Objective To explore the mechanism of hypoxic-ischemic brain damage(HIBD) and provide new ideas for clinical treatment of hypoxic-iscbemic encephalopathy.Methods Neonatal 7-day-old Wistar rats were randomly divided into sham-operation control group,HIBD 6 h,12 h,24 h,48 h and 72 h groups(n =6 per group).The model of HIBD was induced by unilateral carotid ligation followed by timed exposure to 8% oxygen.The expression of MMP-9 mRNA and TIMP-1 mRNA in brain tissue of neonatal rats was measured by Real-time Q-PCR method.Results (1) The ligatod brainhemisphere of HIBD groups showed obvious edema from 12 h to 48 h after hypoxia -ischemia in the neonatal rats.(2) The expression of MMP-9 mRNA was very low in the sham-operation control group,but in HIBD groups,it began to increase at 6 h,and reached a peak at 24 h,then gradually decreased,but still maintained at high level at 72 h(P＜0.01).(3) The expression of TIMP-1 mRNA was aslo very low in the sham-operation control group.But in HIBD group,it increased slightly at 6 h,12 h and 24 h,compared to the sham-operation control group,each group was statistically significant(P＜0.05),with no significant difference among the three groups(P＞0.05),then decreased at 48 h and 72 h,but with no significant difference from sham-operation control group (P＞ 0.05).(4) The ratio of MMP-9 mRNA/TIMP-1 mRNA was normal in the sham-operation control group and HIBD 6 h group,it began to increase at 12 h,and reached a peak at 48 h,then gradually decreased,but still maintained at high level at 72 h(P ＜0.01).Conclusion Hypoxia-ischemia increases the expression of MMP-9 mRNA in brain tissue of nenatal rats,and the imbalance in the expression of MMP-9 mRNA and TIMP-1 mRNA possibly is one cause of brain edema induced by HIBD.

Objective To study the effects of the BanxiaXiexin decoction (BD) on express of glycogen synthesis and the expression of MC4R.and to discuss the mechanism of its maintaining glucose metabolism balance.Methods Diabetic mice were induced and divided into the model group,metformin group(0.3 g/kg)and BD high dose group(1.7 g/kg),medium dose group(0.85 g/kg),and low dose group (0.425 g/kg).In addition,a normal group was set to feed normal diet.Fasting blood glucose,body weight on an empty stomach,express of MC4R and hepatic glycogen were assayed after a week of treatment.Results Body weight of BD high,medium and low dose group was (296.62 ±1.14) g,(293.83 ± 2.1 7 ) g,and (286.75 ± 1.91 ) g respectively,showing a significant increase compared with the model group (284.73± 2.91) g (P＜0.01).MC4R expression of these MD groups were (29.69 ± 1.64) ng/L,(29.38±1.88) ng/L and (30.04 ±1.24) ng/L,showing a significant increase compared with the model group (25.95±2.24) ng/L (P＜0.05).Liver glycogen content of these MD groups were (15.94±0.82)mg/g,(11.52±0.56)mg/g,and (10.48±0.45)mg/g respectively,also showing a significant increase than the model group (9.05±0.44)mg/g(P＜0.01,P＜0.01,P＜0.05).Conclusion BD improves MC4R expression and rat feeding behavior by increasing by peripheral organization utilization of glucose.

Objective To compare the effects of different composite resins as core material on the degree of microleakage in post-core repairation.Methods A total of 46 recently extracted single-rooted mandibular premolars were distributed into different groups according to different core material including ParaCore,AP-X and Ceramage.Then we used direct or indirect forming method to make post-core restorations.All of the specimens were submerged in dyes.Then,they were demineralized,dehydrated and processed to be transparent.The extent of the dye leakage was examined under a stereomicroscope.Results The microleakage value was significantly higher in Group of direct-mold-cement with ParaCore (4.94± 1.71)mm than in Group of indirect-mold-secondary-cement with ParaCore (0.91 ± 0.33) mm,Group of indirect-mold-secondary-cement with AP-X (0.87 ± 0.27) mm,and Group of indirect-mold-secondary-cement with Ceramage (1.02 ± 0.34)mm.Conclusion Different methods of building and cementing FRC post-core restorations,but not different composite resins as core material,have significant effects on the extent of microleakage in post-core repair.

