1.Therapeutic effects of hypobaric hypoxia on asthma in guinea pigs

AIM: To explore the mechanism of the therapeutic effect of hypobaric hypoxia on allergic asthmas guinea pigs. METHODS:After the model of asthma was established with ovalbumin(OA) challenge in OA sensitized guinea pigs, the animals were randomized into the asthmas group(AG), the alleviative group(ALG) and hypobaric hypoxia treated group(HHTG). The levels of plasma cortisol and endothelin(ET), ET in bronchoalveolar lavage fluid(BALF) were determined by radioimmunoassay. The pulmonary pathological changes were observed with optical microscope. RESULTS:(1) Infiltration of eosinophils (EOS) in alveolar septum was found in AG and ALG, while it was reduced in HHTG. (2) The level of plasma cortisol was significantly higher in AG than in normal control group(NCG) (P<0.01).But it was significantly lower in ALG than in NCG(P<0.01), there was no difference in plasma cortisol level between HHTG and NCG. (3) The content of ET in plasma was significantly higher in ALG than that in NCG, AG and HHTG(P<0.01), and ET level in BALF was significantly higher in AG than that in NCG, ALG and HHTG(P<0.05),however, no significant difference was found among the latter three groups, respectively. CONCLUSION: After the treatment with hypobaric hypoxia, the ET levels of the plasma and BALF were decreaced and the content of plasma cortisol was increaced, and infiltration of EOS in alveolar septum was decreaced in asthmatic guinea pigs.That may be one of the mechanisms by which hypobavic hypoxia prevents and cures asthma.

3.Comparative analysis of Brucellosis epidemic between Shanxi province and the country

Objective To analyze the Brucellosis incidence and to predict the trends of the disease in Shanxi province and the national in recent years,which could provide the reference for surveillance,prevention and control of the disease.Methods Brucellosis data which was reported monthly during January 2006 and December 2010 in Shanxi province and the data released by Chinese Center for Disease Control and Prevention during January 2005 and December 2010 were collected.Several indexes,such as the annual increasing number,the development rate,growth rate and other indicators were applied to compare Shanxi province with the national Brucellosis epidemic in recent years.What's more,the seasonal autoregressive integrated moving average model (ARIMA) was fitted respectively with the data of Brucellosis incident number reported monthly,so as to predict the prevalence status in the coming two years by verifying the fitting effect.Results Brucellosis prevalence of Shanxi province reached the peak in 2008,and the incidence number was 5397,which was 900 more than 2007.From the onset of decline after 2008,the prevalence decreased by 17.67% (906/5128) in 2010.However,national incidence of Brucellosis kept increasing before 2009 and the prevalence increased rapidly from 2007 to 2008,and the growth rate reached 39.16% (8442/21 560).Although the number of Brucellosis fell by 2041 cases in 2010 than in 2009,the rate of decline was only 5.14%(2041/37 734).The fastigium of Brucellosis was from May to July yearly whether Shanxi province or the country.The ARIMA models of Shanxi province and the nation were ARIMA [(1,0,1)(1,1,0)12] and ARIMA[(1,0,1)(0,1,1)12],respectively,according to the incidence numbers reported monthly.The fitting effect of models showed that the predicted values of the two models were both consistent with the actual situation and all predicted values fell within the 95% confidence limits,which depicted that they both fitted well.The predicted values depict that the incidence of Brucellosis overall trend was basically stable in Shanxi province,while the numbers in the nation would increase in a small extent in 2011 and 2012.The fastigium of Brucellosis was still from May to July yearly.Conclusions Brucellosis control measures are effective in Shanxi province,incidence of Brucellosis declining.The ARIMA model could predict the number of Brucellosis well,which can provide a valuable reference for the predication and evaluation of Brucellosis epidemic in the future.

6.Relationship of Neural Protection and Expression of P-CREB and C-Jun in Hippocam pal of Neonatal Rats after Hypoxia-Ischemia Brain Damage and Reperfusion

