2.Application of Calcium Phosphate Cement in Bone Defect Repair (review)

In recent years, more and more biological materials are used in bone defect repair. The ideal materials should not only have good biocompatibility, but also have the right space structure, surface morphology and the physical and chemical properties, to adapt the growth of osteoblasts. Calcium phosphate cement was used more widely among biological materials in bone defect repair with its good biocompatibility, degradability, osteoconductivity, injection, plasticity and other advantages. This paper introduced the experimental research and progress of the calcium phosphate cement application in bone defect repair.
3.Clinical Study on Deep Needling at Tianshu (ST 25) and Fujie (SP 14) plus Electroacupuncture for Functional Constipation

Objective To observe the clinical efficacy of deep needling at Tianshu (ST 25) and Fujie (SP 14) plus electroacupuncture in treating severe functional constipation (FC).Method Sixty-three patients with severe FC were randomized into a treatment group of 33 cases and a control group of 30 cases. The treatment group was intervened by deep needling at Tianshu (ST 25) and Fujie (SP 14) plus electroacupuncture, while the control group was by sham electroacupuncture. The Complete Spontaneous Bowel Movement (CSBM), Bristol Stool Scale (BSS), and Patient Assessment of Constipation Quality of Life (PAC-QOL) were evaluated before and after intervention.Result In the treatment group, the CSBM after 2-week, 4-week, and 8-week treatments and CSBM of the 4-week and 12-week follow-up study were significantly different from that before intervention (P<0.05). In the control group, the CSBM after 8-week treatments was significantly different from that before intervention (P<0.05). The CSBM after 2-week, 4-week, and 8-week treatments and CSBM of the 4-week and 12-week follow-up study in the treatment group were significantly different from that in the control group (P<0.05). The BSS scores were significantly changed after 2-week, 4-week, and 8-week treatments in both groups compared to that before intervention (P<0.01,P<0.05). The BSS scores after 2-week, 4-week, and 8-week treatments in the treatment group were significantly different from that in the control group (P<0.01,P<0.05). The item scores of PAC-QOL (physical discomfort, psychosocial discomfort, worries and concerns, and satisfaction scores and global score) were significantly changed after intervention in the treatment group (P<0.05). The item scores of PAC-QOLA including physical discomfort, worries and concerns, and satisfaction and global score were significantly changed after intervention in the control group (P<0.05). After intervention, the PAC-QOL scores of the treatment group were significant different from that of the control group (P<0.05).Conclusion Deep needling at Tianshu (ST 25) and Fujie (SP 14) plus electroacupuncture is an effective method in treating severe FC.
4.Establishment of floxuridine-resistant JeG-3 subline and the role of thymidylate synthetase mRNA expression in chem-resistant-prediction

Objective To establish human choriocarcinoma JeG-3 cell line resistant to floxuridine (FUDR)and describe the characteristics of this FUDR-resistant subline.The thymidylate synthase (TS) expression level in FUDR-resistant subline was also discussed.Methods The FUDR-resistant sub-line JeG-3/FUDRA was established by intermitted exposure to grads increased FUDR.Reversed microscope was used to observe the morphological changes in FUDR-resistant sub-line.Population doubling time was calculated and compared based on the growth curve of these two cell lines,cell cycles and chromosomal ploidy were assayed with flow cytometry methods.The chemo-luminescence assay was used to detect the hormone secretion by two kinds of cell lines.The resistant index (RI) was measured by cell counting kit-8 (CCK-8)assay.Quantitative RT-PCR was used to detect the mRNA expression level of TS and we also detected the TS mRNA expression level in different doses exposed subline.Results The RI of JeG-3/FUDRA was 31.62.Compared with the JeG-3 cell,the FUDR-resistant cell line had gross changes in morphological,cell growth,cell cycles and chromosomal numbers.The ability of human chorionic gonadotrop(hCG) and progesterone secretion was lower in JeG-3/FUDRA subline.The trend of TS mRNA expression was:while exposed to low concentration of FUDR,the TS mRNA expression level was downregulated,then followed the increasing dose of the drug,the expression level of TS mRNA ascended gradually.When the terminal concentration was reached,the expression level of TS mRNA in JeG-3/FUDRA subline was higher than that of JeG-3 cell line (P<0.05).Conclusions We established the FUDR-resistant subline of JeG-3 successfully.The TS mRNA expression level is stage-related to the different concentration and different phase in FUDR exposure.Our data suggested that TS mRNA expression level may not be used as a biomarker to predict the chemosensitivity in FUDR-based chemotherapy.
5.Survey on serum free fatty acids levels in patients with hyperlipidemia

