5.Effect of nicotine on coagulation abnormalities in endotoxemic rats

Objective To investigate the effect of nicotine on coagulation abnormalities in endotoxemic rats.Methods Ninety-six male SD rats weighing 200-250 g were randomly divided into 4 groups (n=24 each): group normal saline (group NS);group LPS;group nicotine(group NIC)and group α-bungarotoxin (α7 nicotinic acetylcholine receptor antagonist, group α-BGT) . Endotoxemia was induced by LPS 10 mg/kg injected via femoral vein in LPS, NIC and α-BGT groups. In group NIC nicotine 400 μg/kg was injected intraperitoneally at 30 min before LPS injection. In group α-BGT α-BGT 1 μg/kg was injected intraperitoneally at 15 min before intraperitoneal nicotine. Prothrombin time(PT),activated partial thromboplastin time(APTT),fibrinogen(Fib),antithrombin (AT),von Willebrand factor(vWF),plasminogen activator inhibitor-1(PAI-1),D-dimer,platelet count and TNF-α were measured before (baseline) and 2, 4 and 6 h after LPS injection.Results PT and APTT were significantly prolonged and plasma Fib and AT concentrations and platelet count were significantly decreased, while plasma PAI-1, D-dimer, vWF and TNF-α concentrations were significantly increased after LPS administration in group LPS as compared with group NS. Nitotine pretreatment significantly attenuated the LPS-induced changes in group NIC.The effect of nicotine was counteracted by α-BGT. Conclusion Nicotine can attenuate coagulation abnormalities induced by LPS by acting on α7 nicotinic acetylcholine receptor.

9.Effect of intravitreal bevacizumab injection before vitrectomy on proliferative diabetic retinopathy

AIM: To evaluate the effect of intravitreal bevacizumab (IVB) injection 1 week before pars plana vitrectomy (PPV) in proliferative diabetic retinopathy (PDR) patients. METHODS: A retrospective research was done on 46 PDR patients who were divided into PPV group (n=28) and IVB group (n=18, PPV with preoperative IVB). Bevacizumab was injected 1 week before PPV. Main outcome measures were visual acuity, incidence of iatrogenic retinal breaks, intraoperative and postoperative bleeding. RESULTS: At 1 month after surgery, visual acuity in PPV (82.1%) and IVB group (88.9%) improved significantly (P<0.01) and the difference between the two groups was not significant. Iatrogenic retinal breaks were reported in 18 cases (64.3%) in PPV group and 4 cases (22.2%) in IVB group (P<0.05). Intraoperative bleeding was encountered in all cases in PPV group and 7 cases (39%) in IVB group (P<0.01). Postoperative bleeding was reported in 9 cases (32.1%) in PPV group and none in IVB group (P<0.01). CONCLUSION: IVB injection before PPV is helpful in reducing iatrogenic retinal breaks, intraoperative and postoperative bleeding in PDR patients.
10. Role of PI3K/Akt pathway in hypoxic-activated calcium sensing receptor mediated A549 cell metastasis

Aim: To explore the role of PI3K/Akt signaling pathway in A549 and A549/DDP cells metastasis mediated by hypoxic-activated calcium sensing receptor (CaSR). Methods: The A549 and A549/DDP cells in the logarithmic growth stage were randomly divided into control, hypoxia, hypoxia + GdCl3 (CaSR agonist), hypoxia + NPS2143 (CaSR inhibitor) and hypoxia + LY294002 (PI3K inhibitor) + GdCl3 group. The protein levels of CaSR, MMP-2 and p-Akt were analyzed by Western blot in A549 and A549/DDP cells under different treatment conditions. The effects of different treatment factors on the ability of cell migration and invasion were measured by wound scratch assay and transwell migration assay. The effects of different treatment factors on the secretion of matrix metalloproteinase-2 (MMP-2) protein by A549 and A549/DDP cells was analyzed by ELISA. Results: Compared with control group, hypoxia increased the protein expression of CaSR, enhanced cell migration ability, increased MMP-2 protein expression in cells and culture medium, and promoted Akt protein phosphorylation in A549 and A549/DDP cells. NPS2143 reduced the effect of hypoxia, GdCl3 amplified the effect of hypoxia, and LY294002 inhibited effect of hypoxia and GdCl3. Conclusions: Hypoxic-activated CaSR promotes A549 cell metastasis, and its mechanism may involve PI3K/Akt pathway.
