2.The clinical progress of antisense technology in the treatment of cancer

According to the base complementation, the principle antisense technology is the sequence specific binding of complementary antisense nucleic acids or their chemical modification to the nucleic acids in cells, resulting in the suppression or prevention of gene translation. For the treatment of cancer, a lot of antisense drugs have been designed, which target the genes involved in the pathogenesis of cancer. And some of them have been in clinical trials. The author reviewed the progress of the clinical trials of these antisense drugs.

4.Effect of hyperbaric oxygen on T-lymphoid cells and cytokines in rats

ObjectiveTo investigate the effects of hyperbaric oxygen therapy on T-lymphocytes 1 and cytokines including IL-6, IL-10 and TNF-α asindicators well representing immune function and inflammation in normal rats and rats with systemic inflammatory response syndrome. Methods Forty Sprague-Dawley male Rats were randomly ( random number) divided into five groups.Rats of group A were treated by saline solution (0.9 % NaCl,5 mL/kg),and served as control group; rats of group B were treated with saline solution and hyperbaric oxygen therapy three times; rats of group C were treated with zymosan (500 mg/kg) ; rats of group D were treated with zymosan and hyperbaric oxygen therapy once; and rats of group E were treated with zymosan and hyperbaric oxygen therapy three times.At 24 hours after saline or zymosan administration,rats were sacrificed and blood samples were collected from the portal vein.CD3 +,CD4+,CD8+ cells were counted by flow cytometer,and plasma levels of IL-6,IL-10 and TNF-αwere measured by ELISA.The results were analyzed by one - way ANOVA and LSD post-hoc test.Results CD4 T cell of normal rats was decreased by hyperbaric oxygen (P<0.01 ),CD4/CD8 T cell ratio was also decreased ( P < 0.05 ).Of rats with zymosan administration,CD4 T cells and CD8 T cells were both decreased on a parallel (P<0.01 ).A single one HBO and HBO for three times both raised the percentage of CD4 T cells which was significantly decreased by zymosan administration ( P < 0.01 and P < 0.05 ).In rats without hyper- inflammation,hyperbaric oxygen therapy showed a pro-inflammatory effect by elevating TNF-α (P<0.01),IL-6 (P<0.05) and reducing 1L-10 (P<0.05).ConclusionsHBO reduces CD4 T cells in normal rats.HBO reduces the production of TNF-α,IL-6 and IL-10 during hyper- inflammation.HBO is pro - inflammatory in normal rats and minor SIRS rats withourt appreciable hyper-inflammation in circulation.
5.Effect of Zedoary Turmerie Oil Injection on Oncosis and Bcl-2 Expression of Human Ovarian Cancer SKOV3 Cell

Objective To discuss the impact of zedoary turmerie oil(ZTO) injection over oncosis index and Bcl-2 expression of human ovarian cancer SKOV3 cell.Methods After ZTO injection acted on human ovarian cancer SKOV3 cells,the changes in the nucleus were observed by fluorescence microscope,oncosis index was counted by projection electron microscope,the situation of cell DNA breakage was observed by agarose gel electrophoresis,and cell Bcl-2 gene expression was observed by immunohistochemical method.Results After treated by ZTO injection for 48 hours,human ovarian cancer SKOV3 cells shows that oncosis cell was swelling in fluorescence microscope,the volume increased,the membrane area narrowed,the nuclear chromatin expansed.The oncosis index increased with dose-effect relationship,DNA electrophoresis showed diffuse type and Bcl-2 gene expression was down-regulated.Conclusion ZTO injection can increase oncosis index of human ovarian cancer SKOV3 cell with the concentration,and lower Bcl-2 gene expression,which may be one of the mechanisms of ZTO injection caused oncosis of SKOV3 cell.

