1.Ecological perspective of integrated TCM intervention model for IGT population

Intervention and prevention of IGT population is crucial for reducing high incidence of diabetic.This paper analyzed the insufficiency of the existing IGT intervention and prevention models by introduing the concept of human health view of ecological theory,and emphasized the close relationship between IGT population and environment.The ecological explanation of TCM etiology of IGT and the ecological evaluation standard of IGT intervention and prevention model were also discussed.This paper further indicated that only by focusing on adapting to the living environment,applying the concepts of "integrated ecosystem management" can we successfully construct a sustainable IGT prevention and intervention model and effectively promote the model into a large-scale community.

Ligusticum Chuanxiong,a traditional Chinese medicine,has the functions of promoting Qi flow and blood,dispelling pathogenic wind and relieving pain.One of effective constituents extracting from Chuanxiong is tetramethylpyrazine,which has several pharmacological activities and anti-tumor function through inhibiting expression of vascular endothelial growth factor(VEGF).Recent relative studies provided scientific basis for a better application oftetramethylpyrazine clinically.
3.Induction of human mesenchymal stem cells in vitro and related identification

OBJECTIVE To establish the isolation and identification system of human mesenchymal stem cells(hMSCs)in vitro, and then to establish a stable cultured system in which the hMSCs can be induced to osteoblasts, chondroblasts and lipoblasts in vitro. To study the possibility if hMSCs can be used as the seed cells in bone tissue engineering.This study would lay a foundation for the clinical application of tissue engineering. METHODS 5ml bone marrow of the patient was aspirated from iliac crest. The human MSCs were isolated using Percoll gradient centrifugation. The hMSCs were cultured in DMEM medium in vitro, FACS(fluorescence axtivated cell sorter)was used to identify the phenotype of hMSCs. The hMSCs were cultured in vitro, and induced to osteoblasts, chondrocytes and lipocytes. The changes of phenotype were tested by mmunohistochemisty and molecular biology technique. RESULTS The hMSCs were isolated by Percoll gradient centrifugation from patient’s bone marrow. The expression rate of MSC markers including CD105, CD166, CD29 were(78?6)%, (43?7)%, (69? 12)% respectively. The phenotype of osteoblasts induced from hMSCs was verified by the mineralized nodes formation when cultured in vitro and Col, and OCN expression showed by immunohistochemistryand RT-PCR. The phenotype of chondrocytes induced from hMSCs was verified by type, collagen, SOX9 expression with immunohistochemical staining technique and type, collagen, aggrecan mRNA expression with RT-PCR. The lipocytes’ phenotype was verified by positive result of oil red O staining and PPAR 2mRNA expression by RT-PCR. CONCLUSION The highly pure hMSCs can be harvested by means of density gradient centrifugation, and the hMSCs have multi-potentiality of cell differentiation via induced culture in vitro. The third passage of hMSCs can be induced to express osteoplast phenotype and meet the qualitative and quantitative demands of seed cells when bone tissue engineering was used in clinical practice.
4.The research on high-density flexible microelectrode array of retinal prosthesis based on MEMS technology.

The paper proposed a new method to design and fabricate a flexible neural microelectrode arrays (MEA) for retinal prosthesis, the ion-beam technology was introduced to decrease the width of conductive wires and the distances between wires, a high density MEA (120 microelectrodes with a matrix of 10 x 12) was fabricated on a single layer of polymer from this. The MEA was proved to possess a fine electrochemical property. In vitro test, the average impedance of MEA on 1 kHz was 16 k omega +/- 2 k omega and the average phase difference was -85 degrees +/-30 degrees.

6.Effects of iptakalim hydrochloride on H_2O_2 induced cytotoxicity in pheochromocytoma(PC12) cells

AIM: To investigate the protective effects and mechanisms of iptakalim hydrochloride(Ipt)on H_2O_2 induced neurotoxity. METHODS: Neurotoxity injury was induced by H_2O_2 in PC12 cells. The cell viability was tested by MTT assay. The glutamate released from PC12 cells was measured by HPLC combined with fluorescent detector analysis. Changes in the intracellular free Ca 2+ concentration ([Ca 2+ ]_i) were determined in fluo-3 AM loaded PC12 cells. RESULTS: Ipt (1, 10 and 100 ?mol?L -1 ) markedly mitigated H_2O_2-induced neurotoxity, 10 ?mol?L -1 Ipt inhibited the release of glutamate and the increase of [Ca 2+ ]_i induced by H_2O_2 .The protective effects was incompletely blocked by 5-HD which is a mitochondrial K_ ATP channels antagnist. CONCLUSION: Ipt provides neuroprotective effects on H_2O_2 induced cytoxixity in cultured PC12 cells and the protective effects may be partially related with mitochondrial KATP channels.
7.In-vitro electrochemical stability evaluation of a flexible MEMS microelectrode.

