1.Effects of Taxol on proliferation, apoptosis, and mRNA expression of α2,6-sialic acid and ST6Gal in cervical carcinoma cell line U14

AIM:To study the effect of Taxol on the proliferation, apoptosis, and mRNA expressions of α2,6-sialic acid (SA) and α2,6-sialyltransferase (ST6Gal) in mouse cervical cancer cell line U14.METHODS:After the U14 cells were treated with Taxol, the IC50 value of Taxol to U14 cells was detected by MTT assay.The expression of α2,6-SA and apoptosis-related factors (Bcl-2, Bax, caspase 8 and caspase 3), the apoptosis rate and cell cycle were determined by flow cytometry.The mRNA expression of ST6Gal1 and ST6Gal2 was detected by qPCR.RESULTS:As compared with control group, Taxol induced obvious U14 cell growth inhibition, reduced α2,6-SA expression, up-regulated Bax, down-regulated Bcl-2, decreased the ratio of Bcl-2/Bax, enhanced caspase 8 and caspase 3 activity, increased the apoptotic rate and cell proportions of Sub-G1 and S phases, and induced G2/M phase arrest.Taxol also down-regulated the mRNA expression of ST6Gal1, and slightly up-regulated the mRNA expression of ST6Gal2.CONCLUSION:α2,6-SA and ST6Gal are involved in the multiple effects of Taxol on modulation of the cell cycle and apoptosis in U14 cells.
2.Clinical Study on NGAL and Iron Status in Patients with Chronic Renal Anemia

Objective To evaluated the association between the NAGL and the chronic kidney disease (CKD) patients with iron status,which was occurred iron scarcity for CKD patient in clinical.Methods 154 CKD patients were included in this study.Serum neutrophil gelatinase-related lipid transport protein(NGAL) were detected in anemia group and non-anemia group of chronic kidney disease.The NGAL,univariate correlation and multivariate regression analysis,as well as ROC curve analysis,were used to assess and compared the diagnostic potential of NGAL,ferritin,EPO and Tfr (transferrin receptor) in CKD patients after analyzing the patient's iron status.Results NGAL levels were markedly higher in CKD patients with anemia than CKD patients without anemia.In the analysis,NGAL was inverse correlated with hemoglobin,hematocrit,MCV,MCH,serum iron and TSAT(correlation relation was-0.36,-0.32,-0.26,-0.31,-0.38,-0.3,respectively).ROC analysis was employed to assess and compare the diagnostic potentials of the NGAL,ferritin,EPO,and Tfr (transferritin receptor) in identifying this condition among CKD patients.The NGAL showed that the area under the curve of iron deficiency in CKD patients (AUC) was 0.713,with good sensitivity and specificity (58.6% vs 74.7 %).Conclusion NGAL might be proposed as a new tool in the assessment of iron deficiency and in the management of iron therapy for CKD patients.
3.Clinical effect and adverse reaction of tiggio combined with oxaliplatin on the treatment of advanced gastric

Objective To explore the clinical effect and adverse effects of tiggio combined with oxaliplatin on the treatment of advanced gastric cancer. Methods From January 2014 to January 2017, 88 cases with advanced gastric cancer were selected as the samples in Yangling demonstration zone hospital. All the patients were randomly divided into the control group (n = 44) and the observation group (n = 44). The control group and the observation group were treated with tiggio and tiggio combined with oxaliplatin respectively, and the clinical effect and the occurrence of adverse reactions in the two groups was compared. Results The total effect in the observation group was 91.0%, which was significantly higher than that in the control group (75.0%), (P<0.05); The incidence of adverse reactions in the control group was 27.3%, while in the observation group was 9.1% the difference was statistically significant(P<0.05). Conclusion Tiggio combined with oxaliplatin in the treatment of advanced gastric cancer, the clinical effect is significant, the incidence of adverse reactions is low, high security, worthy of promotion.

6.Failure of Batch Monitoring System During Sterilization Process:Analysis of Causes

OBJECTIVE To prove the batch monitoring system to be effective and practicable.METHODS The sterilization process of each batch with the batch monitoring system was monitored.RESULTS Among monitored 7057 6 batches failed,the failure rate was 0.09%.CONCLUSIONS By analyzing the failure causes,we should enhance the maintenance of the sterilizator and limit the using times of the monitor device.It proves that the batch monitoring system is effective and practicable to monitor the pulse-vacuum-pressure-steaming sterilizator.

10.Cloning and Optimized Prokaryotic Expression of a pbmag-1 cDNA Fragment from Plasmodium berghei ANKA

Objective To clone and express a novel gene cDNA fragment,pbmag-1,from Plasmodium berghei ANKA strain. Methods The cDNA sequence of pbmag-1 was obtained from the GenBank of P.berghei ANKA genomic databases,with which a pair of primers was designed and RT-PCR was used to get a cDNA fragment of the gene from the parasite. The expanded cDNA 3′ fragment of the gene was obtained by 3′-RACE using the oligo dT primer and a set of specific primers. The intact cDNA 3′ fragment was cloned into a prokaryotic expressional vector and transformed into the BL21-(DE3)-RIL strain of Escherichia coli. The recombinant protein of PbMAg-1 was expressed with an optimized strategy and used to immunize mice. Results The pbmag-1 cDNA fragment obtained was 1 341 bp in length,A/T rich (73%) and with a correct 3′ end sequence. By Western blot,the anti-serum of mice immunized with the recombinant protein of PbMAg-1/GST,which was expressed as inclusion bodies,specifically recognized a band with Mr 64 000 molecule from the protein extracts of P. berghei-infected mouse erythrocytes. Conclusion The pbmag-1 cDNA sequence with intact 3′ has been obtained,which will be used for further study on its role in the immune response of P. berghei infection.
