2.Evaluating the physical activity status of 259 advanced aged people

Objective To evaluate the physical activity status of the advanced aged adults and determine the influence of long-term physical activities on the physical function and health of the advanced aged adults. Methods A survey was carried out from March, 2004 to September, 2004, among those advanced aged intellectuals or cadres taken care chronically by the Patient Clinic for Senior Cadre in PUMC Hospital. Physical activity was assessed using the modified leisure-time physical activity questionnaire. Results There were 259 participants, aged from 80 to 94 (83.92?3.41)years. 85.3%(221/259 cases)took physical activities almost every day for more than eleven years, and 21.7% of them (48/221 case)even stuck to it from their youth. Among these 221 participants, 34.4% took≥ 60 min per day of physical activities,and 51.0% ≥30 min per day. The group that took ≥ 60 min per day held a lower proportion than the group that took ≥ 30 min per day to suffer from agrypnia, debilitation, somasthenia and anorexy (by 20.3%, 24.0%, 21.7% and 13.1%,respectively(P
3.Application value of cardiac activity index on evaluation right ventricular function after neonatal asphyxia

Objective To investigate the roles of cardiac activity index(Tei index) on evaluation the right ventricular function after neonatal asphyxia.Methods Sixty neonatal asphyxia who included 35 cases of mild asphyxia(mild asphyxia group) and 25 cases of severe asphyxia(severe asphyxia group) and 30 cases of normal full-term newborns(control group) were selected.Echocardiographic examinations were performed on 24-48 h after birth,which included pulmonary artery systolic pressure (PASP),right ventricular ejection fraction(RVEF),tricuspid early diastolic peak(peak E) and late diastolic peak(peak A),and E/A ratio was acquired.The right ventricular cardiac activity index (RV-Tei index) was measured by Doppler spectrum.Results There was no significant difference in RVEF,E/A ratio among mild asphyxia group,severe asphyxia group and control group (P > 0.05).RV-Tei index in mild asphyxia group and severe asphyxia group was increased compared with that in control group (0.489 ± 0.090,0.625 ± 0.100 vs.0.345 ± 0.120),and there was significant difference (P< 0.05 or <0.01).There was significant difference in RV-Tei index between mild asphyxia group and severe asphyxia group (P < 0.05).RV-Tei index in neonatal asphyxia was positively correlated with PASP (r =0.950,P < 0.05),and there was no relationship between RV-Tei index and gestational age,weight,heart rate (r =-0.068,-0.280,-0.360,P >0.05).Conclusions Neonatal asphyxia can lead to disorders of the right ventricular function.Tei index can evaluate early overall changes of the right ventricular function and is better than conventional ultrasound technology in neonatal asphyxia.
4.Preparation and Quality Evaluation of Shenhuang Vaginal Thermosensitive in situ Gels

Objective:To prepare and evaluate Shenhuang vaginal thermosensitive in situ gels. Methods:Shenhuang vaginal ther-mosensitive in situ gels( SHV-TISG)were prepared with poloxamer 407 and poloxamer 188 as the materials. The viscosity change at different temperatures and the gelation temperature of SHV-TISG and SHV-TISG diluted by stimulant vaginal fluid were measured,the in vitro drug release of the preparation was evaluated by a membraneless diffusion method,and the safety of SHV-TISG was detected by vaginal irritation test. Results:The preparation showed the vaginal injectability and gelationability in vivo without vaginal irritation. Gel erosion was the principal mechanism of the drug release from the gels. Conclusion:The preparation of SHV-TISG is feasible in tech-nique,and the properties meet the requirements of vaginal topical use.
5.Observation of Clinical Curative Effects of Compound Yimucao Oral Liquid on Postpartum Lochiorrhea

To observe the clinical effect of compound Yimucao oral liquid on postpartum lochiorrhea. Methods:A to-tal of 200 patients with postpartum lochiorrhea were randomly divided into the observation group ( 100 cases ) and the control group (100 cases). The patients in the observation group received 20ml compound Yimucao oral liquid three times a day for seven days, while those in the control group were given 15g Yimucao granules three times a day for seven days. After the treatment, the duration, amount, color and quality of lochia, lower abdominal pain, uterine involution and the status of postpartum infection in the two groups were compared. Results: The total effective rate in the observation (94. 0%) was higher than that in the control group(84. 0%). Compared with that in the control group, less average hemostasis time, better situation of abdominal pain, uterine involution and lower postpartum infection rate in the observation group were observed after the treatment(P<0. 05). Conclusion:Compound Yimucao oral liquid can promote the uterine involution. It’ s effective and safe in the treatment of postpartum lochirrhea.
6.The Clone, Expression and Site-directed Mutagenesis of 3-Ketosteroid-Delta (1)-Dehydrogenase from Arthrobacter Simplex

