1.Screening and preliminary research of an actinomyces strain with antitumor effect

The apoptosis was observed under a fluoroscope when the tested tumor cells were treated with the supernatant derived from the liquid culture of an actinomyces strain AK 11, which had showed cell toxicity by means of MTT assay, and was further confirmed by the correspondent band of ”DNA ladder” in the agarose gel electrophoresis. In a bioassay employed nude mouse, the pure compound produced by the strain AK 11 showed tumor inhibition in rates of 52.01% and 33.64% corresponding to the dosages of 0.016 ?g?kg -1 and 0.008 ?g?kg -1 ip, respectively. According to the indexes IC 50 of human hepatocarcinoma SMMC7721 and stomach cancer MGC 803 cells, it was estimated that the inhibition effect of the pure compound was slightly stronger than that of Adriamycin. The results showed high antitumor effect of the tested compound from the strain AK 11. When the cells were treated with AK 11, it was found by flow cytometry analysis that cell division was ceased at S phase due to the DNA synthesis block .
2.NS-398, a selective cyclooxygenase-2 inhibitor, can down-regulate the expression of P-glycoprotein in the gastric cancer cell

Objective To investigate the effect of NS-398, a selective cyclooxygenase-2 (COX-2) inhibitor, on the expression of P-glycoprotein(P-gp) in a gastric cancer cell line SGC-7901. Methods SGC-7901 cells were treated with NS-398 in different concentrations (0, 10 ?mol/L and 100 ?mol/L) respectively. Prostaglandin E2(PGE2) was detected by ELISA. Twenty four hs and 48 hs later, the expression of mdr1 mRNA were detected by RT-PCR. Objective To investigate whether RNA interference in mediated Bcl-2 gene silence can decrease the gallbladder carcinoma xenograft formation rate and inhibit growth of xenograft in nude mice.Methods In the xenograft formation ability group,the Bcl-2-siRNAs were transfected into the gallbladder carcinoma cell.Transfected gallbladder carcinoma and gallbladder carcinoma were injected into nude mice hypodermically to form a gallbladder carcinoma tumor.The formation rate and volume of the tumors were measured and the tumor growth rates were calculated.In the therapy group,the animal models of human gallbladder carcinoma in nude mouse body were constructed.The nude mice were randomly divided into pSilencerTM-EGFP sh515 group,pSilencerTM-EGFP shCon group,and normal control group.After the formation of tumor knot,siRNA expressing plasmids pSilencerTM-EGFP sh515 and pSilencerTM-EGFP shCon were injected into the knots of pSilencerTM-EGFP sh515 group and pSilencerTM-EGFP shCon group.The tumor volume was measured and tumor growth rate was calculated.All mice were sacrificed after 6 weeks.The distribution of Bcl-2 in the tumors was detected by immunohistochemistry.Results The nude mouse model of human gallbladder cancer xenograft tumor was constructed successfully.Xenograft formation ability:The xenograft formation rate of gallbladder carcinoma and Bcl-2 siRNA transfected gallbladder carcinoma was 100% and 60%,respectively;the average tumor volume in gallbladder carcinoma group and transfected gallbladder carcinoma group was(1 914.6?125.0)mm3 and(629.7?78.9)mm3;the growth rate was 45.58% and 14.99%,and the average tumor weight was(2.24?0.33)g and(0.77?0.12)g,respectively.The tumor volume,weight and growth rate in the transfected gallbladder carcinoma group decreased significantly when compared with those in control group(P
4.Design and Application of a Kind of Controllable Enema Input Apparatus for Patients with Chronic Constipation post Spinal Cord Injury

Objective To design and apply a kind of controllable enema input apparatus in patients with spinal cord injury (SCI). Methods 20 SCI patients with chronic constipation were included. Their defecation time, nature of defecation and usage of enema of 5 times of defecation were recorded before and after using the new apparatus. Results There were significant differences in the defecation time, nature of defecation and usage of enema before and after using the new apparatus (P<0.05). Conclusion The application of the new controllable enema input apparatus may shorten the defecation time, improve the nature of defecation, and reduce the usage of enema in SCI patients with chronic constipation.
5.Pathophysiology and Treatment of Secondary Hydronephrosis after Spinal Cord Injury (review)

