1.Evaluation for the performance of HIV Ag/Ab assay based on ECLIA in blood screening

【Objective】 To evaluate the performance of electrochemiluminescence immunoassay (ECLIA) in detecting HIV antigen/antibody in blood screening. 【Methods】 A total of 128 donors, reactive to anti-HIV(ELISA)testing, from September 2016 to September 2020 were enrolled, and seven samples were reactive to double anti-HIV reagents, among which 6 were confirmed by WB, 1 confirmed by NAT as negative.Two group of donors, reactive to solo anti-HIV reagent but being confirmed negative by WB(n=121) vs.randomly selected donors non-reactive to ELISA + NAT(June to September 2020, n=1360), were subjected to HIV antigen/antibody testing using ECLIA to compare the testing results, including concordance rate, sensitivity and specificity. 【Results】 The ECLIA results remained non-reactive for 1360 samples initially non-reactive to both ELISA and NAT.The concordance rate of anti-HIV reactivity by ECLIA and ELISA+ WB were 100%(6/6). For 122 samples, reactive to ELISA anti-HIV testing but nonreactive to confirmatory testing, 4(3.28%)of them were reactive to HIV antigen/antibody testing and 118(96.72%) nonreactive, with the concordance rate of ECLIA and ELISA at 96.88%(124/128). The sensitivity, specificity and false positive rate of ECLIA and ELISA were 100% vs 100%, 99.73% vs 91.77%, and 0.27% vs 8.23%, respectively. 【Conclusion】 ECLIA for HIV antigen/antibody detection has good sensitivity and specificity, which can meet the requirements of blood screening, and the false positive rate is lower than that of ELISA, adopted commonly in blood bank at present.

3.Research progress of single Chinese exterior-relieving medicine in the treatment of osteoporosis

In recent years, pharmacological researches of Chinese medicine have found that single Chinese exterior-relieving medicines such as Pueraria, Semen Sojae Praeparatum and Cimicifuga had very good anti-osteoporosis effect. They can significantly improve the bone mineral density (BMD) and bone mineral content(BMC), promote bone formation, and prevent bone absorption. Based on relative documents from home and abroad, this paper analyzed the current situation and provided new ideas and methods for prevention and treatment of osteoporosis.
5.ANTI-ARRYTHMIC EFFECTS OF TETRAHYDROBERBERINE

The action of Tetrahydroberberine ( THB ) on arrhythmias of the animal models were studied. The arrhythmias induced by CaCl2 and BaCl2 were significantly counteracted by THB ( 30mg/kg ip ) without markedly changing the hemodynamic parameters of rats, but those evoked by aconitine or ouabain were not inhibited in rats or guinea pigs. THB ( 40mg/kg ip) was used to be prevented the arrhythmias induced by ischaemia-reperfusion of left anterior des-cending coronary artery in rats. It could decrease the incidence of vetricular fibrillation, short restored time of the sinus rhythm and diminish the mortality. THB ( 10mg/kg iv ) effectively reduced the incidence of the ventricular arrhythmia induced by hypothalamic electric stimulation in rabbits. The results indicate that THB possess significant antiarrhythmic effect. It may be concerned the action of THB to blok calcium entry
6.Qualitative research on status of residents' living condition in Third Grade Class A hospitals in Beijing

Objective By exploring the state of residents' living condition, it is helpful to promote the living condition of residents and improve the quality of medical services. Methods A qualitative re-search was conducted on 13 residents in 4 hospitals in Beijing by depth interview during April to May 2016, and the data were collected and analyzed by using Colaizzi seven step method. Results In terms of work, the residents had specific work, and the interpersonal relationship was more harmonious. In learning, residents were able to take the initiative to learn, and hoped to get more help in scientific research. In life, residents faced multiple pressures from the economy and the family. In the subjective cognition, the cogni-tion of resident was rational and objective. Conclusion The residents' work shows a heavy workload, and while working the residents need to take the study and the life pressure into account. And the residents can rationally recognize their role. To improve the living status of the resident physician, we need to integrate multiple forces, to promote the integration of live doctors into the hospital culture, and enhance their sense of existence. We also need to improve the construction of the system and improve the protection of live training physicians to enhance their sense of security. Most important of all, the whole society should pay more attention to the living condition of the residents and enhance their sense of access.
7.Epidemiology and pathogen composition of enterovirus in children with hand, foot and mouth disease in Guangyuan, Sichuan

