1.Effect of retroviral HSVtk gene transfer on mice bladder tumor growth in vivo

Objective:To evaluate the effectiveness of intratumoral and intraperitoneal injection with retrovirus HSVtk gene on mouse bladder cancer.Methods:Subcutaneous and intraperitoneal bladder tumor was respectively established with mouse cancer cell line(T739) in syngeneic mice.Direct intratumor injection of retroviral HSVtk into the subcutaneous tumor was performed.In peritoneum cavity tumor model,HSVtk gene was transferred by intraperitoneal injection of retrovirus vector carrying HSVtk gene.After GCV therapy the tumor volume and animal survival time were monitored.Results:Both animal models were successfully induced in all mice within one week following subcutaneous and intraperitoneal injection of T739 cells.Compared with control animals,the tumor growth in both models was significantly inhibited with HSVtk/GCV system,and also the animal survival time was prolonged with administration of GCV(P
2.Clinical observation of breast-conserving operation combined with radiotherapy for 43 patients with breast cancer

Objective To investigate the efficacy,side effects and cosmetic effects of breast-conserving operation combined with radiotherapy for patients with breast cancer. Methods Forty-three patients with early stage of breast cancer(n=30 for stage I and n=13 for stage II) were performed extended resection plus axillary lymph node dissection or quadrant resection plus axillary lymph node dissection.After operation,tangential field irradiation was conducted with 50 Gy of X ray,tumor bed was irradiated with additional 10 Gy of electron beam,and supraclavicular field irradiation was also administered with 30 Gy of X ray and 22 Gy of electron beam. Results There was no recurrence during the follow up for 14 to 50 months.The acute side effects of radiotherapy included breast complaints(n=10,grade Ⅰ,23.26%),skin erythema and hyperpigmentation(n=19,grade Ⅰ,44.19%),moist desquamation(n=5,grade Ⅱ,11.63%) and radiation esophagitis(n=4,grade Ⅰ,9.30%),while one patient experienced radiation esophagitis one month after radiotherapy.No such complications as skin fibrosis and necrosis,angiotelectasis,breast and upper extremity edema occurred.The short-term cosmetic effects were satisfactory to 93.02% of all the patients. Conclusion For early stage of breast cancer(stage I and stage Ⅱ),breast-conserving operation combined with radiotherapy may yield satisfactory survival rate and short-term cosmetic effects,as well as low incidences of side effects.
3.The perioperative hidden blood loss in intertrochanteric fractures: a comparative study

Objective To determine the perioperative hidden blood loss (HBL) associated with different fixation methods for intertrochanteric fracture.Methods We retrospectively studied 128 patients with 31-A1 and 31-A2 intertrochanteric fractures which underwent internal fixation with dynamic hip screw (DHS,n=35),short reconstruction intramedullary hip screw (IMHS,n=55) or percutaneous compression plate (PCCP,n=38) from January 2008 to January 2012.The HBL were calculated and compared.Results The mean total hidden blood loss (THBL) was more than 7 times than the mean apparent blood loss (ABL) in all the three groups.For the THBL,there were significant statistical differences among three groups; no difference was found between IMHS and DHS group.For the phasic hidden blood loss (PHBL),there was no difference among the three phase of treatment in DHS group.But in IMHS and PCCP group,there were significant differences between the 4th-5th day and the 1st day,the 2nd-3rd day after operation; no difference was found between the Ist and the 2nd-3rd day.For the ratio of PHBL/THBL there were significant differences between the 4th-5th day and the 1st day,the 2nd-3rd day after operation; but no statistical differences were found between the 1st day and the 2nd-3rd day after operation.Conclusion It can be concluded that HBL after surgery for intertrochanic fractures is much greater than that observed intra-operatively.The IMHS and DHS cause more HBL than PCCP.Frequent post-operative measurements of haemoglobin are necessary,especially the first three days after surgery.
4.Progress of adipokines in the treatment of type 2 diabetes by the Roux-en-Y bariatric surgery

The adipokines is a class of active substances secreted by adipose tissue,with the function of regulating the body glucose balance and others.Some experiments prove Roux-en-Y gastric bypass postoperative plasma levels of adipokines will change that play an important role in improving islet function in the treatment of type 2 diabetes mellitus.Here,the author will make a review about the adipokines role in pathways and postoperative level changes,etc,in order to further explore the mechanism of the surgical treatment of diabetes.
5.The killing effects of ganciclovir on HyTK gene-transduced murine bladder cancer cell line T739 cells in vitro

