1.Clinical Study on the Relationship Between the p53 Gene Examination and Colorectal Neoplasms

Objectives To study the clinical significance of the early diagnosis on colorectal cancer and the value of monitoring precancerous lesions of p53 gene examination. Methods Histological specimens of tumor from 110 patients with colorectal neoplasms were collected under the endoscopy, another 10 normal intestinal mucosa tissues with benign enteropathy as a control group. By PCR-SSCP methods, examined p53 exon 5~8 of mutation circumstance, made pathological section at the same time, and combine to track the observation. Results 24 patients p53 gene mutation were detected out in 78 cases colorectal cancers,counted up 32 4%, among 17 patients were moderate,low differentiation, 3 patients of them were mucinous adenocarcinoma.There are 6 patients p53 gene mutation in 36 cases colorectal adenoma,counted up 16 7%, among them an patient of Ⅱ~Ⅲ class atypical hyperplasia adenoma occurrence canceration in the next 10 months.There was no p53 gene mutation in normal intestinal mucosa tissue.Conclusions p53 gene examination combinated with pathological examination could monitor precancerous lesions and early diagnosis of the colorectal cancer.
2.Isolation,purification and identification of human periodontal ligament stem cells

Objective:To isolate and identify the periodontal ligament stem cells.Methods:Periodontal ligament tissues were digested with a mixture of collagenase and Dispase,then the periodontal ligament stem cell clones were isolated by limited dilution method.The transmission electron microscopy(TEM),flow cytometery and immunohistochemistry procedure were employed to study the ultrastructure,cell cycle and surface marker of the cloned cells.Results:The obtained cells showed the characteristics of undifferentiated morphology at ultrastructural level.94.4% of the cells were in phase G_0/G_1.The cells were Vimentin,STRO-1,ON and BSP positive.Conclusion:Cloning incubation may be the effective way to isolate and purify the periodontal ligament stem cells.
3.Effects of Glutamine Combined with Growth Hormone on Cytokine and Amino Acid Metabolism in Septic Rats

Objective To study the effects of glutamine (Gln) combined with growth hormone (GH) on the levels of cytokine (TNF-?, IL-1, IL-6), coritsol and amino acid metabolism in septic rats. Methods Ten out of 79 SD rats were randomly collected as the control group. Thirty of 69 septic SD rats, which were made by cecal ligation and perforation (CLP) method and were given parenteral nutrition (PN) lived to day 6. They were also randomly divided into three groups as follows: septic group (n=10), parenteral supplemented glutamine group (Gln group, n=10), and Gln combined with GH (Gln+GH group, n=10). On the 6th day, blood drew from portal veins of the dead rats was used to detect the levels of TNF-?, IL-1, IL-6 and cortisol by ELISA. The plasma concentrations of free amino acids were determined by amino acid auto-analyzer. The muscle tissue of extensor digitorum longus was used to determine 3-methyl-histidine (3-MH) by high performance liquid chromatographic (HPLC). Results Except for the control group, most rats developed celiac abscess, hepatic abscess and pulmonary infection. Objective To learn the control status of iodine deficiency disorders in Baoji City of Shaanxi Province.Methods From 2009 to 2011,according to the National Iodized Salt Monitoring Program,the iodized salt was monitored,and thyroid was examined in 12 counties(areas) of Baoji City.According to rural and urban area stratification,60 children aged 8 to 10 were selected in each county(district),and their urine samples were collected for determination of iodine content.In 2011,per capita daily salt intake was surveyed by the three weighing method at children's home whose urine was collected.Chencang Area,Fufeng County and Taibai County,representatives of Baoji City different geomorphic features were selected for investigation of water iodine,and urinary iodine of pregnant and lactating women,and 15 people were selected in each county (district).Results From 2009-2011,in the 12 counties(areas) in Baoji City,the coverage rate of iodized salt was all 100.00%(3468/3468); the qualified rate of iodized salt and the consumption rate of qualified iodized salt were all > 99.00%.Children's goiter rate was 3.41%(87/2548),3.06%(77/2520) and 3.33%(84/2520),and they were all less than that of the national standard (< 5%).Medians of urinary iodine of 8-10 years old children were 368.20,293.80 and 332.50 μg/L,respectively,and the ratios of urine iodine ≥300 μg/L were accounted for 66.42% (797/1200),48.05%(692/1440) and 56.67%(816/1440),respectively.Median urinary iodine of pregnant women was 301.81 μg/L and lactating women was 329.79 μg/L.A total of 1116 households were investigated,the median of per capita daily salt intake was 8.9 g.Eighteen water samples were collected,range of water iodine value were 0.60-10.25 μg/L.Conclusions Iodine nutrition in general population of Baoji City is exceeded the optimum level,and the current iodized salt concentration has some down space,but fully consideration should be taken on iodine needs in different iodine deficiency areas and among different groups of people.
8.Effects of enteral and parenteral nutrition on gut epithelial tight junction and immune barrier of rats with abdominal infection

Objective To investigate the effect of enteral and parenteral nutrition on gut microecology in rats with abdominal infection. Methods Fourteen Sprague-Dawley (SD) model rats of abdominal infection, which had survived for more than 6 days were divided into two groups: PN group ( n =7) and PN+EN group ( n =7) via jejunostomy and jugular vein respectively for another 5 days. The nutrition support in the two groups was isonitrogen and isocaloric. At sacrifice on the sixth day, occludin and IgA level in plasma cells of intestine epithelium of the gut were measured by immunohistochemistry. Vena cava blood and homogenated tissue of liver, lung and mesenteric lymph nodes were cultured to determine bacterial translocations, and portal vein blood was tested for endotoxin. 9.Assessment of the effects of mesenchymal stem cell transplantation in the treatment of myocardial infarction at high altitude area

Objective To evaluate the changes of cardiac function and myocardial perfusion by Gated 99Tcm-MIBI myocardial perfusion imaging after autologous mesenchymal stem cell implantation in patients with acute myocardial infarction at high altitude area.Methods 33 patients with anteroseptal myocardial infarction were ran- domly divided into two groups.18 patients (control group) underwent percutaneous tranluminal coronary angioplasty (PTCA) and 14 cases (transplantation group) received additional mesenchymal stem cell transplantation.Myocardi- al perfusion imaging were performed in all patients before and at 6 and 12 months after treatment.Results Com- pared to pre-implantation,LVEF of transplantation group was improved 8%~9%after 6 months.The improving lev- els of control group were lower.However,there were not statistical differences among all data.Conclusion Mesen- chymal stem cell transplantation could improve myocardial systolic function and myocardial perfusion.
10.Postoperative binocular visual function in 28 patients with congenital superior oblique palsy

AIM: To investigate the postoperative changes of binocular visual function in patients with congenital superior oblique palsy.METHODS: Eye position and binocular visual function were examined in 28 patients with congenital superior oblique palsy pre- and post-operatively. The results were statistically analyzed.RESULTS: The normal eye position rate after operation was 89.3%. There were 9 patients who gained simultaneous perception after operation, while 13 patients gained fusion as many as that gained stereoacuity. There was statistically significant difference between postoperative and preoperative eyes (P<0.05). Near stereoacuity was improved after operation. Patients who had better visual acuity and low strabismus degrees as well as fusion had a better recovery.CONCLUSION: The operation could improve the eye position and rebuild the binocular visual function of the congenital superior oblique palsy. The recovery of binocular visual function was related to visual acuity and strabismus degrees of the patients and whether they had fusion function.
