2.Effect of low-dose FK778 on the progression of chronic renal ailograft rejection in a rat model

Objective To study the effect of low-dose FK778 in preventing chronic renal al- lograft rejection in rats.Methods The rat model of chronic renal allograft rejection was established by using micro-surgery technique.The recipients were divided into two groups.The recipients in the study group were treated with FK778 at a dose of 5mg?kg~(-1)?d~(-1)dissolved in carboxymethylcellulose by means of gavage and the controls were treated with carboxymethylcellulose.Urinary protein con- centrations were measured every 4 weeks for 24 weeks.On 24th week after operation,the rats were killed and the kidney grafts were taken out for histological and immunohistological examinations as well as quantitative real-time RT-PCR analysis.Results After 24 weeks of treatment,proteinuria, the severity of chronic rejection,glomeruIosclerosicytes and monocytes/macrophages in the study group were significantly milder than in control group.And the expression of TGF-?mRNA and PDGF-B mRNA was significantly reduced in the study group as compared with that in the control group.Conclusion Low-dose of FK778 might prevent the rats from chronic renal allograft rejection.
3.Comparative study of EEG and CT in early youth with cerebral cysticercosis

Objective To investigate the diagnostic value of EEG and CT in early youth with cerebral cys- ticercosis.Methods The EEG and CT manifestations were studied in 240 early youth with cerebral cysticercosis. Results The abnormal rate of EEG was 86.7 % in children with cysticercosis,which mainly showed the diffuse or focal irregularity complex slow waves in frontal lobe,central lobe and anterior lobe.The abnormal rate of CT was 70.8 % in all patients,and flaky and circular focus were chief manifestations.There was a significant difference be- tween the rates of EEG and CT(P
4.Clinical Characteristics and Diagnosis of Hemiconvulsion-Hemiplegia-Epilepsy Syndrome

Objective To summarize the clinical characteristics and diagnosis of hemiconvulsion-hemiplegia-epilepsy(HHE) syndrome.Methods The clinical data of 5 children with HHE syndrome were analyzed.Cranial magnetic resonance imaging MRI and /or computerized tomography(CT) and electroencephalogram(EEG) were done in all patients.Results The age of onset was less than 4 years in 4 of 5 patients and initial seizures were associated with fever in 3 cases.The longest duration of convulsive seizures lasted more than 2 hours in all cases,followed by the appearance of hemiplegia involving the side of the body where the convulsions were localized.Subsequent epilepsy developed within 2 years of the hemiplegia in all 5 cases,with 4 cases simple partial seizure and 1 case psychomotor seizures.All cases had mental retardation.Cranial MRI showed left hippocampal sclerosis in 4 patients and CT brain scan showed right cerebral hemiatrophy in 1 case.The EEG showed abnormality in all patients.Epileptiform discharges were found in 4 cases,and the damage hemisphere showed slowing and low voltage of background activity in 1 case.All cases received mainly carbamazepine therapy,and the seizures were satisfactorily controlled or partly controlled.Conclusions HHE syndrome is recognized as hemiplegia-epilepsy syndrome after prolonged hemiconvulsion.The hippocampal sclerosis may be the consequence of repeated seizures,but not the cause.Better understanding of the syndrome,early and adequate treatment of prolonged seizures,especially of febrile convulsion,will reduce the incidence of HHE syndrome.
5.Significance of Multidrug Resistance-Associated Protein Gene and P-gp Expression in Malignant Lymphomas

Objective: This study was designed to investigate the relationship between expression level of multidrug resistance-associated protein (MRP) or P-glycoprotein (P-gp) genes and chemotherapy efficacy or clinical drug resistance in the patients with malignant lymphomas. Methods: Using the methods of semi quantitative reverse transcription polymerase chain reaction (RT-PCR) and flow cytometry (FCM), The authors examined the expression level of MRP and P-gp of 46 lymphoma patients. Results: The expression level and positive rate of P-gp in the recurrent patients was higher than that in untreated patients (P<0.001). There was no difference in MRP gene expression level and positive rate between recurrent and untreated patients (P>0.05). Chemotherapeutic effective rate in P-gp positive patients (26.67% ) was lower than that in P-gp negative patients (83.87% )(P<0.001). While there was no difference between MRP gene positive and negative patients, their chemotherapy effective rate had no difference. Relevant analysis showed that there was no correlation between MRP and P-gp (r-0.0808, P>0.05). Conclusion: P-gp expression correlates to multidrug resistance, and is the major mechanism of clinical drug resistance of lymphomas, whereas, MRP gene does not appears to play any role in that course.
7.Modification of culture method of retinal vascular endothelial cells in vitro

