3.Changes of leptin and IGF-Ⅰ in addescents with anorexia nervosa

Objective To study the changes of leptin and insulin-like growth factor-Ⅰ (IGF-Ⅰ) in adoles-cents with anorexia nervosa(AN) before and after three months of therapy. Methods The plasma levels of leptin and IGF-Ⅰ were measured in 11 adolescents with AN before and after three months of therapy and 11 normal ado-lescents of the same age,and their height,weight were examined and their body mass index(BMI) was calculated,and the results were compared before and after therapy, and were compared with those from age-matched normal adolescents respectively. Results (1)The plasma levels of leptin and IGF-Ⅰ , as well as the BMI in the AN patients after three months of therapy were significantly higher, than those before the therapy, and both were significantlylower than those in the controls. (2)Significant correlations between the plasma leptin levels and IGF-Ⅰ and BMI were detected in the AN patients both before and after the therapy, as well as in the controls. (3)Significant correla-tions between the plasma leptin levels and IGF-Ⅰ were detected in the AN patients both before and after the thera-py, as well as in the controls. Conclusion The plasma levels of leptin and IGF-Ⅰ resulted in significant changes, and these changes were consistent with body fat content in adolescents with AN.

Objective To evaluate cortisol level in experimental frequent febrile seizures model rats.Methods Rats were divided into 3 groups: normal control group(n=10,NC group),hyperthermic control group(n=12,HC group) and febrile seizures group(n=21,FS group).Cortisol level in serum and hippocampus of rats were determined by ELISA.Ultrastructural changes of neurons in hippocampal region CA1 was observed by electron microscop.Results 1.No special ultrastructual findings were observed from neurons in hippocampal CA1 in NC and HC group.In FS group,the neuronal degeneration and edema,the mitochondrial degeneration and edema,the mitochondrion ridge obscured or disappeared.2.Cortisol levels in serum and hippocampus in FS group [(26.57 ? 15.94)?g/L and((4.11)?1.74)?g/L,respectively] were significantly lower than those in HC group [(40.04 ? 15.72)?g/L and(6.70?3.45)?g/L,respectively](P0.05).Positive correlations were demonstrated between serum cortisol level and hippocampus cortisol level(r=0.40 P

9. Effect of astragalus polysaccharide on cell proliferation and the relevant gene expression during senescence of zebrafish

OBJECTIVE: To elucidate the anti-aging mechanism of astragalus polysaccharide (AP). METHODS: The effect of AP on cell apoptosis and senescence was evaluated by p-galactosidase (SA-p-gal) and acridine orange fluorescence analysis. In addition, the differential gene expression of AP treated zebrafish embryos was examined by RT-PGR. RESULTS: AP delayed zebrafish embryos apoptosis at the concentration of 0.125 mg · mL-1. Moreover, the expression of mdm2 and tert genes were up-regulated while bax, p21 and p53 genes were down-regulated during the early apoptosis of zebrafish embryos mediated by AP. CONCLUSION: AP may play a role in anti-aging by affecting the expression of some relevant genes.
