3.Human erythropoietin-modified human amniotic mesenchymal stem cellsvia subarachnoid transplantation promote neurologic recovery from brain injury
Chinese Journal of Tissue Engineering Research 2016;20(23):3432-3438
BACKGROUND:Studies have shown that as a regulator of bone marrow functionerythropoietinis a glycoprotein that controls the development of the central nervous system and has neurotrophic and neuroprotective potential. Therefore, transplantation of human amniotic mesenchymal stem cels geneticaly modified by human erythropoietin is a new choice for brain injury treatment.
OBJECTIVE:To observe the effect of transplantation of human amniotic mesenchymal stem cels geneticaly modified byhuman erythropoietin on the functional recovery from brain injury in rats.
METHODS:Eukaryotic expression plasmid pcDNA3.1 carrying erythropoietin was successfuly constructed and transferred into amniotic mesenchymal stem cels culturedin vitro. Expression of erythropoietin was detected using western blot assay before and after transfection. Rat models of middle cerebral arterial occlusion was made and given transplantation of transfected amniotic mesenchymal stem celsviathe tail vein (transfection group). Additionaly, model and simple cel transplantation groups were set in a comparative study.
RESULTS AND CONCLUSION:Findings from western blot detection showed that transfected cels could express human erythropoietin. Compared with the other groups, modified neurologic severity scores, growth-associated protein 43 and aquaporin 9 at mRNA and protein levels were al decreased significantly in the transfection group. Furthermore, the number of cels positive for CM-Dil was highest in the transfectiongroup, folowed by simple cel transplantation group, and lowest in the model group (alP<0.05). Overal findings from this study show that human erythropoietin-modified human amniotic mesenchymal stem cel transplantation promotes neurologic recoveryfrom brain injury through eliciting a reduction in growth-associated protein 43 and aquaporin 9 at mRNA and protein levels as wel as inhibiting cel apoptosis.
4.The experimentation about the expression of Lin28b and its clinical relevance in Pancreatic cancer
Journal of Chinese Physician 2016;(z1):1-4
Objective Through detected the expression degree of Lin28b in three different malig-nant degree pancreatic cancer cell lines and human pancreatic cell line to study the relationship of pancreatic cancer and the express of Lin28b.Methods The RT-PCR was used to detect the expression degree of mR-NA in Lin28b in cell lines of PANC-1,BxPC-3,AsPC-1,HPC-Y5 and Western-blot was used to detect the expression degree of protein in Lin28b in cell lines of PANC-1,BxPC-3,AsPC-1,HPC-Y5,then discovering the differential expression. Results The Lin28b was high expression in three pancreatic cancer cell lines, also higher than human pancreatic cell lines, and the expression of difference had statistical significance. Conclusions The results suggested that Lin28b may play a role in the pathogenesis of pancreatic cancer, and there is a certain relationship between the expression of Lin28b and malignant degree of tumor cells.
5.Arthroscopic core decompression with autologous bone marrow induction material combined with titanium rod in the repair of stage II femoral head necrosis
Chinese Journal of Tissue Engineering Research 2015;(28):4460-4464
BACKGROUND:Although core decompression can contribute to the reconstruction of necrotic bone, its single use appears to have some deficiencies and cannot completely realize the reconstruction of the femoral head.
OBJECTIVE:To explore the effect of arthroscopic core decompression with autologous bone marrow induction material combined with titanium rod in the repair of stage II femoral head necrosis
METHODS:A retrospective analysis was performed on clinical data of 79 cases of stage II femoral head necrosis admitted at the South Branch of Tongchuan People’s Hospital from February 2011 to February 2013. According to the therapeutic methods, these cases were divided into control group (40 cases) and observation group (39 cases), and were given conventional core decompression and arthroscopic core decompression with autologous bone marrow induction material combined with titanium rod. Patients were fol owed up for 24 months, and the range of hip flexion and Harris scores on hip function and adverse events were observed and compared between two groups.
RESULTS AND CONCLUSION:Al the 79 patients successful y completed the 24-month fol ow-up. By the last fol ow-up, the range of hip flexion and Harris score were significantly higher in the observation group than the control group (P<0.05). There was no failure case due to surgical material rejection. To sum up, arthroscopic core decompression with autologous bone marrow induction material combined with titanium rod is safe and effective for repair of stage II femoral head necrosis.

8.Clinical study of chemoradiation therapy in rectal cancer
China Oncology 2001;0(05):-
Rectal cancer is a common malignant tumor in the gastrointestinal system. For the patients with resectable but high-risk rectal cancer, neoadjuvant and adjuvant chemoradiation plus surgery may improve local control.Compared to adjuvant chemoradiation, neoadjuvant chemoradiation showed the advantage of sphincter preservation. When the goal of preoperative chemoradiation was sphincter preservation, conventional radiotherapy was recommended.Through the analysis of the patients with 5-year survivals, the radiation had a similar impact on the survival as the chemotherapy alone for the patients with the intermediate risk factors. The impact on local control need to be further investigated before recommending chemotherapy alone for those subset patients.Advances in preoperative irradiation, postoperative irradiation and the comparison of preoperative irradiation with postoperative irradiation are also reviewed in this article.
9.Advance on the research and clinical treatment of gastrointestinal stromal tumors
China Oncology 2006;0(10):-
Gastrointestinal stromal tumors(GISTs) are rarely one of gastrointestinal carcinomas that originate from mesenchymal tissue.GISTs are characterized by expression of CD117 receptor in cells and have variable biological phenotypes ranging from benign to highly malignant. The most common complaints of patients with GISTs are abdominal pain, peptic ulcer, bleeding or/and obstruction in digestive tract.GISTs have an extraordinarily high rate of recurrence after surgical resection and are highly resistant to radiation and standard chemotherapy. The imatinib mesylate(Gleevec) is a selective inhibitor of the tyrosine kinase receptor. The outcome for the patients with GISTs treated by Gleevec was promising in terms of immediate response and survival fraction.Recent research shows that a new targeting therapeutic agent,SU11248,can prolong the survival time of the patients with GISTs who were resistant to Gleevec therapy.