Objective: To observe the effect of tolvaptan on treating the patients with intractable heart failure (HF) combining hyponatremia for their clinical conditions and improvements. Methods: A total of 100 relevant patients treated in our hospital from 2014-01 to 2015-01 were studied. Based on conventional treatment as cardiac stimulant, dieresis and correction of electrolyte disturbance, the patients were randomly divided into 2 groups: Tolvaptan group, the patients received tolvaptan15 mg/d for 5 days and Control group, the patients had no tolvaptan administration.n=50 in each group. The improvement of clinical symptoms, changes of cardiac, renal functions and blood levels of electrolyte were compared between 2 groups after treatment. Results:① Compared with Control group, Tolvaptan group had significantly improved dyspnea, reduced lung moist rale and low limb edemaP<0.01; Tolvaptan group showed increased 24 h urine volume (2416.0±771.6 vs 1124.6±215.7) ml,P<0.01, decreased plasma levels of NT-proBNP (2678.04 ± 537.09) pg/ml vs (4051.34 ± 306.07) pg/ml,P<0.01 and increased blood levels of sodium (139.08 ± 6.18) mmol/L vs (129.44 ± 2.20) mmol/L,P<0.01. ②In Tolvaptan group, the above improvements were found in 38 patients, and with 1 day tolvaptan therapy, their blood nitrogen dropped from (10.39 ± 1.23) mmol/L to (7.28 ± 1.53) mmol/L,P<0.01; the other 12 patients had no such effect, and with 1 day tolvaptan therapy, their blood nitrogen elevated from (10.39±1.23) mmol/L to (13.38 ± 0.66) mmol/L,P<0.01. Conclusion: Tolvaptan could effectively improve clinical symptom and cardiac function in patients with intractable HF, it is helpful to increase blood sodium level; the efifcacy of tolvaptan might be evaluated by the changes of blood nitrogen at the early stage of treatment.

BACKGROUND: Neonatal hypoxic ischemic encephalopahty(NHIE) is a common neonatal disease, which could cause cerebral palsy, inferior intelligence and convulsion. At present, usual examinations for this disease are CT.MRI and single photon emission computed tomography(SPECT), etc.OBJECTIVE: To investigate the character of SPECT image of NHIE, and compared with CT and MRI as well to provide a theoretical gist for function surveillance and early intervention of NHIE.DESIGN: An observatory comparative study based on NHIE patients and normal neouates as control.SETTING: Department of nuclear medicine in a medical university hospital.PARTICIPANTS: Thirty neonates hospitalized in the Department of Pediatrics, Affiliated Hospital of Jining Medical College between September 1998 and October 2000 were selected as observation group with 23 males and 7 females. Another 10 normal neonates including 7 males and 3 females were randomly selected from same period into control group.METHODS: SPECT brain blood perfusion was conducted in the subjects of two groups and the results were compared with that of CT and MRI.MAIN OUTCOME MEASURES: ① comparison of SPECT brain blood image features between NHIE and normal neonates; ② comparison of SPECT images and CT, MRI results. RESULTS: In 22 NHIE cases who received SPECT, CT and MRI simultaneously, mild group had 7 cases, and positive cases of SPECT, CT or MRI was 7, 3 or 5; moderate group had 10 cases, and positive cases of three methods was 10, 8 or 9; severe group had 5 cases, the all of them were positive in all three examinations. The total positive detectable rate of SPECT, CT or MRI was 100, 72.73% or 86. 36%. There was no significant difference of sensitivity between SPECT and MRI in mild group, while there was significant difference between SPECT and CT(x2 = 3.95, P ＜ 0.05);there was no significant difference among three methods in either moderate group or severe group(x2 =0. 83, P ＞ 0.05).CONCLUSION: SPECT brain blood perfusion image is a quite sensitive method for the surveillance of NHIE, which can provide imageological reference for the evaluation of its prognosis and early intervention.

METHODS: Thirty Sprague-Dawley male rats were selected and performed five-sixths nephrectomy to produce chronic renal failure model. Two weeks after the surgery, the rats were randomly divided into model group, valsartan group and benazepril group, and established a shame group serving as normal control. Objective:To prepare and evaluate Shenhuang vaginal thermosensitive in situ gels. Methods:Shenhuang vaginal ther-mosensitive in situ gels( SHV-TISG)were prepared with poloxamer 407 and poloxamer 188 as the materials. The viscosity change at different temperatures and the gelation temperature of SHV-TISG and SHV-TISG diluted by stimulant vaginal fluid were measured,the in vitro drug release of the preparation was evaluated by a membraneless diffusion method,and the safety of SHV-TISG was detected by vaginal irritation test. Results:The preparation showed the vaginal injectability and gelationability in vivo without vaginal irritation. Gel erosion was the principal mechanism of the drug release from the gels. Conclusion:The preparation of SHV-TISG is feasible in tech-nique,and the properties meet the requirements of vaginal topical use.

