1.A panel of monoclonal antibodies for distinguishing metastasis adenocarcinoma cells from reactive mesothelial cells in serous fluids

Objective To enhance the diagnosis rate for metastasis adenocarcinoma cells in serous fluids and detect a panel of a monoclonal antibodies for distinguishing adenocarcinoma cells from reactive mesothelial cells in serous fluids. Methods Three marks of low molecular weight cytokeratin (CK LMW ), carcinoembryonic antigens (CEA) and mesothelial cells (MC) were used to immunostain the cells in serous fluids from 50 patients and in peritoneal washing from 14 patients. Results Expression rate of three groups of cells positive from CK LMW , CEA and MC in adenocarcinoma cells was 95.83%, 70.83%, 16.67%, in suspicious cancer cells 100%, 50.00%, negative, in reactive mesothelial cells 33.33%, 33.33%, 66.66%. The four suspicious cases and two reactive cases in primary cytopathological classifiecation should be reclassified as adenocarcinoma cases. The sensitivity, specificity and reliability of CK LMW , CEA and MC were 96.30%, 80.00%, 77.10%; 70.37%, 90 00%, 61 60%; 90.00%, 85.20%, 75.30%, respectively. Conclusion Immunocytochemistry, using a panel of CK LMW , CEA and MC antibodies appears to be adjunct of important value for distinguishing adenocarcinoma cells from reactive mesothelial cells in serous fluids.
2.The Cytologic Diagnosis and Analysis of Canceration of Breast Intraductal Papillomas

Objective: The cytologic methods were used in the diagnosis of cancerous change of (intraductal papillomas,IP) and its significance was discussed. Methods: We studied the nipple discharge smear by using Wright-Gimsa staining. Meanwhile, infrared ray scanning B-ultrasound and X-rays were performed. Results: One hundred and twenty-eight patients were with IP, among them, 15 (11.7%)were confirmed as tumors by surgery and pathological examination. The cancerous change of cytomorphology was based on IP. The average age of patients with cancerous change was 48.1.The nipple discharge in patients with cancerous change was primarily bloody(66.7%). Conclusion: Introductal papillomas is carcerous. The possibility of canceration in persons with longer time of nipple discharge is larger than those with shorter time of nipple discharge. Women who are elder than 45 years old, with nipple discharge and post menopause, should be considered to have the possibility of cancerous change.
3.Clinical observation of Xuanfeitongfu method in cluster treatment of severe pneumonia

Objective To observe the clinical efficacy of Xuanfeitongfu method in treatment of severe pneumonia and to explore its clinical value in the management of severe pneumonia. Methods Totally,62 patients with severe pneumonia were randomly divided into the control group (n = 30) and the treatment group (n = 32). The control group was given cluster treatment,including oxygen cure,anti-infection and nutrition support and maintaining a stable internal environment and etc. The treatment group was treated with Tongfu decoction orally(one dose a day,a total of 5 days)on a basis of cluster treatment. The comparison was conducted in the 2 groups in the levels of C reaction protein,calcitonin,the change of blood gas analysis,the time of mechanical ventilation and the mortality of severe pneumonia in ICU at baseline and 3,5 days after treatment. Results The level of C-reactive protein,calcitonin,the change of blood gas analysis were statistically significant (P < 0.05). The time of mechanical ventilation and the mortality were better in the treatment group (P < 0.05). Conclusion Xuanfeitongfu method can effectively improve oxygenation ,assist the anti-infection effect ,reduce the time of mechanical ventilation and ICU retention time ,ultimately improve the outcome of severe pneumonia.
4.Determination of bisphenol A in water by truncated aptamer-fluorescence method

