1.Adrenoceptor and endothelin receptors of artery isolated from colonic cancer

Objective To study the contraction of artery in colonic cancer.Methods Arteries in the cancer part and the cancer ambient part of patients with colonic cancer,the concentration-contractile curves induced by the noradrenaline(NA),sarafotoxin 6c(S6c) and endothelin-1(ET-1) in cumulative application were recorded by myograph.Results The E_(max) and pD_(2)of normal colonic artery contraction induced by NA were(97?19)% and(5.94)?0.17.The E_(max)and pD_(2) of contraction in colonic cancer artery was lower than that in cancer ambient artery(P0.05)).Conclusion The ?-adrenoceptor and ET_(A) receptor are main receptors mediating contraction in colonic cancer artery.
2.Contractile response of artery supplying colon cancer to vasoconstrictors

Aim The present study was designed to investigate the contractile responses to vasoconstrictors in artery supplying colon carcinoma.Methods The artery supplying carcinoma was obtained from patients suffering from colonic carcinoma. The artery supplying carcinoma ambient normal tissue from patients with colon carcinoma and normal artery from patients with accident were used to control. Artery were cut into about 1 mm long cylindrical segments. The concentration-contractile curves induced by 4 vasoconstrictors were recorded. Results In the normal artery, the E_max and pEC_50 of contraction induced by noradrenaline (NA) were 97%?19% and 5.94?0.17(n=5), respectively; while in the artery supplying carcinoma, the E_max and pEC_50 of contraction induced by NA were 74%?5% and 5.54?0.21(n=5). In the artery supplying cancer ambient normal tissue, the E_max and pEC_50 of contraction induced by NA were 119%?11% and 5.84?0.09, respectively. 3.Influence factors of long-term outcomes of mitral valve repair for moderate and severe mitral regurgitation due to myxomatous degeneration

Objective To explore and conclude the influence factors of long-term outcomes of mitral valve repair for moderate and severe mitral regurgitation due to myxomatous degeneration.Methods To review the in-patient data and followup outcomes of 261 patients after mitral valve repair for moderate and severe mitral regurgitation due to myxomatous degeneration from Jan 1993 to Jan 2008 in Changhai Hospital of Second Military Medical University.Results There were 7 perioperative deaths and 254 survivors who obtained satisfactory perioperative outcomes.During the follow-up,24 patients were lost and 230 patients were followed up from 36 months to 174 months (77.3 ±30.3) months and follow-up rate was 90.6%.Multivariate Cox regression shows age ≥ 60 years old,left ventricular ejection fraction < 0.50,undergoing combined coronary artery bypass grafting were the independent risk factors for long-term death after operations and left ventricular ejection fraction < 0.50,New York Heart Association functional classification Ⅲ-Ⅳ,anterior leaflet prolapse were the independent risk factors for long-term recurrent moderate or severe mitral regurgitation after operations and prosthetic ring or band annulopasty was a protective factor.Conclusion The age ≥60 years old,left ventricular ejection fraction < 0.50,undergoing combined coronary artery bypass grafting,New York Heart Association functional classification Ⅲ - Ⅳ,anterior leaflet prolapse,and prosthetic ring or band annulopasty were closely related with long-term adverse events after operations.
4.Experimental research on the characteristics of pancreatic cells regeneration in an animal model for isolated pancreatic trauma

Objective To study the relationships between tissue damage and the ability of the pancreatic cells to regenerate ,and analyze the alteration of the pancreatic cells regeneration .Methods Sixty rats were divided into two groups :impact group(the pan‐creas was injured by a BIM‐Ⅲ biotical impact machine ,40 rats) and control group(sham operated ,20 rats) .All rats were sacrificed at 6 h ,24 h ,72 h ,7 d after operation .The level of AMS ,LPS in the serum were detected by spectrophotometry ,pancreatic cells re‐generation were examined and analyzed by TUNEL staining and flow cytomertry ,and the Bcl‐2 and Bax expression were measured by Western blot .Results In the impact groups ,LPS was activated later than AMS ,and lasted persistently .The results from TUNEL stain ,flow cytometry and Western blot indicated that pancreatic trauma induces cell death and the compensatory prolifera‐tion of pancreatic cells .The characteristics of pancreatic cells regeneration in the animal model of isolated pancreatic trauma indicate that the proper remedial time is in the first 24h after the pancreatic trauma .Conclusion Detecting AMS and LPS at the same time can help us to determine the exocrine function of pancrease .
5.Comparison between MR relaxometry techniques and dual-energy X-ray absorptiometry in evaluation of osteoporotic postmenopausal rats

