1.THE EFFECT OF METOCLOPRAMIDE ON EXPERIMENTAL ARRHYTHMIA
Chinese Pharmacological Bulletin 1986;0(05):-
Metoclopramide ( MCP ) is similar to Procainamide in structure. In rats anaesthetized by ip chloral hydrate 300 mg/kg, arrhythmia induced by BaCl 3 mg/kg was shown to be antagonized by injecting iv MCP 30 mg/kg. It was also found that MCP 10 mg/kg injected iv protected rabbits against a'rrhythmia induced by chloroform-adrenaline . In anaesthetized rats, MCP 30 mg/kg injected iv raised the dose of aconitine required to induce first arrhythmia, ventricular fibrillation ( VF ) and heart arrest, while in anaesthetized guinea pigs, the same amount of MCP injected iv showed no significant action on the dose of ouabain inducing these effects. Ventricular tachycardia produced by occlusion of coronary artery in rats could not be prevnted by injecting iv MCP 30 mg/kg. It was found that MCP has local anaesthesia action.
2.POSITIVE INOTROPIC ACTION OF DIMETHYLTRILOBINE IODIDE ON CARDIAC MUSCLE
Chinese Pharmacological Bulletin 1986;0(06):-
Dimethyltrilobine iodide (DMT) 1.5 M-M increased the contractile force of isolated guinea pig left atria. This positive inotropic action could not be an-tagonized by either propranolol, cimetidine or diphen-hydramine. DMT 1.5 nM also increased the contractile force of isolated guinea pig left atria exposed to low concentration of calcium or to verapamil.
3.EFFECT OF 4-PHOSP ATE-HYDROXYPIPERAQUINE ON THE ISOLATED AURICAL OF GUINEA PIG
Chinese Pharmacological Bulletin 1987;0(03):-
The effect of 4 -Phosphate- lydroxypiperaquine ( PHPQ) on the phsiologic properties in isolated auricle of guinea pig was studied.Twenty minutes after PHPQ 30 ?mol/L was added, the contractility & dT/dtmax of the left atria of guinea pig were decreased to 74.4 ? ( SD ) 5.3%(n=6, P

Journal of Xi'an Jiaotong University(Medical Sciences) 1982;0(01):-
After Sinomenine 27 ?M was given, the contractibility was decreased to 51?11% (SD) and dT/dt max to 50?14% (SD). The automaticity induced by epinephrine was inhibited. Sinomenine (2.7-270 ?M) caused a concentration-dependent prolongation in the functional period. The amplitude of contraction and the heart rate in the right auricle was also inhibited. The results indicate that sinomenine may be antogonistic to both Ca~(++) influx and inward Na~(++) current.

Journal of Xi'an Jiaotong University(Medical Sciences) 1982;0(04):-
The effects of amrinone on the contraction and action potential of isolated guinca pig papillary muscles were studied with intracellular microelectrodes. When the preparations were exposed to 25 ?mol/L amrinone, their pcak developed tension(DT) was inereased by 65.4%, the maximum rate of force development (dT/dt_(max)) was increased by 50.8% the time to peak developed tension (tPT) was shortened by 15.9%. By raising the concentration of amrinone to 250 ?mol/L, DT and dT/dt_(max) were increased by 98.5% and 91.3%, respectively, and tPT was shortened by 24.7%. Concomitantly, amrinone can prolong the durations of action potential (APD50, APD90)and the effective refractory period (ERP) of the ventricular cells. These results suggest that amrinone (made in China) possesses the obvious positive inotropic effect.

Chinese Pharmacological Bulletin 1987;0(02):-
The effects of dl-Tetrahydropalmatine ( THP ) on the physiologic properties in the auricle of guinea pig and on isoproterenol and calcium-mediated positive chronotropic action in guinea pig atria were studied.30 minutes after THP 52 nM was added, the contractility was decreased to 53+ ( SD ) 17%(P0.05) in the left auricle of guinea pig. Furthermore, the contractility and automaticity of the right auricle were decreased to 48+13% (P

Chinese Pharmacological Bulletin 1987;0(03):-
The effects of metoclopramide( MCP ) on the physiologic properties of guinea pig papillary muscles and the hemodynamics in anesthetic dogs were studied. MCP decreased contractility, increased the epinephrine concentration inducing automaticity and shifted the voltage-time plot to the right. But the functional refractory period was prolonged. Furthermore, the contractility and automaticity of the right auricle were decreased. MCP 5 mg/kg iv increased cardiac index (CI), systolic blood pressure ( SBP ), diastolic blood pressure ( DBP ), left ventricular systolic pressure ( LVSP ), maximal rate of change of intraventricular pressure ( dP/dtmax ), and total peripheral resistance ( TPR ) while MCP 10mg/kg iv decreased SBP, DBP, LVSP, dP/dtmax and TPR except increase of CI.

Le LI ; Feier ZHUANG ; Gengsheng ZHAO
Chinese Pharmacological Bulletin 1987;0(03):-
It was reported that Os-thole(Ost) was isolated from the fruits of Cnidium monnieri(L. ) Cusson, possessesanti - arrhythmic action and depressed the aortic pressure action in animal studies. Its calcium -antagonistic effects were studied on the isolated rabbit aortic strips.Ost shifted the dose response curves for NE, CaCl2 and KC1 nonparallelly to the right, and depressed their maximal response; pD2' value is 3. 16, 4. 28 and 4. 39 respectively. It produced neither ?- a-drenoceptic stimulating nor ? - adreno-ceptic blocking effects. At the concentration of 100 ?mol ? L-1, Ost significantly inhibited the intra cellular Ca2+ dependent component of NE - induced contraction of the aortic strips, however, it didn't in-hibit the extra cellular Ca2+ dependent component of NE - induced contraction of the aortic strips, These results indicated that Ost produced relaxation on vascular smooth muscle might result from its antagonistic effect to Ca2+ and mainly blocked calcium influx through PDC, and the mode of its anta -gonism is similar to that of verapamil.

Journal of Xi'an Jiaotong University(Medical Sciences) 1981;0(03):-
Osthole, the coumarin isolated from the fruits of Cnidum monnieri (L.) Cusson. The effects of osthole on the physiological properties in isolated auricle of guinea pig were studied. It possesses dose—dependent negative inotropic effects, the BaCl_2 inducing automaticity was inhibited, and the functional refractory period (FRP) was prolonged, and the frequence of sinoatrial node was slowed The negative inotropism and automatism suggest that sothole mey be antagonis tic to Ca~(2+), while the decrease of excitability and the prolongation of FRP indicate that it may be due to its inhibition on Na~+ current. The decrease of automaticity and excitability and prolongation of FRP are thought to be its antiarrhythmic action.
