1.The differences of platelet PAC-1,CD62P positive rates and serum platelet factor 4 levels in patients with large artery atherosclerotic stroke and cerebral small vessel disease

Objective To investigate the differences of platelet membrane glycoprotein PAC-1 and CD62P expression in patients with cerebral small vessel disease and its subtype and large artery atherosclerotic stroke and to compare the traditional platelet activation markers and inflammatory chemokine platelet factor 4(PF4).Methods Peripheral blood platelet PAC-1,CD62P positive rates and serum PF4 concentration in 30 patients with large artery atherosclerotic stroke,45 patients with cerebral small vessel disease and 30 controls were detected using flow cytometry and enzyme-linked immunosorbent assay.The differences between the groups were compared.Results The PAC-1,CD62P positive rates and serum PF4 concentration in the large artery atherosclerotic stroke group were 63.21% ± 9.78%,55.91% ± 8.17%,and 30.55 ± 15.56 ng/ml,respectively.They were significantly higher than 40.65% ± 17.42%,36.49% ± 14.60%,and 12.59 ±5.57 ng/mlin the cerebral small vessel disease group(all P=0.000),and the latter was still higher than 13.55% ± 3.14%,9.00% ± 2.32%,and 4.95 ±2.82 ng/ml in the control group(all P = 0.000).There were significant differences in all the subtypes groups of cerebral small vessel disease between the PAC-1,CD62P positive rates and serum PF4 concentration.The leukoaraiosis with lacunar infarction group(n = 15;47.72% ±15.52%,43.75% ± 12.54%,and 13.96 ± 5.23 ng/ml)and the simple lacunar infarction group (n = 15;49.87% ± 14.65%,43.98% ± 10.55%,and 14.41 ± 5.53 ng/ml)was significantly higher than the simple lacmar infarction group(n = 15;24.44% ± 8.45%,21.74% ± 7.19%,and 9.40 ±4.99 ng/ml)(P =0.000,0.000,and 0.013,respectively).There was no significant difference between the forrner 2 groups(P = 0.658,0.952,and 0.858,respectively).The peripheral blood platelet PAC-1 positive rate had significant correlation with CD62P positive rate in patients with ischemic cerebral disease(r= 0.964,P= 0.000),and the serum PF4 concentration showed linier correlation with the PAC-1(r =0.846,P =0.000)and CD62P(r =0.857,P =0.000)positive rates.Conelusions The platelet membrane glycoprotein PAC-1 and CD62P expression showed linear correlation,and they were consistent with the changes of PF4 concentration.This sugested that platelet activation and its mediated inflammatory mechanisms played an important pathophysiological role in the processes of atherosclerosis and thrombosis.This mechanism had significant difference between the different lesion types.
2.In silico cloning, expression and bioinformatics analysis of NtODB from common tobacco.

It has been reported that ODB genes play an important role in homologous recombination-directed DNA repair, suggesting their potential applications in plant breeding. To analyze the expression characteristics of tobacco NtODB gene, the cDNA sequence of NtODB was obtained using in silico cloning technique. The physicochemical properties, signal peptide, and advanced structures of the predicted protein were analyzed using bioinformatics tools. The results showed that the NtODB gene has a 579-bp open reading frame which encodes a protein with 192 amino acid residues. The protein NtODB is predicted to be alkaline and hydrophilic. Real-time quantitative PCR showed that NtODB was constitutively expressed in different tissues. Subcellular localization showed that NtODB was mainly expressed in cell membrane and chloroplast. These results may help us to better understand and elucidate the roles of ODB genes in the homologous recombination-directed DNA repair.

