1.Association between cystathionine-beta-synthase gene mutation and ischemic cerebrovascular disease in youths

BACKGROUND: Homocysteine is associated with the attack of cerebral infarction, and cystathionine-beta-synthase (CBS) is a key enzyme of the metabolism of homocysteine, but it is still not clear whether its gene mutation is the potential candidate genic factor of cerebral infarction.OBJECTIVE: To observe the correlation of CBS base mutation at T833C and G919 sites with the attack of ischemic stroke in youths from the angle of genic mutation.DESIGN: A patient-control analysis.SETTING: Department of Neurology, China-Japan Friendship Hospital of Jilin University.PARTICIPANTS: Patient group (n=100): Young inpatients with cerebral infarction ≤ 45 years old in the China-Japan Friendship Hospital of Jilin University between April 2003 and December 2004, admitted within 2 days after attack. Control group (n=100): Normal young physical examinees in this hospital at the same period.METHODS: The levels of fasting and loaded homocysteine in plasma were detected with high performance liquid chromatography (HPLC), CBS genes at T833C and G919A sites with polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) and amplification refractory mutation system in all the subjects.RESULTS: All of the 200 subjects entered the analysis of results. ① Detection of CBS genes at T833C and G919A sites: The distribution of genotype, frequency of homozygote and that of allele had no significant differences between the patient group and control group (P > 0.05). ② Concentration of homocysteine in plasma: There were significant differences among the genotypes at G919A and T833C sites (P < 0.001). The results of LSD-t test of the mutation at the two sites showed that there were significant differences between homozygote and heterozygote, homozygote and wild type,as well as C heterozygote and wild type (P < 0.05).CONCLUSION: ① The CBS gene mutations at both T833C and G919A sites can lead to the obvious increase of the concentration of homocysteine in plasma. ② The CBS gene mutations at T833C and G919A sites had no direct association with the attack of cerebrovascular disease in youths.
2.Study on HPLC fingerprint of Pollen Typhae

bjective:To study and establish the fingerprint of Pollen Typhae.Methods:Hypersil BDS C18(5?m,4.6mm?250mm) chromatographic column mobile phase acetonitrile-0.1 % phosphoric acid solution(10:90)with fiow rate of 1.0 ml/min and UV detector at 254 nm.ResultsTaking Isorhamnetin-3-O-neohesperidoside as the reference peak,11 common peaks were selected as the fingerprint peaks of Pollen Typhae,Technology investigation indicated that the analytical method this study established has desirable precision,reproducibility,and stability.The similarity of Pollen Typhae fingerprints from different batches is better.Conclusion HPLC fingerprint analysis can be a method for quality control of medical material of Pollen Typhae.
3.THE VALUE OF SERUM CA-50 DETERMINATION IN THE DIAGNOSIS OF PANCREATIC CARCINOMA

In this communication, the values of serum CA-50 in a series of patients with benign and malignant GI diseases are reported. It was found that CA-50 was positive in 86.7% of 30 pancreatic cancer patients with an average level of 427.53U/ml, which was statistically different from that found in non-malignant control group. In this series serum CA-50 determination helped to confirm the diagnosis of pancreatic cancer in 6 cases, in which B-mode ultrasono-graphy failed to make a definite diagnosis, and in another 3 cases CT scanning was unable to disclose the tumor. The combination of CA-50, B-mode ultrasonography, and CT made definite diagnosis in 100% of all 30 pancreatic cancer patients. The author discussed the significance of CA-50 determination in the diagnosis and differential diagnosis of pancreatic cancer.
4.CT findings of secondary pulmonary lymphoma:report of 20 cases