Objective To investigate the effects of the HyTK gene transduction on murine bladder carcinoma cells in vitro . Methods The retroviral vector plasmid PL(HyTK)SN was transfected into packaging cell line PA317 using the liposome method. The obtained recombinant retroviral supernatant fluid was used to infect murine bladder cancer cell line T739 cells. B resistant colonies, named as T739 TK, were obtained after hygromycin B selection. The integration and expression of HyTK gene in T739 TK cells were identified by RT PCR. The in vitro killing effects of GCV on T739 TK cells were observed. Results RT PCR showed that HyTK gene was inserted into T739 cells successfully and there was mRNA expression of HyTK gene. In vitro , T739 TK cells expressed the killing effects after treatment with GCV of different concentrations. However, T739 cells grew well at different doses of GCV. Conclusion GCV has obvious killing effect on HyTK gene transfected T739 cells in vitro .
6.Bystander effect induced by suicide gene on mouse bladder cancer

Objective To determine the bystander effect of therapy with herpes simplex virus thymidine kinase ( HSVtk ) gene combined with ganciclovir (GCV) on mouse bladder cancer. Methods Mouse bladder cancer cell line (T739) was transfected with retroviral vector HSVtk gene. The sensitivity of T739TK cells to GCV was detected in vitro . Bystander effect in vitro in co cultured mixtures of T739TK and T739 cells at different ratios or at the same ratio in culture bottles of different size was determined. In the mouse model of bladder tumor, mixtures of T739 and T739TK cells were implanted beneath the peritonea of syngeneic mice. When tumors grew to the size of 0 5-0 8 cm, intraperitoneal administration of GCV was carried out for 6 d. Changes of tumor size and survival rate of mice were observed. Results T739 cells retrovirally transfected with the HSVtk gene became sensitive to low concentrations of GCV. Analysis by RT PCR confirmed HSVtk expression in the transfected T739TK cells. An absolute bystander effect was observed in the mixed culture of cells in vitro when the HSVtk gene transferred cells were at the ratio of above 10%. The tumor growth in the animal model was significantly inhibited by GCV, demonstrating the existence of bystander effect in vivo . The survival time of mice was prolonged after administration of GCV. Conclusion The in vitro and in vivo bystander effects exist in mouse bladder cancer transfected with HSVtk gene, enhancing the role of HSVtk /GCV system for the treatment of bladder carcinoma.
7.Free-ranging dogs infection in the plague natural foci in Karamay, China

We investigated the current situation about free-ranging dogs infection with plague and provided relevant data for plague prevention and control in Karamay City.We examined the dog serum F1 antibody by indirect hemagglutination assay (IHA).Results showed that the F1 antibody was found in 1 of 13 dogs serum samples,and the positive rate was 7.69%.The free-ranging dogs plays an important role in controlling animal plague epidemic,and there are potential risk for local people's infection of plague.

9.Effect of diabetic neuropathy on renal 1-? hydroxylase and bone mineral density in old type 2 diabetic rats

Objective To investigate the change of renal 1-alpha hydroxylase and its effect on bone mineral density in the old rats with type 2 diabetic nephropathy. Methods Forty Wistar rats of 18 months old were randomly divided into normal control (N), diabetes (D), diabetes treated with vitamin D_ 3 (T1) and diabetes treated with 1-?(OH)D_ 3 (T2) groups, respectively. Ten rats in each group. Dual energy X-ray absorption (DEXA) was used to determine bone mineral density (BMD) of lumbar spines and femoral bone. 24 h urinary protein excretion, serum 25(OH)D_ 3 and 1,25(OH)_ 2 D_ 3 were measured by radioimmunoassay. Results Compared with controls, 24 h urinary protein excretion increased remarkably D, T1, T2 groups, while BMD greatly decreased, much lower in D group and T1 group than T2 group (P0.05). Objective To synthesize a multiple antigenic peptide(MAP),observe its humoral immune response and evaluate its antifertility potential.Methods Based on fusogenic epitope mimic peptide and synthetic nonnatural Pan DR T Helper Epitope(PADRE),MAP antigenic peptides were designed and synthesized.After suspending with the equivalent Freund's adjuvant,the antigens were used to immunize the female C57BL/6 mice to evalutate their humoral immune response in animals and efficiency of specific antisera on inhibiting syncytial trophoblast fusion.Results The MAP peptide was successfully synthesized.After purified with HPLC,its purity reached 95%.After immunization,the highest level of specific IgG in mice serum was 1:1 024,and the corresponding antigen of human trophoblastic cell could be identified in the antisera.In presence of mice antiserum by 1:10 dilution,forskolin-induced intercellular fusion of BeWo choriocarcinoma cells decreased remarkably(P
