1.Investigation of TCM Constitutions and Relevant Syndrome Study on Patients with Hormone-dependent Breast Cancer

Objective To study the distribution characteristics of TCM constitutions of hormone-dependent breast cancer patients; To provide references for TCM diagnosis and treatment.Methods TCM constitution differentiation in the 210 patients with hormone-dependent breast cancer were investigated by a questionnaire survey, which is made according to the Classification and Determination of TCM Constitutions invented by professor Wang Qi and four methods of diagnosis of TCM, with a purpose to investigate the correlation of TCM constitutions and TCM syndrome differentiation typing.Results There was statistical significance among TCM constitutions of the 210 patients with hormone-dependent breast cancer (P<0.001), with yin deficiency 43.8% (92 cases), qi stagnation 15.2% (32 cases), qi deficiency 11.4% (24 cases), blood stasis 7.6% (16 cases), and yang deficiency 6.7% (14 cases). There was statistical significance among TCM syndromes (P<0.001), with dysfunction of Chong and Ren channels 63.8% (134 cases), stagnation of liver qi syndrome 16.2% (34 cases), weakness of the spleen and stomach syndrome 11.4% (24 cases). There was statistical significance between dysfunction of Chong and Ren channels and correlation of TCM constitutions (P<0.001).Conclusion Most hormone-dependent breast cancer patients are accompanied with yin deficiency pattern and the syndrome differentiation typing of dysfunction of Chong and Ren channels. There is correlation between dysfunction of Chong and Ren channels and TCM constitutions.

3.Advances in roles of NF-κB in regulating pathways of apoptosis

Apoptosis is a process of programmed cell death that is controlled by genes. Normally, there are three regulation pathways involved in the process of apoptosis, including the signaling of intracel-lular mitochondria, endoplasmic reticula and extracellular death receptors. Recent studies showed that NF-κB is a key regulator in the process of apoptosis. NF-κB plays a promotional and a inhibitory role as well in the regulation of apoptosis, closely related to the the inhibitor of apoptosis proteins family, the B cell lymphoma/ lewkmia-2 family, tumor necrosis factor receptor associated factors, c-Jun N-terminal kinase, tumor necrosis factor related apoptosis inducing ligand and Fas-associated death domain protein-like interleukin-1β. Thus, investigation of the mechanism regarding NF-κB in apoptosis regulation is of great importance for apoptosis-related drug development. The paper reviews the recent research progress in the function of NF-κB in apoptosis pathway regulation.
4.Extracurricular activities in general practice teaching for medical students

One hundred and forty-four medical students were enrolled in the study, the participants were randomly divided into experimental group and control group with 72 in each. All students attended the theoretical lecture of general medicine. Students in experimental group took part in the extracurricular teaching activities including the nutritional course and practice, the clinical case study and problem based learning ( PBL), students in control group did not have the extracurricular activities. The results showed that although there was no difference in general testing scores between two groups, the problem solving ability and assay-writing ability of experimental group were significantly higher than those of control group.

Aim Toassesstheregulatoryeffectsofcar-damonin (CDN ) on toll-like receptor (TLR )-4/MyD88/NF-κB/iNOS signaling pathway in lipopolysac-charide (LPS )-stimulated RAW264. 7 macrophage cells.Methods LPS-stimulatedRAW264.7cells were divided into three groups:vehicle-treated group, LPS-treated group and LPS +CDN-treated group.Cell viability was assessed by CCK-8 assay.The concentra-tion of nitric oxide (NO)in cell culture medium was measured by Griess reagent.The mRNA levels of iN-OS,COX-2,MCP-1 ,TNF-α,IL-6 and IL-1βwere de-termined by reverse transcription real-time quantitative PCR(RT-qPCR).The protein levels of inducible nitric oxide synthase(iNOS),TLR4,myeloid differentiation factor 88(MyD88),nuclear factor κB(NF-κB)phos-phorylated (p )-p65 ,inhibitor κBα(IκBα),and p-IκBαweredeterminedbyWesternblot.Results 1~50 μmol·L-1 CDN had no cytotoxicity in RAW264. 7 cells.However,CDN inhibited the LPS-induced secre-tion of nitric oxide(NO)and mRNA expressions of iN-OS,COX-2,MCP-1 ,TNF-α,IL-6 and IL-1βin a dose-dependent manner.Moreover,50 μmol · L-1 CDN inhibited the LPS-induced up-regulation of iNOS, TLR4,MyD88,NF-κB p-p65,p-IκBαand down-reg-ulationofIκBα.Conclusion Cardamonininhibitsthe production of NO via a mechanism associated with the inhibition of TLR4/MyD88/NF-κB/iNOS pathway.
6.Effects and mechanisms of vitexin against ulcerative colitis in mice

