1.Analysis on the analgesic effect of sufentanil combined with dezocine in endoscopic resection of fallopian

Objective To study the analgesic effect of sufentanil combined with Dezocine in endoscopic resection of fallopian.MethodsA total of 188 patients with tubal pregnancy from April 2013 to December 2014 in Haining central hospital were enrolled in the study, they were randomly divided into two groups (n=94).On the basis of conventional intravenous analgesia, the patients in control group was were received sufentan, the observation group were given sufentan combined with dezocine.The score of pain at different time points after operation, the Ramsay sedation score, and the level of IL-6 and IL-10 were compared.The adverse reactions were compared between the two groups.ResultsThe score of VAS in the observation group was significantly lower than that in the control at 4h, 8h, 12h and 24h after operation (P<0.05).The score of VAS in the two groups were similar at 48h after operation;the Ramsay score in the observation group was significantly higher than that in the control group(P<0.05) at five time points (4h, 8h, 12h, 24h and 48h after operation);when compared Compared with the indexes before operation, the level of IL-6 was significantly increased in the two groups at 24h and 48h, but the observation group increased less, the IL-6 in observation group was lower than that in the control group at the two points(P<0.05);while the IL-10 was higher than that beforeoperation, and the observation group was more higher(P<0.05).The adverse reactions in the two groups were mainly dizziness, nausea, pruritus and lethargy, and the incidence was only 12.8% in the observation group, which lower than that in the control of 29.8%(P<0.05).ConclusionIt which dazocine combined with sufentanil had a good analgesic effect in the process of analgesia in gynecological laparoscopic resection of fallopian tube after the operation.The VAS score and Ramsay sedation score was improved significantly.The levels of inflammatory factors were lower, and the incidence of adverse reaction was significantly reduced.It is a reliable method for clinical intravenous analgesia.
2. Bio-oss combined with fibrin glue in reconstruction of canine mandibular defects

Objective: To evaluate the osteogenic activities of Bio-oss after combining with fibrin glue in reconstruction of canine mandibular defects. Methods: The second and fourth premolar teeth and the second molar teeth were extracted bilaterally, in 9 hibrid canines, resulting in 6 bone defects (1 cm × 1 cm) in each canine. Bio-oss, Bio-oss+FG and FG were implanted into the bone defects of the second, fourth premolar teeth and the second molar teeth, respectively. Canines were executed in group of 3 after 4, 8, and 12 weeks to observe the healing of soft tissues. The bone density was assessed by X-ray, the property of Bio-oss were observed via gross specimen, and the morphology of the newly-formed bone was observed by tissue sections. The proportion of newly-formed bone was obtained by computer image analysis (SAS software, analysis of variance). Results: Stage I healing of soft tissues was achieved in all animals. The bone densities were not significantly different between Bio-oss+FG and Bio-oss groups. The bone in FG group had transparent area. We also found that the bone in Bio-oss+FG group was closely combined and there were sccatered Bio-oss dusts in the soft tissues of the Bio-oss group. The newly-formed bone in the FG group was only found in the border between the defects and FG. The proportion of newly-formed bone was less in the Bio-oss+FG group than in the Bio-oss group at 4, 8, and 12 weeks after extraction(P<0.05). Conclusion: Fibrin glue can well shape the Bio-oss and has satisfactory biocompatibility, but it reduces the new bone forming.

4.Effects of DKK1 on breast cancer cell migration and invasion capa-bility

Objective:To analyze the DKK1 expression in breast cancer cell line MDA-MB-231 and examine its effect on the mi-gration and invasion of the cell line. Method:DKK1 expression in the breast cancer tissues was detected immunohistochemically, and DKK1 distribution in the breast cancer cells was observed using immunofluorescence. Western blot was used to investigate DKK1 ex-pression in breast cancer tissues and their matched normal breast tissues. A eukaryotic expression vector of DKK1 was constructed and transfected into MDA231 cell line. The migration and invasion ability of the cells were observed through scratch assay and Boyden chamber, respectively. Results: Immunohistochemical staining showed that DKK1 in the transfected lymph node or non-lymph node was highly expressed in the breast cancer tissues (P<0.05). The positive DKK1 expression in lymph node metastasis was less signifi-cant than those in non-lymph node transfected cells. Immunofluorescence localization results showed a remarkable bright red granular fluorescence in the cytoplasm of the MDA231 cell line. Western blot analysis showed that the DKK1 protein expression in carcinoma tissues were obviously higher than that in matched normal breast tissues. The migration and invasion ability of DKK1-overexpressed MDA231 cell was lower than those of MDA231 cell transfected with pCMV-Tag-2b empty vector. Conclusion:Negative correlation was found between DKK1 expression and the migration and invasion ability in breast cancer cells.