Objective To investigate the epidemiology and pathogenic characteristics of enterovirus in children with hand, foot and mouth disease (HFMD) in Guangyuan area, and to provide a basis for the development of HFMD prevention and control strategies. Methods A total of 759 children with HFMD clinically diagnosed in Guangyuan area from January 2019 to January 2021 were randomly selected. The seasonal, age, gender, regional distribution and pathogen distribution characteristics of HFMD were analyzed by descriptive epidemiological methods. Results The incidence of HFMD was seasonal and presented a typical bimodal distribution, with the summer peak occurring from April to July (χ2=8.714, P<0.05) and the winter peak from October to December (χ2=7.542, P<0.05). There was a significant difference in the incidence rate between regions, and the incidence rate in urban areas was much higher than that in rural areas (χ2=5.915, P<0.05). P < 0.05). The male-female incidence ratio was 1.63:1. The high incidence group of HFMD was children younger than 5 years old, and the number of reported cases was 730. The high incidence age group was 1-3 years old (χ2=8.455, P<0.05). The incidence rate of HFMD in all age groups ≤5 years old was higher in male than female, and the difference was statistically significant (χ2=5.174, P<0.05). A total of 388 HFMD cases were tested positive for viral nucleic acid, with a positive rate of 51.12%, In mild cases, Cox A16, EV71, and other enteroviruses accounted for 27.64%, 29.54% and 29.54%, respectively. The proportion of EV71 in severe cases was 89.47%. Conclusion HFMD is widely distributed in Guangyuan area, with obvious seasonal and population differences. The prevention and control of HFMD should be strengthened in key areas, high incidence seasons and key populations, and the vaccination of children ≤3 years old should be further promoted to prevent the outbreak of HFMD.
8.Current situation and influencing factors of HPV vaccine hesitancy among parents of primary and secondary school girls in Guangyuan

Objective To investigate HPV vaccine hesitancy and influencing factors among parents of primary and secondary schools in Guangyuan, and to provide scientific countermeasures for reducing the hesitancy rate of HPV vaccine in parents. Methods Using stratified multi-stage cluster random sampling, 1,018 parents of girls in primary and secondary schools in Guangyuan were selected for a questionnaire survey from March to July 2021. The data were analyzed by Chi-square test and multivariate logistic regression model. Results The hesitancy rate of HPV vaccine in parents was 42.95%. Multivariate logistic regression analysis showed that families with low economic income, parents who believed that HPV vaccination would have long-term side effects, and be unsafe and expensive, and parents who concerned with the effect of HPV vaccine on the prevention of cervical cancer and insufficient supply of first doses of vaccine, had positive effects on HPV vaccine hesitancy (OR = 2.02, 1.44, 3.13, 1.53, 3.76, and 2.43, respectively, P < 0.05). Conclusion HPV vaccine hesitancy rate is high among parents of primary and secondary schools in Guangyuan. It is necessary to fully promote school education and increase the publicity of HPV vaccine to improve parents' awareness of HPV vaccine. Government departments need to make an overall plan to reduce vaccine costs and ensure sufficient vaccine quantity, so as to reduce parents' hesitation to vaccinate their children with HPV vaccine.
9.Clinical effi ciency of pemetrexed in the treatment of advanced retreated non-small cell lung cancer

Background and purpose:Pemetrexed is a multitargeted anti-metabolite. Pemetrexed has been approved as the second-line treatment in non-small cell lung cancer. Our aim was to evaluate the clinical effi ciency and toxicity of pemetrexed in treatment of advanced retreated non-small cell lung cancer. Methods:17 advanced retreated NSCLC patients received pemetrexed at a dose of 500 mg/m2, the chemotherapy was repeated every 21 days on two cycles until progressive disease or untolerant adverse effects. Results:Among 17 patients, the overall response rate was 11.8%, the clinical benefi t rate was 76.5%, and the main toxicity was hematological toxicties. Conclusions: Pemetrexed is effective in treatment of advanced retreated non-small lung cancer and the toxicity can be well tolerated.
10.A STUDY OF DETERMINATION OF ASCORBIC ACID IN PLASMA BY FERRIC REDUCTION METHOD

The ascorbic acid in plasma was determined on the basis of reduction of ferric to ferrous ion and its formation of a colored complex with batho-phenanthroline. This method was highly correlated to Roe's dinitrophe-nylhydrazine method in the determination of plasma ascorbic acid of 16 normal adults and the correlation coefficient was 0.90. By animal experiment the new method may be used to discriminate different levels of plasma ascorbic acid. It is rather simple and water bath is not required. Only 0.25ml of plasma is needed for each determination. So this method is useful in assessing the ascorbic acid nutriture in tield survey.