Objective To explore the differences of serum free fatty acids (FFA)between the hyperlipidemia group and the healthy people with different ages and genders.Methods 912 healthy individuals of physical examination (healthy control group) and 1 061 patients with hyperlipidemia were selected and detected the serum FFA,total cholesterol,triglyceride,high density lipo-protein-cholesterol,low density lipoprotein-cholesterol,fasting plasma glucose and body mass index.The covariance analysis was used to investigate the differences in the level of serum FFA among different genders and age groups.The relationship between ser-um FFA and other serum lipids index in the patients with hyperlipidemia was analyzed by the multiple correlation analysis.Results The serum FFA level in the middle-aged females,middle-aged males and young males with hyperlipidemia was higher than that in the corresponding control groups;the serum FFA level in the young females with hyperlipidemia was lower than that in the other groups with hyperlipidemia;in the population groups with normal blood lipid,the serum FFA level in the old-age group was signifi-cantly higher than that in the middle-age and young group.Conclusion There is a difference in the serum FFA level in the hyperlip-idemia group with different ages and genders.
6.Increase of p-P38 and uPA expression in human breast cancer

Objective To study the expression and the significance of phosphorylated P38(p-P38) and uPA in breast cancer tissues.Methods Immunohistochemistry(S-P) was used to test the protein expression of p-P38 and uPA in 60 specimens from 50 patients with breast cancer.Western blotting was adopted to detect the protein expression of p-P38 and uPA in breast cancer cells and uPA protein expression after incubation with SB203580,an specific inhibitor of P38 MAPK blocked P38 MAPK signaling pathway.Results The positive rates of p-P38 protein and uPA protein in breast cancer tissues were 56.7%and 60.0% respectively.The protein expression level of p-P38 and uPA in breast cancer tissues was significantly higher than that in adjacent normal tissues(P
7.Assessment of the performance of red blood cells folic acid analyzer

Objective Through performance analysis of domestic red blood cells folic acid instrument to verify its testing ability. Methods According to national committee for clinical laboratory standards(NCCLS)EP5-A2,the precision was obtained through two level quality product detection within 20 days then the intra-assay variation coefficient(CV),inter-assay CV and CV in labora-tory was calculated and compared with the precision requirement coming from the red blood cells folic acid biological variation.Ac-cording to EP6-A,the linear evaluation was investigated with method of polynomial regression.By measuring the red blood cells fo-lic acid from 20 persons who presented the total healthy people,the reference scope given by the manufacturer was verified.Results Within the group CV was less than 6.67%(1/4 of biological variation allow total error),between the group CV was less than 8.90%(1/3 of biological variation allow total error),the laboratory CV was less than 13.35%(1/2 of biological variation allow to-tal error).At medical decision level,the calculated bias and the confidence limit were smaller than the allow range of error.Both fac-tors of quadratic polynomial and cubic polynomial regression models were not statistically significant compared with zero,so detec-tion system was linear.The reference scope given by manufacturer was not suitable for the local people.Conclusion The precision, accuracy and linearity of domestic red blood cells folic acid analyzer were accord with requirements.The laboratory should establish reference interval suitable for the local population.
8.Analysis of risk factors associated to malnutrition in elderly inpatients

Objective To investigate the risk factors associated to malnutrition in elderly inpatients for supplying clinical evidences for the prevention and therapy of malnutrition. Methods A one-to-one matched case-control study was used to investigate the risk factors associated to malnutrition in elderly inpatients over 60 years old. Results Eight-five pairs were included. Single factor analysis suggested that serum total protein. hemoglobin, serum total cholesterol, non auto -selective food, anorexia, dysphasia, confinement in the bed and infectious diseases might be associated with malnutrition in elderly inpaticnts (all P< 0.05 ). Logistic regression analysis demonstrated that serum total protein, hemoglobin, anorexia, dysphasia and confinement in the bed were the major risk factors (P<0.05). Hosmer-Lemeshow test showed the predicable results and practical results had better similarity (x2 =4. 957, P= 0. 762). Receiver operation characteristic curve ( ROC curve) also revealed the area under curve of serum total protein and hemoglobin were 0 760 and 0.621(P=0.000 and P=0.006 respectiuely), therefore, 56.8 g/L of serum total protein and 108 g/L of hemoglobin might be appropriate reference points for evaluating the malnutrition. Conclusions Serum total protein, hemoglobin, anorexia, dysphasia and confinement in the bed may play crucial roles for malnutrition in elderly inpatients, the status of malnutrition will be improved if the reversible risk factors are amendable.
9.Expression of C1QBP gene and its correlation with drug resistance in human resistance choriocarcinoma cell line

