cAMP response element binding protein(CREB) is a genetranscription regulator locating in the nucleus.The activated CREB by phosphoration plays an important role by binding to the cAMP response elements and modulate the transcription of various target genes.In addition,the activity of CREB is also regulated by lots of transcriptional regulators in signal transduction mechanism.In central nerve system,CREB participates in promoting neuron growth,the processes of plasticity and long term memory.More importantly,researches of CREB in the pathological processes of Alzheimers disease,Hungtingtons disease and HIV-related dementia have made a progress recently,providing a basis for some therapy strategies to the related neurodegenerative disorders.
5. Oxidized low density lipoprotein-induced expression of transforming growth factor beta 1 and production of extracellular matrix in rat glomerular mesangial cells is mediated by lectin-like oxidized low density lipoprotein receptor-1

Objective: To investigate the effect of oxidized low-density lipoprotein (ox-LDL) on expression of lectin-like oxidized low-density lipoprotein receptor-1 (LOX-1), transforming growth factor beta 1 (TGF-β 1), and secretion of extracelluar matrix (ECM) in cultured rat glomerular mesangial cells (GMCs), and to investigate the influence of LOX-1 inhibitor polyinosinic acid (PIA) on the effect of ox-LDL. Methods: Rat glomerular masengial cells were cultured in vitro. RT-PCR was employed to determine the LOX-1 mRNA expression in GMCs incubated with different concentrations of ox-LDL (0, 25, 50, 100 μg/ ml). The expression of LOX-1 and TGF-β1, mRNA was also determined by RT-PCR in the blank control group, ox-LDL (50 μg/ml) group and PIA(50 μg/ml ox-LDL+250 μg/ml PIA) group. The contents of TGF-β1 fibronectin (FN), and collagen IV (Col IV) in the supernatants of the above 3 groups were determined by ELISA. Results: RT-PCR showed that LOX-1 mRNA expression in 25, 50 and 100 μg/ml ox - LDL groups was significantly higher than that of blank control group(P<0.05), with the highest expression found in the 50 μg/ml ox-LDL group; the expression of LOX-I and TGF-β 1 mRNA was significantly higher in 50 μg/ml ox-LDL group than that in the other 2 groups (P< 0.01). ELISA results demonstrated that the supernatant contents of TGF-β1, FN and Col IV were significantly higher in 50 μg/ml ox-LDL group than those in the other 2 groups (P< 0.05). Conclusion: Ox-LDL can upregulate the expression of LOX-1 and TGF-β1, mRNA and the secretion of extracelluar matrix in GMCs. Polyinosinic acid can antagonize the above effect of ox-LDL, suggesting that LOX-1 may participate in ox-LDL-induced GMCs damage and is involved in the development and progression of glomerulosclerosis.
7.Analysis on chlorogenic acid of chaenomeles speciosa and its correlations with anticancer by high performance liquid chromatography-mass spectrometry

Objective:To analyze chlorogenic acid of Chaenomeles speciosa by high performance liquid chromatography-mass spectrometry. Methods:A method for determining chlorogenic acid of Chaenomeles speciosa by HPLC-MS is described. The operating conditions were Diamonsil C18 column (5μm, 4.6×250 mm) at room temperature, acetonitrile-0.05%phosphoric acid solution as mobile phase by step gradients elution at a flow of 0.8 ml/min and UV detection at 325 nm. Results: The content of chlorogenic acid of Chaenomeles speciosa is 1.3 mg/g (RSD 0.4%), respectively. Conclusion:Chlorogenic acid was detected from Chaenomeles speciosa, which may be related to its inhibition of FAS and the potential role of anticancer.

Objective: To detect gallic acid in Paeonia suffruticosa Andr. by high performance liquid chromatography-mass spectrometry. Methods: A method for determining gallic acid in Paeonia suffruticosa Andr. by HPLC-MS is described. The operating conditions were Diamonsil C18 column (5μm, 4.6×250 mm) at room temperature, methanol-0.05% phosphoric acid solution as mobile phase by step gradients elution at a flow of 0.8 ml/min and UV detection at 270 nm. Results:The content of gallic acid is 1.8 mg/g. The relative standard deviation of gallic acid is 1.7%. Conclusion:The content of gallic acid in Paeonia suffruticosa Andr. is high, which may be related to the inhibition of FAS.

10.The analysis of localization and diagnosing of radionuclide myocardial perfusion imaging for stable angina pectoris

Objective: To analyze the application value of radionuclide myocardial perfusion imaging in the localization and diagnosis for stable angina pectoris in clinical practice. Methods: A total of 70 patients who had been diagnosed with stable angina pectoris (SAP) were enrolled, and all patients underwent ATP-loaded ATP-99Tcm-MIBI (99Tcm-MIBI) myocardial perfusion imaging (MPI) and coronary angiography (CAG) were used to compare the accuracy of MPI imaging in localization and diagnosis for SAP. Results:In the 70 cases of patients with two methods of examination: ①In MPI diagnosis, positive patients were 50 cases, and the positive predictive value of SAP, sensitivity, negative predictive value and specificity were, respectively, 92.0%, 90.2%, 75.0% and 78.9%. There were 45 patients were consistently diagnosed as SAP by the two methods and the total compliance rate was 90.0%. ②in 70 patients, 51 cases were diagnosed as SAP by CAG examination, the positive rate was 72.9%; ③in 50 MPI positive patients, 46 patients were consistent with the results of CAG diagnosis, and the positive rate was 92.0%. The difference between the two methods was statistically significant (x2= 5.72, P<0.05). Conclusion: 99Tcm-MIBI myocardial perfusion imaging can be used as a gatekeeper in patients with stable angina pectoris, and has high application value in the location and diagnosis for stable angina pectoris. The diagnosis and treatment links of stable angina pectoris can save costs and provide a reliable evidencet for clinical practice.
