1.Relationship between the pancreatic stellate cells and the angiogenesis of pancreatic carcinoma

Pancreatic cancer is still a dismal disease.Angiogenesis is very important for the development of pancreatic cancer.Pancreatic stellate cells (PSCs) are the main source of extra-cellular matrix of pancreatic cancer and they provide advantageous microenvironment for cancer cells.PSC could promote the angiogenesis of pancreatic cancer both in vitro and in vivo.Further studies of the angiogenesis of pancreatic cancer are helpful in learning the characteristics of development and metastasis of pancreatic cancer,and provide new treatment method in the cellular and molecular levels.
2.Studies on Chemical Constituents from Rubus pinfaensis

Objective To study the chemical constituents from dry roots of Rubus pinfaensis. Methods The compounds were separated from alcohol extracts of the roots of Rubus pinfaensis with silica gel column chromatography,and identified using IR,1 H-NMR and EI-MS methods. Results Five compounds were isolated and identified as oleanolic acid (Ⅰ) ,3-keto,16α-hydroxy,24-noroleanolic acid (II) , 3 '-methoxy myricetin (Ⅲ) , 2 '-deoxyuridine (Ⅳ) and p-hydroxybenzoic acid (Ⅴ) . Conclusion CompoundsⅠ,II,Ⅲ,ⅣandⅤwere isolated from Rubus pinfaensis for the first time.
4.The role of musculoskeletal ultrasound in the treat-to-target in rheumatoid arthritis

The treat-to-target (T2T) strategies has greatly improved the prognosis of rheumatoid arthritis (RA).However,the emergence of musculoskeletal ultrasound has further brought new challenges and opportunities for RA T2T treatment.It has been revealed the subclinical synovitis generally presented in RA patients who achieved clinical remission.The subclinical synovitis has been confirmed to be associated with subsequent progression of bone erosion and flare.Therefore,clinical remission or low disease activity may be inappropriate or insufficient target.The ultrasound evaluation in addition to clinical assessment can help to control the disease activity better,which may eventually improve the long-term outcomes of RA patients.
5.Influencing factors of the formulation and preparation on in vitro drug release from sinomenine hydrochloride delayed-onset sustained-release tablet

AIM: To study the formulation and preparation factors on vitro drug release from sinomenine hydrochloride delayed-onset sustained-release tablet. METHODS: With hydrophilic matrix materials as excipient,the tablets containing hydrochloride sinomenine as a model drug were prepared by direct compression. The effect of the type and amount of tablet core matrix materials(HPMC K15M,HPMC K4M,xanthan gum and carrageenan),the type and amount of coating matrix materials,the preparation of coating materials and the pressure on in vitro drug release of the tablets were studied. RESULTS: The lag time of the tablet was 4 ~ 5 h and drug release slowly in 24 h. 6.Analysis of 60 Operated Cases of Giant—cell Tumor of Bones

60 cases of Giant-cell tumor of bones are reported in this paper.Of the 60 cases, 37 were males and 23 females. Their age was 16-67 andthe average age was 31. 75%. 75% was in the age of 21-40 5 were undertwenty and 4 out of them were females. The knee region was the commonest site. In 16 patients tumors werein the distal part of the femur, 13 in the proximal part of the tibia, 7 inthe proximal part of the femur. According to the different forms of the stromatic cells of the tumor.they were divided into spindle cell type(F) and ovalcell type (H). Thesurvial rate for five years of type F were obviously higher than that of typeH. However, lung metastasis were found in type H than in type F. Onthe other hand, in the pathological pattern of the metastatic cases of thelung 75% got plentiful blood sinusoid structures. All of this group underwent different opeations. Four cases were givenbiopsy. Sixteen cases were curetted and packed with bone chips with therecurrence rate 13. 3%. Sixteen cases were enbloc removal of the tumorand arthrodesis with the recurrence rate 13. 3%. Six cases (grade Ⅲ) wereamputated. All of them died within two and a half years. Fifty-five cases were followed and in two yaers seven out of themdied, and local recurrence was present in one case and lung metastasis wasfound in one case. Fourty-six cases were followed for 2 to 20 yearsand the average period was 7 years. There were 2 deaths, 2 local recurre-nces and one lung metastasis. The total recurrence-death rate was 25. 45%.

