OBJECTIVE: To study the anti-inflammatory effect of the aqueous extracts of Sophorae tonkinensison. METHODS: The auricle edema was induced by xylene in the mice to measure the swelling of ear; the experimental peritonitis of mice and scytitis of rats were induced to measure the absorbability of the effusion. The ear swelling and capillary permeab-ility in drug-treated group was compared with those in the normal control group and hydrocortisone-intervented model group.RESULTS: Mice's swelling induced by xylene was lessened by aqueous extracts of Sophorae tonkinensison(high or med-ium dose), which also showed remarkable inhibitory effects on the experimental peritonitis of mice and scytitis of rats (P
2.Optimization of Semi-bionic Extraction Technology of Forsythia Suspensa by Uniform Design

Objective:To optimize semi-bionic extraction(SBE)of Forsythia suspensa. Methods: The best conditions of semi-bionic extraction of Forsythia suspense were screened by uniform design with the total content of forsythoside A,and forsythin and dried extract weight as the indices. Results:The results of the uniform design were analyzed by DPS data, and combined with the industrial production conditions, the optimal extraction technology was as follows:the pH value of water in the three-time extraction was 2. 0,7. 0 and 10. 0 with the extraction time of 1. 0h,0. 5h and 0. 5h, respectively. Conclusion: The technology provides a theoretical basis for the extraction optimization of Forsythia suspensa.
3.Fingerprint Chromatogram of Forsythia Suspensa Dispensing Granule by HPLC

Objective To establish the fingerprint of Forsythia suspensa dispensing granule by HPLC. Methods The total of 10 samples of Forsythia suspensa were analyzed by HPLC on a Waters SunFire C18 column(4. 6 mm×250 mm,5 μm) with mobile phase of A:acetonitrile and mobile phase B:0. 4% acetic acid solution as gradient elution, with flow rate at 1. 0 mL·min-1 , wavelength at 277 nm and column temperature at 30 ℃. Results There were 13 characteristic common peaks in the Forsythia suspensa dispensing granule, in which two peaks identified as forsythoside A and forsythin, the similarities were more than 0. 999. Conclusion The HPLC fingerprint of Forsythia suspensa dispensing granule is stable, and is easy to manipulate, which can provide more information for the quality control of Forsythia suspensa dispensing granule.
4.Effects of glutamine-enriched early enteral nutrition on intestinal mucosal barrier injury after liver transplantation in rats

Objective To observe the effect of glutamine-enriched early enteral nutrition on intestinal mucosal barrier injury after orthotopic liver transplantation in rats.Method Male Wistar rats,the recipients were randomly divided into three groups:control group (control group,n =10),orthotopic liver transplantation group (OLT group,n =30) and glutamine-enriched early enteral nutrition group (EEN group,n =30).Only dissecting hepatoduodenal ligament was performed in control group,and OLT was performed from Wistar to Wistar rats by modified two-cuff method in OLT group and EEN group.For EEN group,recipients were supplied with Nutrison Fiber (125 ml/kg every day) plus Gln (0.3 g/kg every day) for 3 days before and 3 h after surgery by gastric perfusion.For OLT group,the same volume of normal saline was administered instead of the Nutrison in the same time.No special treatment was given in control group.The levels of plasma endotoxin,D-lactic acid,and TNF-α were determined at different time points in the three groups.The ultrastructural changes of ileal mucosa were observed under the transmission electron microscopy.At the same time,the remaining 5 rats per group were used for observing the lifetime.Result As compared with control group at 12,24 and 72 h,the levels of plasma endotoxin,D-lactic acid,and TNF-α were significantly increased in OLT group and EEN group (P<0.01),and as compared with OLT group,the above-mentioned indexes were obviously decreased in EEN group at 24 and 72 h (P< 0.01).The ileal mucosal epithelial clearance in control was normal,and microvilli arranged neatly under the electron microscope.the ultrastructure damage in OLT group was more serious than in EEN group at 12,24,and 72 h.As compared with control group at 12,24,and 72 h,the expression levels of TNF-α mRNA were significantly increased in OLT group and EEN group (P<0.01),and as compared with OLT group,the expression levels of TNF-α mRNA were obviously decreased in EEN group (P<0.01).The survival time in cntrol group was significantly longer than in OLT group and EEN group (P<0.05),and as compared with OLT group,the survival time in EEN group was obviously extended (P<0.05).Conclusion OLT can lead to the damage of intestinal mucosal barrier,and glutamine-enriched early enteral nutrition is a potent protection against intestinal mucosal barrier injury and prolongs the survival time of rats after OLT.
5.Clinical efficacy of captopril combined with amlodipine in hypertension patients of the influence of high noise

