2.An overview of antibody-based cancer therapy.

The use of monoclonal antibodies (mAbs) for cancer therapy has achieved considerable success in recent years. Approximate 17 monoclonal antibodies have been approved as cancer therapeutics since 1997. Antibody-drug conjugates (ADC) are powerful new treatment options for cancer, and naked antibodies have recently achieved remarkable success. The safety and effectiveness of therapeutic mAbs in oncology vary depending on the nature of the target antigen and the mechanisms of tumor cell killing. This review provides a summary of the current state of antibody-based cancer therapy, including the mechanisms of tumor cell killing by antibodies, tumor antigens as antibody targets, clinical effectiveness of antibodies in cancer patients and nanoparticles-based ADCs.

3.Emergency repair of severe complex defect in forearm by transplantation of free flap and functional reconstruction

Objective To report the outcome of emergency repair of severe complex defect in forearm by transplantation of free flap and simultaneous functional reconstruction.Methods From Mar.1994 to Aug.2003,4 cases with severe complex defect in forearm was repaired by transplantation of free skin flap, free skin flap combined with fibula flap,or fibula osteocutaneous flap in emergency.Simultaneously the flexion and extension function were repaired by muscle transfer and/or tendon grafting,tenonectomy.Results All the cases were successful.Follow-up period ranged from 1 to 3 years postoperatively.The blood-supply,tex- ture and elasticity of transferred flaps were excellent with good bone healing.Opposition of thumb with four fin- gers was good.Sensory recovery of the hand was satisfactory.Conclusion Transplantation of free flap com- bined with simultaneous functional reconstruction is an ideal method in emergency repair of severe complex de- fect in forearm.
4.The study of multi-detector CT on the grouping and measuring of the hepatic veins

Objective To study the three-dimensional topography of the hepatic vein(HV),the inferior vena cava(IVC)and the inferior right hepatic vein(IRHV)in the retrohepatic and pre-IVC tunnel in human beings,and to provide an anatomic reference for liver surgery.Methods One hundred and ten volunteers underwent CT scanning at 60 to 75 s after injection of contrast medium,and their HV,IVC and IRHV were reconstructed into MPR and 3D-MIP images.The hepatic veins were grouped according to the way by which the hepatic vein enters IVC.The angle between the right hepatic vein and the middle hepatic vein or the trunk of hepatic vein was measured,and the width from right hepatic vein to middle hepatic vein or to the trunk of hepatic vein on IVC was recorded.The frequency of IRHV was observed,and the length of the tunnel was measured.Results Among the 110 volunteers,there were 6 cases(5.45%)with three hepatic veins respectively entering IVC,98 cases(89.10%)with right hepatic vein and a common trunk of the middle hepatic vein and the left hepatic vein entering IVC,and 6 cases(5.45%)with left hepatic vein and a common trunk of the right hepatic vein and the middle hepatic vein entering IVC.The mean value of the angles between the right hepatic vein and the middle hepatic vein or the trunk of hepatic vein was(55? 18)?.The width from the fight hepatic vein to the middle hepatic vein or to the trunk of hepatic vein was (21?7)mm.The IRHV was observed in 30 cases(27.27%).The mean value of the tunnel length was (53?11)mm.Conclusion The parameters of the retrohepatic and pre-IVC tunnel in human beings can be measured accurately by the imaging of MPR,which can provide an anatomic reference for the liver surgery.
5.Effect of vitamin C and E on antioxidative enzyme, NOS activity and NO contents in hippocampus of rats with lead poisoning.

