In order to explore the effects of exogenous human telomerase reverse transcriptase (hTERT/hTRT/hEST2) on telomeric restriction fragment (TRF), telomerase activity and its subunits expression in human embryonic fibroblasts (hEFs), hTERT sense eukaryotic expression vector pIRES2 EGFP hTERT was constructed with DNA recombinant technique and then transfected into primary hEFs by Lipofectin method. TRF length, telomerase activity and changes in telomerase subunits expression were examined and evaluated in transfected and untransfected cells. Objective:To investigate the root and canal morphology of mandibular first permanent molars in adolescents by cone-beam computered tomography (CBCT).Methods:366 mandibular first permanent molars in 1 83 adolescents aged 1 2 -1 6 years were examined by CBCT from July,201 2 to December,201 3.The age,sex,the number of roots and canals and canal morphology were re-corded.The data were statistically analyzed by software SPSS 1 9.0.Results:365 mandibular first permanent molars showed single mesial root except one with C-shaped root;282(77.05%)teeth had one distal root,83(22.68%)had two.356(97.27%)teeth had two mesial canals and 9(2.46%)had one.229(62.57%)teeth had one distal root with one canal,53(1 4.48%)had one distal root with two canals.83(22.68%)had two distal roots with one canal in each root,one (0.27%)had C-shaped root with three canals. There were no significant differences between males and females and between the two sides on the number of roots and number of ca-nals.Conclusion:Majority of mandibular first molars have two roots,the root change occurs in distal root.The prevalence of three-rooted mandibular first molars has no significant difference between genders and 2 sides.

Objective To investigate the biological characteristics , therapy selection of tubular breast carcinoma.Relationship between axillary lymph node metastasis and other factors are analyzed retrospectively . Methods Tubular carcinoma patients were collected in our hospital from June ,1987 to March,2014.Paraffin tis-sues were re-checked again by pathology doctors and all patients were performed follow -up by telephone to e-valuate the prognosis .Results Sixty eight cases of tubular carcinoma of the breast were chosen .Axillary lymph node metastasis was closely related to tumor size and tumor grade in our research .In all patients ,only one patient was relapsed ,24 patients died of other diseases ,without distant metastasis or death of breast cancer .Conclusion Tubular breast carcinoma has a good prognosis with low recurrence rate .The treatment of tubular carcinoma is not quite reasonable .Preoperative core needle biopsy and surgery axillary staging should be paid more attention and breast conserving rate is still needed for improvement .

Objective This trial was to observe the effect of Ulinastatin on coagulation functions in patients during operation period,and to investigate the protective mechanisms of Ulinastatin.Methods Forty patients were randomly divided into Ulinastatin group (Group U,n =20) and control group (Group C,n =20).Group U was infused intravenously ulinastatin 4000 U/kg (diluted with saline to 30 ml,20min losers) after anesthesia and before cutting skin,while Group C received the same volume of normal saline.All patients were phlebotomized 1 ml peripheral blood before administered (T0) and after 1 hour (T1),respectively.Coagulation activation time (SonACT),clot rate (CR) and platelet function (PF)were detected by sonoclot coagulation analyzer and platelet function analyzer.Results Compared with group C (controlled group),SonACT of Group U was prolonged significantly at T1 (P ＜ 0.05),and PF were increased at T1 (P ＜ 0.05) ; Compared with T0,SonACT and PF were increased at T1,respectively (P ＜ 0.01).Conclusions Ulinastatin can improve perioperative coagulation function and platelet function.It may reduce intraoperative micro-thrombosis syndrome and postoperative deep vein thrombosis.

Objective To evaluate the relationship between mRNA and protein expression of HtrA and Streptococcus mutans i-solated from the children with different caries experience and to provide the theoretical and experimental basis on prediction of dent-al caries in deciduous teeth .Methods The strains of Streptococcus mutans isolated from children with different carious experiences in the preliminary experiments were divided into three groups :high caries-susceptible group ,middle caries-susceptible group ,caries-free group .All strains were reanimated on the agar plate of MS ,and after smear pure culture examination ,typical bacteria were in-cubated in BHI ,then purified nucleic acid and extracted all the RNA of streptococcus mutans by reverse transcription PCR and de-tected it by agarose gel electrophoresis integrality .Synthetic cDNA and take further PCR amplification with cDNA products .Ob-serve records results by Gel imaging system .HtrA of target gene and electrophoresis image were gray scan by Gel quantitative soft-ware Gel-Pro analyzer 4 .0 was used to analyze relative expression value of gene .After purifying protein ,collected total protein of Streptococcus mutans strains by Western Blot method ,then tested the concentration of total protein sample .The results of Chemilu-minescence imaging were scanned into computer by Bio-Rad analyzing system ,calculated the gray value by software Quantity One 4 .4 .0 which showed the relative expression level of protein .Results There were significant differences in HtrA mRNA and protein expression of different Streptococcus mutans isolated from the children with different caries susceptibility .high caries-susceptible group> middle caries-susceptible group> caries-free group (P< 0 .05) .Conclusion There were significant differences in HtrA mRNA and protein expression of different Streptococcus mutans isolated from the children with different caries susceptibility .The higher caries susceptibility the group was ,the more HtrA mRNA and protein the strain express .