Objective To explore the survival mechanism of hippocampal ne urons after damage of hypoxia-ischemia and reperfusion of brain.Methods Seven days old SD rats(n=56) were randomly divided into hypoxia-ischemia br a in iniury(HIBD) group and sham group.The HIBD and reperfusion model was establis hed.The flowing of blood was de tected by multicolor Doppler.The p-CREB(phosphorylated c-AMP response element bi nding protein)and c-Jun were immunohistochemically evaluated in hippocampus.Thi onin staining was used to observe the apoptosis.Results The expression of p-CREB reache d the peak at 3,24 h postreperfusion in the right hippocampus of HIBD group,and then decreased to the normal level on the 7th day.In contral group the same reg ions showed basic immn-noreactivity.While c-Jun reached the peak at 6 h postreperfusion,then with a slightly decrease at 24 h;and at 48 h the other peak appeared,then with a gradual decline .On the 7th day the mumber of positive cells were still significanthy more than control group(P0.05).The sham animal showed very few apoptosis cells in the regio ns of hippocampus.Conclusions The persistent activation of CREB in the hippocampus regulates,the expression of c-Jun through the signal transductions and is involved in the course of neuron s′ survival and repair during the period of post hypoxia-ischemia reperfusion.I t is very important for the protection of the pyramidal hippocampal neurons on t he damaged side,especially for the sensitive region CA1. 8.Analysis of paper publishing conditions by postgraduate background staff in an upper first-class hospital in Guangxi Province during 2010-2014

Objective Based on the analysis of papers published by postgraduate background staff from an upper first-class hospital in Guangxi during 2010-2014,claimed that the medical scientific research administrators should pay more attention and give more support to such staff.Methods Literature metrology method was used to describe and analyze the hospital staff,postgraduate staff resource composition and published papers from the annual growth rate,per capita,constituent ratio,quantitative analysis of core authors group.Results In the latest five years,the hospital postgraduate staff had published 169 papers,143 papers were published in statistical source journals or SCI;the amount of paper publication increased annually,the average of per author published paper was 0.39;but the quality still has a lot space for improvement.Conclusions Although the amount of paper publication by postgraduate background staff increased annually,the average paper number published per author showed the features of great fluctuation,the quality of those papers should be improved.The core authors were formed,but the quantity of this author group needs to be increased,as well as the scientific quality of those core authors.
9.Mechanism of hepatitis B virus infection of trophoblast cells and hepatitis B virus intrauterine infection

The HBsAg IHC staining positive cells could be observed in 6 placental tissues and 3 fetus' liver tissues,and HBcAg was also positive in 1 case of fetus' liver tissue.After co-incubating the tropho- blastic cells and HBV DNA positive serum in vitro,HBsAg expression and HBV DNA could be detected.Apoptosis of HBV-infected trophoblastic cells increased,which was demonstrated by in vivo and in vitro experiments and the apoptosis of placental cells was correlated with the cord blood HBV DNA level.The results of in vitro experiments showed that the apoptosis of trophoblastic cells increased with the elongation of infection time.After 6 months,1 of 12 newborns was positive for HBsAg,HBeAg and anti-HBc,6 was positive for anti-HBs.Conclusions The mechanism of HBV intra-uterine infection may be that HBV breaches the placental barrier and infects the fetus.The localization and replication of HBV in fetal tissues and organs are probably the important factors of chronic HBV infections in neonates.The apoptosis of trophoblastic cells may be the protective mecha- nism for the placental barrier to block the HBV intra-uterine transmission.
10.Protective Effects of Insulin on Acute Global Cerebral Ischemia Reperfusion Injury in Rats

Objective To observe effects and mechanisms of insulin on reperfusion injury after cerebral ischemia.Methods Sixty-six male Wistar rats were used in this study.All rats were divided into 3 groups as treated group(A),control group(B) and random sham-operated group(C).Four-vessel occlusion was used to establish global cerebral ischemia reperfusion model in study groups.The treated group were divided into 5 groups(A1-A5) and intraperineally injected with biosynthetic human insulin 2 IU/(kg?d) and 50%glucose 2 g/(kg?d) for 7 days,the blood glucose was monitored in preoperative and postoperative 3,6,12,24 h,and the blood glucose was maintained between 3.5-6.5 mmol/L.These animals of control group were given with saline 2 mL/(kg?d) for 7 days in abdominal cavity.All the rats were killed in the seventh day,brain homogenate was collected for detection of neuron specific enolase(NSE)and nitric oxide(NO).The hippocampus was separated for observation of electronic microscope.Results Concentration of NSE in brain tissue in group C was significantly higher than that of group A and group B,while the level in group A was higher than that of group B.Concentration of NO in group C was lower than that of group A and group B while the level of NO in group A was significantly lower than that of group B.Electron microscope showed that the ultrastructure of sham-operated group was nearly normal,damage degree of hippocampal neuron and gliacyte and capillary was gradually worse from group A1,A2 to A4,the damage degree of group B1,B2 and B4 was serious and there was no difference among them.Conclusion Insulin can really promote recovery of the cerebral injury after ischemia reperfusion.