OBJECTIVETo observe the effect of vitamin C and E on blood lead (Pb) levels and SOD, GSH-Px, NOS activity and NO, MDA content in hippocampus of rats with lead poisoning.
METHODSRat lead poisoning model was established by oral administration of 0.615 mmol/L lead acetate in drinking water for 4 weeks; and animals were fed with vitamin C 100 mg/kg. bw and/or vitamin E 100 mg/kg. bw for 1 week. Then blood Pb levels and SOD, GSH-Px, NOS activity and MDA, NO contents in hippocampus of rats were determined by corresponding kits.
RESULTCompared with control group, blood Pb level was decreased significantly (P<0.05) after given vitamin C, vitamin E or combination of vitamin C and E. The concentrations of SOD, GSH-Px, NO and NOS were significantly higher in vitamin C and/or E groups than those in control group (P<0.05). The concentration of MDA in vitamin treatment groups was significantly lower than that in lead control group (P<0.05); furthermore concentration of MDA in combination of vitamin C and E group was significantly higher than that in vitamin C alone group (P<0.05).
CONCLUSIONAdministration of vitamin C and E can decrease blood lead level, alleviate damage of lipid peroxidation in hippocampus by lead toxicity and reverse NO, NOS levels in rats with lead poisoning.

6.Effect of daunorubicin and cytarabine on cell line NB4.

OBJECTIVETo explore whether daunorubicin (DNR) combined with cytosine arabinoside (Ara-C) and DNR alone have similar effect on acute promyelocytic leukemia (APL) cell line NB4 and acute myeloblastic leukemia cell line HL-60 in vitro.
METHODSCell morphology, cells viability, and cell apoptosis (Annexin-V by flow cytometry assay) were analysed.
RESULTSAfter incubation with DNR plus Ara-C for 24 hours,NB4 cell viability [(36.75 +/- 3.82)%] (n = 6) and cell apoptosis rate [(21.24 +/- 5.82)%] (n = 3) did not change significantly compared to that treated with DNR alone for 24 hours [(35.73 + 6.28 )%, (22.55 +/- 3.26)%, respectively] (P > 0.05). However, HL-60 cell viability [(67.17 +/- 2.07)%] and cell apoptosis rate [(48.05 +/- 0.92)%] changed significantly in DNR plus Ara-C group compared with DNR alone [(63.31 +/- 1.80)% ,(41.51 +/- 0.89)%, respectively] (P < 0.01 and < 0.05, respectively).
CONCLUSIONDNR plus Ara-C and DNR alone have similar effect on NB4 cells, but have different effect on HL-60 cells.

7.A prevalence study on mild cognitive impairment among the elderly populations of Mongolian and Han nationalities in a pastoral area of Inner Mongolia

Objective To understand the epidemiological characteristics and distribution of mild cognitive impairment (MCI) in elderly populations from Mongolian and Han nationalities living in the pastoral areas of Inner Mongolia Autonomous Region of China.Methods According to the MCI clinical diagnostic criteria from Diagnostic and Statistical Manual of Mental Disorders 4th revised edition (DSM-Ⅳ ) by American Psychiatric Association,the individuals under study were at the age of 55 or over,with Mongolian or Han ethnicities and living in the pastoral area of Inner Mongolia.Results The crude MCI morbidity rates of Mongolian and Han of the study populations in the pastoral area of Inner Mongolia Autonomous Region of China was 19.48% (1782/9146)and the standardization morbidity was 18.98%.The crude MCI morbidity rates of both Mongolian and Han cthnicities were 17.46% (the standardization morbidity was 16.99%) and 20.60% (the standardization morbidity was 19.98% ),respectively.There showed a significant positive correlation between the crude morbidities and age,also significantly increasing with the latter.In the Mongolian population,the morbidity increased from 12.17% at the age 55-59 to 27.78% at 85 while in the Han population,the morbidity increased from 15.50% at the age 55-59 to 23.53% at 85.In both the populations of Mongolian and Han,there was a statistically difference found between the morbidities of MCI ( x2=13.229,P=0.000).The morbidity was higher in Hans than in the Mongolians.However,there was no statistically significant difference noticed between the morbidities of MCI in the Mongolian males and females ( x 2 =2.376,P =0.123 ).There was statistically significant difference found between the morbidities of MCI in the Han males and females,with females having higher risk than males (x2=24.470,P=0.000).Conclusion The morbidity of MCI in the elderly Mongolian and Han populations from the pastoral area of Inner Mongolia Autonomous Region of China was considered to be quite high and correlated to age and gender.
8.Detection and clinical study on coronavirus HKU1 with acute lower respiratory tract infections of hospitalized children in Changsha