Objective To evaluate influence of general anesthesia on postoperative cognition function in the elderly patients by using different methods and choose a better anesthesia method for elderly patients. Methods Forty elderly patients undergoing selective abdominal surgery were selected and divided into two groups: anesthesia was maintained with general anesthesia combined with epidural anesthesia in observe group and with general anesthesia singlely in control group. Vital signs were observed and recorded during operation and blood pressure varied within the extent of 20% of basic level. Cognition function of each patient 24 hours before and 24,48 hours after operation was e-valuated by MMSE method. Results All the patients maintained steady vital signs and there was no significant differ-ence in MMSE scores between two groups at all the time points. There were 11 cases(55% ) who had acute cognitive dysfunction in observe group and 13 cases(65%) in control group 24h after operation(P>0.05) ,5 cases(25%) in observe group and 9 cases(45%) in control group 48h after surgery(P>0.05). Conclusion Compared with single general anesthesia, general anesthesia combined with epidural anesthesia uses less general anesthetics and has less negative effect on postoperative cognition funetion in the elderly patients,it maybe better in elderly patients undergoing non-cardiac operation.

Objective To study the right ventricular function of patients with pulmonary thromboembolism by using color Doppler ultrasound.Methods 31 patients with acute pulmonary thromboembolism,compared with 31 vohnteers,were enrolled in this study.The right ventricular anterior wall movement,right ventricular end diastolic volume,right ventricular ejection fraction,and myocardial performance index were observed by echocardiography.Resuits The right atrium diameter,right ventricles diameter,right ventrieular end diastolic volume and pulmonary artery inner diameter in study group were much larger than that in control group (P<0.01),and the right ventricular anterior wall movement and right ventrieular ejection fraction decreased in study group (P<0.01).Tricuspidal annular E peak velocity tended to be decreased,isovolumie relaxation time and isovolumic contraction time were prolonged and myocardial performance index was increased (P<0.01).Right ventricular myocardial performance index showed significant correlation with right ventrieular ejection fraction (r=0.78,P<0.01),isovolumic relaxation time and isovolumic contraction time(rl=0.88,r2=0.57,P<0.01).Conclusion The right ventricular function in patients with pulmonary thromboembelism is decreased and myocardial performance index is a sensitive index which can be used to reflect right ventricular function in pulmonary thromboembolism.

Objective To investigate the effect and mechanism of Imatinib mesilate (Imatinib) on intimal hyperplasia of rabbit carotid arteries after balloon injury. Methods Thirty adult Newzealand rabbits were randomly divided into three groups:group A, B and C. Their right carotid arteries were injuried then administered with 0, 25 or 50 mg/kg of Imatinib dai?ly for 14 consecutive days when the rabbits were sacrificed. The carotid arteries were harvested and sectioned for HE-stain?ing and immunohistochemisty staining. Real-Time PCR was used to examine transcription levels of PDGF-B and PDGFR-βmRNA. The plasma level of PDGF-BB was assayed by ELISA. Results Arterial intimal hyperplasia and stenosis following balloon injury were seen in three groups. Thickness and area of neointima, ratio of thickness of intima to media, ratio of area of intima to media and mRNA level of PDGF-β are all higher in group A than those in group B than those in group C (P<0.01). By contrast, the mRNA transcription level of PDGFR-β increased significantly in group C than that in group A (1.236±0.356 vs 0.708±0.372;t=2.91;P<0.01). Plasma level of PDGF-BB increased in all three groups after balloon injury than that in the baseline (P<0.01). The transcription level of PDGF-BB is higher in group A than that in group B and in group C (ng/L:23.464±3.542, 19.504±2.454, 16.588±1.207, F=17.322, P<0.05). There was no difference between group B and C. There was positive correlation between mRNA transcription level of PDGF-B and plasma level of PDGF-BB ( r=0.806, P<0.01). Conclusion Vascular injury can cause intimal hyperplasia and increased PDGF-B mRNA transcription. Imatinib mesilate could inhibit the intimal hyperplasia through down regulating PDGF-B mRNA transcription.