Objective:In this study,a prokaryotic expression of the 3-ketosteroid-Delta (1)-dehydrogenase (KSDD) which came from Arthrobacter simplex was built.Moreover,in order to investigate the catalytic mechanism of KSDD and improve its stability,the structure of KSDD was predicted by computer and the critical sites were confirmed by site-directed mutations.Methods:The recombinant plasmid was constructed by eukaryotic expression vector pET-22b and the recombinant strain was constructed and expressed in Escherichia coli BL21 (DE3).High-performance liquid chromatography was used to determine the transformation rate of 4-AD to ADD.The KSDD structure and key sites were predicted by SWISS-MODEL.Site-directed mutations for the amino acid residues of key sites were constructed and activities of the mutations were detected.Results:The recombinant strain E.coli pET-22-ksdd was successfully constructed.It was induced to express the dehydrogenase by IPTG and the conversion rate of 4-AD to ADD was 27% at 21 ℃.The structure of 3-ketosteroid-Delta (1)-dehydrogenase and the four key sites was analyzed by SWISS-MODEL.Four mutants,Y120R,Y320L,Y488F and G492Y were constructed.Mutants Y120R and Y488F were inactivated,so they were proved to be the key active sites of KSDD.The conversion rate of mutant Y320L was consistent with that of wild type,but the stability at 37 ℃ was improved.The conversion rate of mutant G492Y was 1.2 times of the wild type and the stability has been improved at 37 ℃.Conclusions:At present,there are few studies about the structure and catalytic mechanism of dehydrogenase.The active sites of the enzyme were verified by this study,which laid the foundation for the further study of the properties of the enzyme KSDD.
7.Detection and clinical significance of Th17/Treg in peripheral venous blood of children with Henoch-Schonlein purpura combined with mycoplasma pneumoniae infection

Objective:To discuss the correlation between the immune instability of Th17/Treg and the pathogenesis of the Henoch-Schonlein purpura(HSP)with Mpcoplasma pneumoniae(MP)infection.Methods:From June 2018 to December 2019, children who had been diagnosed as HSP in the Department of Pediatrics at Qinhuangdao First Hospital and Qinhuangdao Maternal and Child Health Care Hospital, as well as in-patients with MP infection and outpatients, who had health check-up were collected.The subjects were divided into HSP group(41 cases), MP group(25 cases), HSP-MP group(19 cases) and control group(20 cases) according to the diagnosis of HSP and MP infection.The percentage of Th17 and Treg cells in peripheral venous blood of four groups were determined by flow cytometry, and the levels of interleukin(IL)-17, IL-6 and transforming growth factor-β(TGF-β)in peripheral venous serum of four groups were detected by ELISA.Results:There were no statistically significant differences regarding age and gender distribution among four groups( P>0.05). The percentages of Th17 cells, IL-17 and IL-6 levels in peripheral blood of the HSP-MP group was significantly higher than those in the other three groups( P<0.05), which in HSP group and MP group were higher than that in control group( P<0.05). There was no statistical difference between HSP group and MP group( P>0.05). The percentage of Treg cells in peripheral blood was significantly lower in HSP-MP group than those in the other three groups( P<0.05), which in the HSP group and MP group were lower than those in control group( P<0.05). There was no statistical difference between HSP group and MP group( P>0.05). There were no statistically significant differences of TGF-β among the four groups( P>0.05). Conclusion:High levels of IL-17 and IL-6 may be an important factor causing severe Th17/Treg immune disorders in children with HSP combined with MP infection, which may play an important role in the pathogenesis of HSP combined with MP infection.
8.Effect of compound Minwei syrup on serum IL-4 and IFN-γ in patients with chronic urticaria

Objective To study the effect of compound Minwei syrup on serum interleukin-4 (IL-4) and interferon-γ(IFN-γ) in patients with chronic urticaria.Methods 189 patients with chronic urticaria were enrolled as the observation group,60 cases of non-allergic diseases were selected as the control group.The observation group was treated by compound Minwei syrup for average 7 days.Serum levels of IL-4 and IFN-γbefore and after treatment were detected.Results After treatment,in the observation group 123 cases recovered,34 cases had marked effect,effective in 23 cases,9 cases without effect,the total effective rate was 95.2%.Serum IL-4 in the observation group before treament was significantly higher than the control group(t =3.729,P < 0.01),IFN-γwas significantly lower than the control group(t =3.064,P < 0.01).After treatment,serum IL-4 in the observation group was significantly lower than before treatment (t =2.778,P < 0.05),1FN-γwas significantly higher than the control group (t =2.467,P < 0.05).Mild drowsiness and sleepiness appeared in 2 patients in the observation group,symptoms disappeared after drug withdrawal.Conclusion Minwei syrup can significantly improve IL-4 and IFN-γ balance in patients with chronic urticaria.

In this study, 38 experimental subjects of diabetes mellitus were given pectin 25g extra per day orally under carefully controlled dietary plan for six weeks. Another 38 cases were selected at random as control group.Pre- and posttreatrnent, blood and urine sugar and serum insulin of each subject were measured and compared. OBJECTIVE:To observe the efficacy and safety of Compound yimucao oral liquid combined with oxytocin in the prevention of postpartum hemorrhage and promotion of uterine involution for puerpera of vaginal delivery. METHODS:240 preg-nants with natural childbirth of vaginal delivery were randomly divided into control group and observation group. Control group was injected oxytocin 20 U in buttock after delivery. Observation group was additionally treated with Compound yimucao oral liquid 20 ml,tid for consecutive 2 weeks. Postpartum vaginal bleeding,uterine involution and incidence of adverse reactions in 2 groups were observed. RESULTS:Compared with control group,postpartum hemorrhage volume after delivery 2-24 h,incidence of post-partum hemorrhage,uterus size,uterine hemorrhage and lochia duration in postpartum 42 d in observation group were significantly better than control group,the differences were statistically significant(P<0.05);there were no significant differences in the hemor-rhage postpartum volume after delivery 1 h and 1-2 h and fundal height in postpartum 42 d in 2 groups(P>0.05). There were no fever,rash,nausea and other obvious adverse reactions in 2 groups. CONCLUSIONS:Compound yimucao oral liquid combined with oxytocin can obviously decrease the postpartum bleeding and promote uterine involution,with good safety.
