected with 250 and 500 mg/kg uric acid.Lesinurad of 250 and 500 mg/kg was ig given to mice,0.5 h later, mice were ip injected with 500 mg/kg uric acid to establish hyperuricemia model, and 1 h later, the established HPLC method was used to detect the plasma uric acid level of mice. Results There was a good linear relationship between peak area and the concentration of plasma uric acid in the range of 7.5-150 μg/mL (r =0.997).The specificity, repeatability, precision, stability, and recovery of the established HPLC method was in accordance with the guiding rules of biological sample determination.Compared with the endogenous serum uric acid concentration of control group mice, serum uric acid concentration of 250 mg/kg dose group was significantly increased 0.5 h after ip administration with uric acid (P < 0.01),and serum uric acid concentration of 500 mg/kg dose group was significantly increased 0.5,1.0,and 2.0 h after ip administration with uric acid.Compared with model group, the concentration of uric acid in plasma decreased significantly in low dosage group administered with Lesinurad (P < 0.05),while decreased more significantly in high dosage group (P < 0.01).Conclusion This convenient, rapid, and accurate method can be applied to the determination of uric acid in mouse plasma and the evaluation of relative drugs, which provide an efficient analysis way for establishing hyperuricemia model and screening relative drugs.

Objective:To investigate the effect of diabetes mellitus on pulmonary uptake of sevoflurane. Methods:Twenty patients with type 2 diabetes mellitus, aged 40-64 yr, with body mass index of 18.5-22.9 kg/m 2, of American Society of Anesthesiologists physical status Ⅰ or Ⅱ, undergoing elective surgery with general anesthesia, served as diabetes group (group D). Twenty non-diabetic patients matched by age, gender and surgery were selected as control group (group C). After anesthesia induction and tracheal intubation, sevoflurane was inhaled at a concentration of 2% (oxygen flow 2 L/min). The inhaled concentration (Fi) and exhaled concentration (Fa) at 1, 3, 5, 10, 15, 20 and 30 min of inhalation of sevoflurane were recorded, and the Fa/Fi ratio was calculated. The time required for the Fa/Fi ratio to reach 0.7 was recorded. Results:Compared with group C, the Fa/Fi ratio was significantly increased at each time point, and the time required for the Fa/Fi ratio to reach 0.7 was shortened in group D ( P<0.05). Conclusion:Diabetes mellitus can reduce pulmonary uptake of sevoflurane in the patients.
