1.Transanal local excision for rectal carcinoma

Objective To evaluate the therapeutic effects and prognostic factors of transanal local excision of rectal carcinoma. Methods We retrospectively analyzed 116 cases who underwent transanal lical excision for rectal carcinoma from 1995 to 2008 with the mented of Cox Regression analysis.Result The survival time of all the patients were from 14 to 160.5 months.median time was 58.5 months.Five-year overall survival rate was 72%.ten-year overall survival rate was 53%.There were 16 cases of local recurrence with a rate of 13.8%.In the univariate survival analysis:histopathology,depth of tumor invasion,radiotherapy,recurrence and metastases were the predictors of survival.In the Cox regression analysis:depth of invasion,recurrence and metastases were the independent prognostic taetors for surwval.For T_1 stage,its overall local recurrence rate was 6.3%,five-year overall survival rate was 93%,ten-year overall survival rate Was 85%:For T_2 stage,its overall local recurrence rate was 14.8%,five-year overallsurvival rate Was 63%.ten.year overall survival rate was 45%.For T_1 stage,there Was no local recurrence with radiotherapy:For T_2 stage,local recurrence rate Was 14.6%.From the surivival curve,there Was no difference betwecn the patient accepted radiotherapy or not(T_1:P=0.260,T_2,P=0.262).But for local recurrence,the differences Was significant(P=0.002). Conclusion The result of transanal local excision of rectal carcinoma is satisfactory with T_1 stage,but it is not suitable for T_2 stage tumors.
2.Effects of Capsaicin on Proliferation,Migration and Invasion of Human Large Cell Carcinoma NCI-H460

Objective To investigate effects of capsaicin on proliferation,migration and invasion of human large cell carcinoma NCI-H460 and discuss the possible mechanisms by which capsaicin inhibits non-small cell lung cancer. Methods NCI-H460 cells were cultured in vitro and treated with capsaicin at various concentrations. Colony formation assay,wound healing assay,and transwell chamber invasion assay were used to detect the effects of capsaicin on proliferation,migration and invasion of NCI-H460 cells. Western blotting was used to detect the protein expression of E-cadherin,N-cadherin,Vimentin and Snail. Results The rate of colony formation of 100,200,300 μmol.L-1 of capsaicin were (91.17±24.38)%,(43.22±12.91)% and (15.71±4.26)%,respectively,the control group was (99.53±21.25)%.the rate of migration of NCI-H460 cells of 25,50, 100 μmol.L-1 of capsaicin were (85.83±17.43)%,(37.92±10.16)% and(21.08±6.39)%,respectively,the control group was (93.04±20.23)%.The number of invasion of NCI-H460 cells of 25,50,100 μmol.L-1 of capsaicin were (99±18.2),(75± 17.9) and( 52 ± 14. 1 ) , respectively, the control group was ( 107 ± 20. 4 ) . The middle and high-dose capsaicincould inhibit proliferation,migration and invasion of NCI-H460 cells ( P<0.05) ,and E-cadherin expression level was significantly up-regulated and N-cadherin, Vimentin, Snail expression level was significantly down-regulated ( P<0. 05 ) . Conclusion Capsaicin can inhibit proliferation,migration and invasion of NCI-H460 cells,the mechanism may be related to up-regulation of E-cadherin and down-regulation of N-cadherin,Vimentin and Snail expression levels.
3.Multiple primary carcinoma in Chinese hereditary nonpolyposis colorectal cancer patients in Northeast China

Objective To investigate the familial incidence of multiple primary carcinoma in Chinese hereditary nonpolyposis colorectal cancer patients (HNPCC) in Northeast China.Methods By family line investigation,multiple primary carcinoma (MPC) spectrum' s characteristics of 509 patients in 85 families registered in strict conformity with the HNPCC Amsterdam criteria Ⅱ were analyzed retrospectively.Results Of the 85 HNPCC families,multiple primary carcinoma developed in 55 patients in 25 families,among them 45 patients had metachronous carcinoma in 17 families,16 patients had synchronous carcinoma occurred in 12 families,6 patients with both synchronous carcinoma and metachronous carcinoma in 4 families.Conclusions Multiple primary carcinoma developed in significantly high incidence in Northeast China in Chinese hereditary nonpolyposis colorectal cancer patients,the most common MPC are colorectal cancer and endometrial cancer.
4.Relationship between expression of DAS-1 protein in intestinal metaplasia and gastric cardia adenocarcinoma

Objective To investigate the relationship between expression of DAS-1 in gastric cardia intestinal metaplasia(CIM)and gastric cardia adenocarcinoma (GCA). Methods The cancerous tissues and CIM tissues (2 cm apart from caneer) obtained from 65 patients with GCA were examined for the expression of DAS-1 protein using immunohistoehemistry. The CIM tissues (<2 cm below Z line) obtained from 15 outpatients and inflammatory mucosa from 25 outpatients were also examined for expression of DAS-1 protein. Results The type Ⅲ IM was accounted for 55.4% (36/65) in GCA patients, which was significantly higher than that in outpatients [13.3% (2/15), P<0.01]. The positive rate of DAS-1 expression in cancerous tissues [78.5 % (51/65)] was also significantly higher than that in CIM tissues [38.8 %(30/80), P<0.01]. The expression of DAS-1 protein in IM tissues was gradually increased from type Ⅰ (0%) to type Ⅲ (71.1%) with positive correlation (P<0.01). Conclusions The type Ⅲ IM with over-expression of DAS-1 is closely related to GCA, which might be one of important precancerous lesions for GCA.
