1.Liposome-encapsulated brain-derived neurotrophic factor targets blood-brain barrier

Objective:To observe the characteristics of the tissue distribution of the blood brain barrier through the modification of the liposome encapsulated brain derived neurotrophic factor.Methods: A total of 32 SD rats were selected from Xi′an Medicine College of Foreign Affairs as the research object,and randomly divided into two groups (n=16) and control group (n=16).To establish a rat model of AD rats in the control group received no treatment,liposome group by rat tail vein injection of 10 μg/kg Tf-pGFAP-BDNF-PEG,analysis of liposome entrapped BDNF targeting liposomes through blood brain barrier tissue distribution characteristics.Results: 40-50 nerve growth factor (1 ml) was collected by using 24mCi99mTC and 20 L 0.5 ml was collected on the top of the column.The concentration of 8-17 tube in the peak was recovered,and the results showed that the labeling rate of neurotrophic factor was 98.9%.Liposome group for tenth days,14 days of BDNF immunoreactive cells,significantly higher than the control group (P<0.05);neurotrophic factor in brain tissue of rats in body lipid gene expression at day fourteenth,significantly higher than the control group (P<0.05);expression of neurotrophic factors in liposome group rat,and neurotrophic factor expression level was significantly higher than the control group (P<0.05).The cerebrospinal fluid was clear,colorless and transparent,and no red blood cells were detected under microscope.Liposome group in cerebrospinal fluid of rats with fecal occult blood detection results of gold colloid showed negative,while the control group was positive.Conclusion: The combination of transferrin and polyethylene glycol modified liposomes combined with brain specific promoter can improve the targeting of liposomes.
2.Preparation of anti-hepatocellular carcinoma scFv immunoliposome and its killing effects on hepatocarcinoma cells in vitro

Objective:To compare the killing effects on HCC SMMC-7721 cells of immunoliposome PE38、immunotoxin PE38 and liposome PE38. Methods:Dissolute PC,CHOL and CHS in chloroform in proportion.Dry membrane was formed after chloroform was removed.Add the protein solution of PE38 to dissolute the dry membrane.Then pass the solution over a Sephadex G-50 column after ultrasoned and filtrated to detect the encapsulation efficiency of the liposome.The solution reacted in EDC,SSNHS and MES for 30 minutes.Then add Ab to the solution in 4℃ over night.MTT method was used to detect the killing effects on HCC cells of immunoliposome PE38、immunotoxin PE38 and liposome PE38 in vitro. Results:The killing effects on HCC cells of immunoliposome PE38 is the best,and that of liposome PE38 is the worst.The respective IC_(50) s are:0.59 ?g/ml、6.04 ?g/ml and 92.07 ?g/ml. Conclusion:Immunoliposome PE38 may be a protential durg in the treatment of hepatocarinoma.
4.The expression of the pancreatic duodenal homeobox-1(PDX-1) in pancreas of experimental type 2 diabetic rat and the effect of rosiglitazone on its expression

Objective To observe PDX-1 expression in the pancreas of type 2 diabetic rat model and effects of rosiglitazone (RGZ) on it. Methods Type 2 diabetic rat model was induced by long-term feeding with high-fat foods followed by a single intraperitoneal injection of low dose of streptozotocin (STZ). The morphological changes of pancreas were examined by microscopy and immunohistochemistry. The mRNA levels of PDX-1 and insulin were determined by RT-PCR. PDX-1 protein expression was detected with Western blot. Results The percentage of insulin-positive cells(12.75±2.18),the average optical densities of PDX-1-positive area(0.240±0.051),PDX-1 mRNA level (0.153±0.071)and PDX-1 protein level(0.253±0.028) were significantly lower in untreated diabetic rats than in the controls(42.61±2.68,0.648±0.087,0.49±0.032,0.720±0.036 respectively)(all P<0.01),and in RGZ-treated versus untreated group those parameters increased significantly (all P<0.05~0.01). Conclusions Rosiglitazone could protect islet cell function,and improve PDX-1 expressions in both mRNA and protein levels.
5.Reconstruction of anti-hepatocellular carcinoma scFv pharmacy and its killing effect on hepatocardnoma cell in vitro

objective To observe whether the killing effect on HCC SMMC-7721 cell of the antihepatocellular carcinoma scFv reconstructed by pharmacy was enhanced or not.Methods Prokarycytic expression vector containing PET32a-RC-RNase was induced to express by IPTG.The inclusion body purified and Western-blotting was used.PC.CHOL and CHS was added in chloroform.Dry membrane was formed after chloroform was removed.RC-RNase protein solution was added to dissolute the membrane.Then pass the solution over a Sephadex G-50 column after ultrasound and filtrated to detect the encapsulation efficiency of the liposome.The solution reacted in EDC.SSNHS and MES for 30 minutes.Then add hdscFv to the solution in 4 ℃ over night.MTT method was used to detect the killing effect on HCC cell of immunoliposome RC-RNase,immunotoxin RC-RNase and liposome RC-RNase in vitro.Resuits The killing effect on HCC cell of immunoliposome RC-RNase is the best.but that of Iiposome RC-RNase is the worst.The respective JC50 are:3.28μg/ml,22.44μg/ml and 98.26μg/ml.Conclusion The anti-hepatocellular carcinoma scFv relomtructed by pharmacy can promote the killing effect on HCC cell and may have potential in the treatment of hepatocarcinoma.
6.Advances in functional genomics studies underlying acetic acid tolerance of Saccharomyces cerevisiae.

