1.Effects of PTEN and its mutants on AKT phosphorylation in gastric cancer cells

Objective To study the effects of PTEN and missense mutations in PTEN phosphotase domain on AKT phosphorylation in AGS and BGC-823 cells. Methods The plasmids of wtPTEN,PTEN-C124S which PTEN mutant is in both lipid and protein phosphotase domain and PTEN-G129E which PTEN mutant is only in lipid phosphotase domain were respectively transfected into AGS and BGC-823 cells. The cells were stimulated with insulin or rhEGF after serum starvation overnight. The levels of AKT phosphorylation were detected by Western blot. Results Both insulin and rhEGF can activate AKT phosphorylation in gastric cancer cells. Overexpressed PTEN inhibitedAKT phosphorylation induced by insulin or rhEGF(P < 0.05). PTEN mutants C124S or G129E could not inhibitAKT phosphorylation(P > 0.05). Conclusions PTEN can inhibit AKT phosphorylation induced by insulin or rhEGF in gastric cancer cells. Missense mutations in the 124th or 129th amino acid of PTEN phospho-tase domain do not exert inhibitive function.
2.Circulating MicroRNAs and Inflammatory Bowel Disease

MicroRNAs(miRNAs)are a group of non-coding RNA molecules having modulating function,and as a post transcriptional modulating factor is involved in the modulation of expression of eukaryotic genes. Inflammatory bowel disease(IBD)is a chronic non-specific intestinal inflammatory disease and its etiology has not yet been fully clarified. Recent studies have shown that circulating miRNAs were specifically expressed in patients with IBD. This article reviewed the advances in studies on circulating miRNAs and IBD.
3.Sensitivity of Plasmodium vivax to Chloroquine in Laza City,Myanmar

Fourty-eight patients infected by Plasmodium vivax were randomly divided into chloroquine regimen groups A and B.Each case in group A received a total dosage of 1 200 mg(base) over a 3-day period(600 mg on the first day then 300 mg daily).Cases in group B each received total dosage of 1 500 mg(base) in 3 days(750 mg on the first day then 375 mg daily).Body temperature and blood smears were examined on days 0, 1, 2, 3, 7, 14, 21 and 28, respectively.All patients were negative for asexual parasites within 3 days.The cure rate on day 28 was 100%.Results showed that P.vivax is sensitive to chloroquine in Laza, Myanmar, and chloroquine can still be used for the imported vivax malaria cases at the China-Myanmar border.
4.Study on the proficiency testing program of pharmaceutical preparation analysis.

This proficiency testing program is established to evaluate the pharmaceutical preparation analysis capacity of laboratories recommended by 18 countries and economies. It was authorized by Asia Pacific Laboratory Accreditation Cooperation (APLAC), and organized by Shanghai Institute for Food and Drug Control (SIFDC) and China National Accreditation Service for Conformity Assessment (CNAS). The 0.3sigma test is used to evaluate the homogeneity and stability of the proficiency testing sample. The results of the laboratories were assessed by Z-score. The robust average and the robust standard deviation of the participants' results were calculated as assigned value and standard deviation for performance assessment of hydrochlorothiazide and captopril using robust statistics. Thirty-three of 38 laboratories recommended by 18 countries and economies sent their results back. Twenty-four laboratories' results were observed as satisfactory. Five laboratories were identified as having reported at least one questionable result. Four laboratories were identified as having reported at least one unsatisfactory result.
5.Cost-effectiveness Analysis of 4 Kinds of Protease Inhibitor Preventing Hyperamylasemia and Pancreatitis after ERCP of Choledocholithiasis Patients

OBJECTIVE:To evaluate the economics of somatostatin,ulinastatin,octreotide and gabexate preventing hyperamy-lasemia and pancreatitis after endoscopic retrograde cholangiopancreatography (ERCP). METHODS:Medical records of 316 cho-ledocholithiasis patients underwent ERCP were selected from our hospital during Jul. 2008-Apr. 2016,and then divided into blank control group(58 cases),somatostatin group(64 cases),ulinastatin group(65 cases),octreotide group(68 cases)and gabexate group (61 cases) according to the use of protease inhibitor. Before ERCP,blank control group received routine treatment as fast-ing,parenteral nutrition support,acid suppression,etc. Based on it,other 4 drug groups received prophylactic drug use according to package inserts 6 h before ERCP. The incidence of hyperamylasemia and pancreatitis after ERCP,VAS scores 3,24,48 h after surgery and the occurrence of ADR were compared among 5 groups. The cost-effectiveness analysis was used to evaluate the eco-nomics of therapy plans in each group. RESULTS:Compared with blank control group,hospitalization time of somatostatin group, ulinastatin group,octreotide group and gabexate group were shortened significantly;the incidence of hyperamylasemia and pancre-atitis were significantly decreased;VAS score 3,24,48 h after surgery were significantly decreased,with statistical significance (P<0.05). Compared among 4 groups,above indexes had no significant difference,and the incidence of ADR was in low level and had no statistical significance(P>0.05). The cost-effectiveness ratio of somatostatin group was the lowest and has cost-effec-tiveness advantage. The results were supported by incremental cost-effectiveness and sensitivity analysis. CONCLUSIONS:Soma-tostatin,ulinastatin,octreotide and gabexate can significantly prevent the occurrence of hyperamylasemia and pancreatitis after ER-CP,and relieve pain with good safety. Somatostatin can achieve the best therapeutic efficacy at the lowest cost,so it is the best plan for hyperamylasemia and pancreatitis after ERCP.
6.MiR-630 inhibits MDA-MB-231 cells migration and invasion by targeting Sox4 in triple-negative breast cancer