Three-electrode testing method was used to investigate the effect of temperature on electrode impedance, and the pH shifts in saline solution resulting from the electrical stimulation were also determined. Experiments in PBS (phosphate buffered solution) solution showed that the electrode impedance was almost invariable at the human body temperature range (35 degrees C-40 degrees C). And the experiments in unbuffered saline solution showed that pH shifts decreased from 0.03 to 0.005 when the frequency of biphasic charged-balanced pulses increased from 1 Hz to 100 Hz. Even stimulated by monophasic pulses (frequency is 15 Hz, amplitude is 50 microA), the stimulus-induced pH shift of electrode only varies 0.15 (anodic pulse current increased 0.15 and cathodic pulse current decreased 0.15).

8.Research on characteristics of liver injury induced by rifampicin in mice

Objective To observe the symptom and characteristics of liver injury induced by different dosage of long-term administration of rifampicin(RIF) in mice.Methods Twenty-four healthy female ICR mice were randomly divided into 4 groups(6 mice in each group):control group,low dosage group,medium dosage group and high dosage group.The four groups were treated with 0,100,200,400 mg·kg-1·d-1 RIF respectively for 2 weeks.Mice blood and liver tissue samples were collected at 6 hours after the last administration for serological test and liver histological observation.Results No mice died before execution.The TBA,DBIL and TBIL of high dosage group all increased compared with the control group, but the ALT,AST and ALP showed no obvious change.The TBA and DBIL of medium dosage group increased compared with the control group, while the TBIL,ALT,AST and ALP showed no obvious change.In the low dosage group,there was no obvious change in terms of TBA,DBIL,TBIL,ALT,AST,and ALP compared with the control group.Obvious pathological change occured in the liver of mice in all the experimental groups.HE staining showed edema and feather steatosis in liver cells, accompanied by a large number of inflammatory cells infiltration and a few sporadic cholestasis.With the increasing of RIF dosage,the liver pathological change became more obviously.In the experimental group,electron microscope showed that there were a lot of fat droplets in the liver cells wrapped slurry,and part of the capillary bile duct were slightly expanded with different electron density and irregular shape of bile sample material inside.The pathologic changes get more obvious with the increase of the concentration of rifampicin as well.Conclusion RIF could induce liver injury after 2 weeks' treatment at different dosages,mainly pathological changes included liver cell steatosis,inflammatory cell infiltration,and cholestasis.Rifampicin induced liver injury in a concentration-dependent manner.However,the mechanism of rifampicin-induced liver injury in mice needs further study.
9.Laparoscopic and thoracoscopic Ivor Lewis esophagectomy using a circular-stapled anastomosis with trans-oral anvil: 30 cases report

Objective To explore the feasibility and short-term efficacy of laparoscopic and thoracoscopic Ivor Lewis esophagectomy using a circular-stapled anastomosis with trans-oral anvil in treating esophageal carcinoma.Methods 30 consecutive patients with esophageal cancer received minimally invasive Ivor Lewis esophagectomy from January 2011 to December 2011.Operations of all the cases comprised of both abdominal and thoracic procedures which were performed with Iaparoscopic and thoracoscopic techniques respectively.The intrathoracic esophago-gastric anastomosis was created using a circular-stapled anastomosis with trans-oral anvil technique.Results There were 24 males and 6 female with median age of 62.5-year-old (from 47 to 82).The lesions located in middle and/or lower portion of the esophagus.25 patients with esophageal squamous cell cancer (n =25) and 5 patients with adenocarcinoma (n =5) underwent an Ivor Lewis esophagectomy.All the operations were successfully performed without intra-operative technical failures of the anastomosis or deaths.The mean operation time was 95 minutes in abdomen and 177 minutes in thorax.The mean blood loss was 310 ml.The mean number of harvested lymph nodes was 10.6 from thorax and 4.9 from abdomen.Postoperalively,there were thoracic wound infections in 4 patients.One patient had a left diaphragmatic hernia and was successfully treated by re-operation 6 days after first operation.One patient had chylothorax and recovered without surgery.There was no anastomotic leak and the patients were able to have spoon meat at 5.6 postoperative days.Conclusion Laparoscopic and thoracoscopic Ivor Lewis esophagectomy can be accomplished smoothly with acceptable occurrence of complications.The circular-stapled anastomosis with the trans-oral anvil is an efficient,safe and reproducible technique for intrathoracic esophago-gastric anastomosis.
