1.Effects of Kangguzhe Formula on Promoting Union of Experimental Rat Fracture

Objective: To investigate the effects of Kangguzhe Formula(KGZF) on promoting union of experimental rat fracture.Methods: The experimental rat fracture models were reproduced by cannon born injure,continually treated with KGZF 21d by oral administration.The X-ray,pathological histology and anti-fracture intension of fracture union tissue were detected to investigate the effects of KGZF.Results: KGZF can significantly improve the osteotylus rebuild,trabeculation of bone and concentrated osteoblasts of fracture tissue;X-ray detection also showed that it can increase the grade of X-ray and anti-fracture intension of fracture union tissue;Compared with model group,it had significant difference.Conclusion: KGZF can obviously improve the union of experimental rat fracture.
2.Establishment of transgenic mice tissue-specifically expressing human decay accelerating factor on the vascular endothelium

Objective To construct transgenic mice tissue-specifically expressing human decay accelerating factor (DAF) on the vascular endothelium for xenotransplantation. Methods Transgenic mice expressing hDAF were generated by microinjection of a hDAF-minigene under the control of the human intercellular adhesion molecule-2 (ICAM-2) promoter. PCR analysis and Southern blot analysis of genomic DNA were used to examine the presence of the transgene in the genome of the offspring, and the expression was detected by RT-PCR and flow cytometry respectively. Immunohistochemical assay was performed to detect the distribution of hDAF on the tissues from these transgenic mice. An ex vivo perfusion model was used to compare hearts from these hDAF mice with wild-type hearts.Results After microinjection of gene, 21 of 133 mice born were shown to be transgenic. Human DAF gene was expressed on the surface of leucocytes in 8 of the 21 hDAF-integrated mice. Expression levels of 8 founders ranged from 70 % to 95 % of that on human leucocytes. Human DAF were strongly expressed on the vascular endothelium of heart, kidney and liver, with little or no positive staining observed on non-endothelial cells. These endothelial-specific hDAF hearts displayed prolonged survival compared to wild-type hearts during perfusion with 20 % human plasma and maintained approximately 20 % maximum work. Conclusions The introduced hDAF gene has been specifically expressed on the vascular endothelium of transgenic mice. The hDAF tissue-specifically expressed on vascular endothelium could enhance xenograft survival.
3.Testing Method for protection capability of skin protection material against nitro oxidizer

The experiment method of "Liquid-vapor" permeation for nitro oxidizer's skin protective materials by the indicator of pH test paper is developed,and the method for cankerous testing and evalution roles are confirmed too.The result of the experiment shows that butyl rubber and halogenated butyl rubber have better protective performances for "liquid-vapor" permeation of nitro oxidizer,while fluorine rubber,polyethylene and polyterafluoroethylene have better cankerous protection capabilities.
4.Method of protection permeation testing on skin protection material for liquid rocket propellant

Objective To explore the method of permeability for skin protective materials of rocket propellant based on the design and make of sealing container. Methods The propellant mass permeated per minute and per square centimeter was calculated by testing the mass through the sample at the fixed time in the sealing container. Results The method of permeability for skin protective materials against rocket propellant was developed by the mass variation of propellant in the sealing container. The limits of 100?g /(cm2?min) was raised to assess if the material had the protection ability. In the meantime, the reliability of material protection behavior was improved. Conclusion The science of the method developed is confirmed by comparing the permeation performance of protection materials at home and abroad, and the method can be the basis to assess the ability of protection materials.
5.Synthesis of A Novel Fluorescence Switch Conjugated Polymers and Its Discrimination Ability Toward S-Adenosylmethionine