Aim To evaluate the effect and mecha-nism of vitexin in a mouse model of DSS-induced ulcer-ative colitis (UC).Methods C57BL/6 mice were randomly placed into three groups: normal control group,DSS group and DSS +Vitexin group.Mice coli-tis was induced by adding 4% dextran sulphate sodium (DSS)into the drinking water for seven days.Vitexin was administered once a day along with DSS treatment. Mice were monitored daily with body weight change and diarrhea symptoms.After sacrifice,colon was re-moved and fixed in 1 0% (W/V)buffered formalin for hematoxylin-eosin (H&E)staining.Histological dam-age was assessed as a combined score of inflammatory cell infiltration and mucosal damage.The remaining colon pieces were collected to measure the activity of myeloperoxidase (MPO)by ELISA method and to de-termine the mRNA expression of TNF-α,IL-6 and COX-2.Results None of the mice receiving vehicle alone exhibited body weight loss and mucosal disrup-tion at any point during the study.Vitexin treatment significantly ameliorated DSS-induced body weight loss and histological score.The activity of MPO and the mRNA expression of TNF-α,IL-6 and COX-2 were markedly inhibited by vitexin treatment.Conclusion Vitexin ameliorates DSS-induced colitis through sup-pressing leukocyte infiltration and pro-inflammatory mediators production.
7.Detection of exhaled nitric oxide of healthy in Nanjing.

OBJECTIVE:
To determine the normal level of nitric oxide through mouth and nose in healthy adult people. From it the normal values were obtained to provide theory support for the prevention and cure of breathe system diseases, such as asthma, allergic rhinitis and so on.
METHOD:
One hundred and eighty two healthy nanjing adults were recruited,including 89 males and 93 females. NIOX system was used to obtain the values of exhaled NO through mouth and nose. The relativity between NO and gender, age, height, body mess index, time, ambient NO were analyzed with multiple linear regression and correlation.
RESULT:
Exhaled NO values were (17 +/- 7) ppb and correlated significantly with height and age. Regression equation: Y (FENO) = -65.784 + 0.102X1 (age) + 0.442X2 (height), P < 0.05. NNO values were (79 +/- 35) ppb.
CONCLUSION
FENO normal values and NNO normal values are easy to obtain and have good compliance and high sensitivity, which correlated with height, age, et al.

8.Liuzijue qigong for stroke survivors with dysarthria

Objective:To document any improvement in the breathing control of stroke survivors with dysarthria after practicing Liuzijue qigong.Methods:A total of 157 stroke survivors with dysarthria and abnormal respiration control were randomly divided into an observation group and a control group. Both groups were given traditional breathing training and basic articulation training (including articulatory organ training and speech training). The observation group also received training in Liuzijue qigong. It requires inhaling through the nose and exhaling through the mouth while producing the speech sounds xu, he, hu, si, chui and xi. The training lasted two weeks. Both groups were then evaluated using the modified Frenchay dysarthria assessment. Maximum phonation time, maximum counting ability and volume were also recorded as secondary indexes.Results:After the 2-week intervention, significant improvement was observed in the average scores on all of the indexes, with all of the observation group′s average scores except for volume significantly better than those of the control group. The average volume scores were significantly improved, but not significantly different.Conclusion:Supplementing basic articulation training with Liuzijue qigong can improve respiratory function and the speaking ability of stroke survivors with dysarthria. It is worthy of wider clinical application.
9. Salubrinal increases the apoptosis of oral cancer cells by inhibiting radiation-induced activation of NF-Κb

Objective:
To explore the mechanism of the role of Salubrinal in regulating the radiation-induced apoptosis of oral cancer cells.
Methods:
Radioresistant KBR cell line was constructed (4 Gy per fraction, every 7-10 d for 4 times). The radiosensitivity of oral cancer cells after Salubrinal pretreatment was measured by colony formation assay. The expression levels of NF-κB-HIF-1α signaling pathway and apoptosis biomarker cleaved PARP in oral cancer cells were measured by Western blot. The apoptosis rate was detected by Annexin V, PI staining and flow cytometry.
Results:
Colony formation assay demonstrated that Salubrinal increased the radiosensitivity of oral cancer cells. The radiosensitization ratios of KB and KBR cells were 1.19 and 1.24. Western blot revealed that the activation of NF-κB-HIF-1α was time-dependent in the radiation-induced oral cancer cells, whereas Salubrinal inhibited the radiation-induced abnormal activation. In addition, Salubrinal increased the expression of apoptosis biomarker cleaved PARP and apoptosis index in radiation-induced oral cancer cells, whereas TNF-α, an activator of NF-κB, reversed the effect, suggesting that Salubrinal increased the apoptosis of radiation-induced oral cancer cells by suppressing the activation of NF-κB. Pretreatment of NF-κB inhibitor Bay11-7082 also increased the cell apoptosis. The expression levels of cleaved PARP of KB and KBR cell lines in the Bay11-7082+ IR group were 2.67±0.26 and 1.91±0.17, significantly higher compared with 2.1±0.16 and 1.44±0.15 in the IR group (both
