1.Effects of dehydroepiandrosterone on amino acids receptor subunit NR2B and GBR1 expressed in primary cultured rat cerebral cortical neurons

Aim To investigate the effect of dehydroepiandrosterone(DHEA)on the levels of NR2B and GBR1 expressed in primary cultured rat cerebral cortical neurons.Methods Primary cultured rat cerebral cortical neurons were treated with DHEA of different concentrations (1,10,100 μmol·L~(-1))and the expression of amino acids receptor subunit NR2B and GBR1 were detected by immunocytochemistry.Results Compared with control group,the expression intensity of NR2B increased by 15.6%,19.9% and 49.4% after DHEA-L,DHEA-M and DHEA-H treatment(P<0.05 or P<0.01);the expression intensity of GBR1 increased by 14.5% and 58.5% after DHEA-M and DHEA-H treatment(P<0.05 or P<0.01).Conclusion DHEA can enhance the expression of neuron receptor subunit NR2B and GBR1.

4.Efficacy of Long xue jie capsule combined with Badger Oil treatment of radiotherapy -induced injuries of skin

Objective To evaluate the effects of Long xuejie capsule combined with Badger Oil on head and neck cancer radiotherapy -induced radiation injuries of skin application value.Methods The received radiation therapy in Ⅱ -Ⅳacute radioactive skin injury in 128 cases were selected.Used the envelope method,they were ran-domly divided into the research group and the control group.Outside the team starts from a radioactive dermatitis with badger oil apply skin irradiation area,with a dragon dragon's blood powder coating outside skin area.Apply 1 -2 times a day,seven days for a course of treatment until wound healing.The control group from a radioactive dermatitis with 0.9% sodium chloride injection,dexamethasone,gentamycin gauze wet moist wound after 30min,1 -2 times a day, 7 days for a course of treatment.Compare the therapeutic effect of the two groups of wet radioactive dermatitis and interruption of radiotherapy and skin lesion healing time.Results The effective rate of the study group was 90.6%, the cure rate was 100%.The effective rate of the control group was 80%,the treatment rate was 62.5%.There was a significant difference between the two groups (χ2 =14.64,11.86,11.48.P <0.01).The vast majority of the study group in a week skin lesions healed,no patients discontinued radiotherapy,the control group 13 cases of termination of radiotherapy,interruption time study group (3.8 ±1.4)d,group interrupt time (8.8 ±1.6)d,lesion healing time of the study group (5.6 ±0.6)d,lesion healing time in the control group (10.6 ±0.8)d.There was a significant differ-ence between the two groups (χ2 =6.68,6.95,11.48,all P <0.05,significant difference).Conclusion Longxuejie capsule combined Badger Oil has significant efficacy in the treatment of radioactive skin injury.It could shorten the healing time,and the method is convenient,economic,security and has certain clinical value.
5.Ultrasonic diagnosis of congenital vaginal reclined septum syndrome

Objective To study the ultrasonic features of the congenital vaginal reclined septum syndrome (CVRS) in order to apply a precise way for operations. Methods To diagnose CVRS accurately before operation, the ultrasonogram and clinical characteristics of 15 cases were studied retrospectively. Results There were duplicated uterus and cervices with ipsilateral renal agenesis on the reclined septum side in all the patients,11 in the right and 4 in the left. All of 10 cases of type Ⅰhad been made final diagnosis before operation according to the specific ultrasonogram and the rest of type Ⅱ and type Ⅲ had been made reliable diagnosis. Conclusions Ultrasonic examination should be the first way to diagnose CVRS.

7.Differentiation efficiency of human umbilical cord mesenchymal stem cells into hepatocytes under two kinds of liver homogenate supernatants: a comparative study