Objective In order to understand the epidemiological and virologic characteristics of coronavirus HKU1 infection in hospitalized children with acute respiratory tract infection (ARTI) in Changsha. Methods 1165 nasopharyngeal aspirates (NPA) specimens were collected from hospitalized children with ARTI between September 2007 and August 2008 in Changsha. Specimens were screened for pol gene of coronavirus HKU1 by polymerase chain reaction. All positive amplification products were confirmed by sequencing and compared with those in GenBank. Results Coronavirus HKU1 were detected in 12patients (1.03%) out of the 1165 children. The patients were from 8 days to 3 years. The most common clinical diagnosis was bronchopneumonia( 83. 33% ). Similarity of coronavirus HKU1 with those published in the GenBank at nucleotide levels was 98. 18%-100%. Conclusion Coronavirus HKU1 may be important pathogens in children with acute lower respiratory tract infection. Coronavirus HKU1 infections are common in children under 3 years old. There is no significant difference in the infectious rate between the boys and the girls. The peak of its prevalence is in spring and winter. A single genetic lineage of Coronavirus HKU1was revealed in human subjects in Changsha.
9.Heterogeneity of Monosymptomatic Resting Tremor in a Prospective Study: Clinical Features, Electrophysiological Test, and Dopamine Transporter Positron Emission Tomography.

BACKGROUNDThe relationship between monosymptomatic resting tremor (mRT) and Parkinson's disease (PD) remains controversial. In this study, we aimed to assess the function of presynaptic dopaminergic neurons in patients with mRT by dopamine transporter positron emission tomography (DAT-PET) and to evaluate the utility of clinical features or electrophysiological studies in differential diagnosis.
METHODSThirty-three consecutive patients with mRT were enrolled prospectively. The Unified Parkinson's Disease Rating Scale and electromyography were tested before DAT-PET. Striatal asymmetry index (SAI) was calculated, and a normal DAT-PET was defined as a SAI of <15%. Scans without evidence of dopaminergic deficits (SWEDDs) were diagnosed in patients with a subsequent normal DAT-PET and structural magnetic resonance imaging.
RESULTSTwenty-eight mRT patients with a significant reduction in uptake of DAT binding in the striatum were diagnosed with PD, while the remained 5 with a normal DAT-PET scan were SWEDDs. As for UPRDS, the dressing and hygiene score, walking in motor experiences of daily living (Part II) and motor examination (Part III) were significant different between two groups (P < 0.05 and P < 0.01, respectively). Bilateral tremor was more frequent in the SWEDDs group (P < 0.05). The frequency of resting tremor and the amplitude of postural tremor tend to be higher in the SWEDDs group (P = 0.08 and P = 0.05, respectively).
CONCLUSIONSmRT is heterogeneous in presynaptic nigrostriatal dopaminergic degeneration, which can be determined by DAT-PET brain imaging. Clinical and electrophysiological features may provide clues to distinguish PD from SWEDDs.

10.The change of serum vascular endothelial growth factor and matrix metalloproteinases-9 in proliferative hemangioma treated with propranolol.

OBJECTIVETo study the level of serum vascular endothelial growth factor, matrix metalloproteinases-9 in the proliferative hemangioma before and after propranolol treatment.
METHODSThe serum VEGF, MMP-9 was detected with ELISA assay before treatment and after 4 weeks and 8 weeks of propranolol treatment. The relationship between the serum VEGF, MMP-9 and the prognosis was analyzed.
RESULTSThe serum VEGF (295.4 +/- 158.1) pg/ml was high before treatment, then decreased after 4 weeks and 8 weeks of treatment (255.7 +/- 130.4) pg/ml, (224.2 +/- 120.6) pg/ml. The serum VEGF was significantly lower after 8 weeks of treatment (P < 0.05). The serum MMP-9 was also decreased after treatment, showing a positive relationship with VEGF.
CONCLUSIONSPropranolol can treat the proliferative hemangioma through decreasing the serum VEGF and MMP-9.