Objective To observed the clinical efficacy of captopril combined with amlodipine in hypertension patients of the influence of high noise.Methods A total of 150 hypertension patients who worked in > 110 dB noise were divided into treatment group 1 (75 cases) and treatment group 2 (75 cases) by random digits table.Patients simply took enalapril in treatment group 1,patients took enalapril combined with amlodipine in treatment group 2.Selected 75 cases common hypertension patients were control group,they took enalapril combined with amlodipine.A year later,compared with three groups of plasma renin activity (PRA) and clinical efficacy.Results The PRA and efficient rate had no significant difference among three groups in half ayear (P >0.05).A year later,the PRA and efficient rate in treatment group 2 and treatment group 1 had obvious difference [(1.89 ± 0.37) ng/ (ml· h) vs.(2.40 ± 0.38) ng/ (ml· h),90.67 % (68/75) vs.72.00 % (54/75)] (P < 0.05 or < 0.01).But compared with control group no significant difference (P >0.05).Conclusion The clinical efficacy of captopril combined with amlodipine in hypertension patients is more apparent.
6.Optimization of Semi-bionic Extraction Technology for Rhubard by Uniform Design

Objective: To optimize semi-bionic extraction(SBE)of rhubard. Methods: The best extraction conditions of semi-bionic extraction for rhubard were optimized by uniform design with the total content of loeemodin, rhein, emodin, chrysophanol, and anthraquinone and the dried extract weight as the indices. Results:According to the optimization and the industrial production condi-tion, the pH values of water in three times of extraction were 2. 0,6. 5 and 9. 0,and the extraction time was 2, 1, 1 h, respectively. Conclusion:This method provides a theoretical basis for the optimization of rhubard extraction.
7.Preventive and Treatment Effects of Xiaochaihu Decoction on Irinotecan-induced Bloodystool in Model Mice with Delayed Diarrhea

OBJECTIVE:To investigate the prevention and treatment effect of Xiaochaihu decoction on irinotecan(CPT-11)-in-duced bloodystool in model mice with delayed diarrhea. METHODS:40 mice were randomly divided into normal control group (equal volume of normal saline),model control group(equal volume of normal saline),Xiaochaihu decoction group(1500 mg/kg, calculated by crude drug)and loperamide group(positive control,0.5333 mg/kg),ig,once a day,for 17 days. Except for normal control group,other groups were intraperitoneally injected CPT-11 to induce delayed diarrhea in 4th-10th day. Body mass changes, bloodystool rate of mice were determined,and pathological changes in large intestine and small intestine were observed. RE-SULTS:After 17 days of administration,compared with normal control group,the body mass in modeling groups was obviously declined (P<0.05),while body mass in Xiaochaihu decoction group was higher than model control group (P<0.05). In the 11th-17th day,bloodystool rate in Xiaochaihu decoction group and loperamide group were significantly lower than model control group(P<0.01),and bloodystool rate in Xiaochaihu decoction group was lower than loperamide group(P<0.05). Pathological re-sults showed,obvious diffuse necrosis and inflammatory cell infiltration were observed in mucosal epithelial cells of large intestine and small intestine in model control group,and the intestinal mucosal tissue in administration groups tended to be normal. CON-CLUSIONS:Xiaochaihu decoction can effectively reduce bloodystool rate,improve intestinal mucosa injury,and has prevention and treatment effect on CPT-11-induced bloodystool in mice,with superior effect to loperamide.

Objective:To study the protective effect of polysaccharide from Rose roxburghii Tratt (RRTP-1) on neural stem cells damaged by glutamic acid (Glu). Method: RRTP-1 was obtained by ion exchange column chromatography and gel chromatography. The neural stem cells were cultured from the striata of fetal mice and damaged by glutamic acid to simulate hypoxic-ischemic brain injury. The cultures were divided into control group, glutamic acid group and test groups in which different doses of RRTP-1 were added with glutamic acid. The death rate of neural stem cells damaged by Glu and LDH leakage were observed. Results:The neural stem cells damage of test groups were alleviated,and the death rate of neural stem cells and LDH leakage of high dose test group were significantly lower than glutamic acid group. Conclusion:RRTP-1 has remarkable protective effect on neural stem cells damage.
