1.PROLIFERATION AND DIFFERENTIATION OF NEURAL STEM CELLS IN THE SUBVENTRICULAR ZONE AND SUBGRANULAR ZONE IN OLD RATS AFTER FOCAL CEREBRAL ISCHEMIA

Objective The proliferation and differentiation of neural stem cells in the subventricular zone(SVZ) and subgranular zone(SGZ) have been observed in normal old rats or treated by middle cerebral artery occluded(MCAO) and sham operation respectively. Methods Old rats were used to make models of middle cerebral artery occluded(MCAO).By BrdU pulse labeling and immunohistochemical single staining,the change of the number of BrdU positive cells was observed in SVZ and SGZ of normal rats or cerebral ischemia rats after MCAO 3d,7d,14d,21d,28d and sham operation respectively.By BrdU accumulating label and immunohistochemical double staining,BrdU/NeuN and BrdU/GFAP positive cells were determined in SVZ and SGZ of aged rats after MCAO 14d. Results BrdU positive cells were found in bilateral SVZ and SGZ of all groups above mentioned.Compared with normal and sham groups,BrdU positive cells of SVZ and SGZ in cerebral ischemia rats were strikingly increased.BrdU positive cells reached the peaks in SVZ at 7d and in SGZ at 14d after MCAO.At 14d after MCAO,BrdU/NeuN(0.98%) or BrdU/GFAP(12.56%) double labled cells were observed in SVZ,but no double labled cell was observed in SGZ.Conclusion The focal cerebral ischemia markedly stimulated proliferation of neural stem cells in SVZ and SGZ in old rats,and some proliferative cells in SVZ can differentiate into neurons or glial cells.

Objective To explore the magnetic resonance imging tracking of superparamagnetic iron oxid(SPIO) labeled embryonic neural stem cells (NSCs) transplantation into corpus striatum of the focal cerebral ischemia(FCI) rats and the effects of learning and memory abilities.Methods NSCs of embryonic rat were cultured in vitro and labeled with Fe2O3-mediated by poly-L lysine (PLL), with Prussian blue staining to evaluate the labeling rate and Trypan blue staining to observe cell viability of NSCs. The rats were randomly divided into groups A (normal control),B (normal labeled NSCs implanted),C (cerebral ischemia),D( labeled NSCs implanted),E (unlabeled NSCs implanted)and F (inactivated labeled NSCs implanted).The FCI models were made in groups C, D, E, F.The labeled NSCs or unlabeled NSCs or inactivated labeled NSCs were stereotaxically injected into the left corpus striatum of groups B,D,E,F respectively. The abilities of learning and memory were measured by the Y-type electronical maze in groups A, C, D, E after 3 d,7 d,2,3,4 weeks of transplantation respectively. MRI scanning was performed to monitor the NSCs in groups B, D, F at corresponding time. After MRI, the rats were killed and undergone Prussian blue staining of the brain histological section to observe the distribution of implanted NSCs.Results The labeling efficiency rate and cell viability of Fe2O3-PLL labeled NSCs were almost 100% and 95% respectively. Objective To investigate the changes of distribution and expression of NADPH\|d positive neurons and Fos like immunoreactive(Fos\|LI) neurons in the rats medullary visceral zone(MVZ) following different time course after global cerebral ischemia reprefussion. Method Fifty adult SD rats were divided into three groups,normal control group( n =2),sham operation group( n =12) and ischemia group( n =36).The 4 brain vessel of ischemia group rats were occluded for 30?min,then reperfused for 2?h,4?h,8?h,12?h,24?h and 48?h respectively.The sections of medulla oblongata were divided into three series.The first and second series were reacted with NADPH\|d histochemical and anti\|Fos immunohistochemical methods respectively,and the third series were double stained with the other two methods mentioned above. Results NADPH\|d neurons and Fos\|LI neurons were distributed in bilateral MVZ(in NTS and VLM predominately).The number of NADPH\|d neurons and Fos\|LI neurons of ischemia group rats were significantly increased than that of control group at 2\|12?h.Approximately 15\|20 percent NADPH\|d/Fos\|LI double labeled neurons were found in MVZ.Conclusion\ The results indicated that NADPH\|d neurons and non\|NADPH\|d neurons could sensitively respond to brain ischemia reperfsion and might associate with regulation of stress and defence responese.
4.Relationship Between the Expression of c-fos Gene and Delayed Neuronal Death in the Neonatal Hippocampus Following Hypoxia-ischamia Insult

Objective To explore the relationship between the expression of c fos gene and delayed neuron death in perinatal hypoxia ischamia encephalopathy (HIE). Methods Terminal deo xynuleotidyl transferase mediated dUTP nick end labeling (TUNEL), immuno histochemistry andreverse transcription PCR(RT PCR) were used to detect the apoptosis and transcription and translation of c fos gene in the hippocampus following HIE. Results Positive signal of apoptosis from CA1 to CA4 sections were observed in experimental hippocampi. 6.Value of Photoshop Software in Differentiating Endoscopic Ultrasonography Image of Gastric Stromal Tumor from Gastric Leiomyoma