8.Construction of vascular endothelial growth factor modified muscle-derived stem cells and its expression

Objective To construct the lentiviral vector encoding vascular endothelial growth factor gene and detect the vascular endothelial growth factor (VEGF) expression in muscle-derived stem cells (MDSCs). Methods To culture MDSCs and detect the CD34,CD45,Bcl-2 and Desmin expression in MDSCs by immunofluorescence. A cDNA encoding VEGF gene was amplified by PCR. This fragment was cut with EcoRI and BamHI and ligated with an EcoRI- and BamHI-reated lentiviral vector pCDH-CMV-MCS-EF1-copGFP. Then DNA sequencing analysis was performed to confirm successful construction of pCDH-CMV-MCS-EF1-copGFP -VEGF. The expression of VEGF was confirmed using enzyme-linked immunosorbent assay (ELISA), Western blot, and Real-time PCR analyses. Results The pCDH-CMV-MCS-EF1-copGFP-VEGF lentiviral vector was constructed successfully. When MOI values in the transfection efficiency MDSCs by FCM. were 1,5,15, the transfection rate reached to 16.7%, 45.6%, 66.3% and 85.6% respectively. When MOI value was of 20, the rate was up to 90.1%. Real-time PCR, Western blot and ELISA showed stable expression of VEGF in MDSCs. Conclusion We successfully constructed lentiviral vector carrying the VEGF and stable expression in MDSCs.

Objective To evaluate the level of granzyme B mRNA and perforin mRNA in urine on the diagnosis of renal acute rejection. Methods A total of 34 cases of renal transplantation were studied.The urine samples were measured with the use of a competitive and quantitative polymerase-chain-reaction assay for granzyme B mRNA and perforin mRNA and the data were analysed by using the SPLM statistical software,mRNA levels were log-transformed. Objective To evaluate the role of autophagy in inflammatory response in the lungs of silicosis rats. Methods The
specific pathogen-free healthy male Wistar rats were randomly divided into four groups with 10 rats in each group. The rats in the
control group were given an equal volume of 0.9% sodium chloride solution; all rats in the silicon dioxide (SiO2
) group, the SiO2
+
3-methyladenine (3-MA) group, and the SiO2
+rapamycin (RAPA) group were given a mass concentration of 0.05 mg/L of SiO2
suspension (1.0 mL/rat) to establish a rat model of silicosis using non-exposed tracheal instillation method. Two days before
SiO2 exposure, the rats in SiO2
+3-MA group were intraperitoneally injected with 3-MA at a dose of 2.5 mg/kg body weight. The rats
in the SiO2
+RAPA group were intraperitoneally injected with a dose of RAPA 1.0 mg/kg body weight, once a day, and once every
other day after modeling, for a total of 10 injections. The pathological changes in the lung tissue of the rats in each group were
evaluated using hematoxylin-eosin and Masson staining. The relative expression levels of transforming growth factor-β (TGF-β) ,
interleukin-1β (IL-1β) and tumor necrosis factor-α (TNF-α) in the lung tissue of the rats in each group were detected using
enzyme-linked immunosorbent assay. The relative expression levels of microtubule-associated protein 1 light chain 3 (LC3) and
yeast autophagy-related gene 6 (Beclin1) in lung tissue of rats in each group were detected by Western blotting. Results The
pathological observation of rat lung tissue showed that scattered inflammatory nodules and interstitial inflammatory cells were
found in the lung tissue of the rats in the SiO2 group. In the SiO2
+3-MA group, the inflammation in the lung tissue was more
severe and the alveolar cavity had viscous secretions. The rats in the SiO2
+RAPA group had less inflammation and smaller
inflammatory nodules than the SiO2 group. Compared with the control group, the TNF-α, IL-1β, TGF-β, Beclin1 protein relative
expression levels and LC3 Ⅱ/Ⅰ ratio in the lung tissue were increased in the SiO2 group (all P<0.05). Compared with the SiO2
group, the relative expression levels of TGF-β and TNF-α in the lung tissue of the rats increased (all P<0.05), and the ratio of
LC3 Ⅱ/Ⅰ and Beclin1 protein relative expression decreased in the SiO2+3-MA group (all P<0.05). Compared with the SiO2+
3-MA group, the relative expression levels of TNF- α, IL-1β and TGF- β in the lung tissue of the SiO2
+RAPA group were
decreased (all P<0.05), and the LC3Ⅱ/Ⅰ ratio and the relative expression level of Beclin1 protein were increased (all P<0.05).
Conclusion Autophagy occurs when inflammatory reaction occurs in the lungs of silicosis model rats; autophagy has inhibitory
effect on pulmonary inflammation.