A novel fluorescent switch was constructed based on poly 3-{[ 1-( 2-hydrazino-2-oxoethyl ) piperidin-4-ylidene] methyl}thiophene ( PMTH ) in the presence of Pb2﹢ or S-adenosylmethionine ( SAM ) . The switch turned off in the presence of Pb2﹢owing to complex effect, and the fluorescence intensity of PMTH solution was efficiently quenched by Pb2﹢ ions. Upon adding SAM to the Pb2﹢-PMTH solution, which was stronger Pb2﹢ chelators, it could form more stable SAM-Pb2﹢, the Pb2﹢ ion was displaced from PMTH and the fluorescence of PMTH was recovered. By triggering the turn-on signal of PMTH, a new discrimination ability toward SAM method for the determination of SAM was established. The effects of the interaction on the fluorescence spectral characteristics of these Pb2﹢-PMTH and Pb2﹢-SAM complexes in H2 O-CH3 CH2 OH(4:1, V/V) solution, suitable reaction conditions and influencing factors were investigated. This method offered good sensitivity and selectivity for detecting SAM in the presence of amino acids and metal ions. Under optimum conditions, the concentration of SAM in the range from 1. 0 × 10-8-2. 0 × 10-6 mol/L exhibited a linear relationship with the relative fluorescence intensity. The regression equation was △I = 68. 51﹢72. 32c (μmol/L), the correlation coefficient r=0. 9982. The limit of detection was 8. 72×10-9 mol/L. The system was successfully applied for detecting SAM in human serum, urine and injection samples.
6.Diagnosis and management of Wunderlich syndrome

Objective To investigate the causes,diagnosis and management of Wunderlich syndrome. Methods A total of 19 cases (15 men and 4 women) of Wunderlich syndrome were reported.Their average age was 47 years (range,11-75 years).Imaging examinations (B-ultrasound,IVU,CT,arteriography,MRI) together with laboratory testing and history supported the etiologic diagnosis of tumorous disease (n=10),non-tumorous disease (n=7) and unknown causes (n=2).Surgical management was performed in 9 cases of kidney tumor and 1 of ureter stone with obstructive hydronephrosis.Conservative treatment was adopted in the rest 9 cases.In 2 of the conservatively treated cases,the bleeding artery was embolized because of persistent bleeding. Results In the 9 tumor cases undergoing surgery,pathology confirmed renal cell carcinoma in 7,malignant fibrous histocytoma in 1 and transitional cell carcinoma of the renal pelvis in 1.These 9 cases all recovered after operation and experienced follow-up for 6 months to 3 years.Of them,5 cases died of tumor recurrence and metastasis;3 survived disease-free for 9 months to 2 years;1 had liver metastasis 1 year after operation and survived with tumor for 11 months.One case of ureter stone got significant improvement.The 9 cases undergoing conservative treatment had satisfactory results. Conclusions The main cause of Wunderlich syndrome is renal tumor.The conservative approach appears to be the most acceptable option for the patients with Wunderlich syndrome who have no malignant tumorous cause.
7.Influence of Gusongbao Capsule on Osteoporosis in Ovariectomized Rats

Objective To observe the therapeutic effect of Gusongbao capsule for osteoporosis. Methods Experimental osteoporosis rat models were established by extirpation of bilateral ovaries of female rats. Bone morphological indexes, blood biochemical indexes, and daily output of calcium and phosphorus in urine in the Gusongbao treatment group and control group were determined. Results Compared with the control, the average width of bone trabecula, relative bone volume, average thickness of bone cortex, bone calcium, and the average gray degree of femoral head and femoral shaft were high, and the serum calcitonin (CT) and blood calcium increased markedly in the treatment group. High-dose and moderate-dose Gusongbao increased number of osteocytes in per unit area of bone trabecula, decreased serum alkaline phosphatase (ALP) level and reduced the daily output of urine calcium. Conclusion Gusongbao capsule can obviously inhibit the excessive bone transformation and promote the re-construction of bone and exerts an antagonistic effect on osteoporosis in ovariectomized rats.
8.Influence of Gusongbao Capsule on the Bone Morphological and Blood Biochemical Indexes in Rats with Osteoporosis Induced by Vitamin A Acid