Objective To analyze the features of CT images of secondary pulmonary lymphoma. Methods The dats of totally 20 patients with secondary pulmonary lymphoma which were proven through lung puncture pathology or clinical treatment were retrospectively analyzed. There were 19 non-Hodgkin lymphoma cases and 1 Hodgkin lymphoma case. All cases received thorax CT scanning, and 13 of them had enhanced CT scanning. Results There were many patterns in our CT findings for secondary pulmonary lymphoma, mixed pattern in 9 cases, mass (nodule) in 4 cases, pneumonic-alveolar in 3 cases, bronchovascular-lymphatic in 3 cases, and miliary in 1 case. There were 13 cases accompanying with mediastinal lymphadenovarix, 5 cases with pleura invasion, 11 cases with pleural effusion, and 2 cases with pericardium involvement. Mild enrichment was found in 7 cases and obviously enrichment was seen in 1 case. Conclusion Diversity is the characteristic feature of secondary pulmonary lymphoma. Combined with other thorax findings, CT scaning can make a definite diagnosis.
5.Anti-depressant-elTect of Xiaoyaofang on mice with behavioral despair

Objective To study the anti-depressant effect of Xiaoyaofang on mice with behavioral despair. Method The mice were given the forced swimming test and tail suspension test to observe the anti-depressant effect of Xiaoyaofang. Result The high dosage of Xi-aoyaofang significantly shortened fast time in the tail suspension test of mice(P < 0.05). And Xiaoyaofang showed the trend of shortening the needed time in the forced swimming test. Conclusion Xiaoyaofang have an anti-depressant effect on mice with behavioral despair.
6.Bone biomaterial composited with human bone marrow mesenchymal stem cells for large costal defects

BACKGROUND:Tissue-engineered biomaterials have the similar structure and function with autologous tissues. OBJECTIVE:To explore the osteoinduction of the bone biomaterial composited with rat bone marrow mesenchymal stem cel s in the treatment of large costal defects. METHODS:Forty Wistar rats were enrol ed used for the preparation of right large costal defect models, and then randomized into two groups, fol owed by the implantation of calcium chloride-sodium alginate gel (control group) or chloride-sodium alginate-bone marrow mesenchymal stem cel s (experimental group). At 2, 4 and 8 weeks after implantation, chest X-ray radiograph and histological examination of the defect region were conducted. RESULTS AND CONCLUSION:X-ray showed that in the experimental group, the defect area had no significant changes at the 2nd week after implantation until the formation of few bones at the 4th week;and at the 8th week, both ends of the defect region gradual y connected, and newly formed bones were ful of the defect. In contrast, the defect region in the control group showed no obvious bone healing, and both ends of the defect closed and osteosclerosis occurred. In the experimental group, there were a smal amount of fibrous tissues and numerous inflammatory cel s infiltratied in the material compartment, and no connection occured between the material and broken ends;there were numerous inflammatory cel s but no bone tissues in the control group at the 2nd week. At the 4th week, the scaffold degraded gradual y and abundant bone tissues were seen in the experimental group;the scaffold degraded little, and bone tissues aggregatied at the both defect ends in the control group. Up to the 8th week, the two kinds of scaffolds degraded mostly. A large number of bone tissues and trabeculae formed and the both defect ends were connected with the newly formed bones in the experimental groups, while in the control group, osteosclerosis appeared at both ends of the defect. To conclude, the bone biomaterial composited with rat bone marrow mesenchymal stem cel s promotes the repair of large costal defects.
7.Clinical and CT features of pancreatic metastasis

Objective To analyze the clinical and CT characteristics of pancreatic metastasis to improve the correct diagnosis rate.Methods The clinical and CT data of 14 patients with pancreatic metastasis were reviewed retrospectively.Results Among the 14 patients,the primary malignancy was lung cancer in 6 cases,renal carcinoma in 3 cases,colon carcinoma in 2 cases,laryngeal cancer in 1 case,gastric cancer in 1 case and ovarian carcinoma in 1 case.Pancreatic metastases from renal carcinomas often developed a long period after initial nephrectomy and survived for a relatively long time.Abdominal CT plain scan and enhanced scan was performed in all cases.Four cases were of solitary type,9 cases were of multiple types and 1 was of diffuse type.Totally 28 lesions were found,which were usually located in the neck and body of the pancreas,the diameter of 28 pancreatic lesions was ranging from 0.5 cm to 11 cm,with a mean diameter of 2.3 cm.On CT plain scan,there were 19 hypodense lesions,7 slightly hyperdense lesions and 2 isodense lesions,5 necrotic lesions.On enhanced CT,the lesions showed moderate enhancement in 7 cases,circular enhancement in 6 cases,moderate and circular enhancement in 6 lesions,obvious enhancement in 8 lesions and no enhancement in 1 lesion.All patients had extra-pancreatic metastases,and 2 cases had peripheral vascular invasion.Conclusions Pancreatic metastases are rare,often accompanied with multiple lesions and extra-pancreatic metastases.The CT features of pancreatic metastases from renal carcinomas are slightly hyperdense and obvious enhancement; in other metastases,there are hypodense,morderate and/or circular enhancement lesions.Accurate diagnosis can be achieved by knowledge of patients' medical history.
8.Association of killer immunoglobulin-like receptors with leukemia

