1.The therapeutic effect of folic acid on transient ischemic attack patients with homocysteinaemia

Objective To observe the therapeutic effect of folic acid on transient ischemic attack(TIA)patients with homocysteinaemia (Hcy ). Methods 129 patients of primary TIA with Hcy were divided into two groups randomly. The observation group ( n = 65 )was administered with conventional therapy and folic acid, and the control group ( n=64 ) was only given conventional therapy. The variances of the plasma HCA level three months later were compared, and remission rate of TIA and complete stroke incidence one year later were analyzed between two groups. Results The Hcy incidence rate of TIA patients was up to 41.4%. Three months later, the plasma HCA level of observation group was lower than control group( ( 14.27 ± 6. 13 ) μmol/L vs (24.99 ± 6.87 )μmol/L, t=2.799, P<0. 01 ) ,and much lower than that of the control group post-treatment ( ( 14. 27 ±6. 13)μmol/L vs (24.68 ± 6.89) μmol/L, t = 2.735, P < 0.01 ). One year later, the complete stroke incidence of TIA in observation group was lower than that of the control group(9.8% vs 25.0%, P<0.05 ) ,and complete remission rate was higher than the latter(73.8% vs 50.0%, P < 0. 01 ). Conclusion Folic acid can decrease the plasma HCA level of TIA patients with Hcy efficiently,and improve the prognosis of such patients.
2.Protective Effect of 3-hydroxybutyrate Derivate on the Apoptosis of PC12

Objective To investigate the protective effect of 3-hydroxybutyrate derivate( methyl-3-hydroxybutyrate and ethyl-3-hydroxybutyrate)on the apoptosis of PC12 induced by FBS deficiency. Methods Methyl-3-hydroxybutyrate and ethyl-3-hydroxybutyrate were prepared by chemical degradation. PC12 cells were divided into different groups:medium with FBS as negative control,medium without FBS as positive control,medium with different concentrations of methyl-3-hydroxybutyrate or ethyl-3-hydroxybutyrate without FBS as sample groups. The shape and viability of cells in each group were analyzed by optical microscope and MTT. Results Compared with the negative control,cells in the positive control group demonstrated a large number of apoptosis,smaller and thinner morphology,and lower activity. However,the activity of cells was improved in the sample groups.Low concentration(0. 01 and 0. 001 mg·mL-1)of methyl-3-hydroxybutyrate and ethyl-3-hydroxybutyrate showed a protective effect on cell apoptosis,and 1 mg · mL-1 had the best protection effect. Conclusion High purity methyl-3-hydroxybutyrate and ethyl-3-hydroxybutyrate can be produced by chemical method,and both chemicals present good effect on protecting the PC12 from apoptosis.
3.Simultaneous Determination of Three Kinds of Components in Compound Xiaozhi Suppository by HPLC

OBJECTIVE:To establish a method for the simultaneous determination of gallic acid,rhein and emodin in Com-pound xiaozhi suppository. METHODS:HPLC was performed on the column of Kromasil C18 with mobile phase of 0.5% phosphor-ic acid-Acetonitrile solution(gradient elution)at a flow rate of 1.0 ml/min,the detection wavelength was 273 nm for gallic acid and 254 nm for rhein and emodin,the column temperature was 28 ℃,and the injection volume was 20 μl. RESULTS:The linear range was 0.296 4-14.82 μg/ml for gallic acid(r=0.999 6),0.215 0-10.75 μg/ml for rhein(r=0.999 9)and 0.307 2-15.36 μg/ml for emo-din(r=0.999 9);RSDs of precision,stability and reroducibility tests were lower than 3.0%;recoveries were 95.1%-97.2%(RSD=0.64%,n=6),95.4%-97.2%(RSD=0.42%,n=6) and 96.5%-99.4%(RSD=1.10%,n=6),respectively. CONCLU-SIONS:The method is simple and accurate,and can be used for the contents determination of gallic acid,rhein and emodin in Compound xiaozhi suppository.
4.Extraction Optimization by Response Surface Method and Content Determination by HPLC of Four Effec-tive Components in Fuke Yangkun Pills