Background The in vitro culture of retinal vascular endothelial cells is the foundation of experimental study of retinal vascular disease. Shortage of human donor eyeballs is a main limiting for the laboratory work. The culture method of rat-derived vascular endothelial cells has been established. However, this method is not enough effective because of severer cellullar injury. Objective Present study was to establish a simple and high effective method for the culture of vascular endothelial cells in vitro. Methods The retinas from 5 SPF SD rats was digested by 0. 1% collagenase and cultured with explant culture method. 20% fetal bovine serum, vascular endothelial growth factor ( VEGF) , insulin-transferrin-selenium( ITS) were composed into the endothelial cell culture medium, and enough blowing was performed to get the cells and fragments from retinal tissue. The cellular suspension was prepared and cultured consequently on human fibronectin-coated culture flasks. Cultured vascular endothelial cells were identified by anti-von Willebrand staining factor. Results The cells emerged from the tissue mass,and cells and some tissue fragments attached to the wall after 24 hours of seeding. The cells grew to show the fusiform in 4 days and merged together in 5 to 6 days,and a cell monolayer was seen in the 14th day after culture. The endothelial cells showed the positive response for von Willebrand factor. After passage, the merging-growth statue of the cells was regained in 2 hours after culture. Conclusion Use of retinal pieces and collagenase-digestion can get the vascular endothelial cells with better activity in vitro. The culture method based on highly selective endothelial cell culture medium associated to FN adhesion-promoting is helpful for gaining the purified of endothelial cells.
8.Effect of continuous quality improvement in shortening the patients’ waiting time in transfusion room

Objective To explore the effect of the continuous quality improvement ( CQI) on shortening the patients ’ waiting time and improving the satisfaction of the waiting time in transfusion room . Methods The CQI procedure including setting up CQI groups , collecting and analysis the data , analyzing the cause, setting goals, making improvement plan and implement method , evaluating effect, summarizing and formulating the consolidation measures etc were carried out , and the patients’ waiting time and the satisfaction of the waiting time in transfusion room were compared before and after the implementation of the CQI . Results The average patients’ waiting time in transfusion room was (18 ±3) min after the implementation of the CQI, and was significantly lower than (30 ±5 ) min before CQI, and the difference was statistically significant (t=20.580, P<0.05).The satisfaction of the waiting time in transfusion room was 89% after the implementation of the CQI, and was significantly higher than 65% before CQI, and the difference was statistically significant (χ2 =9.416, P=0.024).Conclusions The nurses find out the best way to solve the problem and strengthen the team cooperation spirit through the implementation of the CQI and the utilization of team wisdom.The nurse work flow is smoothly, and the performance evaluation mechanism is reasonable , which greatly enhance the work enthusiasm and efficiency , shorten the patients ’ waiting time, and improve the patients’ satisfaction.
9.Microarray data analysis of genes related with erectile dysfunction in diabetic rats.

OBJECTIVETo investigate the changes of gene expression profiles associated with erectile dysfunction in diabetic rats.
METHODSAffymetrix Gene Chip arrays from the Gene Expression Omnibus (GEO) were used to examine the alterations in the gene expression profiles between streptozotocin-induced diabetic rats and littermate controls, and the data were analyzed with GeneSifter microarray analysis software.
RESULTSA total of 661 differentially expressed genes were identified, including 280 up-regulated and 381 down-regulated ones. Among the differentially expressed genes, kruppel-like factor 5 (klf5) was upregulated by 4.01 folds and ceruloplasmin(cp) by 5.14 folds; collagen, type XI, alpha1 was down-regulated by 5.84 folds and collagen, type I, alpha1 by 5.77 folds. The 661 differentially expressed genes involved such functional processes as glycoprotein biosynthesis, collagen fibril organization, angiogenesis in wound healing, triglyceride metabolism, cell proliferation and other important biological processes, and some pathways also involved such as fatty acid metabolism, neurodegenerative disorders, and ECM-receptor interactions.
CONCLUSIONSome genes such as klf5, cp, and collagen play important roles in the pathophysiology of diabetes-induced erectile dysfunction. Bioinformatic approaches offer a new means for identifying candidate genes and pathways relevant to the pathophysiology of diabetes-induced erectile dysfunction, highlighting also the potential complexity of this disorder.

10.Standardization of rat stable orthotopic liver transplantation model and comparison of the effect of two liver graft perfusion methods.

OBJECTIVETo standardize the establishment of a stable rat model of orthotopic liver transplantation and surgical technique of two-cuff technique for improving the operation success rate, and compare the effect of graft perfusion via the portal vein and the abdominal aorta on the graft function.
METHODSTotally 100 cases of orthotopic liver transplantation was performed in rats under microscope according to modified Kamada's two cuff technique. The rats were divided into 2 groups with the donor liver perfused through the portal vein with 10 ml cold lactated Ringer's solution, and via the abdominal aorta with 20 ml cold lactated Ringer's solution, respectively. The postoperative function recovery and pathological changes of the liver grafts were evaluated by serum ALT detection and histopathological examination. The operation success rate, 3-month survival rate of the rats and the complications were observed.
RESULTSNo significant differences was noted in the liver function, operation success rate and 3-month survival rate between the 2 groups, and histopathological examination also showed similar findings. The success rates of the two groups were 98% and 96%, with 3-month survival rate of 93.5% (29/31) and 93.3% (28/30) (P>0.05), respectively.
CONCLUSIONThe liver transplantation models with portal vein or abdominal aorta graft perfusion both serve well their respective purposes. Good microsurgical skills, standardized performance and shortened anhepatic period are the keys to improved stability and survival rate and reduced operative complications.
