1.Percutaneous lumbar diskectomy: the relationship of imaging features with clinical efficacy

Objective To analize the relationship of imaging findings of lumbar disc herniation with the clinical efficacy after percutaneous lumbar diskectomy (PLD). Methods 52 patients were undergone PLD, includinog 40 cases of disk confined protrusion, 4 with calcification of posterior edge of the disc and posterior longitudinal ligament, 3 severe degeneration, 2 shown narrowing of the intervertebral space, 2 mild herniation and 1 obvious herniation. Results The total effective rate reached 84.6%, including markedly improved 50.0%, moderately improved 34.6% and no change in 15.4%. No complication occurred. Conclusions PLD is an efficient, safety method to treat the protrusion of lumbar intervertebral disc. Imaging is helpful for exploring the indication and evaluating efficacy of PLD.
2.Comparison of the clinical efficacy of mammotome minimally invasive biopsy and conventional breast tumor resection for benign breast tumor

Objective To explore the treatment effect and application value of ultrasonic guided mammotome minimally invasive whirling surgery and open surgery for benign breast tumor.Methods 264 patients with benign breast diseases were randomly divided into observation group and control group,132 cases in each group.The control group took open surgery,the observation group used mammotome minimally invasive surgery,rotary cutting system.Recorded clinical surgery treatment of the two groups.Results The operating time,intraoperative blood loss,operation scar size of the observation group were (18.27 ± 3.05) min,(10.86 ± 1.69) mL,(2.76 ± 0.85) mm respectively,those of the control group were (53.48 ± 6.17) min,(97.54 ± 14.28) mL,(27.63 ± 4.31) mm respectively,the differences between the two groups were statistically significant (t =58.7753,69.2560,65.0429,all P < 0.05).In the observation group,breast deformation in 0 case,postoperative occurrence skin pigmentation in 1 case,itchy skin in 2 cases,with lateral swelling in 1 case,the patients' satisfaction score was (93.42 ± 3.89) points.In the control group,breast deformation in 5 cases,postoperative occurrence skin pigmentation in 8 cases,skin itching in 9 cases,lateral swelling in 7 cases,the patients' satisfaction score was (88.16 ± 1.45) points,the differences between the two groups were statistically significant(x2 =5.096,5.636,4.648,4.604,t =14.557,all P <0.05).Conclusion Using ultrasonic guided mammotome minimally invasive rotary surgical treatment for benign breast diseases can shorten the operation time,reduce intraoperative bleeding,reduce the incidence of postoperative complications,patients' satisfaction is higher,and it is worthy of popularization and application in clinic.
3.Expression of Musashi 1 correlates with microvessel density and prognosis in colon cancer

Objective:To investigate the Musashi 1 expression, and its correlation with clinical variables, such as clinical pathologic factors,angiogenesis and prognosis in colon cancer.Methods: Musashi 1 mRNA level was determined by quantitative real-time PCR( qRT-PCR) in 36 pairs of matched colon cancer tissues and adjacent non-tumorous tissues.The expression of Musashi 1 and mi-crovascular density ( MVD) were tested by immunohistochemical in 96 cases of colon cancer.Combined with postoperative follow-up survival situation, the correlation between the expression of Musashi 1, pathological parameters, prognosis and MVD were statistical analylised.Results:Musashi 1 mRNA level of colon cancer tissues determined by PCR was significantly higher than adjacent non-tumorous tissues( P<0.05).Positive Musashi 1 immunoreactivity was seen in 63.5%of colon cancer specimens,which was correlated with the depth of invasion,Duckes stage,lymph node metastasis,and TNM stage (P<0.05).Compared with Musashi 1 negative patients, Musashi 1 positive patients had significantly shorter survival time ( 23.0% vs 60.0%, P<0.01 ) .High intratumor blood microvessel density patients had significantly shorter survival times than low intratumor blood microvessel density patients(P<0.05).Intratumor blood microvessel density correlated with Musashi 1 expression (P<0.01).Conclusion:Musashi 1 mRNA level of colon cancer tissues was higher than adjacent non-tumorous tissues,the difference has statistically significant.We conclude that Musashi 1 expression correlates with the depth of invasion,Dukes stage,lymph node metastasis,and TNM stage and microvascular density ( MVD) in colon cancer.We propose that synthesized Musashi 1 increases intratumor angiogenesis,thus promotes tumor progression.Musashi 1 expression may be a good biomarker for poor prognosis in colon cancer.
4.Efficacy observation of glutathione on prevention of acute radiation enteritis in pelvic radiation therapy