Objective:To investigate the effect of host derived immune deficient dendritic cell(DC) on immune tolerance in transplantation of xenoislet.Methods:Mature or immune deficient DC were cultured from BALB/c mice and loaded with MHC antigen of Wistar rat. The DC were injected into the diabetes mice through tail venous, then islets of Wistar or SD rats were implanted beneath the capsule of renal after one week. Survival time of grafts was monitored. 7.Follow up Screening of Extraction Process of Volatile Oil in Supplemented Xanthium Powder by GC MS

Objective: To screen the extraction process of volatile oil in Supplemented Xanthium Powder.Methods: The main compositions of volatile oil contained by every herb in prescription were followed up by GC MS. Results: The extraction process of volatile oil in Supplemented Xanthium Powder was established, that is, Flos Magnoliae and Cortex Moutan Radicis were powdered, their volatile oils were extracted with Herba Menthae together by the steam distillation; then 13 fold first fraction of distillata and 3 fold redistillation liquid were collected respectively; the volatile oil of Rhizoma Liqustici Chuanxiong was extracted with alcohol. Inasmuch as Fructus Xanthii and Radix Angelicae Dahuricae contained less volatile oil, at the same time, to avoide that Fructus Xanthii produced toxicity by water extraction and coumarinic lactones of Radix Angelicae Dahuricae was destroyed their oils were considered unfit for extraction. Conclusion: GC MS is a useful way for follow up optimization of the extraction process of volatile oil in compound prescription of Chinese medicinal and worthy of popularization.
8.Effects of Zn~(2+) on cell cycle changes induced by multiglucosidum tripterygii

Objective To explore the effect of Zn 2+ on human leukemia promyelocyte (HL 60)cycle changes induced by multiglucoside tripterygii (MT).Methods ① HL 60 phenotype and membrane protein Fas expression was detected.② HL 60 was cultured with MT observing the cell forms and cell cycle changes,DNA electrophoresis results and the effect of Zn 2+ on the cell cycle change.Results ① The HL 60 were CD4 +CD8 -CD19 -IFN ? +IL 8 -,and the Fas protein expression was poorer.② Following culture of HL 60 with MT,the apoptosis relative cell cycle change appeared in HL 60.This change mainly affected G 2/M and S stage cells,and presented dose time correlation.③ Zn + nearly all blooked the effect of MT induced HL 60 cell cycle change.Conclusion ① HL 60 has Th1 lymphocyte like characteristics,and the Fas protein expression is poorer.② Zn + affects the MT induced HL 60 cell cycle change.It suggests that Zn + might affect the therapeutic and side effects during clinical treatment of many autoimmune diseases with MT.
9.Assay of ?-asarone and ?-asarone in Rhizoma acori tatarinowii by HPLC

Objective: To determine ? asarone and ? asarone in Rhizoma acori tatarinowii(RAT) . Methods: HPLC condition consists of ODS C 18 column(150mm?4.6mm, 5?m), methanol: water(6∶4) as mobile phase containing potassium dihydrogen phosphate 1.4g and sodium lauryl sulfate 1.2g per 1000mL, detective wavelength at 257nm, flow rate at 1.0mL?min -1 . Results: For RAT the mean recovery of 99.02%( RSD =1.03%) for ? asarone, 101.26%( RSD =3.57%) for ? asarone are obtained, respectively. Conclusion: The method is sensitive, rapid and accurate.
10.Practice and thinking of LBL combined with PBL in teaching cancer pain standardization training for residents

Resident doctor standardization training program is the necessary and important part of medical education after graduation.The Hainan branch of PLA General Hospital combines scientific and practical medical conditions and discipline characteristics of Hainan to scientifically plan the standardized treatment of cancer pain in training.This model combines lecture-based learning (LBL) teaching method and virtual demonstration for theoretical training.In practical training,we conduct problem-based learning (PBL) teaching method,situational teaching and standardized examination to systemically improve resident doctors' skill for cancer pain management.This combined teaching strategy can help resident doctors to establish correct and standard concept in future clinical practice,and enable them to establish a correct concept of treatment,and adopt two-way evaluation to continuously improve teaching quality before the end of training.