Background:Endoscopic ultrasonography( EUS)is the first choice for examining submucosal protuberant lesions in the gastrointestinal tract,however,it is very difficult to differentiate gastric stromal tumor from gastric leiomyoma. Photoshop software now is tried to differentiate some easily misdiagnosed diseases. Aims:To investigate the value of Photoshop software in differentiating EUS image of gastric stromal tumor from gastric leiomyoma. Methods:EUS image of 118 cases of gastric stromal tumor and 42 cases of gastric leiomyoma confirmed by pathological and immunohistochemical examination were collected,mean gray value( intensity of echo)and gray value standard deviation( uniformity of echo)of EUS image of lesion were determined by Photoshop software,and the differentiation value was analyzed. Results:The mean gray value of EUS image of gastric stromal tumor was significantly higher than that of gastric leiomyoma(71. 94 vs. 48. 99,P<0. 01). The gray value standard deviation of EUS image of gastric stromal tumor was significantly higher than that of gastric leiomyoma(16. 63 vs. 9. 80,P<0. 01). Conclusions:Under EUS,gastric stromal tumor exhibits higher intensity of echo and less uniformity of echo than gastric leiomyoma. EUS combined with Photoshop software is helpful for differentiating gastric stromal tumor from gastric leiomyoma by analyzing mean gray value and gray value standard deviation of the lesion.
7.Application of CT perfusion imaging in radiotherapy for lung cancer

Objective To investigate the value of CT perfusion imaging in evaluation of therapeutic effect and prognosis in radiotherapy for lung cancer.Methods Fifty-one cases of lung cancer who were unable or refused to be operated on,36 males and 15 females,aged 37 - 80,underwent CT perfusion imaging,29 of which only before radiotherapy and 22 before and after radiotherapy twice.The images were collected by cine dynamic scanning (5 mm/4 slices ) and input into the GE AW4.0workstation for data processing.The slice positions of CT imaging were determined according to the largest tumor size in CT scan.Regions of interest of tumor were drawn at the region corresponding to the original images of CT perfusion.Radiotherapy was performed after CT perfusion imaging.Relevant parameters,including blood flow ( BF),blood volume ( BV),mean transit time ( MTT),and permeability surface (PS) were calculated.The treatment response after radiotherapy was evaluated by RECIST.At 2 -4 weeks after the treatment,CT examination was conducted once more.Results The tests of the 51 patients showed that the BV was 13.6 ml·100 g-1,the BF was 129.5 ml·min-1 ·100 g-1,the MTT was 9.1 s,and the PS was 10.0 ml· min- 1· 100 g-1 before radiotherapy.The tests of the 22 of the 51 patients showed that the values of BV and BF after radiotherapy were 7.6 ml· 100 g-1 and 97.8 ml·min-1· 100 g-1,respectively,both lower than those before radiotherapy (11.2 and 108.7 ml·min-1·100g-1,respectively),however,both not significantly ( t =1.28,0.40,P > 0.05 ) ; and the values of MTT and PS after radiotherapy were 8.9 s and 7.8 ml·min-1· 100 g-1,respectively,both not significantly higher than those before radiotherapy ( 7.2 s and 6.8 ml· min -1· 100 g-1,respectively,t =- 1.15,- 0.57,P >0.05 ).The mean area of tumor after radiotherapy was 1189.6 mm2,significantly less than that before radiotherapy ( 1920.3 mm2,t =3.98,P <0.05).The MTT of the SCLC patients was 12.9 s,significantly longer than that of the NSCLC patients (6.5 s,t =2.54,P <0.05).The MTT of the tumor with the area ≤ 10 cm2 was 11.2 s,significantly longer than that of the tumors with an area > 10 cm2(5.8 s,t =2.59,P < 0.05 ).The BV of the responder group was 19.2 ml· 100 g- 1,significantly higher than that of the nonresponder group (4.6 ml· 100 g - 1,t =3.62,P < 0.05 ).There were not significant differences in all the perfusion characteristics between the cases with the disease-free advanced survival time ≤ 10 months and those with disease-free advanced survival time > 10 months.Conclusions CT perfusion imaging helps in diagnosis and radiotherapy of lung cancer to a certain degree.
8.Therapeutic effect of three-dimensional conformal involved-field radiotherapy combined with chemotherapy on limited disease stage small cell lung cancer