Objective:
To establish a fluorescence method based on turncated aptamer for the determination of bisphenol A in water.
Methods:
The bisphenol A truncated aptamer containing 38 bases was selected as a recognition module, and was modified with the fluorophore 6-FAM at the 5'end. The 3'end of the complementary sequence cDNA was modified with the quencher DABCYL. The standard solutions of bisphenol A and interfering compounds were configured. The detection system was established after optimizing the number of bases in cDNA, the concentration ratio of truncated aptamer to cDNA, the incubation temperature and time, and the pH of the buffer. The specificity and recovery experiments were carried out.
Results:
When the complementary sequence cDNA included 9 bases, the concentration ratio of the truncated aptamer to cDNA was 1:1.5, the pH value of the buffer solution was 7.5, the cDNA was incubated at 55 ℃ for 60 minutes, in the concentration range of 10-75 pmol/L, the linear regression equation was y=2 230.7x+110 825, the correlation coefficient was 0.926. The limits of detection was 3.3 pmol/L. The difference values of fluorescence intensity between tetrabromobisphenol A, estradiol, estriol, bisphenol S and bisphenol A were obviously different, so there was no significant interference to the test result. The recovery rates were 97.8%, 98.8% and 102.3% with the spiked concentrations of 20.0, 40.0 and 60.0 pmol/L. The relative standard deviations were 4.4%, 2.1% and 2.6% (n=5), respectively.
Conclusion
The fluorescence method based on turncated aptamer has the advantages of easy operation, high sensitivity and specificity, which can be used for the determination of bisphenol A in water.
5.Combined strategies in treatment of breast ptosis and breast hypoplasia

Objective To seek an effective surgical procedure to treat patients with the varying degrees of breast ptosis and micromastia.Methods Patients were classified into Ⅰ-Ⅳ degrees based on different breast ptosis,and treated by different methods.Degree Ⅰ was treated with implanting prosthesis to the post-pectoralis major space; degree Ⅱ patients were repaired with avulsion of deeper mammary gland and pectoralis major and hanging fixed breast tissue,based on the degree Ⅰ procedure; degree Ⅲ was fixed with breast tissue flap,excised redundant epidermis by the method of double-rings and the complex of nipple and areola was shifted,based on the degree Ⅱ procedure; degree Ⅳ was repaired with implanting prosthesis and the method of lines to hanging fixed breast.Results In 116 cases of this study,there were no hematoma,infection and nipple and areola necrosis by the combined strategies.The follow-up period after the surgery was 6-31 months (mean 13.3 months).All cases had voluptuous and upright breasts,rectified breast ptosis,with the normal sense of nipple and areola.Conclusions For breast ptosis and hypoplasia,the combined strategies have better clinical therapeutic effects.
6.Effect of health belief model on compliance behavior of PICC patients after discharge

Objective To explore the effect of health belief model on compliance behavior of patients with PICC(peripherally inserted central catheter).Methods According to admission order,75 cases of PICC were divided into control group(n=36)and observation group(n=39).Patients in the control group received conventional health education and regular visits after discharge and besides the regular methods,patients in the observation group were intervened by health belief model for the compliance behavior.All patients of two groups were investigated with self-designed scale for compliance behavior.Result The compliance behavior of the observation group was better than that of control group with significant difference(P<0.05).Conclusion The health belief model of education can improve the compliance behavior of PICC patients after discharge from hospital.
7.Effects of Simvastatin Intensive Treatment before PCI on the Postoperative Related Indexes of Patients with Acute Coronary Syndrome