Hydronephrosis after spinal cord injury (SCI) may result in renal insufficiency which ranks as the most common late cause of death in SCI patients. SCI may cause vesicourethral dysfunction of nerve regulation, which in turn brings about functional obstruction, high intravesical pressure, increase of the incidence and frequency of detrusor uninhibitory constractions, leading to hydronephrosis. The paramount principle of prevention and treatment of hydronephrosis is keeping low vesical pressure (storage pressure <40 cmH2O, voiding pressure <60 cmH2O). Oral anticholinergics combined with intermittent catheterization are the best choice for the slight hydronephrosis, urinary operations such as botulinum toxin injection into detrusor, transurethral sphincterotomy, ileocystoplasty may be needed for the moderate and severe hydronephrosis.
7.Relationship between NF-κB p65 activation and k-ras gene mutation in colorectal cancer

Objective To investigate the corresponds between NF-κB p65 activation and k-ras mutations.Methods NF-κB p65 activation was analyzed by immunochemistry in 167 colorectal cancer specimens in which the k-ras mutation status was confirmed.Resluts Among 167 colorectal cancer specimens screened,59 (35.3 %) had k-ras mutations and 63 (37.7 %) had NF-κB p65 activation.k-ras mutations and NF-κB p65 nuclear expression were no significant difference in different sex,age,ECOG score,pathological types,degrees of pathological differentiation,TNM stage,and metastases on lymph nodes (P> 0.05).The positive nuclear expression rate of NF-κB p65 was 50.8 % (30/59) in specimens with k-ras mutations and 30.6 % (33/108) in specimens with wild type k-ras.There was obviously statistical difference between them (P =0.010).Conclusion NF-κB p65 activation in coloretal cancer was associated with k-ras mutations.
8.Study on Living Related Liver Transplantation for Treating Wilson′s Disease in Children

Objective To explore the liver pathology from live related liver transplatation(LRLT) of Wilson′s disease(WD) in children,and evaluatethe indication of LRLT.Methods The sample of this study,including the donater and patient,came from the LRLT. It was observed with HE,MASSON,Timm′s and Rubeanic staining.Results With HE and MASSON stainning,hepatocyte showed degeneration,fiber hyperplasic and false lobule formed;Timm′s and Rubeanic stainning showed typical black deposit of granules and conglomerations.Liver pathology revealed that liver cirrhosis appeared,and it was diffrentent from Child Pugh.Conclusions It is of great significance that the liver pathology is useful for the treatment of WD.If liver pathology supportes,the indication of LRLT can be measurably broadened.

Objective To observe the change of platelet activity marker in hypertensive patients complicated with ischemic cardiovascular and cerebrovascular disease ,and assess it s value in the happening of thrombotic disease. Method The platelet activity marker (CD62p,CD63) were measured by flow cytometry in 40 hypertensive patients complicated with ischemic cardiovascular and cerebrovascular disease (group C ),46 hypertensive patients not complicated with ischemic cardiovascular and cerebrovascular disease (group B),and 30 cases of healthy control (group A). 10. Chemical constituents from roots of Breynia fruticosa

Objective: To study the chemical constituents from the roots of Breynia fruticosa. Methods: The compounds were isolated by comprehensive column chromatography, and the structures were elucidated by spectral methods. Results: Thirteen compounds were isolated and elucidated, including four triterpenoids, friedelin (1), friedelinol (2), lupenone (3), and glochidiol (4); three steroids, including β-sitosterol (5), stigmastane-3β, 6β-diol (6), and β-sitosterylglucoside-6'-octadecanoate (7); two cerebrosides, including 1-O-β-D-glucopyranosyl-(2S, 3R, 4E, 8Z)-2-[(2-hydroxyoctadecanoyl) amido]-4, 8-octadecadiene-1, 3-diol (8) and 1-O-β-D-glucopyranosyl-(2S, 3S, 4R, 8Z)-2-[(2R)-2-hydroxypentacosanoylamino]-8-octadecene-1, 3, 4-triol (9); and four other compounds, including (-)-epicatchin (10), ε-caprolactone (11), aviculin (12), and vanillin (13). Conclusion: Compounds 3, 4, 6-9, and 11 are isolated from the plant of Breynia J. R. et G. Forst. nom. cons. for the first time, and compound 11 is isolated from the natural product for the first time.
