Objective: To investigate the intervention effect of astragaloside on the expression of nuclear factor??(NF-?B) injured by hypoxia/reoxygenation(H/R) in vascular endothelial cells.Methods: The human umbilical vein endothelial cells(hUVECs) were cultured and passaged.After the third generation,hUVECs were randomly divided into three groups: control group,H/R group and astagaloside group.The cells in the control group were cultured under normal conditions;the cells of H/R group were cultured under hypoxia,in a closed container with oxygen concentrations below 1% for 1 hour,and then under normal conditions for 1 hour in a CO2 foster box;the hUVECs in astragaloside group were pretreated by different dose of astragaloside(the final concentrations of 20,40,80 mg/L) at the time of pre-hypoxia,after 12 hours of the pretreatment,they suffered from H/R.The concentration of intercellular adhesion molecule-1(ICAM-1) and content of malondialdehyde(MDA) in supernatant were detected respectively by ABCenzyme linked immunosorbent assay(ELISA) and thio-barbituric acid method.The expression of NF-?? of the vascular endothelial cells was analyzed by immunohistochemistry.Results: Compared with the control group,the content of MDA and concentration of ICAM1 in the supernatant fluid were remarkably increased in the H/R group(MDA content(6.98?1.15) ?mol/L vs.(2.38?0.49) ?mol/L,ICAM-1 concentration(3 169.01?132.75) ng/L vs.(995.14?74.93) ng/L,both P

Objective To observe the effects of different folate levels on inorganic arsenic-induced cytotoxicity and oxidative stress status. Methods Chang human hepatocytes were cultured with 20 ?mol/L sodium arsenite (NaAsO2) in medium containing 0 ?mol/L, 2.3 ?mol/L and 10 ?mol/L of folate, respectively. After 24 h of treatment with NaAsO2, cell viability was analyzed with 3- (4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) method, apoptosis was assessed by Annexin V/PI double staining, levels of ROS were detected by 2’,7’-dichlorodihydrofluorescein diacetate (DCFH-DA) probe, and intracellular reduced glutathione (GSH) and malondialdehyde (MDA) content were determined by DTNB method and formation of thiobarbituric acid reactive substances, respectively. 3.Laparoscopic treatment of cholelithiasis merging extrahepatic and acalculous and benign obstructive jaundice

Objective To evaluate the possibility and reliability of laparoscopic management of cholelithiasis merging extrahepatic and acalculous and benign obstructive jaundice. Methods Eleven patients with cholelithiasis merging extrahepatic and acalculous and benign obstructive jaundice who underwent laparoscopic management were retrospectively studied.Results Ten cases were successful with laparoscopic management,among the total,4 patients were underwent laparoscopic cholecystectomy (LC),6 cases were experienced LC and laparoscopic common bile duct exploration (LCBDE).One case was converted to open operation.No serious complications were observed.All the patients were cured after the operative treatment.None of the complications could be traced after follow-up for 6 months. Conclusions Cholelithiasis merging extrahepatic and acalculous and benign obstructive jaundice is a challenge to laparoscopic surgeons.Intraoperative diagnosis is very difficultly,but very important.It is better prognosis by careful and skilled operators and reasonable operative method to be chose.
5.Analysis on influencing factors on plasma homocysteine level in elderly people in Tianjin

Objective To investigate the influencing factors on serum homocysteine level in healthy retired-elderly people in Tianjin,in order to provide theoretical basis and guidance for clinical intervention therapy.Methods A total of 184 participants meeting the standard of Chinese healthy elderly people (2013) were selected from 1247 retired-elderly people who took healthy physical examination in three grade Ⅲ-A hospitals in Tianjin from June to August in 2014.They were divided into two groups based on plasma Hcy level:hyper-homocystinemia group (≥15 μmol/L) and normal plasma Hcy level group (<15μmol/L).The influencing factors on plasma Hcy level were analyzed by multiple stepwise regression and logistic regression analysis.Results The proportion of healthy retired-elderly people accounted for 14.76 %,with men at 82.61% and women at 17.39 %,mean age (78.37±6.88) years.The Hcy level was higher in healthy males than in healthy females [(16.90± 6.86) μmol/L vs.(12.89 ± 5.97) μmol/L,P<0.005].The proportion of people with hyper-homocystinemia accounted for 45.65%,and the mean Hcy concentration was (21.39±6.98) μmol/L.The age of people with hyper-homocystinemia was higher than that with normal Hcy [(80.58±6.09) years vs.(76.56±7.04) years,P<0.005].There were no statistically significant differences in blood pressure,lipid and blood glucose levels and the proportion of patients with coronary heart disease,hypertension,type 2 diabetes mellitus,gout,periodontitis and other diseases between the two groups (all P>0.05).Spearman correlation analysis showed that the level of serum Hcy was positively correlated with serum uric acid levels (P=0.000).Multiple stepwise regression and logistic regression analysis showed that age,gender and serum uric acid level were risk factors for plasma Hcy level in healthy elderly people.Conclusions Age,gender and serum uric acid level are risk factors for serum Hey level in healthy elderly people.
7.Brachial-ankle pulse wave velocity is a predictor of recurrent cardiovascular events in elderly male patients with acute coronary syndrome

