Objective To explore the role of granulocyte macrophage colonystimulating factor(GM-CSF),C reaction protein(CRP)and tumor necrosis factor-?lpha(TNF?)in infant cytomegalovirus hepatitis.Methods Cytomegalovirus DNA of 56 cases of children with cytomegalovirus hepatitis and 51 normal controls in blood were detected using FQ-PCR.The levels of GM-CSF and TNF?in serum were assayed with ELISA,and CRP was measured with immunoturbidmemetry assay.Results The levels of M-CSF,CRP and TNF? in serum of infant cytomegalovirus hepatitis were higher obviously than those in serum of the normal control group.Conclution M-CSF,CRP and TNF? participate in pathological immune response and anti-infective immunity of cytomegalovirus hepatitis.

Objective:To observe the effect of CD34 immunoaffinity column on the isolation of hematopoietic stem and progenitor cells of cord blood and the proliferatory and expansion characteristics of CD34 + cells in vitro.Methods:CD34 +cells were isolated from cord blood using CD34 immunoaffinity column.Cell surface antigens were analysed by FACS.The separated and un separated cells were cultured with human hematopoietic growth factors(HGFS)in liquid culture system and CFU GEMM culture system.Results:CD34 + cells were enriched with a purity of 49.62%?17.69% and a recovery of 54.38%?11.91% using the immunoaffinity column.The separated CD34 +cells and cord blood MNC expanded to 561.00 folds and 44.44 folds respectively after cultured with HGFS for 20 days.The percentages of CD34 + cells in separated group and control were 53.38% and 7.91% respectively after cultured for 12 days.The number of CFU GEMM in separated group was significantly higher than that in control(P

AIM: In order to explore the mechanism of the GC - like effect of Tsiao Shaihutang (TSS), the down - regulation of glucocorticoid receptor and mRNA by TSS were studied. METHODS: GR sites were determined by receptor radio ligand binding assay method. At the same time, GR mRNA level was determined by quantitative reverse- transcriptive polymerase chain reaction (RT- PCR) method. RESULTS: (1) GR sites and GR mRNA level were down - regulated significantly after GC, TSS (P < 0.01 ) treatment . (2) GR sites and GR mRNA level in GC plus TSS group were obviously higher than those in GC group (P < 0.01 ). CONCLUSIONS: These data suggested that TSS can significantly down- regulate GR and GR mRNA level.
5.Evaluation of Intracoronary Electrocardiogram on Myocardial Ischemia During Percutaneous Transluminal Coronary Angioplasty

The changes of intracoronary electrocardiogram (IC-ECG) and surface electro-cardiogram (S-ECG) were recorded simultaneously during percutaneous transluminal coronary angioplasty(PTCA) in 33 cases of coronary heart diseases with 41 vessel lesions. The results sug-gested that ST-segment change rate on IC-ECG(90. 2%) was higher than that on S-ECG(56%). ST-segment elevation on IC-ECG was earlier and more significant than that on S-ECG. The IC-ECG appeared to be more sensitive than S-ECG for detecting myocardial ischemia. The technique for recording IC-ECG is simple and it can help to perfome the PTCA successfully.
6.Study on the effect of Klotho gene interferred by plasmid-mediated short hairpin RNA (shRNA) on sinoatrial node pacing channel gene.

The study was aimed to assess the effect of Klotho gene and sinoatrial node pacing channel gene (HCN4 and HCN2) for studying sick sinus syndrome, with Klotho gene under the interference of Plasmid-mediated short hairpin RNA. Twenty-five C57BL/6J mice were divided into four groups, i. e, plasmid shRNA 24h group, plasmid shRNA 12h group, sodium chloride 24h group and sodium chloride 12h group. Plasmid shRNA 50microL (1microg/microL) and sodium chloride 50microl were respectively injected according to mice vena caudalis into those in plasmid shRNA group and sodium chloride group. After 12h or 24h respectively, all mice were executed and their sinoatrial node tissues were cut. The mRNA of Klotho, HCN4 and HCN2 gene were detected by RT-PCR. The results of RT-PCR showed that Klotho, HCN4 and HCN2 mRNA levels were lower compared with those in sodium chloride 12h group after 12h interference interval. The results indicated that there might be the a certain relationship between Klotho gene and sinoatrial node pacing channel gene.

7.Survival properties of ETEC surface-displayed K88ac-LT(B) on Lactobacillus casei.