Objective To study the correlation between the genes of killer immunoglobulin (Ig)-like receptors (KIR) in natural killer cells and leukemia. MethodsPrimers of 16 KIR genes, including KIR2DL1-5, KIR3DL1-3, KIR2DS1-5, KIR3DS1 and 2 pseudogenes 2DP1 and 3DP1, were designed and synthesized. A polymerase chain reaction with sequence-specific primers (PCR-SSP) was developed to detect the mRNA expressions of the 16 genes. Totally 46 healthy individuals and 46 leukemic patients (15 cases of acute myelocytic leukemia, 13 cases of chronic myelocytic leukemia and 18 cases of acute lymphocytic leukemia) were detected and the frequencies of the KIR genes were analyzed. ResultsThe frequency of 2DL4, 3DL2, 3DL3 and 2DP1 in healthy and leukemic patients were 100%, and were detected in all patients and healthy individuals. The other KIR genes had similar frequencies except those of 2DL2 and 2DS2, which were 47.8% of the healthy individuals and 89.1% in leukemic patients. There was a significant difference between the 2 groups.ConclusionThere may be an association between pathogenesis of leukemia and 2DL2 and 2DS2 genes.
9.HPLC fingerprint of ultramicro decoction piece of Fructus gardeniae

AIM: To determine the HPLC fingerprint of ultramicro decoction piece of Fructus gardeniae. METHODS: Chromatographic conditions were as follows: column: Hypersil BDS C_ 18 (4.6 mm?200 mm,5 ?m), mobile phase: acetonitrile-water (10∶90), column tempemture: 25 ℃,flow rate: 1 mL?min -1 , wavelength: 238 nm, and inject volume: 10 ?L. RESULTS: The fingerprint of ultramicro Fructus gardeniae was established and 7 common peaks in the fingerprint were indicated. CONCLUSION: It can nearly completely reflect intrinsic quality of ultramicro decoction piece of Fructus gardenian.
10.Effects of lactate peritoneal dialysis solution on apoptosis of human peritoneal mesothelial cells,expressions of bcl-2,bax and activity of caspase-3

Objective To study the effects of lactate peritoneal dialysis solution(L-PDS) with different concentrations on apoptosis of human peritoneal mesothelial cells(HPMC),the expressions of bcl-2,bax and activity of caspase-3.Methods HPMC were separated using enzyme digestion and cultivated stably in vitro.After HPMC were co-cultivated with different concentrations(1.50%,2.50%,4.25%) L-PDS,flow cytometry was used to test the apoptosis of HPMC,RT-PCR was used to observe the expressions of bcl-2 and bax,fluorometric method was used to detect the activity of caspase-3.Results Compared with control group, L-PDS could induce the apoptosis of HPMC,especially in high concentration.With the increasing of L-PDS concentration,the expression of bcl-2 mRNA decreased,the expression of bax mRNA increased,the activity of caspase3 raised.There were significant differences of the indexes mentioned above between 4.25%,2.50% L-PDS groups and control group(P0.05).Conclusion L-PDS could induce HPMC apoptosis,which may be executed by alternating of the expressions of bcl-2,bax and activating of caspase3.