Objective To verify whether miR-630 could inhibit MDA-MB-231 cells migration and invasion by targeting Sox4 in triple-negative breast cancer(TNBC).Methods Collection normal breast tissue and breast cancer tissue from patients undergoing breast cancer resection.RT-PCR were used to test the expression of miR-630,miR-21,miR-195,miR-134,miR-200a,miR-381 and miR-1228.Western blot were used to test the expression of COL1A1,COL1 A5,MMP-2,MMP-9 and Sox4.In vitro experiment,after miR-630 was transfected into MDA-MB-231 cells,wound healing were employed to test the migratory ability of MDA-MB-231 cells,and transwell were used to test the invasion ability of MDA-MB-231 cells.Western blot were used to investigate the expressions of COL1 Al,COL1 A5,MMP-2,MMP-9 and Sox4 in MDA-MB-231 cell.Luciferase assay was used to confirmed whether Sox43'-UTR the target gene of miR-630.Results Compared with normal breast tissue,the expression of miR-630 was decreased(P<0.01),meanwhile the expression of COL1A1,COL1A5,MMP-2,MMP-9 and Sox4 were significantly increased in the triple-negative breast cancer tissue(P<0.01).In the vitro experiment,compared with the control group,the expression of COL1A1,COL1A5,MMP-2,MMP-9 and Sox4 were decreased in the miR-630 group (P<0.05);The migration activity of MDA-MB-231 cells was decreased in the miR-630 group (P<0.01);The Luciferase activity of the Sox4-3'-UTR plasmid was significantly suppressed by miR630 (P<0.05);Over expression of Sox4 could reverse the effect of miR-630 on MDA-MB-231(P<0.05,P<0.01).Conclusion In triple-negative breast cancer tissue,the expression of miR-630 decreased;miR-630 inhibits triple-negative breast cancer cells migration and invasion by targeting Sox4-3’-UTR.
7.Role of miR-188-5p in oxygen-glucose deprivation and restoration injury to mouse N2a cells: relationship with SENP3

Objective:To evaluate the role of miR-188-5p in oxygen-glucose deprivation and restoration (OGD/R) injury to mouse neuroblastoma (N2a) cells and its relationship with small ubiquitin-like modifier-specific proteases 3 (SENP3).Methods:N2a cells were cultured and divided into 5 groups ( n=23 each) using a random number table method: control group (group C), OGD/R group, group NC, transfection of mir-188-5p agonist group (group M) and transfection of mir-188-5p inhibitor group group (group I). Cells in group C were cultured routinely.Cells in group NC, group M and group I were transfected with mir-188-5p negative control miRNA, agonist and inhibitor, respectively.N2a cells were subjected to OGD for 3 h followed by restoration of oxygen-glucose supply to establish the model of OGD/R injury.At 24 h of oxygen-glucose restoration, the cell viability was recorded by the cell counting kit-8 assay, the amount of lactic dehydrogenase (LDH) released was detected, the expression of miR-188-5p and SENP3 mRNA was detected by quantitative real-time polymerase chain reaction, and SENP3 expression was determined by Western blot.The targeting relationship between miR-188-5p and SENP3 mRNA was detected using dual luciferase reporter assay. Results:Compared with group C, the cell viability was significantly decreased, amount of LDH released was increased, and expression of SENP3 and its mRNA was up-regulated in the other 4 groups, miR-188-5p expression was down-regulated in OGD/R and I groups, and miR-188-5p expression was up-regulated in group M ( P<0.05 or 0.01). Compared with group OGD/R, the cell viability was significantly decreased, amount of LDH released was increased, and expression of SENP3 and its mRNA was up-regulated, and miR-188-5p expression was down-regulated in group I, and the cell viability was increased, amount of LDH released was decreased, expression of SENP3 and its mRNA was down-regulated, and miR-188-5p expression was up-regulated in group M ( P<0.05 or 0.01). The dual luciferase reporter assay showed that miR-188-5p could act directly on SENP3. Conclusion:miR-188-5p is involved in OGD/R injury, which is associated with targeted down-regulation of SENP3 expression in N2a cells.
8.Neuronal differentiation of adipose tissue-derived stromal cells