Industrial microorganisms are subject to various stress conditions, including products and substrates inhibitions. Therefore, improvement of stress tolerance is of great importance for industrial microbial production. Acetic acid is one of the major inhibitors in the cellulosic hydrolysates, which affects seriously on cell growth and metabolism of Saccharomyces cerevisiae. Studies on the molecular mechanisms underlying adaptive response and tolerance of acetic acid of S. cerevisiae benefit breeding of robust strains of industrial yeast for more efficient production. In recent years, more insights into the molecular mechanisms underlying acetic acid tolerance have been revealed through analysis of global gene expression and metabolomics analysis, as well as phenomics analysis by single gene deletion libraries. Novel genes related to response to acetic acid and improvement of acetic acid tolerance have been identified, and novel strains with improved acetic acid tolerance were constructed by modifying key genes. Metal ions including potassium and zinc play important roles in acetic acid tolerance in S. cerevisiae, and the effect of zinc was first discovered in our previous studies on flocculating yeast. Genes involved in cell wall remodeling, membrane transport, energy metabolism, amino acid biosynthesis and transport, as well as global transcription regulation were discussed. Exploration and modification of the molecular mechanisms of yeast acetic acid tolerance will be done further on levels such as post-translational modifications and synthetic biology and engineering; and the knowledge obtained will pave the way for breeding robust strains for more efficient bioconversion of cellulosic materials to produce biofuels and bio-based chemicals.

7.Effect Observation of Azithromycin Combined with Bailing Capsules on Chronic Obstructive Pulmonary Disease

Objective:To observe the efficacy and safety of azithromycin combined with Bailing capsules in the treatment of chron -ic obstructive pulmonary disease ( COPD) in stable phase .Methods:Totally 154 cases of COPD patients were randomly divided into the treatment group(80 cases)and the control group(74 cases).Both groups were treated with the routine symptomatic treatment , and at the same time, the control group was treated with Bailing capsules (1 g, po, tid)and the treatment group was treated with azithromy-cin tablets (250 mg, po, qd)combined with Bailing capsules(1 g, po, tid).The course of treatment was 6 months and all the patients were followed up for 6 months after the treatment.The clinical efficacy, lung function, exercise tolerance, quality of life, the times of acute exacerbation and the incidence of adverse drug reactions of the two groups were observed and compared .Results:After the treat-ment, the lung function, exercise tolerance and quality of life were improved significantly in the two groups (P<0.05 or <0.01), and all the indices in the treatment group were significantly better than those in the control group (P<0.05 or <0.01).The total ef-fective rate in the treatment group was higher than that in the control group (P<0.05).The times of acute exacerbation in the treat-ment group were lower than those in the control group (P<0.01).The adverse drug reactions showed no significant difference between the groups (P>0.05).Conclusion:Azithromycin tablets combined with Bailing capsules can improve lung function , enhance exer-cise tolerance , improve the quality of life and reduce the frequency of acute exacerbation effectively with good security in the treatment of patients with COPD in stable phase .
8.Study on Quality Standard of Huangdi Cataplasm

Objective To develop the quality standard for evaluating Huangdi cataplasm. Methods Thin layer chromatography (TLC) was used to qualitatively identify Astragalus membranaceus (Fisch.) Bunge,Rheum palmatum Linn,Rhizoma Chuanxiong,Angelica sinensis and Resina Draconis in Huangdi cataplasm.HPLC method was used to determine astragaloside A and loureirin B in Huangdi cataplasm. Results The Astragalus membranaceus (Fisch.) Bunge,Rheum palmatum Linn,Rhizoma Chuanxiong,Angelica sinensis and Resina Draconis were well separated by TLC without interference in the negative control.content of Astragaloside A and loureirin B showed good liner relationships with respective peak area within the range of 6.96-23.2 μg,and 0.072-0.648 μg,with r = 0.999 5,r = 0.999 9, respectively;and the average recovery was 97.18%,and 96.93%,RSD was 1.21%(n= 6),1.53% (n = 6 ), respectively. Conclusion The established qualitative and quantitative detection method is simple, specific, reproducible, accurate and reliable, which can be used for quality control of Huangdi cataplasm.
9.Perioperative nursing for patients with hepatic cancer adjacent to bile duct treated with microwave ablation by using bile duct cooling technique

Objective To discuss the perioperative nursing care for patients with hepatic cancer located near the central bile duct, who receive microwave ablation treatment by using bile duct cooling protection technique. Methods A total of 21 patients with hepatic cancer located near the central bile duct received percutaneous microwave ablation treatment (PMWA) using bile duct cooling protection technique. Preoperative routine nursing and psychological nursing, close cooperation, close postoperative observation and prompt nursing measures were strictly carried out. Results PMWA was successfully accomplished in all the 21 patients, and no biliary complications occurred. All 21 patients were cured at the time of discharge, no nursing-related complications or death occurred. Conclusion In treating hepatic cancer located near the central bile duct, bile duct cooling protection technique is a new treatment technology. Effective perioperative nursing care can improve the operative success rate and reduce the incidence of complications.
10.Observation of Therapeutic Effects of Needle Warming Moxibustion for Chronic Diarrhea

Objective: To observe the therapeutic effects of needle warming moxibustion for chronic diarrhea. Methods: Two-hundred cases with chronic diarrhea were divided randomly into a treatment group and a control group. 100 cases in the treatment group were treated with needle warming moxibustion on specific acupoints. 100 cases in the control group were treated by oral administration of Smecta. Results: The therapeutic results showed cure in 52 cases, remarkable effect in 34 cases, improvement in 14 cases and the total effective rate in 100.0% in the treatment group, and cure in 5 cases, remarkable effect in 23 cases, improvement in 33 cases, failure in 39 cases and the total effective rate in 61.0% in the control group. The comparison of the total effective rate between the two groups was statistically significant (P<0.05). Conclusion: The needle warming moxibustion on specific acupoints is better than oral administration of Smecta in the therapeutic effects.