Objective To evaluate MR relaxometry techniques and dual-energy X-ray absorptiometry (DXA) for the diagnosis of osteoporotic diseases in rats. Methods Thirty 3-month-old female rats were randomly divided (using completely randomized grouping method) into two groups (each contained 15 rats). Animals in group A without osteoporotic castration were included as normal controls, whereas osteoporotic castration was created in each animal in group B. Three parameters (BMC, BMD, Hbmdl)was measured for both groups by DXA at two time points, one immediately before the castration and another at the 12 th week after the castration. Then animals from the control group and the osteoporotic group went through the following three diagnostic procedures using a 1.5 T MR system: (1) A fast multi echo gradient echo (MEGRE) pulse train sequence with different inter-echo intervals (1000, 500, 400, 300, 200, 100) to obtain the T_2~* value. (2) A multi-echo fast spin echo sequence to obtain the T_2map. (3) A conventional spin-echo (CSE) sequence to obtain the T_1map. The statistical difference between group A and group B was tested by t-test to analyze parameters. And, the most significant parameter for diagnosis ofosteoporotic diseases was picked out from all parameters by Fisher Sequential diseriminant analysis. At the end of experiments, animals were killed and histopathological examination was performed on the femurs of animals from both control and osteoporotic groups. Results (1) Histopathological examination confirmed the presence of osteoperosis in all animals in group B. (2) BMD was picked out from 3 DXA parameters (BMC,BMD,Hbmdl) by fisher stepwise discriminant analysis, and its discriminant rates was 87.6%. (3) All 2-sample t-test results(t=6.20, 4.79, 5.18, 5.22, 5.59, 4.37, 6.14, 5.12, 5.09, 4.99, 5.57, 4.84, 4.07, 2.98, 6.75 individually) for MR relaxometry parameters(T_2~* 1000,R_2~* 1000,T_2~* 500,R-2~* 500,T_2~* 400,R_2~* 400,T_2~* 300,R_2~* 300,T_2~* 200, R_2~* 200, T_2~* 100, R_2~* 100, T_2map, R_2map, T_1map) showed statistically significant differences between groups A and B (P=0.01 for T_2~* map, P=0.00 for all other parameters) except the R_2map(P=0.07). (4) Using fisher stepwise discriminafion method in the analysis of 14 parameters of MR relaxometry techniques and 3 parameters of dual X-ray absorptiometry(T_2~* 1000,T_2~* 500,T_2~* 400,T_2~* 300,T_2~* 200,T_2~* 100,T_2map, R_2~* 1000, R_2~* 500, R_2~* 400, R_2~* 300, R_2~* 2OO, R_2~* 100,T_1map,BMC,BMD,Hbmdl), we found that the most significant difference was from the T_2map and T_1map. Conclusions The MR relaxometry parameter-T_2map in the present study is shown to be appropriate parameter for the diagnosis of osteoperotie diseases, and stability of magnetic field plays an important role in this process. It would be the optimal method to make a diagnosis of osteoporotic diseases with both MR relaxometry and DXA technological means.
6.The feasibility of dual-source CT using low voltage scan and iterative reconstruction combine with split-bolus injection to reduce radiation dose in urography