Protein tyrosine kinase genes are the largest family of oncogenes. This is not surprising since protein tyrosine kinases are important components of signal transduction pathways that control cell shapes, proliferation, differentiation, and migration. This review will address the recent progress in understanding the function of Eph receptor and ephrin ligands in normal development and how disregulation of these functions could promote tumorigenesis.
6.Culture of outer root sheath cells of murine vibrissa follicles in vitro and expression of uPA/uPAR

Objective To establish a method for the cultivation of hain follicle outer root sheath (ORS) cells of murine vibrissa and to study their expression of uPA/uPAR. Methods The ORS cells of murine vibrissa were cultured by combination of enzyme digestion and tissue culture with the feeder layer of dermal sheath cells of the hair follicle. The cells and their uPA/uPAR expression were identified by immunocytochemistry (ICC). Results The feeder layer of DS was suitable for the culture of ORS cells. ORS cells expressed uPA/uPAR protein. Conclusion The DS of hair follicle is a better feeder layer for the growth of ORS cells. ORS cells have the ability to migrate and proliferate.
7.Determination of Hyperoside in Jintu Liangguang Capsules by RP-HPLC

Objective To establish a determination method for the hyperoside in Jintu Liangguang Capsules. Methods Reverse-phase HPLC with Dikma Diamons C18 column was applied. The chromatographic conditions were as follows:methanol-0.025 mol/L of thophosphoric acid (40 ∶60) as the mobile phase,detection wavelength at 370 nm,column temperature set at 40 ℃and the flow rate being 0.8 mL/min. Results The linear range of calibration curve was 0.303 ~1.818 ug/uL for the hyperoside (r=0.999 6). The average recovery was 95.2 %and RSD was 1.3 %. Conclusion The method is simple and accurate,and can be used for quantitative analysis of Jintu Liangguang Capsules.
8.Research and Enlightenment on the Drug Safety Supervision Mechanisms in USA

OBJECTIVE:To provide references for perfecting the drug safety supervision mechanisms in China. METHODS:Based on the theory of“market failure”“contract failure”and“government failure”,representative drug safety supervision mecha-nisms were selected from USA for analysis and evaluation. RESULTS & CONCLUSIONS:USA has made some achievements in the implementation and application of drug safety supervision mechanisms. The representative drug safety supervision mechanisms included prescription drug applicant payment mechanism,drug safety information communication mechanism,punishment and in-centive mechanism, public-private partnership mechanism, passive monitoring and active monitoring combination mechanism. These mechanisms relieved“government failure”of FDA,and solved the problems of“market failure”and“contract failure”to cer-tain extent. In China,drug safety supervision mechanisms should be perfected from refining the legal guarantee,mobilizing exter-nal forces completely,absorbing and applying modern information technology,focusing on both prevention and disposal,paying at-tention to the coordination of different mechanism,striving for open procedure and transparent result.
9.Assessment of antegrade/retrograde cardioplegia by magnetic resonance technique

Objective To compare the efficacy of retrograde cardioplegia for myocardial perfusion with that of antegrade cardioplegia at the same flow rate using magnetic resonance techniques. Methods Myocardial perfusion was evaluated with magnetic resonance imaging in isolated pig hearts during antegrade and retrograde cardioplegia. Phosphorus 31 magnetic resonance spectroscopy was used to determine the efficacies of both cardioplegic techniques in sustaining myocardial energy metabolism. Results Magnetic resonance images showed that contrast-enhanced signal-time courses obtained from different regions of the myocardium during retrograde cardioplegia varied significantly, whereas signal-time courses acquired during antegrade cardioplegia appeared more uniform. Localized phosphorus 31 spectra showed that retrograde cardioplegia resulted in myocardial ischemia at the flow rate of 10 ml/min. However, no significant changes were observed in myocardial energy metabolites until the flow rate was decreased to 3 ml/min during antegrade cardioplegia. Conclusion Microvascular perfusion provided by retrograde cardioplegia varies more significantly among various small areas of the myocardium than that did by antegrade cardioplegia. Moreover, the efficacy of retrograde cardioplegia in sustaining normal myocardial energy metabolism is lower than that of antegrade cardioplegia.
10.Isolation and culture of rat hair follicle Bulge cells and its growth character in vitro

Objective To isolate and culture the hair follicle Bulge cells of rats and study their morphological, immunological characteristics. Methods Hair follicle Bulge obtained by micromanipulation and enzyme digestion was cultured in intro. The morphological features and numbers of Bulge cells were identified and counted with light microscopy. Immunocytochemical staining was used to detect expressing of K19 and ?_ 1integrin in cultured cells. Results Bulge cells after enzyme digestion pasted rapidly. Cells were small and round, paving stone-shape, and had a large ratio of nuclear to cytoplasm. The characteristics suggested a primitive morphologic feature. Cells growth well and could maintain low differentiation and higher reproductive activity, they co-expressing K19 and ?_ 1integrin, immunofluorescence staining showed the cells expressed ?6-integrin strongly, weakly or no expressing CD71. Conclusion This culture system can amplify a lot of pure hair follicle Bulge cells in short time. We successfully isolate and culture the hair follicle Bulge cells of rats in intro and can keeping its property for a long time.