Objective To observe the prevention effect of glutathione on acute radiation enteritis in pelvic radiation therapy.Methods All 80 pelvic tumor patients treated with radiotherapy were randomly assigned to the control group (40 patients)and the treatment group (40 patients)by the number table method.40 cases in the con-trol group treated with radiation were not treated with preventive drugs,but the patients in the treatment group were treated with glutathione.The occurrence time of acute radiation enteritis and the severity of acute radiation enteritis after treatment were evaluated.Results 15.0% of the treatment group suffered from acute radiation enteritis in the second week and 72.5% in the third week.however,62.5% of the control group were suffered from acute radiation enteritis in the second week and 27.5% in the third week.The difference was statistically significant (χ2 =18.775, 15.998,all P <0.001).Glutathione delayed the occurrence time of acute radiation enteritis.The grade 1 and grade 2 acute radiation enteritis effective rate in the treatment group were 77.5% and 17.5%,and that in the control group were 20.0% and 72.5%,the difference between the two groups was significant(χ2 =26.136,24.139,all P <0.001).The glutathione could reduce the incidence of acute radiation enteritis extent.Conclusion Glutathione could delay the occurrence time of acute radiation enteritis and reduce the incidence of acute radiation enteritis extent.It is worth clinical application.
5.Risk factors of nosocomial infections in patients with head and neck cancer during radiotherapy

Objective To investigate the risk factors of nosocomial infections in patients with head and neck cancer during radiotherapy .Methods Clinical data of 189 patients with head and neck cancer receiving radiotherapy in Huzhou Central Hospital during August 2011 and December 2014 were retrospectively studied .Chi-square test and multivariate Logistic regression analysis were performed to explore the risk factors of nosocomial infections in patients during radiotherapy .Results Among 189 patients with head and neck cancer receiving radiotherapy , nosocomial infection was observed in 64 (33.86%) patients.Univariate analysis showed that radiation-induced oral mucositis (χ2 =11.47, P<0.01), underlying disease (χ2 =6.59, P<0.05), stage of tumor Ⅲ-Ⅳ (χ2 =4.36, P<0.05), whole-neck radiation (χ2 =7.64, P <0.05), dose of radiation ≥50 Gy(χ2 =8.92, P <0.05), combined chemotherapy (χ2 =6.53, P<0.01), invasive operation (χ2 =4.21, P<0.05), PS score=1 (χ2 =3.58, P<0.05) and low body mass index (χ2 =9.56, P <0.05) were risk factors of nosocomial infections . Multivariate Logistic regression analysis indicated that radiation-induced oral mucositis ( OR=3.46, χ2 =8.72, P<0.01),underlying disease (OR=1.91, χ2 =4.66, P<0.05), stage of tumor Ⅲ-Ⅳ (OR=2.01,χ2 =5.22, P<0.05), whole-neck radiation (OR=1.78,χ2 =4.89, P<0.05), dose of radiation ≥50 Gy (OR=1.69,χ2 =3.25, P<0.05), combined chemotherapy (OR=1.84, χ2 =6.91, P<0.05) and low body mass index (OR=1.53, χ2 =2.59, P<0.05) were independent risk factors of nosocomial infection.Conclusions Nosocomial infections are popular in patients with head and neck cancer during radiotherapy.Radiation-induced oral mucositis, underlying disease, stage of tumor, radiation field, dose of radiation, combined chemotherapy and body mass index are associated with the occurrence of nosocomial infections in these patients .
6.Diagnostic value of ultrasound-guided puncture biopsy in breast masses