Objective To evaluate the image quality,diagnosis accuracy and dose reduction of split-bolus CT urography (CTU) with low voltage scan and sinogram affirmed iterative reconstruction (SAFIRE).Methods A total of 80 cases of consecutive patients with confirmed or suspected urinary system disease needed CTU examination were divided into two groups (control group and test group) by using random number table.In control group,convention scan (120 kV) with one time injection was used.But low voltage scan (80 kV) with SAFIRE algorithm and split-bolus injection (SBI) was used in experiment group.The radiation dose,image quality and diagnosis accuracy were compared.Results A total of 77 cases completed CTU examination successfully in the two groups,including 39 cases in control group and 38 cases in test group.The effective dose reduced from (26.68 ± 4.07) in control group to (3.93 ± 0.85) mSv in test group (t =-33.78,P < 0.05).Subjective image quality score was (4.49 ± 0.79) in control group and (4.39 ± 1.53) in test group,with no significantly statistical difference (Z =2.71,P > 0.05).Signal-to-noise ratio (SNR) of objective image quality in test group was higher than that in control group (127.3±15.9 vs.109.6 ± 13.2,t =4.49,P<0.05).But there was no significantly statistical difference in contrast-to-noise ratio (CNR) between control group(100.8 ± 12.9)and test group (109.0 ± 14.4,P > 0.05).For diagnosis accuracy,no statistical difference were found between two groups(84.62% and 81.58%,P > 0.05).Conclusions The combination of low voltage scan with SAFIRE algorithm and split-bolus injection CTU could reduce the radiation dose significantly,but the objective image quality,CNR (except SNR) of subjective image quality and diagnosis accuracy were all unaffected obviously.

Objective To observe whether bone marrow mesenchymal stem cell transplantation can improve vasospastic rats sense and motor function.Methods Rats grouped with randomized number method as Control group,Subarachnoid hemorrhage group.Stem cell culture media group and Stem cell transplantation group.Subarachnoid hemorrhage model were made with tail artery blood twice injection,2 days after 2' nd injection.Bone marrow mesenchymal stem cell were transplanted to lateral cistern.Subarachnoid hemorrhage(SAH) group didn' t transplant stem cell.Stem cell culture media group injected DMEM media as DMEM group.Stem cell transplantation group injected 30μl Bone Marrow mesenchymal stem cell suspension,so called BMSCs group.Neurofunctional score and learning memory expression were detected with morris mazer and Neurofunctional Score Scale in each group.Results After transplantation for 7 d,functional score of Control,SAH,DMEM and Stem cell group were 3.95 ±2.51,7.20 ± 1.03,7.23 ± 1.79 and 5.81 ± 1.11 respectively.Compared with others groups,Stem cell group score was significantly decrease(P=0.017).After transplanting stem cell for 14 d,the mean spanning plate time in Control group,SAH group,DMEM group and Stem cell group were 7.38 ± 1.73,4.52 ± 0.90,5.11 ± 1.93 and 7.32 ± 2.16 respectively,SAH and DMEM group vs other 2 groups,there were clearly statistically differences (P =0.009),while between control group and stem cell group,there were no statistically differences (P =0.14).Conclusion SAH rat transplant stem cell can improve sense,motor and learning expression in certain level.
8.Islet-like cells derived from mesenchymal stem cells in Wharton's Jelly of the human umbilical cord co-cultured with rat pancreatic cells for transplantation to control type Ⅰ diabetes mellitus

BACKGROUND: Mesenchymal stem cells in Wharton's Jelly of the human umbilical cord can induce differentiation into islet-like cells.OBJECTIVE: To verify the possibility of human umbilical cord derived mesenchymal stem cells co-cultured with rat pancreatic cells differentiate into islet-like cells, and to observe the effects of transplantation of islet-like cells on blood glucose of diabetic rats.METHODS: Mesenchymal stem cells in Wharton's Jelly of the human umbilical cord was separated, induced, passaged, and co-cultured with pancreatic cells to induce differentiation into islet-like clusters. Rats were divided into the normal control, model and experimental groups. Rats in the model group were prepared for diabetic models, and those in the experimental group were transplanted islet-like cells after model preparation. RESULTS AND CONCLUSION: There were cells crawled out of cultured Wharton's Jelly of the human umbilical cord, and morphology of adhered cells turned into fusiform shape at 7 days. The isolated cells are characterized by expressing specific surface markers of mesenchymal stem cells, such as CD44, CD29, CD105, but not expressing CD34, CD45 or CD14. The cells were strongly stained by PDX-1 and human insulin at 7 and 10 days. Compared with the simple culture group, the expression of human insulin and concentration of C-peptide were obviously increased; PDX-1 and human insulin mRNA expressions were highly expressed at 7 and 10 days after induction. Compared with the model group, the streptozotocin test of rats in the experimental group was obvious decreased (P < 0.01), but extremely higher than that of the normal control group at 1 week after transplantation (P < 0.01). Brdu positive nuclei and insulin positive kytoplasms could be seen in the experimental group at 8 weeks after transplantation. The results demonstrated that, umbilical cord derived mesenchymal stem cells existed in Wharton's Jelly. The co-cultured cells promote mesenchymal stem cells differentiating into islet-like cells, which can dramatically decrease blood glucose in diabetic rats.
9.Analysis of Causes of Death in Elderly Inpatients Aged 65 Years and Over in Dongguan City