BACKGROUND:Previous studies have demonstrated that normal rat liver homogenate supernatant can induce human umbilical cord mesenchymal stem cels to differentiate into hepatocyte-like cels with partial hepatocyte functions. However, whether fibrotic liver homogenate supernatant can work or how the inducing effect is remains unclear. OBJECTIVE:To investigate the differentiation potential of human umbilical cord mesenchymal stem cels into hepatocytes under the normal liver and fibrotic liver microenvironment in vitro. METHODS:Liver fibrosis was induced in the SD rats by repeated intraperitoneal injections of 3% thioacetamide at a dose of 200 mg/kg body mass, twice a week for 4 weeks, and then fibrotic liver tissues and normal liver tissues were used to prepare liver homogenate supernatants. Passage 3 human umbilical cord mesenchymal stem cels were used and divided into standard control group (cels were cultured in DMEM/F12 with 10% fetal bovine serum), fibrotic liver homogenate supernatants group (cels were cultured in DMEM/F12 with 10% fetal bovine serum and 50 g/L fibrotic liver homogenate supernatants), normal liver homogenate supernatants group (cels were cultured in DMEM/F12 with 10% fetal bovine serum and 100 g/L normal liver homogenate supernatants). The morphological changes of the cels in each group were recorded under inverted microscope; the protein levels of CK18, AFP, CYP3A4, CYP2E1, CYP2D6 and TPH2 were evaluated using western blot assay. Furthermore, the concentration of albumin in the cels was measured. RESULTS AND CONCLUSION:After a 7-day inducement, the stem cels in liver homogenate supernatants groups lost their fusiform shape and changed into hepatocyte-like cels with the morphous of round shape. Compared with the standard control group, the hepatocyte-like cels in the two liver homogenate supernatants groups exhibited human hepatocyte biomarkers, CK18 and AFP. The standard control group cels could express a little amount of CYP2E1, while cels in the two liver homogenate supernatants groups could express CYP3A4, CYP2E1, CYP2D6, TPH2. Compared with the standard control group, the expression level of CYP2E1 in the two liver homogenate supernatants groups increased significantly (P < 0.01), and however, the relative levels of CYP3A4, CYP2E1, CYP2D6, TPH2 in the two liver homogenate supernatants groups showed no statistical significance (P > 0.05). At the same time, compared with the standard control group, the concentration of albumin in the two liver homogenate supernatants groups markedly increased (P < 0.01), but there was no difference between the two liver homogenate supernatants groups (P > 0.05). Experimental findings demonstrated that both of normal liver tissue and fibrotic liver tissue microenvironments could induce human umbilical cord mesenchymal stem cels to differentiate into hepatocyte-like cels. To achieve the same effect, compared with normal liver tissue, fibrotic liver tissue required lower concentrations, suggesting that fibrotic liver tissue microenvironment may be more conducive to differentiation of umbilical cord mesenchymal stem cels into hepatocytes.
8.Constitution Factors for Metabolic Syndrome: A Case-control Study

Objective To investigate the constitution factors for metabolic syndrome(MS). Methods 271 MS cases and 271 non-MS controls were selected. They were assessed according to the diagnosis standard of constitution of TCM. Results Multiple logistic regression analysis revealed that phlegm-dampness constitution and dampness-heat constitution were dangerous factors for MS, normal constitution was protective factor for MS (P<0.05). Conclusion Research on constitution play a role in investigating pathogenesis of MS.

10.The expression of stromal cell-derived factor-1 and CXCR4 in pancreatic carcinoma tissues, cell lines and pancreatic stellate cells

Objective To investigate the expressions of stromal cell-derived factor1 (SDF-1) and its receptor CXCR4 in human pancreatic carcinoma tissues, cell lines and pancreatic stellate cells (PSCs). Methods SDF-1 /CXCR4 and α-SMA protein expression levels and SDF-1 and CXCR4 protein in AsPC-1 and PSCs were detected by immunohistochemical staining in 10 cases of peri-eareinoma tissues and 37 cases of pancreatic carcinoma tissues. The expression of SDF-1 and CXCR4 mRNA in pancreatic cell lines and PSCs were detected by RT-PCR. Results CXCR4 were positively expressed in all pancreatic carcinoma tissues [(+) 8 cases, (+ +) 20 cases, (+ + +) 9 cases]; and there were no CXCR4 expression in 2 cases of pori-careinoma tissues and CXCR4 were positively expressed in 8 cases [(+) 7 cases, (+ +) 1 cases]; with significant difference (P <0.01). And the expression of SDF-1 protein in carcinomatous stromal tissues was much higher than that in the stromal tissues of peri-carcinoma (P < 0.01), and it corresponded to the increase of α-SMA expression. The CXCR4 protein expression was found in AsPC-1, while SDF-1 protein expression was found in PSC. The CXCR4 mRNA expression was found in AsPC-1, BxPC3, SW1990, while there were no SDF-1 mRNA expression in the above mentioned cell lines. SDF-1 mRNA was expressed in PSC and CXCR4 mRNA was weakly expressed in PSC. Conclusions The expression of CXCR4 mRNA and protein were found in pancreatic carcinoma specimens and cell lines. PSCs expressed SDF-1 mRNA and protein. PSCs may promote the invasion and metastasis through SDF-1/CXCR4 axis.