Objective: To develop an HPLC method for the simultaneous determination of paeoniflorin , liquiritin, baicalin and costunolide in Fuke Yangkun pills , and optimize the extraction technology by response surface methodology ( RSM).Methods: The separation of targeted compounds were performed on a Kromasil C 18 column (250 mm ×4.6 mm, 5 μm).The mobile phase consisted of acetonitrile (A) and 0.2%phosphoric acid (B) with gradient elution at a flow rate of 1.0 ml min-1.The detection wavelength was set at 230, 276, 280 and 225 nm, respectively.The column temperature was 28℃.Using the contents of the four components as the indices, the extraction process was optimized by a response surface method with methanol concentration , solid-liquid ratio and ex-traction time as the influencing factors .Results: The linear range of paeoniflorin , liquiritin, baicalin and costunolide was 1.616-161.600, 0.432-43.180, 2.045-204.500 and 0.518-51.840 μg ml-1, respectively.The average recovery (n=9) was 98.3%, 99.6%, 97.9%and 98.1%, respectively.The optimum conditions of extraction process were as follows:the methanol concentration was 64%, the solid-liquid ratio was 1 ∶51, and the extraction time was 25 min.Conclusion: The response surface methodology is convenient and highly predictive in optimizing the extraction process of the four effective components in Fuke Yangkun pills .The devel-oped content determination method is simple and accurate , which can be used for the quality control of Fuke Yangkun pills .
5.Diagnostic Value of 64-slice Spiral CT Angiography in Aortic Diseases

Objective To explore the value of the 64-slice spiral CT angiography(SCTA)in diagnosis of aortic disease.Methods 32 cases with aortic diseases confirmed by operation underwent 64-slice spiral CT enhanced scan,raw data were dealed with multiplanar reformation(MPR),curved plannar reformation(CPR),maximum intensity projection(MIP),volume rendering(VR)and advantage vessel analysis(AVA).Results The aortic disease in 32 cases included aortic dissection in 16 cases,pseudoaneurysm in 7 cases,true aneurysm in 4 cases,narrowing of the aortic arch in 3 cases and amputation of aortic arch in 2 cases.The endometrial break and mural thrombus better showed with MPR and the detecting rat of intimal flake and the initial break was 81%(13/16),while for the periphery thrombosis in 7 cases with pseudoaneurysm,the detecting rate was 100%(7/7).The showing rate for displaying the whole with CR was 100%(32/32).In showing calcification and accurate of vessels with MIP,the detecting rate was 84%(27/32).The showing rate of the extent of the disease and the relationship between peripheral vascular was 100%(32/32).AVA was of significance in the measurement of vascular diameter and vascular cross-sectional area,the showing rate was 44%(14/32).Conclusion 64-slice spiral CT angiography is of significance in diagnosing aortic diseases.
6.Establishment of a real-time fluorescent quantitative RT-PCR rapid detection method for human astrovirus

Objective:To establish a rapid detection method for human astrovirus based on TaqMan-probe real-time fluorescent quantitative reverse transcription-polymerase chain reaction (RT-PCR).Methods:According to the conservative sequence of human astrovirus ORF1 b gene, we designed the amplification primers and specific fluorescent probe to establish the human astrovirus TaqMan real-time fluorescent quantitative RT-PCR rapid detection method. The specificity, sensitivity and stability of the method were evaluated. We also used this method to detect human astrovirus in clinical samples. Results:The established human astrovirus TaqMan real-time fluorescent quantitative RT-PCR detection method has good specificity and repeatability for human astrovirus, and the sensitivity can reach 10 2 copies/μl. After testing the clinical samples, the detection rate of human astrovirus by our method was 100%. Conclusions:The human astrovirus TaqMan real-time fluorescent quantitative RT-PCR detection method established in this study is simple, rapid, sensitive, specific and stable. It can be used for clinical human astrovirus detection and epidemiological investigation.
7.Numerical simulation on evolution and control of spiral wave in heart.

In this paper, the numerical simulation for evolution and control of spiral waves in 2D cardiac excitable media is performed; it can be represented as modified FitzHugh-Nagumo (FHN) model by selecting suitable parameter values. When the plane wave being cut, the system can evolve into a spiral wave. The excitability of media can produce effect on the stability of spiral wave. When the excitability is greater than the critical value, the spiral wave core and period tend towards infinity and disappear. The drifting ways of spiral wave tip are different (meandering) in various driving frequencies when one spiral wave is driven by uniform periodic small current. When period T is below or above the resonance period of spiral wave, the spiral wave is in its pedaflower or pediflower orbit, and if the driving period precisely equals the resonance period, the spiral wave tip drifts along a straight line.