Objective To explore the diagnostic value of ultrasound-guided puncture biopsy in breast masses.Methods 50 patients were enrolled in this study for breast mass examination and ultrasound presentation of BI-RADS class Ⅳ.All the patients underwent ultrasound-guided biopsy and pathological examination.The pathological results of the biopsy were compared with the final results of pathological diagnosis.The sensitivity and specificity of ultrasound-guided biopsy,and the accuracy of each type of breast mass examination were observed.Results 50 cases of breast masses were successfully punctured under ultrasound guidance,the postoperative pathological results were the diagnostic criteria.The puncture biopsy results showed that 16 cases of benign lesions and 34 cases of non-benign lesions.The postoperative pathological examination results showed that 13 cases of benign lesions,37 cases of non-benign lesions,the sensitivity and the specificity of the biopsy was 89.19% and 92.31%.Pathological puncture results showed 16 cases of benign tumors with an accuracy rate of 81.25%,27 cases of malignant tumors,the accuracy rate was 92.59%,7 cases of high risk,the accuracy rate was 57.14%.Compared with the accuracy of the diagnosis of benign and malignant results,the diagnostic accuracy of high-risk results was the lowest(x2 =20.802,P < 0.01).Conclusion Ultrasound-guided puncture biopsy can better identify the benign and malignant nature of breast masses,with high accuracy,small trauma,it is safe and reliable,and has important clinical value for the diagnosis of breast mass,which can provide a reliable basis for further clinical treatment scheme.
7.Advantages of Restoring miR-205-3p Expression for Better Prognosis of Gastric Cancer via Prevention of Epithelial-mesenchymal Transition

Purpose:
miR-205 is a tumor suppressor and plays an important role in tumor invasiveness. However, the role of miR-205 in human gastric cancer (GC) epithelial-mesenchymal transition (EMT) remains unclear. The aim of this study was to investigate the molecular mechanism of miR-205 in the regulation of EMT in GC invasion.
Materials and Methods:
Quantitative polymerase chain reaction (qPCR) was used to detect the expression of miR-205 in GC. Further, the correlation between the pathological parameters and prognosis of GC was statistically analyzed. A transwell model was used to evaluate the effect of miR-205-3p on the invasion and migration of GC cells. qPCR, western blotting, and luciferase assay were performed to analyze the relationship and target effects between miR-205-3p and the expression of zinc finger electron box binding homologous box 1 (ZEB1) and 2 (ZEB2).
Results:
We found that the levels of miR-205-3p were significantly lower (P<0.05) in GC tissues than in matched normal tissues. Additionally, the expression of miR-205-3p was related to the tumor invasion depth, lymph node metastasis, lymph node invasion, and tumor, node, metastasis stage. Patients with lower miR-205-3p expression levels in the tumors had a poorer prognosis. The in vitro assays indicated that miR-205-3p could affect the invasion ability and EMT of GC cells by targeting the expression of both ZEB1 and ZEB2.
Conclusions
miR-205-3p promotes GC progression and affects the prognosis of patients by targeting both ZEB1 and ZEB2 to directly influence EMT.
8.Effect of corilagin on IκB-α and nuclear factor-κB P65 in U251 glioma cells and glioma stem cells

Objective To explore the effect of corilagin on proliferation of glioma U251 cells and glioma stem cells and IκB-α and nuclear factor (NF)-κB P65 protein expressions in these cells.Methods The glioma stem cells were isolated from glioma U251 cells by using immune magnetic beads.The cells were intervened by different corilagin concentrations (0,25,50 and 100 μg/mL) for 48 h,respectively.Cell morphology changes were observed by microscope;cell counting kit (CCK)-8 assay was used to detect the cell proliferation;dual-luciferase reporter assay was employed to detect the P65 gene promoter expression;Western blotting was used to investigate the protein expressions of Iκ B-α in cytoplasm and NF-κB P65 in nucleus.Results (1) Cell morphology observation results showed that the cells became shrunken,cell density was decreased,and cell structure was destroyed with a great deal of cell debris.(2) CCK-8 assay results showed that as compared with those in the 0 μg/mL corilagin group,the survival rates ofU251 glioma cells and glioma stem cells were significantly decreased in the 25,50 and 100 μg/mL corilagin groups (P<0.05);while in the presence of the same corilagin concentration,the survival rate of U251 glioma cells was significantly higher than that of glioma stem cells (P<0.05).(3) Dual-luciferase reporter assay results showed that as compared with the 0μg/mL group,the P65 gene promoter expressions of U251 glioma cells and glioma stem cells in the 25 μg/mL corilagin group were significantly increased (P<0.05),but with increasing concentrations of corilagin,the expressions were gradually decreased.(4) Western blotting results showed that the IκB-α expressions in cytoplasm of U251 cells and U251 stem cells were significantly increased,but the NF-κB P65 expression in nucleus of U251 cells and U251 stem cells was significantly decreased with increasing concentrations of corilagin (0,25,50 and 100 μg/mL),with signficant differences between each two groups (P<0.05).Conclusion Corilagin could inhibit the expression of P65 gene promoter,promote the IκB-α protein expression in cytoplasm,reduce NF-κB P65 protein into the nucleus,thereby to inhibit the NF-κB signaling pathway,and it is likely to be one of the important mechanisms to inhibit the proliferation of glioma cells and glioma stem cells.
9.Chemical Constituent Analysis of Tongxie Yaofang Decoction by UPLC-LTQ-Orbitrap-MS

