1.Clinical analysis of six cases of prurigo pigmentosa associated with ketogenic diet and bariatric surgery

Objective:To analyze clinical characteristics of ketosis-associated prurigo pigmentosa after ketogenic diet and bariatric surgery.Methods:Clinical data were collected from patients with ketosis-associated prurigo pigmentosa, who were diagnosed and treated in Department of Dermatology, Peking University People′s Hospital from September 2018 to September 2020. The clinical characteristics, sequelae and therapeutic effect of dietary modification were analyzed and summarized.Results:A total of 6 patients with ketosis-associated prurigo pigmentosa were collected, including 5 females who developed prurigo pigmentosa after ketogenic diet, and 1 male who developed prurigo pigmentosa after bariatric surgery. The skin lesions mainly involved the chest, back, waist and abdomen, and rarely involved the eyelids, axillae, elbows and mons pubis. Common skin lesions included urticaria-like erythema, papules and pigmentation arranged in a reticular distribution, and rare skin lesions included mung bean- to soybean-sized blisters, whose walls were liable to break. Among 5 patients undergoing routine urine analysis, 4 were positive (from + to ++++) for ketone bodies in the urine, and 3 were positive for urinary protein (+) . Pathological examination in 2 patients showed epidermal spongiosis, scattered necrotic keratinocytes, basal cell liquefaction, lymphocyte infiltration in the superficial dermis, and erythrocyte extravasation. The 6 patients were advised to eat staple foods. After dietary modification, 5 patients were nearly cured within 1 week; 1 patient, who continued ketogenic diet for weight loss, still received marked improvement after the treatment with minocycline at a dose of 100 mg/d in spite of restriction of carbohydrate intake. The levels of urinary ketone bodies and urinary protein in the 6 patients all returned to normal within 1 week after treatment.Conclusions:Ketosis plays an important role in the occurrence of prurigo pigmentosa. Dietary modification alone or adjuvant medical treatment such as minocycline is effective for the treatment of ketosis-related prurigo pigmentosa.
2. Effects of sodium quercetin-7-sulfate on recombinant human protein kinase CK2 holoenzyme

AIM: To study the effects and kinetics of sodium quercetin-7-sulphate (SQMS) on recombinant human protein kinase CK2 holoenzyme. METHODS: The recombinant human CK2 holoenzyme activity was assayed by detecting incorporation of 32p of [γ-32P]ATP into the substrate at various conditions. RESULTS: SQMS was shown to strongly inhibit the holoenzyme activity of recombinant human protein kinase CK2 with an IC50 of 1.37 μmol·L-1, Kinetic studies of SQMS on recombinant human CK2 showed: the inhibition was competitive with ATP and noncompetitive with casein. CONCLUSION: SQMS is an inhibitor of protein kinase CK2, and the inhibitory action of SQMS on CK2 is competitive with ATP and noncompetitive with casein.
3. Analysis and Comparison of HPLC Fingerprints of Natural and Artificial Agarwood

OBJECTIVE: To establish the HPLC fingerprint analysis method for agarwood with compare the natural agarwood and artificial agarwood combined with the contents of ethanol-soluble extraction (E%) and agarotetrol (A%). METHODS: The E% and A% were determined according to the ChP(2015). The chromatographic fingerprints of agarwood were established on a Platisil ODS C18 column (4.6 mm×250 mm, 5 μm) with mobile phase consisting of acetonitrile-0.1% formic acid. Gradient elution was performed at a flow rate of 0.7 mL•min-1, the detection wave length was set at 252 nm, and the column temperature was maintained at 31 ℃. The similarity evaluation system of traditional Chinese medicine chromatographic fingerprint (2004A) was used to establish the common pattern and calculate the similarity of chromatograms. Principal component analysis (PCA) was used with multivariate statistical analysis software for relative peak area of common chromatographic peak. RESULTS: The chromatographic fingerprint similarity of 13 batches of natural agarwood was 0.021-0.856, the E% was 10.1%-31.0%, and the A% was 0.04%-2.83%, respectively. The chromatographic fingerprint similarity of 11 batches of artificial agarwood ranged from 0.079 to 0.453, and the E% and A% were 10.3%-31.3% and 0.14%-1.02%, respectively. Thirteen batches of natural agarwood and 11 batches of artificial agarwwod were divided into two groups respectively, and the reference crude drug was gathered in the natural agarwood group. The similarity between natural and artificial agarwood chromatographic fingerprint was significantly different (P<0.05). Meanwhile, the similarity of artificial agarwood chromatographic fingerprint was positively correlated with the E% (P<0.05). CONCLUSION: The chromatographic fingerprint analysis combined with multivariate statistical analysis can easily and quickly distinguish natural and artificial agarwood, which provides a reference for the comprehensive evaluation of the quality of agarwood.
4. Network Pharmacology Study of Compound Danshen Dropping Pill Based on Molecular Docking and Recognition of Biological Network Function Modules