8.Effective control of excitable waves in 2D cardiac excitable media.

We propose a method for effective control of patter on dissipative system by use of little perturbation analysis, and apply this nonuniform feed back method to control the polarization wave of heart represented by FHN equation. In the numerical experiment, we successfully alter the propagating direction of planar waves and drift the spiral waves to boundary without resonant repulsion. The effective control of the excited system will be used to study the mechanism of defibrillation and that is our interesting work.

9.Preliminary studies on effects of methylprednisolone and dexamethasone on Th cytokines in asthmatic children

Objective This study was carried out to compare chnical efficiency of methylprednisolone (MP) and dexamethasone (Dex) and their effects on Th cytokines in asthmatic children.Methods A total of 39 children with moderate and severe asthma at acute exacerbation were randomly divided into two groups, one (A) with MP 1 -2 mg/kg (n=21) and the other (B) with Dex 0.25 -0.75 mg/kg (n= 18),every 12 -24 h by intravenous drip.Scores of respiratory effort and peak expiratory flow rate prior to and three days after treatment in the asthmatic children aged over five years were evaluated,respectively,as well as the time of wheezing vanishing after treatment.Serum levels of interleukin-2 (IL-2) and IL-4 were determined by radioimmunoassay (RIA) prior to and three days after treatment,respectively.Results There was statistically significant difference in the time of wheezing vanishing between groups A and B (P = 0.042).Three days after treatment,PEER was significantly higher in group A than that in group B (P = 0.025).No statistically significant difference in serum IL-2 and IL-4,as well as ratio of IL-2/IL-4,was found in group B prior to and three days after treatment (P>0.05).Statistically significant difference in serum IL-2 was not observed in group B prior to and three day after treatment (P>0.05),and serum IL-4 decreased significantly (P=0.001) and ratio of serum IL-2/IL-4 (P=0.027) increased significantly three days after treatment than those prior to treatment.No significant correlation between respiratory-effort scores and ratio of serum IL-2/IL-4 prior to treatment was found (P=0.613).Conclusions Up-regulation of IL-2,or inhibition of release of IL-4,probably is not the main anti-inflammatory mechanism of dexamethasone.Methylprednisolone has little effect on serum IL-2,but can effectively reduce serum IL-4, thus increasing the ratio of serum IL-2/IL-4 and counterbalancing function of the Thl/Th2 cells.
10.Research on the association between glutathione S-transferase P1 genic polymorphism and heart rate and blood pressure.

Oxidative stress may reduce cardiovascular function. Glutathione Stransferases(GSTs) play an important role in cell defending against oxidative stress. Glutathione S-transferase P1 (GSTP1) gene is one of the most intensively investigated glutathione S-transferase genes in epidemiologic studies. The GSTP1 gene displays a polymorphism at codon 105 (Ile105 Val), which results in an enzyme with altered substrate affinity. To date, there have been few studies evaluating whether Ilel05Val polymorphism of GSTP1 gene has an effect on cardiovascular function in the broad masses of people. In this study, we investigated the relationship between Ile105 Val polymorphism of GSTP1 gene and heart rate and blood pressure in 197 unrelated adult males of Han nationality. It was found that there were two types of the GSTP1 genotypes, Ile105/Ile105 and Ile105/Val105, but genotype Val105/Val105 was not found, and the frequencies of IleIes/Ileos and Ile105/Val105 genotypes were 78% and 22% respectively. Comparison with individuals with lie105/Val105 genotype showed that those with Ile105/Ile105 genotype had higher rest heart rate and maximal heart rate mean values. However, whether for rest heart rate and maximal heart rate or for heart rate reserve, no significant differences were found between the two genotype groups (P>0.05). Compared with individuals with Ile105/Val105 genotype, those with Iler105/Ile105 genotype had higher systolic blood pressure and pulse pressure mean values and lower diastolic blood pressure mean value. However, for systolic blood pressure, diastolic blood pressure and pulse pressure, no significant differences were found between the two genotype groups (P>0.05). The results suggested that Ile105 Val polymorphism of GSTP1 gene may not be associated with heart rate and blood pressure in the broad masses of people.
