1.The expression of Pim-1 in non-small cell lung cancer and its relationship with c-Myc

Objective To investigate the expression of Pim‐1 in non small cell lung cancer and adjacent normal tissues ,and study the relationship between c‐Myc and Pim‐1 in the corresponding tissue gene expression .Methods Totally 30 cases of non small cell lung cancer tissue and adjacent normal lung tissues were collected by surgical operation in department of thoracic surgery . Clinical data were statisticed and tracking late pathologic results ,using RT‐PCR ,qRT‐PCR and immunohistochemical method to de‐tect Pim‐1 mRNA ,c‐Myc and Pim‐1 protein expression in lung cancer and adjacent normal tissue ,and to analyze the relationship be‐tween the expression of Pim‐1 and c‐Myc .Results The positive rate of Pim‐1 mRNA and protein expression in non small cell lung cancer was obviously higher than that in adjacent normal tissue ,the mRNA expression levels were 0 .798 ± 0 .083 and 0 .394 ± 0 .107 (P<0 .01) ,the protein positive cases were 18 cases and 6 cases (P=0 .002) .The expression of Pim‐1 protein had no relationship on age ,gender ,smoking history ,pathological types and degree of differentiation of patients with non‐small cell lung cancer (P>0 .05) ,bue had related to ymph node metastasis and TNM stages of tumor (P<0 .05) ,with lymph node metastasis and TNM sta‐ges increases ,its expression quantity also rise .There was a positive correlation between Pim‐1 and c‐Myc protein expression ,corre‐lation coefficient (r) was 0 .433 (P=0 .017) .Conclusion High expression of Pim‐1 in non small cell lung cancer gene and is also increased with lymph node metastasis and TNM stages ,Pim‐1 and c‐Myc expression has positive correlation ,this could provide clues to the early diagnosis and prognosis evaluation of non small cell lung cancer ,and also provides a new train of thought and to find a new target for gene therapy of lung cancer .
2.Effectiveness and Safety of Yinzhihuang Oral Liquid in the Adjuvant Treatment of Neonatal Jaundice:A Sys-tematic Review

OBJECTIVE:To systematically review the effectiveness and safety of Yinzhihuang oral liquid in the adjuvant treat-ment of neonatal jaundice,and provide evidence-based reference for clinical treatment. METHODS:Retrieved from CJFD,VIP Da-tabase,Wanfang Database,Medline and EMBase,randomized controlled trials(RCT)about the effectiveness of Yinzhihuang oral liquid(test group)based on the canventional treatment(control group)in the adjuvant treatment of neonatal jaundice were collect-ed,and Meta-analysis was performed by using Rev Man 5.2 software after data extracting and quality evaluating by modified Jadad. RESULTS:Totally 12 RCTs were enrolled,involving 1585 patients. Results of Meta-analysis showed total effective rate [OR=0.19,95%CI(0.12,0.30),P<0.001],serum total bilirubin levels [MD=-40.78,95%CI(-42.68,-38.89),P<0.001] and time of bilirubin decreased to normal [MD=-2.56,95%CI(-2.72,-2.40),P<0.001] in test group were significantly better than control group,the differences were statistically significant between 2 groups;3 reports of adverse reactions showed scme children had vomiting and mild diarrhea,they were improved after symptomatic treatment,and it did not affect the treatment. CONCLU-SIONS:The effectiveness of Yinzhihuang oral liquid is good in the adjuvant treatment of neonatal jaundice,with mild adverse reac-tions.
3.Efficacy of Yixinshu Capsule in the Adjuvant Treatment of Angina Pectoris:A Meta-analysis

OBJECTIVE:Tosystematically evaluate the efficacy of Yixinshu capsule in the adjuvant treatmentof angina pecto-ris,and provide evidence-based reference for clinical treatment. Meta-analysis was performed bu using Rev Man 5.2 software after data extraction and quality evaluation by modified Jadad. RESULTS:Totally 11 RCTs were enrolled,involving 1 827 pa-tients. Results of Meta-analysis showed the total effective rate of angina pectoris improvement [OR=0.30,95%CI(0.23,0.40),P<0.001] and electrocardiogram improvement [OR=0.52,95%CI(0.41,0.66),P<0.001] in test group were significantly higher than control group,there was significant difference between 2 groups. CONCLUSIONS:Yixinshu is effective for the adjuvant treatment of angina pectoris.
4.Efficacy of Qijiao Shengbai Capsule in the Adjuvant Treatment of Leucopenia:A Meta-analysis

OBJECTIVE:To systematically review the efficacy of Qijiao shengbai capsule in the adjuvant treatment of leukope-nia,and provide evidence-based reference for the clinical treatment. METHODS:Retrieved from CJFD,VIP,Wanfang Database and Medline and EMBase,randomized controlled trials(RCT) about the efficacy of Qijiao shengbai capsule (test group) versus blank control(control group) in the adjuvant treatment of leucopenia were collected. Meta-analysis was performed by using Rev Man 5.2 software after the quality assessment and data extraction. RESULTS:A total of 15 RCTs were included,involving 1 383 patients. Results of Meta-analysis showed the total effective rate [OR=0.31,95%CI(0.23,0.43),P<0.001],obvious effective rate [OR=0.31,95%CI(0.24,0.41),P<0.001],leukocyte count [MD=-1.04,95%CI(-1.10,-0.98),P<0.001] and myelosuppres-sion reduction [OR=0.27,95%CI(0.14,0.52),P<0.001] in test group were significantly higher than control group,there were sig-nificant differences. Only 9 patients had adverse reactions of mild diarrhea,nausea and other symptoms,and it relieved after symp-tomatic treatment. CONCLUSIONS:Qijiao shengbai capsule has achieve good efficacy in the adjuvant treatment of leukopenia. Due to the limits of quality and sample size,more strict designed,long-term follow-up of large scale RCTs are needed for the fur-ther verification of the conclusion.
5.Study on screening for 274 suspected cases of corona virus disease 2019