Objective To analyze the therapeutic effect of three-dimensional conformal involvedfield radiotherapy (3D-CRT)combined with chemotherapy on limited disease stage small cell lung cancer (LD-SCLC).Methods The clinical data of 85 patients of LD-SCLC treated with 3D-CRT at the dose of 2 Gy/fraction,5 fractions per week for 5 - 7 weeks,with the median dose of 50 Gy ( 46 - 66 Gy),combined with 4 -8 cycles chemotherapy,64 males and 21 females,aged 29 -76,were collected and analyzed.Results The complete remission rate,partial remission rate,stability rate,and total effective rate were 36.5%,52.9%,10.6%,and 89.4%,respectively.The median survival time was 18 months,with the 1-,2-,and 3-year overall survival rates of 65.9%,33.8%,and 15.9%,respectively.The local recurrence rate,distant metastasis rate,and local recurrence + distant metastasis rate were 15.2% (9/85),49.2% (29/85),and 35.6% (21/85),respectively.Body weight,response to therapy,cycles for chemotherapy,and concurrent chemo-radiotherapy were all independent prognostic factors for LD-SCLC.Cox muhivariable regression was used to analyze the prognostic factors.Conclusions Involved-field radiotherapy is effective for LD-SCLC.Distance metastasis is the main cause of treatment failure.
9.Photoshop combined with Endoscopic Ultrasonography in grading invasive risk of gastric stromal tumors

Objective To investigate the application value of Photoshop in grading invasive risk of gastric stromal tumors (GSTs). Methods EUS image of 97 cases of GSTs confirmed by pathological and immunohistochemical examination were collected. GSTs were divided into four groups (very low risk, low risk, intermediate risk, high risk) by tumor size, mitotic count and rupture of tumor. Mean gray value (intensity of echo) and gray value standard deviation (uniformity of echo) of EUS images of the lesions were determined by Photoshop and then the differences of each group were found by statistical analysis. Results It is difficult to differentiate EUS images of GSTs from each group by visual observation. The mean gray value of EUS image of very low risk group,low risk group, intermediate risk group and high risk group of GSTs respectively were (56.54 ± 6.10), (59.20 ± 7.51), (77.77 ± 10.90) and (83.43 ± 12.47). There was no significant difference between very low risk group and low risk group (P > 0.05). There was no significant difference between intermediate risk group and high risk group (P > 0.05). In addition, the others all had significantly different from that of each group (P < 0.05). The mean gray value standard deviation of EUS image of very low risk group, low risk group, intermediate risk group and high risk group of GSTs respectively were (8.46 ± 2.59), (12.57 ± 5.89), (12.84 ± 4.15) and (16.69 ± 4.69). There was no significant difference between low risk group and intermediate risk group (P > 0.05). In addition, the others all had significantly different from that of each group (P < 0.05). Conclusions The higher risk of GSTs, the higher of echo intensity and the worse of echo uniformity under EUS. Photoshop combined with EUS is helpful for differentiating different risk of GSTs by analyzing mean gray value and gray value standard deviation of the lesions.
10.The study of SWI combined with 3D-PCASL on the prognosis analysis of acute cerebral infarction

Objective To investigate the clinical value of susceptibility weighted imaging (SWI)combined with three dimensional pseudo continuous arterial spin labeling (3D-PCASL)on the prognosis analysis of acute cerebral infarction.Methods Thirty cases with acute cerebral infarction (< 72 h)underwent conventional MRI,MRA,3D-PCASL and SWI.NIHSS scores were performed at the time of examination and 3 months later.The correlation between the collateral blood vessels,regional cerebral blood flow (rCBF)detected by combination of SWI and 3D-PCASL with clinical prognosis were analyzed.Results Twenty-three cases showed collateral blood vessels in the lesions with 1 grade in 14,and 2 grade in 9.The average rCBFs in grade 0,1,2 infarction areas were (22.69±11.94)mL·100 g-1 ·min-1 ,(25.10±16.55)mL·100 g-1 ·min-1 and (33.04±24.24)mL·100 g-1 ·min-1 ,respectively.Collateral blood vessels,rCBF were positive correlated with the NIHSS scores (r=0.989,P< 0.01).18 cases showed multiple vessels around the lesions. The average rCBFs in the infarction area with or not with periphery collateral blood vessles were (28.33±24.24)mL·100 g-1 ·min-1 and (22.69±11.94)mL·100 g-1 ·min-1 ,respectively.There was a positive correlation between rCBF and NIHSS scores (r=0.897,P<0.01). Of 30 cases of acute cerebral infarction,the average CBFs in the infarct areas and the contralateral mirror areas were (26.92±18.22)mL·100 g-1 · min-1 and (34.22±12.37)mL·100 g-1 ·min-1 .There was significant difference (t=8.093,P<0.01).Conclusion The combination of SWI and 3D-PCASL can display the collateral blood vessels in the lesions and soft meninges,and provide the quantitative analysis of rCBF,which has important clinical significance for prediction of the prognosis of acute cerebral infarction.