Objective To observe the anti-osteoporosis effect of Gusongbao capsule (GC).Methods Osteoporosis rat models were induced by gastric infusion of vitamin A acid at a dose of 70 mg/kg for 14 days. The bone morphological and blood biochemical indexes and the amount of calcium and phosphorus in urine excreted every day were determined in both treatment group (treated with Gusongbao) and model control group.Results In the treatment group,the average width of bone trabecula,relative bone volume,average thickness of bone cortex,bone calcium,cell number in per unit area of bone trabecula,and the average gray degree of caput femoris and femoral shaft were higher than those in the moderate group. 9.Research on Polyamidoamine Dendrimers Mediated Foreign Gene for Xenotransplantation Transfected Pig Sperm Cells

Objective:To investigate the effect of the new nanomaterials,polyamidoamine dendrimers PAMAM-D,mediated foreign gene human decay accelerating factor(hDAF) for xenotransplantation transfected pig sperm cells.Methods:PAMAM-D/hDAF cDNA compounds were made.The compounds were divided into 0.2 μg,0.4μg,0.6μg,0.8 μg and 1.0μg groups (each group adding corresponding dose of PAMAM-D in accordance with the N/P ratio 10:1,20:1,40:1),then were digested by restriction enzymes.The compounds were incubated with washed pig sperm cells.Then the transfection efficiency was detected by in situ hybridization in the different groups.Results:The PAMAM-D molecule can prevent electrophoretic migration of DNA in the compound.After digested the compounds by restriction enzymes,DNA can not be degraded.The transfection efficiency was different in different groups.Among the total,the efficiency was higher in both groups of 0.4 μg and 0.6μg than that of others.The top was the group of 0.4 μg linear plasmid plus PAMAM-D when the N/P ratio was 20:1(47.5%±O.2%,167% vs control group).Conclusion:PAMAM-D can improve the efficiency of exogenous gene transfeeted pig sperm cells,which can reinforce the stable binding of exogenous DNA to sperm cells.
10.Clinical comparison of modified postauricular transverse incision and traditional vertical incision for microvascular decompression in the treatment of hemifacial spasm

Objective:To compare the effect of modified postauricular transverse incision and traditional vertical incision for microvascular decompression in the treatment of hemifacial spasm.Methods:Prospective study method was used. A total of 116 patients with hemifacial spasm in Handan Central Hospital from January 1, 2019 to January 1, 2020 were selected, and divided into two groups according to the admission order. Both groups underwent microvascular decompression; control group (57 cases) received traditional vertical incision, while treatment group (59 cases) received modified postauricular transverse incision. The brainstem auditory evoked potential (BAEP), pain degree, surgical indicators, facial aesthetic satisfaction and complications were compared between two groups.Results:After treatment, the BAEP of latency, wave interval and wave amplitude in the two groups increased compared with that before treatment, and the BAEP of latency, wave interval and wave amplitude in the treatment group were higher than those in the control group: (1.89 ± 0.15) ms vs. (1.62 ± 0.21) ms, (7.89 ± 0.15) ms vs. (6.25 ± 0.41) ms, (1.79 ± 0.19) ms vs. (1.54 ± 0.11) ms ( P<0.05). After treatment, the visual analogue score (VAS) of patients in the two groups decreased compared with that before treatment, and the VAS of patients in the treatment group was lower than that in the control group: (1.15 ± 0.27) points vs. (2.18 ± 0.24) points ( P<0.05). The operation time, intraoperative bleeding volume and postoperative scar length of patients in the treatment group were less than those in the control group: (60.41 ± 3.81) h vs. (76.87 ± 3.87) h, (30.18 ± 4.19) ml vs. (56.87 ± 4.15) ml and (4.18 ± 1.07) cm vs. (6.87 ± 1.05) cm ( P<0.05). The satisfaction rate of patients in the treatment group was higher than that in the control group: 91.53% (54/59) vs. 71.93% (41/57) ( P<0.05). The complication rate of patients in the treatment group was lower than that in the control group: 5.08% (3/59) vs. 21.05% (12/57) ( P<0.05). Conclusions:Compared with traditional vertical incision, the modified transverse incision for microvascular decompression in the treatment of hemifacial spasm can reduce intraoperative blood loss and postoperative scar area, enhance brainstem auditory evoked potential, and improve facial aesthetics, which is worthy of recommendation.
