2.Pharmacokinetics of Fagopyrum tataricum Total Flavonoids in Beagle Dogs

OBJECTIVE:To study the pharmacokinetic features of Fagopyrum tataricum total flavonoids. METHODS:6 Beagle dogs were intragastrically given single dose of 80 mg F.tataricum total flavonoids (with content of quercetin at 1.2% and the content of eldrin at 24.7%). HPLC-UV spectrophotometry was adopted to determine the content of total quercetin in the hydrolyzed plasma samples. RESULTS:Taking the total quercetin as the index of the pharmacokinetic features of the F. tataricum total flavonoids in the Beagle dogs,the main pharmacokinetic parameters were as follows:AUC0→t:(9.92?1.49)?g?h?mL-1;AUC0→∞:(10.81?1.86)?g?h?mL-1;Cmax:(1.07?0.19)?g?mL-1. CONCLUSION:The pharmacokinetics of the F.tataricum total flavonoids in the Beagle dogs conforms to two-compartment model,and the concentration-time curves are characterized by multiple peak pharmacokinetics.
5.Diagnosis, treatment, and prevention of iatrogenic distal common bile duct injury

Objective To review the diagnosis and causes of iatrogenic injury to the distal choledochus at operation. Method The case notes of the patients with bile duct injuries that were treated in my Department from 1990.2-2005.2 were reviewed. Results To detect distal bile duct injuries, a sound in the bile duct had an accuracy rate of 95 % while injection of water into the bile duct to detect leakage had an accuracy rate of 100%. Using a long arm T tube in the common bile duct was an effective method to treat the injury. In 18 patients with an average follow-up time of 20. 8 months, the results were satisfactory. Conclusions Injecting water into the bile duct to diagnose distal common bile duct injury at operation was an effective way to detect distal bile duct injury. Adequate exposure of the opeative field is the best method to prevent bile duct injury.

The endophytic bacterum 01-144 was marked by using the method of antib iotic-resistance. Colonization of 01-144 in tomato root and stem was investig ate d. Result showed that 01-144 colonized in the root and stem and the colonizing a bility in the root was stronger than in the stem after dipping seed or watering root treatmeat, It was also found that this bacterium could more easly colonized in the low stem than in the upper stem. The population fluctuation of 01-144 ha d the same trend in both root and stem i.e.first increased then decreasing, an d the fluctuation in the root was more even than in the stem.
7.Review of Studies on Azo Dyes Decolorization

Biological process is an important approach to treat the dye wastewater. The azo dyes decolouration by special azoreductase of different aerobic bacteria and fungi was summarized. Under anaerobic condition, reductive decolourization of azo dyes was carried in the presence of redox mediators which act as electron shuttle. It was also pointed out that azo dye reduction occurred mainly under anaerobic condition. Different electron donor resulted in different decolourization rate. Problems of current biotechnology were analyzed and corresponding measures were discussed.

Objective To study the expression of hMLH1, hMSH2 and their association with clinico-pathological characteristics in hepatocellular carcinoma(HCC). Methods The expression of hMLH1 and hMSH2 in 37 HCC tissues was detected by SABC immunohistochemistry. Results The positive rate (score) of hMLH1 and hMSH2 expression in HCC was 48.7%(1.65?1.70) and 59.5%(2.30?1.96), respectively, which was obviously lower than that in the adjancent tumor tissues with 83.8%(3.30?1.37), and 86.5%(4.30?2.01), and in the normal liver tissues with 88.2%(3.88?1.98) and 82.3%(4.29?1.83), respectively(P
9.Effect of propofol on vascular reactivity in rat septic shock

Objective To study the effect of propofol on vascular reactivity and plasma malondialdehyde (MDA) and nitric oxide (NO) in septic shock.Methods Forty male SD rats weighing 200-250 g were randomly divided into 4 groups (n = 10 in each group) : group Ⅰ control; group Ⅱ septic shock; group Ⅲ septic shock + propofol and group Ⅳ septic shock + melatonin. The animals were anesthetized with intraperitoneal pentobarbital 40 mg?kg-1. The femoral artery and vein were connulated for MAP monitoring and drug administration. The animals were breathing spontaneously. Septic shock was induced by intravenous LPS 15 mg?kg-1 . In group Ⅲ a bolus of propofol 10 mg?kg-1 was given i.v. at 1 h after intravenous LPS followed by intravenous propofol infusion at 10 mg?kg-1?h-1. In group Ⅳ melatonin 10 mg was given intraperitoneally at 1h after LPS i.v. . Six hours after LPS administration 4 doses of phenylephrine (PE) 0.5, 1, 2, 2.5 ?g?kg-1 were given i.v. in succession. The next dose was given when MAP returned to the baseline level after previous PE. The percent change in MAP after each dose was recorded. Blood samples were taken at 6 h after LPS administration for determination of plasma MDA and NO concentrations. After the in vivo experiment the animals were sacrificed and thoracic aortas were removed and cut into segments of 3 mm in length which were bathed in Krebs buffer aerated with 95% O2 and 5% CO2 at 37℃. The aortic rings were stretched to a resting tension of 2.0 g. The segments were then exposed to increasing concentrations of PE (from 1 nmol?L-1 to 30 ?mol?L-1). The dose-response curves were obtained. Emax and EC50 were calculated. Results The percent increase in MAP induced by PE was significantly reduced by septic shock (groupⅡ,Ⅲ,Ⅳ) as compared with control group (Ⅰ), but was significantly larger in propofol and melatonin groups (Ⅲ,Ⅳ) than in group Ⅱ.