3.RISK FACTORS ASSOCIATED WITH THE DEVELOPMENT OF INTESTINAL BACTERIAL TRANSLOCATION IN THERMALLY INJURED RATS

The current study was conducted to analyze the risk factors associated with the development of intestinal bacterial translocation in rats following major burns. Wistar rats were subjected to 30% total body surface area skin full thickness thermal injury, and samples were collected on postburn days 1, 3, and 5. Univariate analysis and multivariate logistic regression analysis were performed to evaluate the correlations among intestinal mucosal barrier, biological barrier and immunologic barrier associated with gut derived bacterial translocation. The results showed that the total bacterial translocation rate was 53 9% (69 in 128) after burns. According to univariate analysis, the levels of plasma endotoxin and interleukin 6, the counts of mucosal fungi and E. coli, and the score of intestinal lesion markedly increased in animals with bacterial translocation compared with those without ( P =0 000~0 005). But intestinal mucus sIgA and the mucosal bifidobacteria count were significantly reduced in animals with bacterial translocation compared with those without ( P =0 000). Multivariate logistic regression revealed that intestinal lesion scores (odds ratio 45 52, 95% confidence interval 5 25~394 80) and the counts of mucosal bifidobacteria (OR 0 039, 95% CI 0 0032~0 48) were independent predictors associated with gut derived bacterial translocation secondary to severe burns. These results suggested that the increase in intestinal lesion score and decrease in bifidobacteria count in mucosal flora were independent risk factors related to bacterial translocation, and specific interventions targeting these high risk factors should be instituted to attenuate gut derived bacterial translocation.
4. Preparation optimization of cationic liposome-siRNA complexes and in vitro therapeutic research on prostate cancer

OBJECTIVE: To investigate the preparation of novel cationic liposomes encapsulating siRNA, and evaluate their silence efficiency on target mRNA in LNCap cells. METHODS: Reverse evaporation technique was used to prepare the cationic liposomes. Protamine and calf thymus DNA were added to increase the encapsulation efficiency. The physicochemical property of the liposomes was evaluated. Human prostate cancer cells, LNCap, were used as the cell model. The silence efficiency on target gene in LNCap cells was evaluated with lipofectamine 2000 as the positive control. RESULTS: The optimum liposomes were obtained when the ratio of DOTAP and cholesterol was 3:1 and the ratio of liposomes and siRNA was 300:1. The size distribution and Zeta potential of the liposomes were (117 ± 4.3) nm and (43 ± 3.6) mV, respectively. The liposomes significantly improved siRNA accumulation in cells. CONCLUSION: The novel cationic liposome showed high transfection efficiency and distinct silence efficiency for target gene, which are expected to become a highly effective drug delivery system for anti-cancer drugs.
5. Synthesis of matrine derivative ZS10 and its mechanism of apoptosis induced by PI3K/AKT signaling pathway in human hepatocellular carcinoma cell line BEL-7402

Aim To explore the signaling pathway of matrine derivative ZS10 in inhibiting proliferation and inducing apoptosis of BEL-7402 cells. Methods ZS10 was synthesized by organic synthesis. The inhibitory effect of ZS10 on the proliferation of BEL-7402 cells was analyzed by MTT method at the time of 24 h, 48 h and 72 h, respectively, and IC50 was calculated. DAPI staining was used to observe the state of BEL-7402 cells. Clone formation method was used to observe the colony formation of BEL-7402 cells, flow cytometry was used to observe the cell cycle arrest and apoptosis of BEL7402 cells, and Western blot was used to detect the expression level of PI3K/AKT pathway and related proteins. Results MTT results showed that the IC50 was(6.62±1.11)μmol·L-1; DAPI staining showed that the cell state changed significantly with the increase of drug concentration, and the results of colony formation showed that ZS10 significantly inhibited the colony formation of BEL-7402 cells. The results of flow cytometry showed that ZS10 induced S phase arrest and cycle apoptosis of BEL-7402 cells. Western blot showed that ZS10 at the concentration of 08 μ mol·L-1 could regulate the PI3K/AKT pathway and its related proteins in a dose-dependent manner. Compared with the control group, the expression of PI3K, AKT, P-AKT and anti-apoptotic protein Bcl-2 significantly decreased, the expression of pro-apoptotic protein Bax significantly increased, the expression of Cyclin D1 and CDK2 significantly decreased, and the expression of EGFR and N-cadherin, Vimentin significantly decreased in the treatment group. The expression of E-cadherin increased. Conclusions Matrine derivative ZS10 can inhibit the growth and proliferation of hepatocellular carcinoma cell line BEL-7402.
6.Replication and Pathology of Duck Influenza Virus Subtype H9N2 in Chukar.

To investigate the susceptibility of Chukars to duck avian influenza virus H9N2 and explore their role in interspecies transmission of influenza viruses. Chukars were inoculated with duck avian influenza viruses H9N2. The present study demonstrated that inflammatory lesions and virus antigen were present in the trachea, bronchus, and parabronchus, and the viruses could be isolated from throat swabs and lung tissue homogenate supernatants. At 14 d post virus inoculation, anti-H9 influenza virus antibody in the serum was detected. The results indicated that Chukars are susceptible to duck avian influenza virus and serve as an intermediate host, thereby facilitating viral gene evolution and supporting the need for continued surveillance of epidemiology and evolution of the influenza virus in Chukars.