OBJECTIVE:To investigate the effects of simvastatin intensive treatment on the Postoperative Related Indexes of patients with acute coronary syndrome(ACS) underwent percutaneous coronary intervention(PCI). METHODS:106 patients with were included in the study and randomly divided into observation group(53 cases)and control group(53 cases). Both groups were given aspirin 100 mg,qd+clopidogrel 75 mg,qd before PCI for 4 weeks;observation group was additionally given Simvastatin tablet orally 20 mg before supper 15 d before surgery. TC,TG,LDL-C,HDL-C,hs-CRP,IL-6 and IL-18 levels,LVEF,the occurrence of coronary artery restenosis were detected in 2 groups before surgery and 6 months after surgery. The occurrence of ADR was recorded during treatment. RESULTS:There was no statistical significance in the levels of TG,TC,LDL-C and HDL-C between 2 groups before surgery and 6 months after surgery (P>0.05). There was no statistical significance in hs-CRP,IL-18, IL-6 and LVEF levels between 2 groups before surgery(P>0.05). 6 months after surgery,hs-CRP,IL-6,IL-18 and LVEF levels of 2 groups were significantly higher than before treatment;hs-CRP,IL-6 and IL-18 levels of observation group were significantly lower than those of control group,and LVEF was significantly higher than control group,with statistical significance (P<0.05). The incidence of coronary artery restenosis in observation group was significantly lower than control group, with statistical significance (P<0.05). No ADR was found in 2 groups during treatment. CONCLUSIONS:Preoperative simvastatin intensive treatment can effectively reduce cardiovascular inflammation degree in patients with ACS after PCI,prevent the formation of coronary artery thrombus,and reduce the incidence of coronary artery restenosis so as to effectively improve the prognosis and don' t increase the incidence of ADR.
8.Effect of point massage combined with foot bathing in hot water on sleep quality of patients after gynecological operation

Objective To investigate the effect of acupoint massage combined with foot bathing in hot water on sleep quality of patients after gynecological operation. Methods Ninety-nine gynecological patients were divided into the control group (n=49) and the observation group (n = 50) after gynecological operation according to time of admission. The control group received the routine nursing and the observation group was treated with acupoint massage and foot bathing in hot water besides routine nursing. Pittsburgh sleep quality index was used to investigate the sleep quality at the day before operation 1 and 8 days after operation. Result The sleep quality of postoperative patients in the observation group was significantly better than that of the control group (P < 0.05). Conclusions The acupoint massage with hot bathing in hot water can improve the quality of sleep of the patients after gynecological operations. For its convenience in manipulation, it is popular among patients and their families.
9.ANALYSIS OF CT IMAGE AND EFFECT OF ANTI-CYSTICERCUS THERAPY FOR 300 PATIENTS WITH CEREBRAL CYSTICERCOSIS

Objective To understand the effect of anti-cysticercus therapy for patients with cerebral cysticercosis and the changes of cysticercus on CT image after treatment. Methods The patients with cerebral cysticercosis were classified by the presentation of their brain CT image before treatment, then the effect of anti-cysticercus therapy on them after treatment was analyzed and the presentations of their brain CT images between before and after treatment were compared. Results There were different changes on CT image of cysticercus in brain tissues after anti-cysticercus therapy for different types of patients with cerebral cysticercosis. Type Ⅰ: the focus was absorbed completely after treatment in the majority of patients and calcificated in the minority. Almost all the patients were cured clinically after anticysticercus therapy. Type Ⅱ: the focus was absorbed completely in the minority, and one to two or more calcification dots were observed in the majority of patients. Anti-cysticercus therapy was effective. Type Ⅲ and Ⅳ: the absorption of focus was not very good and the effect of anti-cysticercus therapy was lower relatively. Conclusion The changes of CT image such as absorption, calcification, has important significance in forecasting prognosis and instructing clinical usage.
10.The Effect of Candida albicans on the Expression Level of Human ?-defensin-2 mRNA in Keratinocytes in vitro

Objective To investigate the effect of Candida albicans and its components on the expression level of human beta-defensin-2 mRNA (HBD-2) in keratinocytes in vitro. Methods Different components of Candida albicans were isolated by lyticase, repeated freezing and thawing, sonication, and centrifugation. The keratinocytes and HaCaT cell lines were co-cultured with Candida albicans and its cellular components for 24 h. The expression level of HBD-2 mRNA was detected by reverse-transcription polymerase chain reaction (RT-PCR). Results Low expression level of HBD-2 mRNA in the unstimulated keratinocytes and HaCaT cells was detected. The HBD-2 mRNA expression levels in the keratinocytes stimulated by Candida albicans, the extract of its cell wall, and pure mannan were significantly increased (P 0.05). Conclusions Candida albicans, the extract of cell wall of Candida albicans, and commercial mannan can increase the expression level of HBD-2 mRNA in keratinocytes.
