4.Application of self-expanding stent and seperable detachable coil for interventional therapy in patients with wide-necked intracranial aneurysm

Objective To explore the application of self-expanding stent and seperable detachable coil for interventional therapy in patients with wide-necked intracranial aneurysm.Methods 32 patients with wide-necked intracranial aneurysm received the interventional therapy by self-expanding stent and seperable detachable coil.After operation,the situation of the aneurysm occluded were observed by DSA immediately and followed up.Results The stents were successfully deployed in the 32 patients.The seperable detachable coils were occluded in the aneurysms in 29 cases.After operation immediately,the DSA showed that the aneurysms in 18 cases were completely occluded by seperable detachable coils,7 cases were 95% occluded,4 cases were 90% occluded.In the other 3 cases,the aneurysms were too small to be filled by seperable detachable coil,the stents were used only.All the parent arteries were no stenosis.18 h after operation,1 case died from multiple hemorrhage caused by thrombasthenia.Follow-up from 3 months to 1 year,DSA re-examination in 21 cases.Compared with the image immediately after operation,the imagings of DSA were no change in 12 cases,residual necks were disappeared in 5 cases and increased in 1 case.In the 3 cases which stents used only,the aneurysms were no development completely in the 2 cases and decreased in 1 case.6~18 months followed up in 26 cases,the rebleeding and cerebral infarction were no occurred.Conclusions The self-expanding stent is very flexible,secure,and effective.It can good support for the wide-necked aneurysms to be occluded by seperable detachable coils and effective promoting the small aneurysms healing.
5.Effects of artemisunate on anoikis resistance in human breast cancer cell MCF-7

AIM: To study the effects of artemisunate on anoikis resistance in human breast cancer cells. METHODS: After breast cancer MCF-7 cells were treated with artemisunate, the growth of anchorage independence of breast cancer MCF-7 cells was examined in soft agar colony formation, and apoptosis of breast cancer were evaluated by terminal deoxynucleatidyl transferase-mediated deoxyuridine triphosphate nick end labeling (TUNEL) and DNA ladder. RESULTS: Artemisunate significantly inhibited the growth of anchorage independence of breast cancer MCF-7 cells in soft agar colon formation in a dose-dependent manner. Artemisunate induced apoptosis in breast cancer cells was conformed by DNA ladder and TUNEL assay, which was in a time-and dose-dependent manner. CONCLUSION: Artemisunate inhibits the growth of anchorage independence of breast cancer MCF-7 cells, which is related to anoikis.
7.Role of PI3K/Akt pathway in effect of paeoniflorin against Aβ25-35-induced PC12 cell injury.

OBJECTIVETo study the role of PI3K/Akt pathway in the neuroprotective effect of paeoniflorin on PC12 cells.
METHODThe paeoniflorin group (5, 10, 20 μmol · L(-1)) was pretreated for 30 min, and then added with Aβ25-35 (20 μmol · L(-1)) for interaction for 24 h. Inhibitor LY294002 (10 μmol · L(-1)) was pretreated for 30 min before the action of paeoniflorin (10 μmol · L(-1)). The MTT colorimetric method was used to detect the cell viability. The apoptosis rate was tested by the FITC-Annexin V/PI staining. The protein expression of p-AKT, Bax, Bcl-2 and cleaved caspase-3 protein were detected by Western blot analysis.
RESULTPaeoniflorin could significantly inhibit the Aβ25-35-induced PC12 cell toxicity and apoptosis. Its protection effect may be achieved by up- regulating AKT phosphorylation level, increasing Bcl-2 protein expression, reducing Bax protein expression, inhibiting the activation of caspase-3. Inhibitor LY294002 could weaken the above protective effects of paeoniflorin.
CONCLUSIONPaeoniflorin could activate PI3K/Akt signaling pathway to protect the PC12 cell injury induced by Aβ25-35.

8.Network pharmacology study on major active compounds of siwu decoction analogous formulae for treating primary dysmenorrhea of gynecology blood stasis syndrome.