To observe the clinical effect of compound Yimucao oral liquid on postpartum lochiorrhea. Methods:A to-tal of 200 patients with postpartum lochiorrhea were randomly divided into the observation group ( 100 cases ) and the control group (100 cases). The patients in the observation group received 20ml compound Yimucao oral liquid three times a day for seven days, while those in the control group were given 15g Yimucao granules three times a day for seven days. After the treatment, the duration, amount, color and quality of lochia, lower abdominal pain, uterine involution and the status of postpartum infection in the two groups were compared. Results: The total effective rate in the observation (94. 0%) was higher than that in the control group(84. 0%). Compared with that in the control group, less average hemostasis time, better situation of abdominal pain, uterine involution and lower postpartum infection rate in the observation group were observed after the treatment(P<0. 05). Conclusion:Compound Yimucao oral liquid can promote the uterine involution. It’ s effective and safe in the treatment of postpartum lochirrhea.

Objective To observe the effect of Antivirus Compound Capsule in protecting liver and reducing enzyme level in chronic type B hepatitis and acute hepatic injury caused by D-GlaN.Methods Mice model was established by intraperitoneal injecting of D-GlaN 800mg/Kg.The level of ALT and AST was obviously increased after 48hours.Pathological test proved to be acute liver cell damage.Clinically 117 cases of chronic hepatitis B was recruited into a control group(57 cases)and a treatment group(60 cases).The treatment group was treated with Antivims Compound Capsule and Silymarin tablet,and the control group was treated with Silymarin tablet.Besides,both groups were infused with diammonium Compound Capsule Can obviously reduce level of AST(P＜0.001、＜0.01、＜0.01、＜0.001);Three dosage levels of Antivirus Compound Capsule can obviously reduce the degree hepatic pathological changes caused by D-GlaN induced(P＜0.01、＜for the treatment group andtlle control group) between the two groups with P＞0.02.while there was obvious difference of negative conversion rate of AST.[92.98%(53/57)and 78.9%(45/57)for the treaunent group and the consul group]between the two groups with P＜0.01.Conclusion Antivirus Compound Capsule is effective in protecting liver and reducing enzyme level of patients with chronic hepatitis B and acute hepatic injury caused bv D-GlaN.

Objective:To study the effect of airway IL-4RA gene transfer on asthma-associated expression of STAT6 in the mice of allergic asthma.Methods:Forty BALB/c mice were randomly divided into four groups:blank control,asthma model,asthma model administered by pLNC-laz,and asthma model administered by glucocorticoid and pLNC-IL-4RA.The ten mice in each groups were sensitized by i.p.injection of 10 ?g of OVA emulsified in 40 mg of aluminum hydroxide in a total volume of 200 ?l on the days of 1st,7th and 14th.On the 15th day,the mice were challenged via airways with OVA for 7 days,20 min a day,blank control were sensitized by i.p.injection of 200 ?l pH=7.4 PBS on days 7 and 14.Asthma model was administered by pLNC-laz or pLNC-IL-4RA for 3 days beginning at the 12th day.The mice of the group with glucocorticoid was administered with glucocorticoid on the 15th day for 7 days.24 hours after the last challenge,the mice were sacrificed by bloodletting.Serum OVA-specific IgE and blood Eosinophils were measured.The lungs were fixed by immersion in 10% formalin.Expression of STAT6 was identified by immunohistochemistry.Results:It was shown that pLNC-IL-4RA was integrated to the genomic DNA of pneumonic tissues.Retrovirus-mediated delivery of IL-4RA to airways of the asthma mice weakened airway eosinophilia triggered by either IL-13 or IL-4.Furthermore,IL-4RA delivered by retrovirus was expressed in airways of the mice with allergen sensitization,resulting in significiant reduction of expressing level of asthma-associated STAT6 in the experimental mice of allergic asthma.Conclusion:Retrovirus mediated delivery of IL-4RA to airways reduces expression of STAT6 in pneumonic tissues of asthmatic mice.Thus,the gene theraphy can be a potential therapeutic option to treat and control chronic airway inflammation and asthmatic symptoms.