7.Effects of 4-hydroxy-2(3H)-benzoxazolone on Inflammatory and Apoptosis Signaling Pathways in Non-alcoholic Fatty Liver Disease Model Rats

OBJECTIVE:To inv estigate the effects of 4-hydroxy-2(3H)-benzoxazolone on inflammatory and apoptosis signaling pathways in non-alcoholic fatty liver disease (NAFLD)model rats. METHODS :SD rats were divided into normal control group(10 rats)and modeling group (50 rats). Normal control group was given basic diet ,and modeling group were given high-fat diet to induce NAFLD model. After modeling ,the rats were divided into normal control group ,model group ,silibinin group (26.25 mg/kg),and 4-hydroxy-2(3H)-benzoxazolone high-dose ,medium-dose and low-dose groups (100,50,25 mg/kg),with 8 rats in each group. Normal control group and modeling group were given 0.6% CMC-Na intragastrically ,and other groups were given relevant medicine 10 mL/kg intragastrically ,once a day ,for consecutive 4 weeks. After last medication ,the serum levels of albumin(ALB),total protein (TP),globulin(GLB),ALB/GLB and free fatty acid (FFA)were detected ;TUNEL staining was used to observe the apoptosis of rat hepatocytes. Western blot assay was used to detect the protein expression and phosphorylation level of inflammatory signaling pathway related proteins [Toll-like receptor 4(TLR4),myeloid differentiation factor 88(MyD88), nuclear factor-κB p65(NF-κB p65),NF-κB inhibitor protein(IκBα)] in liver tissue as well as the expression of apoptosis signaling pathway related proteins [B cell lymphoma 2(Bcl-2),Bax,caspase-3]. RESULTS :Compared with model group ,serum levels of TP (except for low-dose group ),GLB and FFA ,the protein expression of TLR 4(except for low-dose group ),MyD88 (except for medium-dose group )and caspase- 3,the phosphorylation levels of NF-κB p65 and IκBα protein were decreased significantly(P<0.05 or P<0.01). The ratio of A LB/GLB in serum and the ratio of Bcl- 2/Bax in liver tissue were significantly increased(P<0.05 or P<0.01),and the phenomenon of hepatocyte apoptosis was improved. CONCLUSIONS :4-hydroxy-2 (3H)-benzoxazolone can ameliorate NAFLD in rats ,and the mechanism may be associated with inhibiting the expression TLR 4/ MyD88/NF-κB signaling pathway-related proteins and apoptosis-related proteins in liver tissues.
8.Automatic Adjustment Algorithm of Bed Height for Multi-position Lower Limb Rehabilitation Robot

Objective:To develop an automatic adjustment algorithm of bed height of multi-position lower limb rehabilitation robot, to meet the variety of leg lengths and training modes to avoid the collision between robot and ground. Methods:Six mathematical models of robot bed body height were established for six training modes of multi-position lower limb rehabilitation robot, which were described with leg length and bed tilt angle. The influence was analyzed that mechanical clearance and deflection as well as the jitter error of leg bracket during movement. Furthermore, a software related to these models was developed to automatically adjust the bed height for training. Volunteers were recruited to test actually. Results:The test data of bed height are consistent with the theoretical calculation of six mathematical models. Clearance and deflection did not affect the theoretical results of bed height. The end of robot's lower limb was always above the safe height during rehabilitation training. Conclusion:The automatic adjustment algorithm of bed height has been established, which can ensure that the rehabilitation robot runs at a safe height.

10.Effects of Zhuang Medicine Jib on Spastic Hemiplegia after Stroke

Objective To observe the effect of Zhuang Medicine Jib on spasm in hemiplegics after stroke. Methods From January, 2014 to December, 2015, 55 patients with spastic hemiplegia after stroke were divided randomly into treatment group (n=27) and control group (n=28). Both groups accepted routine rehabilitation and acupuncture, while the treatment group accepted Zhuang Medicine Jib in addition. They were assessed with modified Ashworth Scale (MAS), Barthel index (BI), Simpling Fugl-Meyer Assessment (FMA) before and four weeks after treatment. The side-effects within three months after treatment were recorded. Results The scores of MAS, BI and FMA improved in both groups after treatment (t>3.32, P<0.01), and improved more in the treatment group than in the control group (t>2.286, P<0.05). No side-effect was found in the follow-up. Conclusion Zhuang Medicine Jib can further relieve spasm after stroke, and improve the motor function and activities of daily living.