Objective To examine the complement component 1 Q subcomponent-binding protein (C1QBP) gene expression in human resistance choriocarcinoma cell lines and its parental cell line JeG-3,and to investigate whether silence C 1QBP by small interference RNA could reverse the resistance of human resistance choriocarcinoma cell lines to its relevant chemotherapy drugs.Methods Expression of C1QBP mRNA and protein in cells were detected by real-time fluorogenic quantitative PCR and western blot,respectively.The difference of C 1QBP expression was compared between human resistance choriocarcinoma cell lines and its parental cell line JeG-3.Sub-cellular location was proved by confocal immunofluorescence microscopy.A lentiviral vector containing short hairpin RNA (shRNA) targeting C 1QBP was constructed and cotransfected with the packaging plasmid mixture into 293T cells by lipofectamine 2000.The human resistance choriocarcinoma cell lines were infected with the packaged lentivirus.Real-time fluorogenic quantitative PCR and western blot were used to validate whether the C 1QBP gene expression was silenced.The cell counting kit 8(CCK8)was used to determine the drug sensitivity.Results (1)The C1QBP mRNA expression levels among four human resistance choriocarcinoma cell lines[JeG-3/floxuridiuum (FUDR),JeG-3/methotrexate (MTX),JeG-3/etoposide (VP),JeG-3/dactinomycin (KSM)] were 2.520±0.680,1.770±0.230,1.940±0.090 and 1.740±0.350 folds compared to that in JeG-3 cells.The C1QBP protein was higher expression level in human resistance choriocarcinoma cell lines than that in JeG-3.The immunofluorescence methods and confocal analysis showed that C1QBP localized predominantly in the mitochondrial matrix.(2)The C1QBP mRNA expression in JeG-3/FUDR cells after infected with lentiviral vector were decreased by 93.1% (P<0.01).The protein expression of C 1QBP in JeG-3/FUDR cells after infected with lentiviral vector were almost completely suppressed.The resistance indexes of four human resistance choriocarcinoma cell lines(JeG-3/FUDR,JeG-3/MTX,JeG-3/VP,JeG-3/KSM) were respectively 86.3%,93.9%,92.8% and 89.9%,which were decreased remarkably by knockdown the C 1QBP expression (P<0.05).Conclusions C1QBP is overexpressed in human resistance choriocarcinoma cell lines compared with parental cell line JeG-3.Inhibition of C 1QBP by lentivirus-mediated small interference RNA could effectively reverses the resistance of human resistance choriocarcinoma cell lines to its relevant chemotherapy drugs.
10.CT features of hepatic echinococcosis in children

Objective To investigate the CT features of hepatic echinococcosis in children.Methods Clinical and CT data of 27 cases with hepatic echinococcosis confirmed by pathology and/or serology were retrospectively analyzed.Results Totally there were 26 cases of cystic echinococcosis and 1 case of alveolar echinococcosis.Ten cases were with simple hepatic echinococcosis and 17 cases with other organic echinococcosis,18 cases of intrahepatic single hydatid and 9 cases of intrahepatic multiple hydatid.A total of 1 alveolar lesion and 47 cystic lesions were found,including unilocular cyst accounted for 51.06% (24/47),cysts with collapse of the internal capsule accounted for 42.55% (20/47),cysts with wall calcification accounted for 4.26% (2/47) and multiple cysts accounted for 2.13% (1/47).There was no solid lesions.The cyst diameters which were larger than 5 cm accounted for 74.47% (35/47).Conclusion The CT can qualitatively diagnose hepatic echinococcosis in children.It is a reliable method of examination.