ObjectiveTo rapidly identify the chemical constituents in Tongxie Yaofang decoction by ultra-performance liquid chromatography-linear ion trap-electrostatic field orbitrap high-resolution mass spectrometry(UPLC-LTQ-Orbitrap-MS). MethodChromatographic conditions were ACQUITY UPLC BEH C18 column(2.1 mm×100 mm, 1.7 μm), mobile phase of 0.1% formic acid aqueous solution(A)-acetonitrile(B) for gradient elution (0-4 min, 5%-15%B; 4-10 min, 15%-25%B; 10-15 min, 25%-60%B; 15-20 min, 60%-90%B; 20-25 min, 90%-100%B; 25-27 min, 100%B; 27-30 min, 100%-5%B; 30-32 min, 5%B), flow rate of 0.3 mL·min-1, column temperature at 35 ℃ and injection volume of 3 μL. UPLC-LTQ-Orbitrap-MS was equipped with an electrospray ionization(ESI), the MS and MS/MS data were collected in positive and negative ion modes, and detection range was m/z 100-1 250. Combining the reference substance, chemical databases and related literature information, TraceFinder 4.1 and Xcalibur 2.1 were used to identify the chemical constituents of Tongxie Yaofang decoction. ResultA total of 90 compounds, mainly including flavonoids, coumarins, monoterpene glycosides, chromones and lactones, were identified from Tongxie Yaofang decoction. By attributing the sources of Chinese medicines for all identified compounds, 9 of them were found to be derived from Atractylodis Macrocephalae Rhizoma, 21 from Paeoniae Radix Alba, 24 from Citri Reticulatae Pericarpium, 29 from Saposhnikoviae Radix, and 7 from at least two Chinese medicines. ConclusionThe method can effectively, quickly and comprehensively identify the chemical components of Tongxie Yaofang decoction, and clarify the chemical composition. These identified compounds cover the main active ingredients of the four herbs with high abundance, which indicates that the extraction method and the ratio of the medicinal materials of Tongxie Yaofang are scientific, and can provide a reference for the research on the material basis and quality evaluation of this famous classical formula.
10.Pluripotent stem cells secrete Activin A to improve their epiblast competency after injection into recipient embryos.

It is not fully clear why there is a higher contribution of pluripotent stem cells (PSCs) to the chimera produced by injection of PSCs into 4-cell or 8-cell stage embryos compared with blastocyst injection. Here, we show that not only embryonic stem cells (ESCs) but also induced pluripotent stem cells (iPSCs) can generate F0 nearly 100% donor cell-derived mice by 4-cell stage embryo injection, and the approach has a "dose effect". Through an analysis of the PSC-secreted proteins, Activin A was found to impede epiblast (EPI) lineage development while promoting trophectoderm (TE) differentiation, resulting in replacement of the EPI lineage of host embryos with PSCs. Interestingly, the injection of ESCs into blastocysts cultured with Activin A (cultured from 4-cell stage to early blastocyst at E3.5) could increase the contribution of ESCs to the chimera. The results indicated that PSCs secrete protein Activin A to improve their EPI competency after injection into recipient embryos through influencing the development of mouse early embryos. This result is useful for optimizing the chimera production system and for a deep understanding of PSCs effects on early embryo development.