OBJECTIVE: To study material basis and mechanism of compound danshendropping pills (CDDP) in the treatment of atherosclerosis (AS) using network pharmacology techniques such as molecular docking and recognition of biological network function modules in this paper. METHODS: Firstly, interactions between ingredients of CDDP and targets were determined through molecular docking technique; secondly, 2-layered heterogeneous network (2-HN) was established from compounds-compounds interaction networks and targets-targets interaction networks, which were constructed by their molecular similarity and from UniHI database, respectively; finally, 2-HN was divided into different modules with various functions through module analysis. RESULTS: The mechanism of CDDP for AS treatment was correlated with inflammation, immune dysfunction and oxidative stress. Pathway analysis was further employed, and showed that main components of CDDP are danshensu, protocatechuic aldehyde, tanshinone VI, etc. CONCLUSION: The present study provides a reference for similar studies such as exploration of multi-component, multi-target, multi-pathway mechanism of traditional Chinese medicine.
5. Isolation and antifungal activities of a pair of isomers of phloroglucinol derivatives from Dryopteris fragrans

Objective: To study the chemical constituents and antifungal activitiy of phloroglucinol derivatives from Dryopteris fragrans. Methods: The phloroglucinol derivatives were isolated by silica gel column chromatography, Sephadex LH-20 column chromatography, and pre-HPLC. The chemical structures were elucidated on the basis of physicochemical and spectral properties. The antifungal activities against Trichophyton rubrum and Microsporum gypseum were tested by CLSI M38-A2 broth microdilution methods. Results: Two phloroglucinol derivatives were isolated and identified as disflavaspidic acid PB (1) and flavaspidic acid PB (2). The minimum inhibitory concentration (MIC) of compounds 1 and 2 were 13.33 and 1.875 μg/mL against T. rubrum respectively, 10 and 1.325 μg/mL against M. gypseum. Conclusion: Compound 1 is a new compound named disflavaspidic acid PB. Compound 2 is isolated from D. fragrans for the first time. Compounds 1 and 2 are a pair of isomers with promising antifungal activity.
6. Superb microvascular imaging in diagnosis of benign and malignant breast lesions

Objective To explore the value of superb microvascular imaging (SMI) for differentiating benign and malignant breast lesions. Methods Totally 281 patients (300 lesions) confirmed with pathology were enrolled, and the data of 2D gray-scale, CDFI and SMI were analyzed retrospectively. Adler's grading was applied to classify blood flow in breast lesions, and the ability of CDFI and SMI in detecting blood flow was compared. The vascular morphology features of breast lesions were also observed with CDFI and SMI, and then were classified and compared between benign and malignant lesions. Results The detection rate of blood flow with CDFI was 78.33% (235/300), with SMI was 90.33% (271/300), and the difference was statistically significant (χ2=116.066, P<0.001). Both CDFI and SMI showed statistically significant differences in blood flow Adler grading between benign and malignant lesions (both P<0.001). The difference of Adler grades with CDFI and SMI was significant (Z=-9.722, P<0.001). The differences in vascular morphology between benign and malignant breast lesions detected with CDFI and SMI were statistically significant (both P<0.001). The benign lesions mostly presented non-vascular, linear or curvilinear and tree-like, whereas enlarged and twisted penetrating and radial vessels were found more common in malignant lesions. Conclusion SMI can increase blood flow detection of breast lesions. SMI Adler grading of blood flow and vascular morphology are helpful to identifying benign and malignant breast lesions.
7. Influence of negative emotion and social support on professional identity of nurses in Guangdong Province during COVID-19 pandemic

OBJECTIVE: To explore the influence of negative emotion on the professional identity of nurses in Guangdong Province and the moderating role of social support during COVID-19 pandemic. METHODS: A convenience sampling method was used to select 436 nurses in Guangdong Province as study subjects. The Depression-Anxiety-Stress Scale, Nurse Professional Identity Scale and Perceived Social Support Scale were used to investigate the negative emotion, professional identity and social support of these nurses. RESULTS: The median, the 25 th and 75 th percentile scores of the negative emotion total scores of nurses was 10.0(3.0,17.8). The total scores of professional identity and social support were(106.6±16.9) and(56.9±13.2), respectively. The professional identity and social support were negatively correlated with negative emotion(P<0.01), while the professional identity was positively correlated with social support in these nurses(P<0.01). The negative emotion of nurses can negatively predict their professional identity, and social support plays a moderating role in it.CONCLUSION: The higher the degree of social support, the less negative emotional impact on professional identity of nurses during COVID-19 pandemic. Nurses should be given more social supports.

Objective:To investigate the difference of isokinetic angle-specific moment curves between anterior cruciate ligament (ACL)-injured patients with and without patellofemoral cartilage injury (PFCI). Methods:A retrospective analysis was performed on patients underwent knee arthroscopy and isokinetic muscle strength testing before surgery from September, 2018 to September, 2019. Seventeen ACL-injured patients with PFCI and 17 ACL-injured patients without PFCI who matched in age, sex and meniscus injury were selected. Before arthroscopy, isometric and isokinetic strength of knee flexion and extension at velocity of 180°/s and 60°/s was tested by isokinetic dynamometer. Normalized torque-angle curves (torque/body mass) were generated in steps of 1° and the differences in angle-specific moment curves between two groups were compared.