BACKGROUND: Appropriate seed cell is important for transplantation in the treatment of cerebrovascular disease and other central nervous system disease.OBJECTIVE: To investigate the capacity of human adipose tissue-derived stromal cells (ADSCs) to differentiate into neurons.METHODS: The fatty tissue was harvested from removed abdominal unnecessary fat of healthy adult with no communicable disease or endocrine disease. Human ADSCs were isolated from human liposuction tissues and cultured in neural induction medium with GM1. Invert phase-contrast microscopy was used to observe morphology changes of ADSCs. The expression of nestin, neuron specific enolase (NSE) and microtubule-associated protein 2 (MAP2) were identified by immunocytochemistry.RESULTS AND CONCLUSION: The majority of cells displayed typical appearance of neuronal-like cells following induction. Following 1 hours of induction, some cells began to express nestin, and NSE and MAP2-positive cells were observed at 5 hours. ADSCs can differentiate into neurons, and the differentiated neurons have the capacity of expressing nestin, NSE and MAP2.
9.Clinical effects of laparoscopic therapy and duodenoscopic therapy in the treatment of biliary severe acute pancreatitis

Objective To discuss the clinical effects of laparoscopic therapy and duodenoscopic therapy in the treatment of biliary severe acute pancreatitis.Methods A total of 140 patients with biliary severe acute pancreatitis(BSAP) in our hospital was assigned into laparoscopic therapy group(60 cases) and duodenoscopic therapy group(80 cases) according to the treatment plan,and indicators in both groups were compared,including acute physiology and chronic health evaluation(APACHE Ⅱ score),WBC,CRP,the operation time,intraoperative blood loss,hospitalization time,hospitalization fee,complication rate,cure rate,mortality rate,abdominal pain disappearing time,body temperature recovered to normal time,blood amylase recovered to normal time,amino acid transaminase(ALT) recovered to normal time,total bilirubin(TBIL) recovered to normal time,alkaline phosphatase(ALP) recovered to normal time.Results APACHE Ⅱ scores and the levels of WBC,TBIL,ALT and CRP in both groups were significantly decreased after surgery(P<0.05),but there were no statistical differences between the two groups at the same time point after treatment(P>0.05).The operative time and the intraoperative blood loss in the duodenoscopic therapy group were significantly less than those in laparoscopic therapy group(P<0.05).There were no statistical differences between the two groups of hospitalization time,complication rate,cure rate,mortality rate,abdominal pain disappearing time,temperature returned to normal time,blood amylase recovered to normal time,ALT recovered to normal time,TBIL recovered to normal time,ALP recovered to normal time(P>0.05).Conclusion In early stage,both laparoscopic therapy and duodenoscopic therapy can treat BSAP effectively,and their therapeutic effects are nearly similar.
10.Open reduction and internal fixation via two lateral approaches in treating Sanders type III intra-articular calcaneal fractures: a comparative analysis

Objective:To compare the post-operative outcomes between extended lateral approach and L-shaped lateral approach in surgical treatment of Sanders type III intra-articular calcaneal fractures.Methods:A retrospective case-control study was made on clinical data of 34 patients with fresh Sanders type III intra-articular calcaneal fractures hospitalized in Langfang Hospital of Traditional Chinese Medicine from October 2014 to October 2017. There were 33 males and 1 female, with the age of 18-56 years [(42.5±9.4)years]. All fractures were fresh. Seventeen patients were fixed by the extended lateral approach (extended approach group) and seventeen patients by L-shaped lateral approach (L-shaped approach group). Operation duration, intraoperative bleeding volume, length of hospital stay, one-stage wound healing rate and complication rate were observed. B?hler and Gissane angle were measured before operation, one week after operation and 12 months after operation. American Orthopedic Foot and Ankle Society (AOFAS) ankle-hindfoot scale was applied to evaluate function recovery.Results:There were no significant differences between groups in operative time and intraoperative bleeding volume ( P>0.05). There were significant differences between extended approach group and L-shaped approach group in length of hospital stay [(15.7±4.0)days vs.(22.4±9.6)days], one-stage wound healing rate [88%(15/17) vs. 47%(8/12)] and postoperative complication rate [12%(2/17) vs. 47%(8/17)] ( P<0.05). The B?hler and Gissane angles did not differ significantly between groups before operation and one week and 12 months after operation ( P>0.05). At postoperative 12 months, the B?hler and Gissane angles were improved from preoperative (8.2±6.0)°, (85.3±10.5)° to (23.9±6.1)° and (119.3±6.2)° respectively in extended approach group, and those from preoperative (9.4±3.9)°, (85.5±7.1)° to (25.8±3.7)° and (122.2±5.6)° respectively in L-shaped approach group (all P>0.05). At 12 months after operation, the AOFAS score was (86.9±7.5)points in extended approach group, with the excellent and good rate of 88%; and was (83.3±12.5)points in L-shaped approach group, with the excellent and good rate of 76% ( P>0.05). While the score of hindfoot joint activity in extended approach group was (5.8±0.7)points, significantly higher than (3.4±1.0)points in L-shaped approach group ( P<0.05). Conclusion:For Sanders III intraarticular calcaneal fractures, compared with L-shaped approach group, plate fixation via extended approach can promote fracture healing, shorten hospital stay, reduce incidence of complications and facilitate function recovery of subtalar joint.