Objective To understand the epidemiological characteristics and hospitalization death disease composition of elderly hospitalized patients aged 65 years and over in Dongguan city, and provide scientific basis for the health prevention and health care work of relevant departments. Methods Through the "Guangdong province medical institutions medical records statistics management system" selected in 2015 in Dongguan city, 65 years old and over hospitalized patients with death of 959 cases, investigated the hospitalization death disease composition, analysis of its age distribution, gender distribution, the distribution of the season of death and so on. Results The top five hospitalization death disease composition were Malignant neoplasms, Ischaemic heart diseases, Cerebrovascular diseases, Pneumonia and Injury, accounting for 65. 17% of the deaths. There were significant differences in mortality among different gender groups(P<0. 05). There were significant differences in mortality among different age groups(P<0. 05). The distribution of death season is the highest in spring and summer. Conclusion To master the hospitalization death disease composition of elderly hospitalized patients, strengthen health education, reduce mortality and improve the quality of life of the elderly.
10.Accuracy of endoscopic ultrasonography for evaluating T3 esophageal squamous cell carcinoma

Objective To explore the accuracy of endoscopic ultrasonography (EUS) for evaluating T3 esophageal squamous cell carcinoma (ESCC).Methods The retrospective cross-sectional study was conducted.The clinicopathological data of 733 patients diagnosed with T3 ESCC by preoperative EUS who were admitted to the Sun Yat-sen University Cancer Center from January 2003 to December 2015 were collected.All the patients underwent radical resection of ESCC.The postoperative pathological stage as a gold standard,the accuracy,overstaged and understaged rates of clinical staging by preoperative EUS were assessed.Observation indicators:(1) comparison between clinical T staging evaluated by preoperative EUS and postoperative pathological T staging;(2) follow-up and survival situations.Follow-up using outpatient examination and telephone interview was performed to detect patients' diseases and postoperative survival up to December 30,2016.Overall survival time was from operation time to death or last effective follow-up.Measurement data with normal distribution were represented as (x)±s.Measurement data with skewed distribution were described as M (range).Count data were represented as cases and percentage.The survival curve was drawn by the Kaplan-Meier method,and survival analysis was done using the Log-rank test.Results (1) Comparison between clinical T staging evaluated by preoperative EUS and postoperative pathological T staging:all the 733 patients were confirmed as T3 ESCC by preoperative EUS.Postoperative pathological diagnosis showed that 9 patients were detected in pT1b,87 in pT2,630 in pT3 and 7 in pT4a.The accuracy,overstaged and understaged rates of preoperative EUS in evaluating T3 ESCC were 85.95%(630/733),13.10%(96/733) and 0.95%(7/733),respectively.N0,N1,N2 and N3 of postoperative pathological N stage were respectively detected in 329,247,110 and 47 patients.Twenty-seven,323 and 383 patients were in stage Ⅰ,Ⅱ and Ⅲ of TNM stage,respectively.The high-,moderate-and lowdifferentiated tumors were respectively detected in 125,403 and 205 patients.(2) Follow-up and survival situations:among 733 patients,639 were followed up for 1.0-153.0 months,with a median time of 29.0 months.The median survival time,1-,3-,5-year overall survival rates were 53.0 months (range,37.7-68.3 months),85.3%,58.1% and 48.2% in 733 patients,respectively.The 5-year overall survival rate was 75.2% in 9 patients with pT1b,63.0% in 87 patients with pT2,46.3% in 630 patients with pT3 and 0 in 7 patients with pT4a,respectively,with a statistically significant difference (x2=24.089,P<0.05).Conclusion There is a higher accuracy of EUS for evaluating T3 ESCC,however,the stage migration should be noted.