Objective:To evaluate the diagnostic efficacies of computed tomography (CT), clinical manifestations and 2019 novel coronavirus (2019-nCoV) nucleic acid test results for the screening and diagnosis of novel coronavirus pneumonia.Methods:The clinical data of suspected cases with corona virus disease 2019 (COVID-19) visited fever clinic or stayed in emergency room of the First Affiliated Hospital of Nanchang University from January 23 to February 9, 2020 were collected. Totally 274 cases who met the inclusion criteria were enrolled. Four screening schemes including chest CT screening, scoring, chest CT and scoring series screening, chest CT and scoring parallel screening were designed. The statistical analysis was performed by using chi-square test. The sensitivities, specificities and the areas under the receiver operator characteristic curve of the four screening schemes were calculated, and the diagnostic efficacies were evaluated.Results:Among the 274 cases, 93.80%(257/274) presented with typical clinical symptoms, 47.81%(131/274) had epidemiological history, and the blood routine examination results of 45.26%(124/274) cases met the positive criteria of the scoring , and chest CT results of 43.07%(118/274) cases met the positive criteria of chest CT screening. The 2019-nCoV nucleic acid test positive rate of cases with epidemiological history was 30.53%(40/131), which was higher than that of cases without epidemiological history (9.09%(13/143)), and that of cases with typical imaging performance on chest CT was 40.68%(48/118), which was higher than cases without typical imaging performance (3.21%(5/156)). The differences between the above groups were both statistically significant ( χ2=20.150、60.468, respectively, both P<0.01). The positive rates of viral nucleic acid detection in cases with positive findings of chest CT screening, scoring, series screening, and parallel screening were 40.68%(48/118), 23.74%(47/198), 44.68%(42/94) and 23.87%(53/222), respectively, while those in cases with negative findings of the four screening schemes were 3.20%(5/156), 7.89%(6/76), 6.11%(11/180) and 0(0/52), respectively. The positive rates of the four screening schemes were all significantly different from that of viral nucleic acid detection ( κ=0.402, 0.100, 0.431, 0.106, all P<0.01). The chest CT screening method had a sensitivity of 90.57%and a specificity of 68.33%, and an area under the operating characteristic curve of the subject was 0.794, which had higher diagnostic value than those of the other three screening schemes. Conclusions:For the screening and diagnosis of COVID-19 cases, epidemiological history, positive 2019-nCoV nucleic acid test with ≥2 typical clinical manifestations have highly diagnostic value. On the basis of the preliminary screening of chest CT examination, flexible analysis of the diagnostic results could improve the diagnostic value of each detection method.
6.Interferon-gamma inhibits aldehyde dehydrogenasebright cancer stem cells in the 4T1 mouse model of breast cancer.

BACKGROUND:
Despite improvements in disease diagnosis, treatment, and prognosis, breast cancer is still a leading cause of cancer death for women. Compelling evidence suggests that targeting cancer stem cells (CSCs) have a crucial impact on overcoming the current shortcomings of chemotherapy and radiotherapy. In the present study, we aimed to study the effects of T cells and a critical anti-tumor cytokine, interferon-gamma (IFN-γ), on breast cancer stem cells.
METHODS:
BALB/c mice and BALB/c nude mice were subcutaneously injected with 4T1 tumor cells. Tumor growth and pulmonary metastasis were assessed. ALDEFLOUR™ assays were performed to identify aldehyde dehydrogenasebright (ALDHbr) tumor cells. ALDHbr cells as well as T cells from tumor-bearing BALB/c mice were analyzed using flow cytometry. The effects of CD8+ T cells on ALDHbr tumor cells were assessed in vitro and in vivo. The expression profiles of ALDHbr and ALDHdim 4T1 tumor cells were determined. The levels of plasma IFN-γ were measured by enzyme-linked immunosorbent assay, and their associations with the percentages of ALDHbr tumor cells were evaluated. The effects of IFN-γ on ALDH expression and the malignancy of 4T1 tumor cells were analyzed in vitro.
RESULTS:
There were fewer metastatic nodules in tumor-bearing BALB/c mice than those in tumor-bearing BALB/c nude mice (25.40 vs. 54.67, P < 0.050). CD8+ T cells decreased the percentages of ALDHbr 4T1 tumor cells in vitro (control vs. effector to target ratio of 1:1, 10.15% vs. 5.76%, P < 0.050) and in vivo (control vs. CD8+ T cell depletion, 10.15% vs. 21.75%, P < 0.001). The functions of upregulated genes in ALDHbr 4T1 tumor cells were enriched in the pathway of response to IFN-γ. The levels of plasma IFN-γ decreased gradually in tumor-bearing BALB/c mice, while the percentages of ALDHbr tumor cells in primary tumors increased. IFN-γ at a concentration of 26.68 ng/mL decreased the percentages of ALDHbr 4T1 tumor cells (22.88% vs. 9.88%, P < 0.050) and the protein levels of aldehyde dehydrogenase 1 family member A1 in 4T1 tumor cells (0.86 vs. 0.49, P < 0.050) and inhibited the abilities of sphere formation (sphere diameter <200 μm, 159.50 vs. 72.0; ≥200 μm, 127.0 vs. 59.0; both P < 0.050) and invasion (89.67 vs. 67.67, P < 0.001) of 4T1 tumor cells.
CONCLUSION
CD8+ T cells and IFN-γ decreased CSC numbers in a 4T1 mouse model of breast cancer. The application of IFN-γ may be a potential strategy for reducing CSCs in breast cancer.