OBJECTIVETo explore the molecular mechanisms of main active ingredients of Siwu decoction analogous formulae for treating primary dysmenorrhea of gynecology blood stasis syndrome by network pharmacology study, and to investigate the correlations between multi-compounds, multi targets and multi pathways.
METHODMajor active compounds from Siwu decoction analogous formulae, including ligustilide, butylidene phthalide, senkyunolide, ferulic acid, gallic acid, peoniflorin, jioglutin A, catalpol, transanethole, zingiberone, commiphoric acid, eugenol, isorhamnetin-3-O-neohesperidoside, wulingzhic acid, alpha-cyperone, cyperene, costunolide, costic acid, tetrahydropalmatine, protopine, amygdalin, 24-methylene cycloartanol, oleic acid, linoleic acid, 3-p-coumaroylquinic acid, hydroxysafflor yellow A, coptisine, berberine, jatrorrhizine, baicalein, baicalin, wogonin were collected to build component-protein networks based on PharmMapper database. The targets information access was used to construct and visualize components-targets-pathways network model using the kyoto encyclopedia of genes and genomes (KEGG) pathway database and Cytoscape software.
RESULT AND CONCLUSIONSerine threonine protein kinases play an important role in the process of cells. They were potential targets in the effect of Siwu decoction analogous formulae. The effect of main active ingredients involved 51 the pathway. Besides the same ones, Shaofu Zhuyu decoction had more effect on lipid metabolism, Xiangfu Siwu decoction on amino acid metabolism pathways, Taohong Siwu decoction on carbohydrate metabolism, while, Qinlian Siwu decoction on ErbB, VEGF signal transduction pathway. Siwu decoction and its derived formulae not only had common targets and pathways, but also had their own emphasis. This reflected the formulae effect mode of multi-ingredients, multi-targets and multi-pathways. It may provide clues to deeper study of molecular mechanism of Siwu decoction analogous formulae action.

10.Comparison of escape learning and memory function between marginal division of striatum and hippocampus in rats

BACKGROUND: Hippocampal structure in brain is the division related to learning and memory, generally it is closely relevant to spatial cognitive activity. Marginal division of striatum is a latest discovered subdivision related to learning and memory function of brain, whether is its learning and memory function different from that in hippocampus?OBJECTIVE: To compare the difference, function and importance in learning and memory function between marginal division of striatum and hippocampus in brain and observe the difference in escape learning and memory between marginal division of striatum and hippocampus.DESIGN: Completely randomized controlled experiment.SETTING: Institute of Neuroscience in Zhujiang Hospital Affiliated to First Military Medical University of Chinese PLA.MATERIALS: The experiment was performed in Institute of Neuroscience in Zhujiang Hospital Affiliated to First Military Medical University of Chinese PLA from March 2002 to July 2003. Totally 109 normal male adult SD rats were employed and 75 rats of them were screened as the qualified animals by twice Y-maze test. Randomly, 25 rats were divided into damaged marginal division of striatum group (DMD group), 10 rats were into bilateral fimbria-fornix transection group (FFT group), 30 rats were into the control of marginal division of striatum (MD control) and 10 rats were into the control of bilateral fimbria-fornix group (FF control). After 24 hours training in Y-maze,In DMD group, 10 g/L kainic acid 0.1 to 0.2 μL was used to damage bilateral marginal division of striatum of rats. In MD control, physiological saline of minim dose was injected in bilateral marginal division of striatum of rats. In FFT group, bilateral fimbria-fornix was transectioned. In FF control, the cortical tissue of the superficial layer of bilateral fimbria-fornix was transectioned. The operation was done on the second day after the 2nd screening. The behavior of rats in learning and memory was observed in Y-maze on the 5th day after operation (during 30 times of maze test, if success frequency ≥ 15,normal capacity of learning and memory was identified.).MAIN OUTCOME MEASURES: Success frequency of rats in different groups in Y-maze learning before and after operation.RESULTS: Of 109 normal male adult SD rats, 75 rats were screened to be qualified after twice Y-maze test. During the experiment, 3 rats were died and other 32 rats fell into disuse for the drug or physiological saline was not injected accurately to the marginal division of striatum. Terminally, 40 rats entered the analysis in total, of which, 11 rats were in DMD group, 9 rats in MD quency in Y-maze learning of rats in DMD group after operation was lower than MD control, FFT group and FF control [(9.27±4.29) times, (22.56±4.25)frequency in Y-maze learning of rats in DMD group after operation was also significantly lower than that before operation [(9.27±4.29) time, (18.27±3.07)FFT group was basically same as FF control and MD control (P=0.660 and P=0.489) and it was basically same to the success frequency in Y-maze learning before operation (P=0.700).CONCLUSION: The learning of Y-maze in rats with damaged marginal division of striatum was remarkably reduced and there was no obvious change in learning and memory between the rats with fimbria-fornix transaction and without transaction. It is verified that marginal division of striatum can reflex complex learning and memory behavior in electric Y-maze test, which cannot be achieved in hippocampus. It is further explained the difference of the two divisions in controlling learning and memory in cerebrum, the marginal division of striatum is able to control hippocampus in learning and memory function.