Objective To explore whether brachial-ankle pulse wave velocity (baPWV) can be used as a predictor of recurrent cardiovascular events in elderly male patients with acute coronary syndrome (ACS). Methods The baPWV and biochemical indicators were measured in 118 elderly men with ACS, aged 61-92 years (mean age 76.6±6.7 years). During the follow-up period (617±297 days), all cardiovascular events (unstable angina, heart failure, cardiac death and stroke, etc.)were recorded. Results The baPWV was (17.48±3.54)m/s in non-event group(n=90),(21.91±4.86) m/s in event group and (24.48±4.39) m/s in major endpoint event group, respectively (both P<0. 05). There were 30 cardiovascular events that occurred in 28 patients, including 12 major endpoint events. The receiver operating characteristic curve demonstrated that the cut-off point of baPWV for predicting a cardiovascular event was 19.51 m/s and that for predicting a major endpoint event was 20.52m/s. The multivariate Cox proportional hazards model demonstrated that the above cut-off points had significantly relative risk for cardiovascular event: 1.37(95% CI: 1.16-1.63)and for major event:1.71(95% CI: 1.23-2.36). Conclusions The baPWV can be used as a predictor of recurrent cardiovascular events in elderly men with ACS.
8.Thioredoxin inhibits human vascular endothelial cell adhesion molecules expression via Smad3/AP-1 pathway

Objective To investigate the molecular mechanisms of protective effects of thioredoxin (Trx) on human vascular endothelial cells in atherosclerosis.Methods The cell models of Trx-overexpressing cells (Ad Trx) and the control cells (Ad-con) were established by adenovirus vector gene transfer technology in human umbilical vein endothelial cells (HUVECs).The oxidized low density lipoprotein,a risk factor of atherosclerosis,was used as a stimulator.Western blot and indirect immunofluorescence were used to detect the protein expression levels and the cellular localization of Trx,adhesion molecules (ICAM-1,VCAM-1) and the upstream signal pathways.Trx activity was detected by insulin disulfide reduction assay,and cellular reactive oxygen species (ROS)production was detected by fluorescent probe DCFH-DA.Results As compared with control group,Trx protein expression level was enhanced in Ad-trx group and the Trx activity in Ad-Trx group was upregulated by (26.2 ±3.3)%.The result of ROS detection showed that overexpression of Trx significantly inhibited the cellular ROS generation.As compared with control group,overexpression of Trx obviously inhibited the adhesion molecules expression but markedly promoted the phosphorylation of Smad3 in endothelial cells with or without oxLDL stimulation (P<0.05).Pretreatment of cells with SIS3,a specific inhibitor of Smad3 phosphorylation,reversed Trx-induced inhibition of adhesion molecules expression.Further studies showed that pretreatment of cells with SIS3 enhanced oxLDL-induced AP-1 subunit c-fos nuclear expression.Conclusions The enhancement of Smad3 phosphorylation and c-Fos nuclear expression are mainly responsible for the Trx-induced downregulation of adhesion molecules.
9.Downscaling research of spatial distribution of incidence of hand foot and mouth disease based on area-to-area Poisson Kriging method

Objective To understand the spatial distribution of incidence of hand foot and mouth disease (HFMD) at scale of township and provide evidence for the better prevention and control of HFMD and allocation of medical resources.Methods The incidence data of HFMD in 108 counties (district) in Shandong province in 2010 were collected.Downscaling interpolation was conducted by using area-to-area Poisson Kriging method.The interpolation results were visualized by using geographic information system (GIS).The county (district) incidence was interpolated into township incidence to get the distribution of spatial distribution of incidence of township.Results In the downscaling interpolation,the range of the fitting semi-variance equation was 20.38 km.Within the range,the incidence had correlation with each other.The fitting function of scatter diagram of estimated and actual incidence of HFMD at country level was y=1.053 1x,R2=0.99.The incidences at different scale were consistent.Conclusions The incidence of HFMD had spatial autocorrelation within 20.38 km.When HFMD occurs in one place,it is necessary to strengthen the surveillance and allocation of medical resource in the surrounding area within 20.38 km.Area to area Poisson Kriging method based downscaling research can be used in spatial visualization of HFMD incidence.