K88ac-LT(B) gene derived from pQE30-K88ac-LT(B) was cloned into the expression vector pLA and then the recombinant vector was transformed into the competent cells Lactobacillus casei 525. The recombinant bacteria were grown at 37 degrees C, in MRS broth. Western blotting analysis with rabbit-anti-K88ac-LT(B) polyclonal serum indicated that the recombinant protein reacted with the specific antibodies. The results showed that the molecular weight of the recombinant protein was about 71.2 kD. The K88ac-LT(B) fusion protein on the cell surface was confirmed by immunofluorescence mciroscopy and flow cytometric analysis. In addition, the survival of recombinant Lactobacillus casei 525 was studied in imitative gastrointestinal environments such as artificial gastro fluid (pH 1.5-5.5), artificial intestinal fluid, bile(0.3-3.0 g/L). The results indicated that the recombinant strain survived well in artificial gastric fluids at pH 2.5-4.5 in 5 h. The recombinant Lactobacillus casei 525 could slowly grow in the artificial intestinal fluid for different time, and could survive in 0.3% bile.

8.Clinical effect of montelukast combined with sidenafil in the treatment of chronic obstructive pulmonary disease complicated with pulmonary hypertension

Objective To study the clinical effect of montelukast combined with sidenafil in the treatment of chronic obstructive pulmonary disease ( COPD ) complicated with pulmonary hypertension .Methods 94 COPD patients with pulmonary hypertension were selected ,and they were randomly divided into two groups according to the digital table,47 cases in each group .The conventional group was treated with conventional treatment ,the combination group was treated with montelukast sodium and sildenafil on the basis of the conventional treatment .The effect,safety, quality of life(SF-36),heart and lung function were compared in two groups .Results The total effective rate of the combination group was 95.74%,which was significantly higher than 78.72%of the conventional group ,the difference was statistically significant (χ2 =6.114,P <0.05).Compared with the conventional group,6 minutes walking distance ,mean pulmonary artery pressure , forced expiratory volume in one second of the combination group were significantly improved,the differences were statistically significant (t =6.244,7.182,7.034,all P <0.05).After treatment,the SF-36 score of the combination group was (84.78 ±13.61)points,which was significantly higher than (63.47 ±9.62)points of the conventional group (t=9.343,P<0.05).The incidence rate of adverse reaction had no statistically significant difference between the two groups (χ2 =0.000,P >0.05).Conclusion Montelukast and sildenafil in the treatment of COPD patients complicated with pulmonary hypertension has good clinical effect ,and it can effectively improve patients'cardiopulmonary function , exercise endurance and quality of life , has no significant side effects.
9.Utility of failure mode and effect analysis to improve safety in patients with continuous renal replacement therapies

Objective To identify the potential risks in patients receiving continuous renal replacement therapies (CRRT). Methods The concept of failure mode and effect analysis (FMEA) was used to analyze the management of CRRT, to find out the potential failure risks in all of the steps, risk priority numbers (RPNs) of each failure mode were calculated and countermeasures were implemented. Results Twenty-eight failure modes were analyzed, of which 8 modes RPN>125. After improvement of the nursing process in patients with CRRT, the RPN was lowered. The incidence of unplanned end of the CRRT was decreased from 10.86% (24/221) to 5.81% (14/241) (χ2=3.896, P<0.05). Conclusions Application of FMEA in risk management for CRRT can help to standardize the workflow and guarantee the safety of patients during CRRT, and is beneficial to continuous improvement of nursing.
10.Co-word clustering analysis for hot research topics in enteral nutrition in critically ill patients based on PubMed

Objective:To identify hot research topics about enteral nutrition in critically ill patients in PubMed from 2011 to 2020, and to explore the current research landscape and development directions.Methods:Literature about enteral nutrition published from 2011 to 2020 were retrived in the PubMed database. Subject headings in publications were extracted using BICOMB and then two-way cluster analysis was performed using gCLUTO to cluster the subjects and generate cluster curve, matrix and tree diagram.Results:A total of 5,686 publications were included, mostly published in Unites States and United Kingdom and with an increasing trend. The researches focused on five aspects including feeding practice in very low birth weight infants, prevention and control of aspiration pneumonia in critically ill patients, catheterization methods in enteral nutrition, protein intake in critically ill patients, and clinical implementation of enteral nutrition guidelines in critically ill patients.Conclusions:Much importance has been attached to enteral nutrition researches over the recent decade. Analysis of hot research topics in enteral nutrition is beneficial to understanding the current research landscape and development directions and providing references for practice, research and education of enteral nutrition.
